#1 - Immunomodulatory effect of Bacillus toyonensis BCT-7112 supplementation in puppies vaccinated against canine parvovirus

Abstract in English:

Bacillus toyonensis is a probiotic microorganism that for decades has been used in animal nutrition around the world. The objective of this work was to evaluate the immunomodulatory effect of oral B. toyonensis supplementation in dogs vaccinated against canine parvovirus. Puppies were randomly selected and divided in two groups, one received B. toyonensis at a concentration of 2x108 viable spores per day and another group without supplementation was left as control. The puppies were vaccinated against canine parvovirus type 2. B. toyonensis supplementation was efficient in stimulating specific IgG for parvovirus with titers of 2, 3, and 2.5-fold higher than controls at 7, 21, and 35 pos-vaccination days respectively. Peripheral blood mononuclear cells (PBMCs) from dogs were cultured and stimulated with B. toyonensis DNA, vegetative cell and spores. The mRNA transcription of cytokines IL-4, IL-17, and IFN-γ up modulated by the stimuli. Thus, we conclude in this study that B. toyonensis supplementation may amplify the vaccine immune response against canine parvovirus.

Bacillus toyonensis é um micro-organismo probiótico que há décadas é utilizado na nutrição animal em todo o mundo. O objetivo deste trabalho foi avaliar o efeito imunomodulador da suplementação oral de B. toyonensis em cães vacinados contra o parvovírus canino. Os filhotes foram selecionados aleatoriamente e divididos em dois grupos, um recebeu B. toyonensis na concentração de 2 × 108 esporos viáveis por dia e outro grupo sem suplementação como controle. Os filhotes foram vacinados contra o parvovírus canino tipo 2. A suplementação com B. toyonensis foi eficiente em estimular IgG específica para parvovírus com títulos de 2, 3 e 2,5 vezes maior que os controles aos 7, 21 e 35 dias pós-vacinação, respectivamente. Células mononucleares do sangue periférico (PBMCs) de cães foram cultivadas e estimuladas com DNA de B. toyonensis, células vegetativas e esporos. A transcrição do mRNA das citocinas IL-4, IL-17 e IFN-γ foi modulada pelos estímulos. Assim, concluímos neste estudo que a suplementação com B. toyonensis pode amplificar a resposta imune da vacina contra o parvovírus canino.


#2 - Defined and undefined commercial probiotics cultures in the prevention of Salmonella Enteritidis in broilers

Abstract in English:

This study aimed to evaluate the efficacy of probiotics from different formations, defined and undefined cultures, applied in the control of Salmonella Enteritidis in broilers, identifying the compositions and states for which the probiotics are more effective. For that, 390 broilers were inoculated orally with 1.00 ml of Salmonella Enteritidis at a concentration of 1.2x109 CFU (Colony Forming Units). The experimental design used was randomized blocks with 5 treatments and 6 replications, totaling 30 boxes with 13 birds/box (13 birds/m2). The treatments were provided via drinking water 1 hour after inoculation, keeping a daily treatment of 12 hours with probiotics, for 3 consecutive days (birds at 1, 2 and 3 days of age). In general, the five treatments conducted were: T1 - Control without probiotic, T2 - Probiotic A (defined culture - lyophilized form, strain 7), T3 - Probiotic B (defined culture - lyophilized form, strain 11), T4 - Probiotic C (undefined culture liquid form), T5 - Probiotic D (undefined culture - liquid form). After treatments, performance was evaluated through average body weight, feed conversion and mortality counting. Microbiological analysis and Salmonella isolation were performed using MPN (Most Probable Number) and selective enrichment technique methods, respectively. Samples of ileum and liver pool, cecal tonsils, cecum, heart and spleen pool were collected at 5 and 31 days of age. No differences were observed on growth performance and isolation of Salmonella Enteritidis (p≥0.05). All probiotics applied were effective on reducing Salmonella Enteritidis colonization in the ileum, cecal tonsils, and cecum at 5 days of life. Probiotics T2 and T5 has shown effectiveness in reducing colonization at 31 days, being considered the most efficient on Salmonella Enteritidis control, for the intestines segments evaluated. It was not possible to affirm which probiotics formation, defined or undefined, is more efficient for Salmonella Enteritidis control.

O objetivo deste trabalho foi avaliar a eficácia dos probióticos de diferentes constituições: de culturas definidas e de culturas indefinidas no controle de Salmonella Enteritidis em frangos de corte, identificando qual a constituição e qual ou quais probióticos testados é mais eficaz. Foram inoculados 390 frangos de corte com 1ml de Salmonella Enteritidis, via oral, na concentração de 1,2 x 109 UFC (Unidades Formadoras de Colônia). O delineamento experimental utilizado foi o de blocos casualizados com 5 tratamentos e 6 repetições cada, totalizando 30 boxes com 13 aves/boxe (13 aves/m2). Os tratamentos foram fornecidos via água de bebida 1 hora após a inoculação, com 12 horas de tratamento com probióticos por dia, durante 3 dias consecutivos (1º, 2º e 3º dia de idade das aves). Os cinco tratamentos foram: T1 - Controle sem probiótico, T2 - Probiótico A (cultura definida - forma liofilizada, 7 cepas), T3 - Probiótico B (cultura definida - forma liofilizada, 11 cepas), T4 - Probiótico C (cultura indefinida - forma líquida), T5 - Probiótico D (cultura indefinida - forma liofilizada). O desempenho zootécnico foi avaliado usando o peso médio, a conversão alimentar e a mortalidade. Análises microbiológicas foram realizadas utilizando o método NMP (NMP/g)e isolamento de Salmonella através técnica de enriquecimento seletivo. Amostras de pool de íleo, tonsilas cecais e cecos e pool de fígado, coração e baço foram coletadas aos 5 dias e aos 31 dias de idade. Para desempenho zootécnico e isolamento de Salmonella Enteritidis não foram observadas diferenças (p≥0,05). Todos os probióticos utilizados foram eficazes na redução da colonização de Salmonella Enteritidis no íleo, tonsilas cecais e cecos aos 5 dias de idade e somente os probióticos do T2 (cultura definida) e T5 (cultura indefinida) reduziram a colonização aos 31 dias sendo considerados os mais eficazes no controle de Salmonella Enteritidis nestes segmentos intestinais avaliados. Não se pode afirmar quais das constituições de probióticos, culturas definidas ou indefinidas, são mais eficazes no controle de Salmonella Enteritidis.

E-mail: mtcvsoares@yahoo.com.br Colon cancer is one of the most common types of cancer in the world and the second leading cause of death related to the disease in developed countries. Up to 75% of cases are associated with eating, indicating that a person can reduce their risk simply through dietary modification. Studies in animals show that various strains of lactic acid bacteria protect against colon cancer in rodents although data in humans are limited and conflicting. The aim of this study was to investigate the efficacy of acid lactic bacteria in the treatment and reduction of colon cancer in animal models. Systematic searches were conducted in electronic databases reaching 1079 related articles, only six articles were elected according instead of to the eligibility criteria for analysis. All reviewed articles showed satisfactory results on the inhibition of colon cancer in rats and mice when using predominantly Lactobacillus strains. This study can answer hypothesis that acid lactic bacteria has antitumor effect against colon cancer.

Até 75% dos casos estão associados com a alimentação, indicando que uma pessoa pode reduzir o seu risco simplesmente através de modificação na dieta. Estudos em animais demonstram que várias cepas de bactérias ácido lácticas protegem contra o câncer de cólon em roedores, embora os dados em humanos sejam limitados e conflitantes. O objetivo deste estudo foi investigar a eficácia das bactérias ácido lácticas no tratamento e redução do câncer de cólon em modelo animal. Foram realizadas buscas sistemáticas em bases de dados eletrônicas alcançando 1079 artigos relacionados, entretanto apenas 6 artigos foram eleitos de acordo com os critérios de elegibilidade para análise. Todos os artigos avaliados apresentaram resultados satisfatórios quanto à inibição do câncer de cólon em ratos e camundongos ao utilizarem cepas predominantemente do gênero Lactobacillus. Este estudo pode responder a hipótese de que as bactérias ácido lácticas apresentam efeito preventivo e antitumoral contra o câncer de cólon.

We used 720 chicks with a day of life, divided into two groups: GA (control group) - birds fed diet without supplementation of Bacillus subtilis strain C3102 (1 to 35 days) and GB (treated group) - birds fed with supplementation of Bacillus subtilis strain C3102 (1 to 35 days). On the 7th and the 35th day, 2cm of jejunum were collected and submitted to histological processing. 1μm cross sections were stained by Alcian blue (AB) for detecting of sialomucins (pH 0.5) and sulphomucins (pH 2.5) as well as to count goblet cells. Histochemical analysis revealed a greater amount of sialomucins in the jejunum of animals that were supplemented with probiotic Bacillus subtilis strain C3102 (GB), when compared with the control group (GA). The number of goblet cells in the treated group (GB) was higher than the control group (GA).

O número de células caliciformes presentes no grupo tratado (GB) foi superior ao grupo controle (GA).

E-mail: janessabreu@yahoo.com.br This study evaluated the histomorphometric characteristics of tambaqui intestinal mucous after using Bacillus spp based probiotics, incorporated in feed and dissolved in water during transport. It was used a completely randomized design, with three treatments and seven replications, and a control treatment (fish fed commercial feed without probiotic) was compared with two other treatments involving the use of commercial probiotic incorporated in the feed or in the transport water. A total of 510 juvenile tambaqui (average weight and total length 83.26±28.14g and 17.39±1.90cm, respectively) was distributed in three cement tanks. During 60 days, fish of two tanks were fed with commercial feed and fish of the third tank received commercial feed supplemented with probiotic. After this period, the fish from each tank were divided into plastic bags and transported for 4 hours according to the following treatments: T1 = feeding with commercial feed (control); T2 = feeding with commercial feed and probiotic dissolved in the water during transport (20mg/L); T3 = feeding with commercial feed supplemented with probiotic (1.0g/kg feed). Before transport (basaline), 24 and 96 hours after transport, eight fish of each treatment were submitted to euthanasia and the intestine was removed for weighing and length measuring to establish the relation between length/gut ratio. Then, the anterior and posterior portions of the intestine were collected for evaluation of morphological and histological characteristics of intestinal mucous. The use of probiotic during transport did not affect the intestine weight of tambaqui. Regardless of the treatment, the intestine length increased as the fish increased in size. The supplementation with probiotics (Bacillus spp.) had no effect on height and length villus of tambaqui’s intestine during transport challenge.

Durante 60 dias, os peixes de dois tanques foram alimentados com ração comercial e os do terceiro tanque com ração comercial suplementada com probiótico. Após este período, os peixes de cada tanque foram divididos em sacos plásticos e transportados por 4 horas de acordo com os seguintes tratamentos: T1 = alimentação com ração comercial (controle); T2 = alimentação com ração comercial e probiótico adicionado na água durante o transporte (20mg/L); T3 = alimentação com ração comercial suplementada com probiótico (1,0g/kg de ração). Antes do transporte (basal), 24 e 96 horas após o transporte, oito peixes de cada tratamento foram submetidos a eutanásia e o intestino retirado para pesagem e mensuração do comprimento para estabelecimento da relação comprimento corporal/ intestino. Em seguida, foi realizada coleta da porção anterior e posterior do intestino, para avaliação das características morfo-histológicas da mucosa intestinal. O uso de probiótico durante o transporte não afetou o peso do intestino de juvenis de tambaqui. O comprimento do intestino do tambaqui não foi alterado pelo tratamento, sendo observada somente relação linear entre o comprimento intestinal e o comprimento corporal (CC) dos peixes. A suplementação com probiótico (Bacillus spp.) não exerceu nenhum efeito na altura e comprimento das vilosidades do intestino do tambaqui desafiados com o transporte.

E-mail: julietaengracia@gmail.com The aim of this study was to evaluate the survival rate, the intestinal microbiota, the mucosal integrity, and the carcass quality of juvenile Nile Tilapia, Oreochromis niloticus, after 80 days being fed on a diet containing probiotic additive (Bacillus cereus 4.0x108 CFUg-1 and Bacillus subtilis 4.0x108 CFUg-1), at the ratio of 4g/kg of pelleted feed. The completely randomized design with two treatments was used: one control group and one group fed on the mentioned diet. The evaluation of survival rate, the intestinal microbiota analysis by microbiological culture, histomorphometrical analysis of intestinal mucosa and chemical analysis of carcass was performed. The results showed that tilapias from the treated group had higher relative survival rate (P<0.05) than the control group, higher number of colony-forming units (P<0.05) regarding intestinal colonization by B. cereus and B. subtilis, and higher rates of intestinal mucosal integrity (P<0.05), evaluated by histomorphometry. As for the latter, the group being fed on feed with probiotic additive was observed to have higher and larger villi, besides having a higher number of goblet cells than the control group. Concerning the carcass quality, the results showed that there was positive interference (P<0.05) of the probiotic on the treated group in comparison to the control group as in regard to levels of protein and ether extract. These results allow the inference that the supplementation with probiotic, as tested in this experiment, led to the intestinal colonization by beneficial bacteria and resulted in higher relative survival rate, decreased the mucosal desquamation and helped in the increase of the number of goblet cells.

O delineamento experimental foi inteiramente casualizado com dois tratamentos, sendo um grupo controle e outro alimentado com dieta adicionada de probiótico. Foram realizados os cálculos do percentual de sobrevivência relativa, análise da microbiota intestinal por cultura microbiológica, análise histomorfométrica da mucosa intestinal e análise químico-bromatológica da carcaça dos peixes. Os resultados demonstraram que as tilápias do grupo tratado apresentaram percentual de sobrevivência relativa maior (P<0,05) que o do grupo controle e colonização intestinal por B. cereus e B. subtilis com maior (P<0,05) número de unidades formadoras de colônia em relação ao grupo controle. A análise histomorfométrica demonstrou que o grupo alimentado com aditivo probiótico apresentou vilosidades mais altas e mais largas, além de maior número de células caliciformes que o observado no grupo controle (P<0,05). Em relação à qualidade de carcaça os resultados demonstraram que houve interferência positiva (P<0,05) do probiótico no grupo tratado em relação ao controle quanto aos teores de proteína e extrato etéreo. Estes resultados permitem inferir que a suplementação com probiótico, como testado neste estudo, induziu a colonização intestinal por bactérias benéficas e promoveu maior percentual de sobrevivência relativa, diminuiu a descamação da mucosa, e favoreceu o aumento do número de células caliciformes.


#7 - Use of probiotics on the T cells activation and Salmonella Minnesota control in broiler chickens, 33(1):11-14

E-mail: santin@ufpr.br To evaluate the effect of probiotics on the immune response of broiler chickens challenged with Salmonella Minnesota (SM), 60 chickens were divided into three groups: CN - Birds that were not inoculated with SM (negative control), CP - birds inoculated with SM (positive control) and Probiotic- birds supplemented with probiotic consisting of Lactobacillus acidophilus, L. plantarium, L. rhamnosus, L. bulgaricus, Enterococcus faecium, Streptococcus thermophilus and Bifidobacterium bifidum in diet and inoculated with SM. At 14 days the birds were challenged with SM, and at 7 and 35 days were quantified goblet cells, CD4 + and CD8 + intestinal mucosa of the ileum and cecum. Birds supplemented with probiotics, at 7 days of age showed a significant increase (P&#8804;0.05) of goblet cells in the ileum and CD4 + and CD8 + cells in the cecum. At 35 days there were significant (P&#8804;0.05) of CD8 + cells in birds inoculated on CN and Probiotic. The use of probiotics provided a significant reduction (P&#8804;0.05) of Salmonella sp. counts.

E-mail: santin@ufpr.br Para avaliar o efeito do probiótico sobre a resposta imunológica de frangos de corte desafiados com Salmonella Minnesota (SM), 60 frangos foram divididos em três grupos: CN- (controle negativo) aves que não foram inoculadas com SM, CP- (controle positivo) aves inoculadas com SM e Probiótico- aves suplementadas na ração com probiótico composto de Lactobacillus acidophilus, L. plantarium, L. rhamnosus, L. bulgaricus, Enterococcus faecium, Streptococcus thermophilus e Bifidobacterium bifidum e desafiadas com SM. Aos 14 dias foi realizada a inoculação com SM e aos 7 e 35 dias foram quantificadas células caliciformes, CD4+ e CD8+ na mucosa intestinal do íleo e ceco. Aves suplementadas com probióticos aos 7 dias de idade apresentaram aumento significativo (P&#8804;0,05) de células caliciformes e CD4+ no íleo e de células CD8+ no ceco. Aos 35 dias houve aumento significativo (P&#8804;0,05) das células CD8+ nas aves inoculadas do CN e Probiótico. A utilização de probióticos proporcionou redução significativa (P&#8804;0,05) da contagem de Salmonella sp.

IgA production, coliforms analysis and intestinal mucosa morphology of piglets that received probiotics with viable or inactivated cells. Pesquisa Veterinária Brasileira 27(6):241-245. Email: fjhblazq@usp.br Two types of probiotics were used in piglets. One product is a mixed culture of viable Lactobacillus acidophilus, Enterococcus faecium e Bifidobacterium bifidum. The second product is composed of inactivated Lactobacillus acidophilus cells. The piglets received two weekly oral doses for 30 days while a control group did not receive probiotics. All piglets were euthanized at the 30th day of life and the mesenteric lymph nodes, the small intestine, and blood samples were collected. The tissue samples were studied by light microscopy and the blood serum was analyzed by ELISA method. The treatment with the probiotic with viable cells produced higher serum levels of IgA (P<0.05) and more IgA expressing cells were found in the mesenteric lymph nodes than observed in the inactivated cells treatment or control groups (P<0.05). Also, intestinal villi were longer, crypts were deeper (P<0.05) and fecal coliform count was lower than found in the inactivated product (P<0.05). These results suggest that viable probiotics are more efficient than inactivated probiotics to induce immunostimulation and intestinal modifications in piglets, thus improving their health and development.
