#1 - Iron interference in hemoglobin production in piglets from birth to weaning

Abstract in English:

Iron deficiency anemia (IDA) in humans is defined as the decrease of total hemoglobin concentration and the non-production of the adult hemoglobin subtype 2 – HbA2 (α2δ2 chains), which is considered a marker of IDA severity in humans, dosed together with the iron serum. This study aimed to determine the standard of hemoglobin types in piglets induced to experimentally IDA in the first 21 days of life (delivery to weaning). In the present study, 40 piglets born from four naïve gilts, were randomly and equally assigned among the gilts. On the third day after delivery, the groups were randomly distributed in different environments (cement and clay floors) and according to the iron supplementation (iron dextran and placebo). Erythrocyte parameters, serum iron, and hemoglobin trait were analyzed at four moments between birth and weaning days. The group of piglets that did not receive iron dextran supplementation on the third-day post-birth and were placed in the pen without soil did not present HbA2 from the seventh day onwards on the agarose electrophoretogram (pH 8.6) and this observation was correlated to decrease of serum iron (ρ: 0.156, p=0.003) when compared to the other groups’ piglets that did not present iron deficiency. In the present study was possible to determine the swine hemoglobin pattern in IDA, since HbA2 was absent in piglets with IDA in comparison to the non-ferropenic groups and the correlation between the reduction of iron levels and the absence of HbA2.

A anemia por deficiência de ferro (ADF) em humanos é definida como a diminuição da concentração de hemoglobina total e a não produção da hemoglobina adulta subtipo 2 – HbA2 (cadeias α2δ2), que é considerada um marcador de gravidade de IDA em humanos, dosado em conjunto com o soro de ferro. Este estudo teve como objetivo determinar o padrão dos tipos de hemoglobina em leitões induzidos experimentalmente à IDA nos primeiros 21 dias de vida (parto ao desmame). Quarenta leitões, nascidos de quatro marrãs nulíparas, foram distribuídos aleatoriamente e igualmente entre as leitoas. No terceiro dia após o parto, os grupos foram distribuídos aleatoriamente em diferentes ambientes (piso de cimento e barro) e de acordo com a suplementação de ferro (ferro dextrano e placebo). Parâmetros eritrocitários, ferro sérico e traço de hemoglobina foram analisados em quatro momentos, entre o nascimento e o desmame. O grupo de leitões que não recebeu suplementação de ferro dextrano no terceiro dia pós-parto e foi colocado em baia sem solo não apresentou HbA2 a partir do sétimo dia no eletroforetograma de agarose (pH 8,6) e esta observação foi correlacionada com diminuição da concentração sérica ferro (ρ: 0,156, p=0,003) quando comparados aos demais grupos leitões que não apresentavam deficiência de ferro. No presente estudo foi possível determinar o padrão hemoglobinêmico suíno na IDA, uma vez que, a HbA2 estava ausente nos leitões com ADF em comparação aos grupos não ferropênicos e há correlação entre a redução dos níveis de ferro e a ausência de HbA2.


#2 - Influence of organic chromium supplementation on the performance of beef calves undergoing weaning-related stress

Abstract in English:

Dietary chromium supplementation before, during, and after weaning was conducted to evaluate the hypothesis that chromium supplementation could reduce weaning-induced cortisol release in beef calves. We examined the effects of chromium supplementation in 150 crossbred calves (male and female) between five and six months of age. The calves were randomly divided by sex and breed into two equal homogeneous groups (n=75). One group was used as the control, and the other experimental group received supplementation with 0.9mg of chromium carbon-amino-phospho-chelate per 100kg BW. The chromium supplement was mixed with mineral salt for the consumption of 0.1% of BW, and the supplement was administered via creep feeding 60 days before and 60 days after forced weaning. Calves were weighed, and their blood and urine samples were obtained at four time-points: T0 (60 days before weaning), T1 (at weaning), T2 (48 hours after weaning), and T3 (60 days after weaning). Blood samples were used to determine chromium, cortisol, total protein, and albumin concentrations, and urine samples were used to determine urinary creatinine and chromium levels. Cumulative weight gain was higher in calves supplemented with chromium before weaning and during the experiment (P<0.05). In addition, weaning-related stress caused an increase in chromium excretion in the urine, and chromium supplementation reduced stress, which resulted in lower cortisol and total protein levels during weaning.

O estudo foi realizado para avaliar a hipótese de que a suplementação dietética com cromo antes, durante e após a desmama possa diminuir a concentração de cortisol causado por este processo em bezerros de corte. Para tal, foram utilizados 150 bezerros mestiços, machos e fêmeas, entre cinco e seis meses de idade. Os animais foram divididos randomicamente por sexo e grupo genético em dois grupos homogêneos (n=75), um mantido como controle e outro suplementado com 0,9mg de carboaminofosfoquelato de cromo/100 kg PV misturado a um sal proteinado para ser consumido na base de 0,1% do PV via creep feeding, no decorrer de 60 dias antes e 60 dias após à desmama forçada. Os animais foram pesados e foram coletadas amostras sanguíneas e urinárias no M0 (60 dias antes da desmama), M1 (desmama), M2 (48 horas após a desmama) e M3 (60 dias após à desmama) para determinação de cromo, cortisol, proteína total e albumina no sangue e da concentração urinária de creatinina e cromo. O ganho acumulado de peso foi superior nos bezerros suplementados com cromo antes da desmama e no decorrer de todo o experimento (P<0,05). A suplementação com cromo reduziu os teores de cortisol e de proteína total durante a desmama. O estresse da desmama provocou aumento da excreção de cromo pela urina.

E-mail: crisgtitto@usp.br Weaning is a critical phase in pigs’ life due to maternal separation and the introduction of a new diet. Infrared thermography measurement taken in the ocular region appears to be a reliable indicator of the stress condition of the pig. The aim of this study was to evaluate the relationship between ocular surface temperature by infrared thermography and cortisol in piglets post weaning. Sixty-six piglets were evaluated once a week, during 7 weeks, in two periods of the day (7am and 15pm) and ocular surface temperature and dorsal surface temperature were collected using a thermographic camera and a laser surface thermometer, respectively. Saliva was also collected to determine salivary cortisol. Statistical analysis included fixed effects of week and period of the day and their interaction, and relationship between thermography, dorsal surface temperature and cortisol were done by Pearson’s correlations with 5% significance level. Salivary cortisol did not differ between periods, but it was higher in the first three weeks after weaning (P<0.05). During the first two weeks after weaning cortisol presented high and positive correlation (P<0.05) between ocular surface temperature (0.89) and dorsal surface temperature (0.80). The two superficial temperatures had a moderate and positive association (r=0.41; P<0.0001) during all experiment. This study highlights that the ocular surface temperature obtained through infrared thermography can be a superficial body temperature indicators, besides being a non-invasive and fast method of measurement. However, more research is needed to deepen the relationship between ocular surface temperature and cortisol during chronic stress.

A termografia infravermelha medida na região ocular se mostra como um indicador confiável para a condição de estresse pontual de suínos. O objetivo deste estudo foi determinar a relação entre medidas de termografia infravermelha da superfície ocular e de cortisol em situações de estresse em leitões na pós desmama. Foram avaliados 66 leitões, uma vez por semana, durante sete semanas, em dois períodos do dia (7h e 15h) com medidas de temperatura superficial ocular, temperatura superficial do dorso e amostras de saliva para determinação de cortisol salivar. A análise estatística contemplou os efeitos fixos de semana e período do dia e sua interação e correlações de Pearson para relação entre termografia ocular, temperatura superficial e cortisol a 5% de significância. Cortisol salivar não diferiu entre os períodos, mas foi superior nas três primeiras semanas após o desmame (P<0,05). Nas duas primeiras semanas após a desmama o cortisol apresentou correlação alta e positiva (P<0,05) com a temperatura ocular máxima (0,89) e a temperatura superficial do dorso (0,80). As duas temperaturas superficiais apresentaram uma associação moderada positiva (r=0,41; P<0,0001) durante todo o período experimental. Este estudo destaca que a temperatura de superfície ocular obtida por meio da termografia infravermelha pode ser um indicador de temperatura de superfície corporal e estado de bem-estar de leitões em fase de creche, além de ser um método não invasivo e de rápida mensuração. Entretanto, mais pesquisas são necessárias para aprofundar a relação entre temperatura ocular e cortisol durante estresse crônico.

E-mail: wdzlopes@hotmail.com The main importance of eimeriosis in cattle is due to lower performance shown with the disease in its sub-clinical form. This study evaluated the efficacy of lasalocid used against Eimeria spp. parasitizing calves. We also evaluated the weight gain of calves submitted to different treatments and analyzed some epidemiological factors that might interfere with Eimeria infection; 288 calves were used in the study. The calves of treatment 1 received protein mineral salt in low consumption without lasalocid, while the calves of treatment 2 received protein mineral salt on low consumption with lasalocid, administered orally to 4 to 10-month-old calves. Harvest of feces and weight control was made on days 0 (before the start of the experiment), at weaning, and 30 and 60 days after weaning (DAW). Evaluation of some epidemiological factors which could be related to infection by Eimeria spp. of the calves, such as weaning, sex and time of year, were analyzed, taking into account the results regarding the 144 calves of the control group. Nine species of Eimeria were identified in descending order: E. brasiliensis, E. wyomingensis, E. bovis, E. canadian, E. zuernii, E. auburnensis, E. ellipsoidalis, E. pellita and E. cylindrica. Unexpectedly, decrease in parasite load could be observed after weaning. Even the farm did not adopt management measures aimed for greater productivity, as Artificial Insemination in Fixed Time, which in turn ends up with increase of the number of births and animal unit per hectare at a certain period of year, high parasitism of coccidia was diagnosed in calves of the control group. Oocyst counts per gram (OPG) of calves treated with lasalocid were significantly lower (P&#8804;0.05) in the control group. The compound achieved &#8805;95% efficacy against the parasite in question. At the end of the study, calves fed lasalocid gained on average 7.2kg (p&#8804;0.05) more than calves in the control group. For a farm that aims to sell calves soon after weaning, is recommended to start treatment with lasalocid, with the creep-feeding, from an age of three months on, since the weight gain calves treated with lasalocid was significantly (p=0.05) higher compared with the weight gain of the control group after five months of treatment. The difference in weight gain of calves treated with lasalocid compared with caves in the control group may also be partially related to the infection by Eimeria spp., as discussed in this paper.

E-mail: wdzlopes@hotmail.com A principal importância da eimeriose em bovinos, se deve ao baixo desempenho produtivo que os animais demonstram quando esta enfermidade apresenta-se sob a forma sub-clínica. Como objetivos, o presente trabalho avaliou a eficácia do uso da lasalocida sódica contra espécies de Eimeria spp. parasitando bezerros; avaliou também o desempenho ponderal dos animais submetidos aos diferentes tratamentos e analisou alguns fatores epidemiológicos que possam interferir na infecção por Eimeria nos bezerros. Foram utilizados 288 bezerros no dia 0 do estudo. Os animais pertencentes ao tratamento 01 receberam sal mineral proteinado de baixo consumo sem adição de lasalocida, enquanto que os bezerros do Tratamento 02 sal mineral proteinado de baixo consumo, com adição de lasalocida sódica, administrado via oral para bezerros dos quatro/cinco/seis meses até dez meses de idade. Colheita de fezes e pesagem dos animais foram realizadas nos dias 0 (antes do início do experimento), na desmama, 30 e 60 dias após desmama (DPD). A avaliação de alguns fatores epidemiológicos que pudessem ser relacionados com a infecção por Eimeria spp nos bezerros, como o desmame, sexo e época do ano, foram analisados neste estudo, levando-se em consideração os resultados encontrados durante todo estudo, para os 144 animais pertencentes ao grupo controle. Foram identificadas nove espécies de Eimeria nos bezerros em ordem decrescente: E. brasiliensis, E. wyomingensis, E. bovis, E. canadenses, E. zuernii, E. auburnensis, E. ellipsoidalis, E. pellita e E. cylindrica. Inesperadamente, diminuição na carga parasitária dos animais pode ser observada após o desmame. Mesmo a fazenda não adotando medidas de manejo que visam maior produtividade como a Inseminação Artificial em Tempo Fixo, que por sua vez acaba aumentando o número de nascimentos e unidade animal/hectare em uma determinada época do ano, elevado parasitismo pelo coccídio em questão foi diagnosticado nos bezerros pertencentes ao grupo controle. Talvez a época do ano em que o estudo foi realizado pode ter influenciado neste aspecto. As contagens de oocistos por grama (OoPG) de fezes para Eimeria dos animais tratados com lasalocida foram estatisticamente inferiores (P&#8804;0,05) as do grupo controle após o início do estudo. O composto alcançou eficácia &#8805;95% contra o parasito em questão. No final do estudo, os animais que receberam lasalocida ganharam em média, 7,2kg a mais (P&#8804;0,05) que os bezerros pertencentes ao grupo controle. Em propriedades que tem como objetivo a venda de bezerros logo após a desmama, recomenda-se o início do tratamento com a lasalocida, junto ao creep-feeding, a partir de três messes de idade, uma vez que diferencial no ganho em peso médio dos bezerros tratados foi significativamente (P&#8804;0,05) mais elevado, em comparação ao grupo controle, após cinco meses de tratamento com o referido composto. Apesar de a lasalocida ser utilizada como um aditivo alimentar para animais, a diferença no ganho em peso vivo médio entre animais tratados com a lasalocida, em comparação a animais pertencentes ao grupo controle, também pode ser relacionada, em partes, a infecção dos animais por Eimeria spp., conforme discutido neste artigo, entretanto, futuros estudos devem ser conduzidos para comprovar esta hipótese.

E-mail: davidbarcellos@terra.com.br The aim of this study was to assess the frequency and association of virulence factors of Escherichia (E.) coli isolated from weaned piglets with diarrhea and to correlate it with fecal consistency. A total of 152 rectal swabs were collected from 25-40 day-old piglets with diarrhea, in farms of Southern Brazil. Phenotypical and molecular techniques were used for bacterial isolation, characterization and classification of enterotoxigenic E. coli (ETEC) pathotypes. Statistical analysis was carried out to determine the frequency of virulence factors and virotypes, of fimbriae F4, F5, F6, F18, F41 and toxins LT, STa, STb and STx2e. Out of 456 E. coli isolates, 287 (62.9%) samples showed significant growth of E. coli. Among them, 194 (67.6%) samples showed at least one virulence factor, indicating that ETEC is an important etiological agent of diarrhea in weaned piglets. Higher frequencies were found of fimbria F4 and F18 and enterotoxins LT, STa and STb. Significant association was found to F4, LT, STa and STb; between F18 and STa and STx2e; between F5 and LT, STa and STb. The most frequent virotypes were F18-STa, F4-LT-STa-STb, F4-STa, F4-LT-STb and F18-STa-STx2e. Beta-hemolysis was observed in 47.4% of samples and there was significant association between hemolytic samples and virulence factors F4, F18, STa and STx2e. Regarding fecal consistency, there was significant association of liquid feces and F4 fimbria, STa toxin and virotypes F4-STa and F4-F5-LT-STa-STb. Since there was significant association of ETEC and liquid feces in nursery piglets, it is important to prioritize the sampling of liquid feces for the diagnosis etiologic cause of diarrhea.

Foram utilizadas técnicas fenotípicas e moleculares para isolamento bacteriano, caracterização e classificação de patotipos de E. coli enterotoxigênica (ETEC). A análise estatística foi realizada para determinar a frequência de fatores de virulência e virotipos, de fímbrias F4, F5, F6, F18, F41 e toxinas LT, STa, STB e STx2e. Duzentas e oitenta e sete (62,9%) amostras apresentaram crescimento significativo de E. coli. Entre os quais, 194 (67,6%) amostras apresentaram pelo menos um fator de virulência, indicando que ETEC é um importante agente etiológico de diarreia em leitões desmamados. As frequências mais elevadas foram encontradas para as fímbrias F4 e F18 e enterotoxinas LT, STa e STb. Associação significativa foi encontrada para F4, LT, STa e STb; entre F18 e STa e STx2e; entre F5 e LT, STa e STb. Os virotipos mais frequentes foram F18-STa, F4-LT-STa-STb, F4-STa, F4-LT-STb e F18-STa-STx2e. Beta-hemólise foi observada em 47,4% das amostras e houve associação significativa entre amostras hemolíticas e fatores de virulência F4, F18, STa e STx2e. Quanto consistência fecal, houve associação significativa de fezes líquidas e fímbria F4, toxina STa e virotipos F4-STa e F4-F5-LT-STa-STb. A associação significativa da ETEC e fezes líquidas em leitões de creche, é importante para priorizar a amostragem de fezes com essa consistência para no diagnóstico etiológico da diarreia.

E-mail: jlacerda@fcav.unesp.br The perinatal phase of foal development is one of the most challenger period for the immune system. The present study has analyzed serum protein profile, considering variables related to immunity in foals at the perinatal phase, verifying passive immunity transfer. The group 1 (G1) contained eight foals evaluated one day after weaning, and group 2 (G2) included twenty foals at more than thirty days after weaning. Total protein concentration was determined by means of refractometry. The concentration of serum proteins was determined through sodium dodecyl sulphatepolyacrylamide gel electrophoresis. Results were submitted to analysis of variance and Tukey test (P<0.05). IgA concentration showed difference (P<0.05) between the two studied groups, however data were within adult healthy horses normal values. IgG didn’t show statistical difference (P>0.05). The cellular immunity establishment occurs in the neonatal phase, in foals that suckled colostrum and milk properly. The present study showed differences in serum protein profile, considering variables related to immunity, in foals immediately after weaning comparing to foals at more than 30 days after weaning. According to the observed values, we conclude that foals, even early weaned, showed proper passive immunity tranfer.

O objetivo do presente estudo foi verificar o perfil protéico sérico de parâmetros relacionados à imunidade de equinos jovens no período perinatal, verificando-se a transferência de imunidade passiva. Oito animais desmamados há um dia, formaram o Grupo 1 (G1), enquanto vinte animais desmamados há mais de trinta dias formaram o Grupo 2 (G2). A concentração sérica de proteína total foi determinada por refratometria. Para o fracionamento das proteínas, utilizou-se eletroforese em gel de acrilamida. Os resultados obtidos foram submetidos à análise de medidas repetidas e ao teste Tukey (p<0,05) para comparação das médias. As concentrações de IgA apresentaram diferença (p<0,05) entre os grupos, porém os valores observados encontravam-se dentro do considerado normal para equinos adultos. Não houve diferença (p>0,05) nas concentrações de IgG. O estabelecimento adequado da imunidade celular ocorre durante a fase neonatal, nos animais que ingerem adequadamente o colostro e o leite. O presente estudo determinou diferenças no perfil protéico sérico de parâmetros relacionados à imunidade de equinos jovens no período imediato ao desmame, comparados com animais desmamados há mais de 30 dias. De acordo com os valores observados, concluiu-se que os animais, mesmo desmamados precocemente, obtiveram transferência adequada de imunidade passiva.

Blood, serum, nasal and rectal swabs samples were collected of sows, farrowing piglets, nursery, growing and finishing pigs. In E1, were sampled 40 animals in each category (G1a and G2). In E2, (G1b and G3), 35 farrowing piglets were identified and sampled along the production cycle. Porcine circovirus type 2 (PCV2) antibodies were assayed. A PCR was used to detected PCV2 genome in blood and swabs. In E1, sows had high rates of viremic and seropositives animals, with percentage of sows with high antibodies titers greater than G2. Passive antibodies decline occurred between nursery and growing area with increased viral shedding in swabs and subsequent seroconversion in G1. In G2, the passive antibodies decay occurred in nursery, with a reduction in viral shedding. In E2, the decline of maternal immunity occurred between the 1st and 2nd collection in G1b, and between 2nd and 3rd collections in G3. In both herds, the decay of passive immunity coincided with increased viremia and viral shedding; and seroconversion occurred between the 3rd and 4th collection in both herds with decline of viremia. Viremia and viral shedding was detected in all samples days, 42% of animals sampled in E2 were viremic and all tissue samples collected at slaughter were positive for PCV2. This study confirms the persistence of viremia even in the presence of high titers of antibodies and the serological profile in a herd with or without PMWS may be different, especially with regard to the passive immunity duration.

Em E1, nas granjas G1a e G2, foram amostrados 40 animais de cada categoria. Em E2, nas granjas G1b e G3, 35 leitões na maternidade foram identificados e amostrados ao longo do ciclo produtivo. O soro foi avaliado para presença de anticorpos contra circovírus suíno tipo 2 (CVS2) e sangue e swabs para presença do ácido nucléico viral. Em E1, a categoria porcas possuía altas taxas de animais virêmicos e soropositivos, com porcentagem de porcas com títulos altos superior a G2. Em G1a a queda de imunidade passiva ocorreu entre o final da fase de creche e início da recria com aumento da eliminação viral em swabs e subsequente soroconversão. Em G2 a queda ocorreu entre a fase final da maternidade e início da creche, com diminuição da eliminação viral. Em E2, a queda da imunidade materna ocorreu entre a 1ª e 2ª coleta em G1b; e em G3, entre a 2ª e 3ª coleta. Em ambas as granjas, a queda de imunidade passiva coincidiu com o aumento da viremia e eliminação viral e a soroconversão ocorreu entre a 3ª e 4a coleta em ambas as granjas com aumento da média de título de anticorpos e declínio da viremia. Viremia e eliminação viral foram detectadas em todas as coletas realizadas; 42% dos animais amostrados em E2 foram virêmicos em todas as coletas e todas as amostras de tecido coletadas no abate foram positivas para o CVS2. Este estudo confirma a persistência da viremia mesmo em presença de altos títulos de anticorpos e que o perfil sorológico em um rebanho com e sem a presença da síndrome pode ser diferente, principalmente em relação à duração da imunidade passiva.

Selected samples were processed for paraffin embedding and stained with hematoxylin and eosin. Immunohistochemistry using anti-PCV2 (porcine circovírus type 2) antibody, anti-Helicobacter pylori antibody and a wide-spectrum anti-cytokeratin antibody was performed. Gross changes in pars oesophagea were classified according to the severity of lesions as score 3, 2, and 1 in 8, 6, 5 stomachs respectivelly. Microscopically, hyperplastic lymphoid follicles, lymphohistiocytic inflammatory infiltrates and focci of necrosis in the gastric mucosa were common findings. Large amounts of PCV2 antigen were observed in the cytoplasm and nuclei from intralesional cells and debris from the gastric glandular mucosal zone; however, in the fundus, anti-PCV2 immunostaining was restricted to the surface mucosal cells and foveolar compartment. All gastric lymph nodes were positive for PCV2 antigen. Anti-H. pylori immunostaining was seen in eleven cases, mainly in the antrum, on the mucosal surface and foveolar compartment. The association of the anti-PCV2 immunostaining with the glandular mucus-producing cells suggests a role for PCV2 as an additional factor for the swine ulcer development.

PCV2 antigen was immunohistochemically demonstrated in the cytoplasm and nuclei from intralesional epithelial cells, histiocytes, and endothelial-like cells in intestinal tissues. Together these findings imply an association with PCV2. The presence of co-infections by Lawsonia intracellularis, Brachyspira spp., Mycobacterium spp., Salmonella spp., rotavirus, parvovirus, coronavirus and enteric calicivirus with PCV2 in the intestinal lesions was investigated.

The second product is composed of inactivated Lactobacillus acidophilus cells. The piglets received two weekly oral doses for 30 days while a control group did not receive probiotics. All piglets were euthanized at the 30th day of life and the mesenteric lymph nodes, the small intestine, and blood samples were collected. The tissue samples were studied by light microscopy and the blood serum was analyzed by ELISA method. The treatment with the probiotic with viable cells produced higher serum levels of IgA (P<0.05) and more IgA expressing cells were found in the mesenteric lymph nodes than observed in the inactivated cells treatment or control groups (P<0.05). Also, intestinal villi were longer, crypts were deeper (P<0.05) and fecal coliform count was lower than found in the inactivated product (P<0.05). These results suggest that viable probiotics are more efficient than inactivated probiotics to induce immunostimulation and intestinal modifications in piglets, thus improving their health and development.
