#101 - Nuclear and mitochondrial DNA markers in traceability of retail beef samples, 30(9):782-786

E-mail: meirellf@usp.br Several characteristics are important in a traceability system of animal products, such as age at slaughter, breed composition, besides information of the productive chain. In general, the certification agent records information about the animals and the system which it came from, although cannot guarantee that the slaughtering, meat processing and distribution are error proof. Besides, there is a differential price, at least at the international market, based on sex and breed composition of the animals. Genetic markers allow identification of characteristics controlled in the beef cattle traceability program, as sex and breed composition, in order to correctly identify and appraise the final product for the consumer. The hypothesis of this study was that the majority beef samples retailed in the local market originate from female with a great participation of zebu breeds. Therefore, the objective of this work was to characterize retail beef samples with DNA markers that identify cattle sex and breed composition. Within 10 beef shops localized in Pirassununga, SP, Brazil, 61 samples were collected, all were genotyped as harboring Bos taurus mitochondrial DNA and 18 were positive for the Y chromosome amplification (male). For the marker sat1711b-Msp I the frequency of the allele A was 0.278 and for the marker Lhr-Hha I the frequency of the allele T was 0.417. The results of sat1711b-Msp I and Lhr-Hha I allelic frequencies are suggestive that the proportion of indicus genome compared with the taurine genome in the market meat is smaller than the observed in the Nellore breed. The procedure described in this study identified sex and subspecies characteristics of beef meat samples, with potential application in meat products certification in special as an auxiliary tool in beef cattle traceability programs.

E-mail: meirellf@usp.br Várias características são importantes no sistema de rastreabilidade, como o sexo, a idade, a raça e/ou a composição racial dos animais, além de dados da cadeia produtiva. Em geral, a empresa certificadora dispõe das informações do animal que está sendo abatido, porém não tem condições de garantir se houve erro entre abate, desossa, processamento e a distribuição dos produtos. Existe diferenciação no custo e na qualidade dos produtos cárneos, especialmente no mercado internacional, em virtude do sexo e composição racial dos animais. Os marcadores genéticos permitem identificar as características que são controladas num programa de rastreabilidade bovina tais como sexo e composição racial, permitindo identificar e avaliar corretamente para o consumidor, o produto final. A hipótese deste estudo foi que a maioria das amostras de carne bovina vendida no mercado local seria proveniente de fêmeas e com grande participação de raças Zebu. O objetivo deste trabalho foi caracterizar amostras de carne bovina com marcadores de DNA para identificar o sexo e a composição racial. Em dez pontos comerciais da cidade de Pirasssununga, SP, Brasil, foram coletadas 61 amostras e todas foram genotipadas como possuindo DNA mitocondrial Bos taurus e 18 foram positivos para amplificação do cromossomo Y (macho). Para o marcador sat1711b-Msp I a frequência alélica do A foi 0.278 e para o marcador Lhr-Hha I a frequência alélica do T foi 0.417. Os resultados das frequências alélicas do sat1711b-Msp I e Lhr-Hha I apresentaram menor proporção do genoma Bos indicus em relação ao Bos taurus quando comparado ao rebanho Nelore. Com a metodologia descrita neste trabalho foi possível avaliar o sexo e as características de subespécie das amostras de carne bovina, tendo uma importante aplicação para a certificação de produtos cárneos especialmente, em programas de rastreabilidade animal.

E-mail: hartman@usp.br The use of specific anti-mastitis drugs is indicated in dry cow therapy to prevent infections in the following lactation. Nonetheless, cells active in the dry period are of fundamental importance for the involution of the mammary gland and its recovery for the following lactation. Since there are no specific drugs for dry goat therapy, the dry cow therapy drugs tend to be misused in goats despite of the anatomical and physiological differences apart from the particularities of the two species. The objective of the present study was to evaluate the influence of five drugs specific for dry cow therapy on the function of goat milk phagocytes. Somatic cells were isolated from 20 milk samples of 10 lactating goats that had not been treated for mastitis during the previous 30 days. Milk samples were collected properly and microbiological culture yielded negative results. Cells adherent to glass coverslips were treated with commercially available dry cow therapy drugs containing active principles such as Gentamicin (M1), Cephalonium anhydrous (M2), Ampicillin (M3), Cloxacillin benzathine (M4) and Cephapirin benzathine (M5). Phagocytosis of Zymosan particles was evaluated. Mean phagocytosis indexes of cells treated with M2 (15.12% ± 16.22), M3 (6.02% ± 7.96), M4 (4.54% ± 5.45) and M5 (2.47% ± 4.64) were lower (p<0.001) than mean phagocytosis index of the control group (40.67% ± 19.68). Mean phagocytosis index of cells treated with M2 was greater (p<0.05) than those treated with M3, M4 and M5, whereas means of the latter three treatments were statistically similar. M1-treated cells did not adhere adequately to the cover slips, making it impossible to evaluate phagocytosis in this group. The results obtained enable the conclusion that drugs used affected milk phagocytes negatively. However, interference on somatic cell function is not the sole factor determining an unsuccessful dry period therapy, since the efficacy of the active principle on pathogens responsible for infectious processes also has to be considered.

Não obstante, a ação das células envolvidas no período de secagem tem fundamental importância para a involução da glândula mamária e seu restabelecimento para a lactação subseqüente. A indisponibilidade de tais medicamentos para uso em cabras tem resultado na extrapolação do uso de produtos recomendados para vacas sem que se considerem as particularidades e diferenças anátomo-fisiológicas entre as espécies bovina e caprina. O presente estudo teve por objetivo avaliar a influência de cinco antimamíticos específicos para vacas secas sobre a função dos fagócitos provenientes de leite caprino. Para tal, fez-se o isolamento de células somáticas de 20 amostras de leite provenientes de 10 cabras lactantes, sem antecedentes de tratamento de mamite nos últimos 30 dias, sob condições higiênico-sanitárias de colheita e com resultados negativos ao cultivo microbiológico do leite. As células aderidas a lamínulas de vidro foram confrontadas com formulações contendo princípios ativos disponíveis no mercado como Gentamicina (M1), Cefalônio Anidro (M2), Ampicilina (M3), Cloxacilina Benzatínica (M4) e Cefapirina Benzatínica (M5). Avaliou-se, por microscopia, a fagocitose de partículas de Zymosan. As médias dos índices de fagocitose das células submetidas ao tratamento com M2 (15,12% ± 16,22), M3 (6,02% ± 7,96), M4 (4,54% ± 5,45) e M5 (2,47% ± 4,64) foram menores (p<0,001) que a média dos índices de fagocitose do grupo controle (40,67% ± 19,68). A média dos índices de fagocitose das células submetidas ao tratamento com M2 foi maior (p<0,05) que as médias dos tratamentos com M3, M4 e M5 enquanto estas foram estatisticamente iguais entre si. As amostras celulares submetidas ao medicamento M1 exibiram adesão insuficiente ou ausente às lamínulas, inviabilizando a avaliação da fagocitose por meio da técnica utilizada. Os resultados obtidos permitem concluir que os medicamentos estudados exerceram influência negativa sobre os fagócitos do leite, porém, esta interferência sobre as funções das células somáticas não pode por si só determinar o insucesso da terapia proposta para o período seco, pois deve ser considerada, outrossim, a eficácia do princípio ativo sobre o patógeno causador do processo infeccioso.

E-mail: savasco@usp.br Risk factors associated with leptospirosis were identified in cows in the state of Bahia, northeastern Brazil. A total of 10,823 cows with > 24 months of age from 1,414 herds were randomly sampled. For the serological diagnosis of Leptospira spp. infection, the microscopic agglutination test (MAT) was carried out using 24 serovars as antigens. A herd was considered positive when presented at least one seropositive animal. Of the 1,414 investigated herds, 1,076 (77.9%; 95% CI = 75.7-80.0%) presented at least one reactant animal at MAT to any serovar. Serovar Hardjo (Hardjoprajitno) was the most prevalent, with 34.49% (95% CI = 31.97-37.14%) of the positive herds. Presence of more than 28 bovine females in reproductive age (OR=2.11; p<0.001), presence of cervids (OR=2.02; p=0.010), animal purchase (OR=1.57; p<0.001), to slaughter animals in the property (OR=1.58; p=0.030) and to share pasture (OR=1.63; p<0.001) were identified as risk factors for leptospirosis due to any serovar. Risk factors for leptospirosis due to serovar Hardjo (Hardjoprajitno) were presence of swine (OR=1.28; p=0.040) and animal purchase (OR=1.48; p<0.001).

Um rebanho foi considerado foco quando apresentou pelo menos um animal soropositivo. Das 1.414 propriedades investigadas, 1.076 (77,9%; IC 95% = 75,7-80,0%) apresentaram pelo menos um animal reagente na SAM para qualquer sorovar. O sorovar Hardjo (Hardjoprajitno) foi o mais prevalente, com 34,49% (IC 95% = 31,97-37,14%) das propriedades positivas. Presença de mais de 28 fêmeas bovinas em idade reprodutiva no rebanho (OR=2,11; p<0,001), presença de cervídeos (OR=2,02; p=0,010), compra de animais (OR=1,57; p<0,001), abate de animais na própria fazenda (OR=1,58; p=0,030) e utilização de partos compartilhados (OR=1,63; p<0,001) foram identificados como fatores de risco para leptospirose por qualquer sorovar. #104 - Inefficacy of albendazole sulphoxide and ivermectin for the treatment of bovine parasitic otitis caused by rhabditiform nematodes, 29(11):910-912

E-mail: gverocai@gmail.com The purpose of this study was to evaluate the efficacy of orally administered albendazole sulphoxide and pour-on ivermectin for the treatment of bovine parasitic otitis caused by rhabditiform nematodes. Eighteen Gyr cows presenting clinical otitis were divided in three groups with six animals each. The first one did not receive any treatment (control group). The second one was treated with 0.5% pour-on ivermectin, 500µg/kg of body weight, and the third group was treated with oral 6% albendazole sulphoxide, at 6.0mg/kg. Both ear canals of each animal were reexamined on days 7 and 21 post treatment. The animals in the control group remained infected throughout the days of observation. Ivermectin treatment did not show effectiveness on days 7 or 21 post treatment. The albendazole sulphoxide treatment had an efficacy of 16.7 and 25% on days 7 and 21, respectively. Further studies are required to assess an effective treatment for this parasitic disease, especially via alternative administration routes, because of its significant impact on Bos taurus indicus cattle breeding in Tropical and Subtropical Regions.

O segundo foi tratado com ivermectina “pour-on” a 0,05% na dose de 500µg/kg de peso vivo. O terceiro grupo foi tratado com sulfóxido de albendazol oral a 6% na dose 6,0mg/kg. Os condutos auditivos de todos os animais foram reexaminados nos dias 7 e 21 pós-tratamento. Os animais do grupo controle permaneceram infectados nos dias de observação. O tratamento com ivermectina não demonstrou eficácia alguma para os dias 7 e 21 pós-tratamento. O tratamento com sulfóxido de albendazol obteve 16,7 e 25% de eficácia nos dias 7 e 21, respectivamente. Mais estudos são necessários para determinação de tratamentos eficazes para tal doença parasitária, especialmente através de vias alternativas de administração, por causa de seu significante impacto na criação de Bos taurus indicus nas Regiões Tropical e Subtropical.


#105 - Shedding and transmission of bovine viral diarrhea virus by persistently infected calves, 29(9):736-742

E-mail: eduardofurtadoflores@gmail.com Persistently infected (PI) calves born to cows infected with non-cytopathic bovine virus diarrhea virus (BVDV) represent the main reservoir of the virus in nature. We herein report an investigation on the patterns of virus shedding and transmission by five PI calves produced experimentally through inoculation of pregnant cows with Brazilian BVDV isolates. Five calves that survived intrauterine infection were born healthy, lacking neutralizing antibodies to BVDV and harboring virus in the blood. After weaning - and following the disappearance of colostral antibodies - PI calves were monitored for virus in serum and body secretions (ocular, oral, nasal and genital) at weekly intervals for up to 150 days. For each animal, the virus titers in serum showed minor variations throughout the collections (with one exception that presented an increase late in infection), yet the titers varied widely among animals (from 102 to 106TCID50/mL). Virus shedding in secretions was detected steadily during all the observation period with minor titer variations for each particular animal. The highest titers were generally detected in nasal and ocular secretions (titers 104 to 106TCID50mL) whereas genital and oral secretions usually contained low amount of virus (102 to 103TCID50mL). To evaluate the kinetics of virus transmission by these animals, one PI was introduced on a group of 10 seronegative calves maintained with a high animal density simulating the conditions of an intensive management. All 10 contact calves seroconverted to BVDV by day 30. Another PI calf was introduced into a 48-head herd kept under a low animal density, extensive grass management. Among these animals, 8/48 (16.6%) seroconverted by day 10, 26/48 (54.1%) by day 40 and 37/48 (77%) were seropositive at day 100, when the monitoring was discontinued. These results show that continuous viremia and virus shedding in high titers in secretions by PI animals assure an efficient and rapid virus transmission to contact animals, being the kinetics of transmission much faster under intensive conditions.

E-mail: eduardofurtadoflores@gmail.com Bezerros persistentemente infectados (PI) nascidos de vacas infectadas com amostras não-citopáticas do vírus da diarréia viral bovina (BVDV) se constituem nos principais reservatórios do vírus na natureza. Este trabalho relata uma investigação do padrão de excreção e transmissão viral por cinco bezerros PI produzidos experimentalmente pela inoculação de vacas prenhes com isolados brasileiros do BVDV. Cinco bezerros que sobreviveram a infecção intrauterina nasceram saudáveis, soronegativos e com a presença de vírus no sangue. Após o desmame - e desaparecimento dos anticorpos colostrais - os bezerros PI foram monitorados semanalmente durante 150 dias para a presença de vírus e títulos virais no soro e em secreções (ocular, oral, nasal e genital). Os títulos virais no soro de cada animal apresentaram pequenas variações durante o período (com exceção de um animal que apresentou um aumento de título tardiamente), mas os títulos variaram amplamente entre os animais (entre 102 e 106TCID50/ml). O vírus também foi excretado continuamente nas secreções de todos os animais, com pequenas variações de título entre as coletas. Os maiores títulos virais foram geralmente detectados nas secreções nasais e oculares (títulos de 104 a 106TCID50/mL), enquanto as secreções orais e genitais usualmente continham títulos virais baixos (102 a 103TCID50/mL). Com o objetivo de avaliar a dinâmica de transmissão viral, um bezerro PI foi introduzido em um grupo de 10 bezerros soronegativos, mantido com uma alta densidade animal e submetido a manejo diário para simular as condições de manejo semi-intensivo. Após 30 dias de convívio com o bezerro PI, todos os demais animais haviam soroconvertido ao BVDV. Para investigar a transmissão viral sob condições extensivas, outro bezerro PI foi incorporado a um rebanho de 48 animais mantido a campo, com baixa densidade animal e submetido a manejo extensivo. Dentre estes animais, 8/48 (16,6%) foram soropositivos para anticorpos no dia 10, 26/48 (54,1%) no dia 40 e 37/48 (77%) haviam soroconvertido no dia 100, quando encerrou-se o monitoramento. Estes resultados demonstram que a viremia e excreção viral contínua em altos títulos por animais PI assegura a transmissão rápida do BVDV a animais mantidos em contato, sendo a transmissão notadamente mais rápida em condições intensivas e de alta densidade animal.

E-mail: cocaarduino@yahoo.com.br In the investigation 100 heifers were used, divided into 5 groups of 20 animals each. The four experimental groups were vaccinated using distinct commercial polyvalent bacterines: A, B, C and D, and a group was the control. Samples were collected at days 0, 3, 7, 14, 21, 28, 35, 42, 42, 56, 63, 70, 77, 84, 91, 120, 150 and 180 from the first injection of the vaccine. The selection of the animals for the experimental groups was done based on a serological screening with 6 antigens of Leptospira sp. constituted by non-reagent animals. The vaccine titers were monitored using the microscopic agglutination test (MAT) for Canicola, Grippotyphosa, Hardjo, Icterohaemorrhagiae, Pomona and Wolffi serovars. The selection of the animals for the experimental groups was done based on a serological screening with 6 antigens of Leptospira sp. constituted by non-reagent animals. The vaccine titers were monitored using the microscopic agglutination test (MAT) for Canicola, Grippotyphosa, Hardjo, Icterohaemorrhagiae, Pomona and Wolffi serovars. Cows (n=19) were vaccinated twice, with a 34-days-interval, with the experimental vaccine and together with non-vaccinated controls (n=18), were mated and challenged between days 60 and 90 of gestation by intranasal inoculation of four heterologous BVDV-1 and BVDV-2 isolates. The antibody response was evaluated by serum-neutralization tests performed at different intervals after vaccination (days 34, 78 and 138 post-vaccination [pv]). Fetal protection was monitored by ultrassonographic and clinical examination of the dams and fetuses during the rest of gestation; and through virological and serological examination of pre-colostral blood obtained from aborted and/or recently born fetuses/calves. At the day of challenge (day 138 pv), all vaccinated cows had neutralizing antibodies in high titers against BVDV-1 (1,280- ³10,240), and with one exception (titer 20), presented moderate to high titers to BVDV-2 (80-1,280). At the end of the monitoring, only three out of 18 control cows (16.6%) delivered healthy, virus-free calves. Fifteen non-vaccinated cows (83.3%) presented signs of fetal infection and/or had reproductive losses. Seven of these cows (38.8%) delivered virus-positive calves; five were healthy and survived (27.7%); two were premature or weak and lasted three and 15 days, respectively. The other eight cows (44.4%) aborted between day 30 post-challenge and the parturition; or delivered premature or stillbirth calves. In contrast, 17 out of 19 (89.4%) vaccinated cows delivery virus-free, healthy calves. One vaccinated cow aborted around day 130 post-challenge, yet this fetus could not be examined for the presence of virus. Another cow delivered a virus-positive calf (5.2%). In summary, the experimental vaccine induced adequate antibody titers in most animals and the immunological response induced by vaccination was able to prevent fetal infection and reproductive losses upon challenge with a pool of heterologous BVDV isolates. Hence, this experimental vaccine may be an attractive alternative for the prevention of reproductive losses associated with BVDV infection.

In small ruminants that area is also characterized by macroscopic blood extravasation (hemophagous areas). This occurs possibly because the iron is transferred to the embryo by trophoblastic erytrophagocytosis in these hemophagous placental areas. In order to investigate the hemophagous placental areas in cattle, placentomes of 34 pregnant Bos indicus cows (2-3, 4-6, 7-8 and 9 months of gestation) were analyzed. They were fixed by perfusion with 10% formaldehyde aqueous solution and 4% paraformoldehyde in PBS, pH 7.4, 0.1M, processed and stained for light microscopy and histochemistry. The methodology used allowed to observe placental haematomes between the uterine and trophoblastic epithelium since 3 months of pregnancy. Erythrocytes had been found in the trophoblastic cells, elucidating the erytrophagocytosis. Through Perl’s histochemical reaction it was possible to prove the existence of ferric iron in the trophoblastic cells. The PAS reaction was positive staining mucoid substance in the epithelial cells, especially in the binucleate cells of the fetal epithelium. Based on histology and histochemistry, it can be inferred that the hemophagous areas are important sites for iron transfer in the bovine placenta.

Hence the aim of this investigation was to evaluate the stability of this antigen under refrigeration for use in the complement fixation test. Fourteen batches of antigen prepared with Brucella abortus strain 1119/3, produced from 9 months to 23 years and 11 months before, were analysed. One hundred and sixty-seven cattle sera with varying titres of antibodies to Brucella were tested through the warm complement fixation microtechnique with five 50% haemolytic units of complement. Sera with at least 25% of complement fixation in dilution 1:4 were considered positive. The results with 13 of the antigen batches were compared with the results obtained with the batch produced 9 months before by the McNemar c2 test and kappa statistic. The oldest antigen batch gave a higher proportion of sera titres which were exactly the same observed with the 9-month-batch (90.4%), and the antigen produced 4 years and 3 months before the test gave de lowest proportion of sera with the same titre of the 9-month-antigen (73.7%). The comparison of the results after being classified as positive and negative showed that the highest proportion of agreed results was observed with the antigen produced 21 years and 4 months before (98.8%, kappa 0.98). The antigen with the lowest proportion of agreed results was the one produced 3 years and 2 months before (91.6%, kappa 0.84). The results of the study show that most sera gave very similar results with all antigen batches evaluated, and that there was no relationship between the period of antigen production and the difference in test results.
