#1 - Diagnosis of Brachyspira pilosicoli, Brachyspira hyodysenteriae and Brachyspira intermedia in hens and laying hens in the western region of Paraná through bacterial isolation and identification in qPCR

Abstract in English:

Bacteria of the genus Brachyspira can cause enteric diseases in poultry causing a decrease in productivity. The occurrence of this disease in chickens has already been verified in countries such as Australia, Italy, and the United States, but in Brazil, until now, epidemiological studies about Brachyspira sp. frequency were only carried out on pig farms. The objective of this study was to evaluate the presence of bacteria of the genus Brachyspira sp. through isolation and confirmation of the species Brachyspira pilosicoli, Brachyspira hyodysenteriae and Brachyspira intermedia using the qPCR technique. Samples from 110 hens aged from 35 to 82 weeks were collected, 40 were from commercial egg farms and 70 were from laying hens matrices. For the first evaluation, bacterial isolation was performed from the feces. Positive samples were submitted to qPCR to identify the three species proposed. Cecum fragments of the birds were collected and fixed in formaldehyde for histological evaluation and counting of goblet cells. Of the 110 samples, 48 characteristic isolates of Brachyspira (43.6%) were obtained and of these in qPCR 13 identified as B. hyodysenteriae (11.8%) and 5 all from the same farm as Brachyspira intermedia (4.5%), 2 samples were positive for both agents (1.8%) and 28 were not characterized by qPCR (25.5%). None histopathological lesions were observed in the chicken cecum and no significant statistical difference was noticed in the count of goblet cells of the positive hens. It can be evidenced by the occurrence of Brachyspira sp. in laying farms and hens in Brazil, with special relevance to Brachyspira intermedia that can be potentially pathogenic for these animals.

Bactérias do gênero Brachyspira podem ocasionar enfermidades entéricas em aves acarretando a queda de produtividade. A ocorrência desta enfermidade em galinhas já foi verificada em países como a Austrália, Itália e Estados Unidos, porém no Brasil, até o momento, trabalhos epidemiológicos sobre a frequencia de Brachyspira sp. só foram realizados em granjas de suínos. O objetivo deste estudo foi avaliar a presença de bactérias do gênero Brachyspira sp. através do isolamento e confirmação das espécies Brachyspira pilosicoli, Brachyspira hyodysenteriae e Brachyspira intermedia utilizando a técnica de qPCR. Foram coletadas amostras de 110 aves com idade entre 35 e 82 semanas, sendo 40 de granjas de postura comercial e 70 de granjas de matrizes de corte. Para avaliação primeiramente procedeu-se o isolamento bacteriano a partir das fezes. As amostras positivas foram submetidas a qPCR para identificação das três espécies propostas. Fragmentos de ceco das aves foram coletados e fixados em formol para avaliação histológica e contagem de células caliciformes. Das 110 amostras foram obtidos 48 isolamentos característicos de Brachyspira (43,6%) e destes na qPCR 13 identificadas como B. hyodysenteriae (11,8%) e 5 sendo todas da mesma granja (4,5%) como B. intermedia, 2 amostras foram positivas para ambos os agentes (1,8%) e 28 não foram caracterizadas através da qPCR (25,5%). Não foram observadas alterações histopatológicas no ceco e diferença estatística significativa na contagem de células caliciformes das aves positivas. Conclui‑se que a Brachyspira sp. é frequente em granjas de poedeiras e matrizes de corte no Brasil, com especial relevância para a B. intermedia que possui potência patogênico para estas aves.

E-mail: juliavetufmg@yahoo.com.br Seventeen Holstein newborn calves were used with the objective of evaluating the influence of milk replacer volume in the pattern of pathogens causing neonatal diarrhea. The animals were divided into two groups, 8 calves in group 1 and 9 calves in group 2. The calves were fed twice daily in the total of 4 liters of milk replacer to group 1 and 6 liters to group 2. From the 1st day of arrival of the calves feces were evaluated daily after the morning feeding for the classification of diarrheal feces or without diarrhea. The first day of diarrhea until the seventh day, feces were collected on alternate days (1st, 3rd, 5th and 7th day) directly from the rectum to evaluate enteropathogens. Were collected blood samples from calves with five days of age for determination of total protein. The average total protein was 6.33 and 6.21g/dL in groups 1 and 2, respectively. The group 2 tended (p<0.1) higher consumption of milk replacer during the study period. The volume of milk replacer did not influenced the incidence of diarrhea and the frequency of positive samples for each etiologic agent between the two groups (p>0.05). Also, there was no difference (p>0.05) on the pattern of the frequency of positive samples for evaluated pathogens. The frequency of pathogens in the samples was 100 and 75% for Cryptosporidium, 28.5 and 43.7% for Salmonella spp., 28.5 and 15.6% for E. coli pathotypes, 3.5 and 6.2% for Rotavirus and 10.7 and 9.4% for Giardia in groups 1 and 2, respectively. Serotypes of Salmonella infantis and muenster were found. The isolated pathotypes of E. coli isolates were classified as Escherichia coli enteropathogenic, enterotoxigenic and Shiga-toxin-producing 1 and 2. Associations between Cryptosporidium spp. and E. coli pathotypes, and between Cryptosporidium spp. and Salmonella spp. were found in 30% of the samples in group 1 and in 45.5% in group 2, respectively. Our results showed that the different volumes of milk replacer did not influence the incidence and etiology of neonatal diahrrea. Longitudinal evaluation of enteropathogens during patency demonstrated that the association between the etiologic agents starts from the first day of disease. This result highlighted the great importance of the infection by Cryptosporidium spp. which was present in every moments and animals evaluated.

E-mail: juliavetufmg@yahoo.com.br Foram utilizados 17 bezerros, recém nascidos, da raça Holandesa, com o objetivo de avaliar a influência do volume de sucedâneo nos principais patógenos causadores de diarreia neonatal. Os animais foram distribuídos em dois grupos, 8 bezerros do grupo 1 e 9 bezerros do grupo 2. Os animais foram alimentados duas vezes ao dia totalizando 4 litros de sucedâneo diários para o grupo 1 e 6 litros para o grupo 2. A partir do 1° dia de chegada dos bezerros foram avaliadas as fezes diariamente após o aleitamento da manhã para a classificação das fezes em diarreicas ou não diarreicas. Do primeiro dia de diarreia até o sétimo dia, as fezes foram coletadas em dias alternados (1º, 3º, 5º e 7° dia) diretamente da ampola retal para avaliação dos enteropatógenos. Foram coletadas amostras de sangue dos bezerros com cinco dias de idade para dosagem da proteína total. A média da proteína total foi 6,33 e 6,21g/dL nos grupos 1 e 2 respectivamente. O grupo 2 apresentou tendência (p<0,1) de maior consumo de sucedâneo no período avaliado. A quantidade de sucedâneo oferecida aos animais não influenciou a incidência de diarreia e sua etiologia, ou seja, não foi observada diferença (p>0,05) na frequência das amostras positivas para cada agente entre os grupos. A frequência dos enteropatógenos nas amostras foi de 100 e 75% para Cryptosporidium spp.; 28,5 e 43,7% para Salmonella spp.; 28,5 e 15,6% para patotipos de E. coli; 3,5 e 6,2% para Rotavírus e 10,7 e 9,4% para Giardia sp. nos grupos 1 e 2 respectivamente. Foram encontrados os sorotipos de Salmonella infantis e muenster. Os patotipos de E. coli isolados foram classificados como E. coli enterohemorrágica, enteropatogênica, enterotoxigênica e produtoras de toxinas Shiga 1 e 2. Foi observada associação entre o Cryptosporidium spp. e os patotipos de E. coli em 30% das amostras do grupo 1 e Cryptosporidium spp. e Salmonella spp. em 45,5% no grupo 2. Os resultados do presente trabalho demonstraram que o fornecimento de diferentes volumes de sucedâneo não apresentou influência sobre a incidência e etiologia da diarreia neonatal. A avaliação longitudinal dos enteropatógenos durante o período de patência da diarreia demonstrou que a associação entre eles ocorre a partir do primeiro dia da doença e destacou a importância da infecção pelo Cryptosporidium spp. agente encontrado em todos os momentos e animais.


#3 - Levantamento dos enteropatógenos de leitões do nascimento a sete dias de idade no Brasil, 33(8):963-969

E-mail: guedesufmg@gmail.com The purpose of the study was to evaluate the real importance of anaerobic enteropathogens and rotavirus in contrast to more common agents as cause of diarrhea in piglets within the first week of life. Sixty 1- to 7-day-old piglets, 30 diarrheic and 30 non-diarrheic (control), from 15 different herds were selected, euthanized and necropsied. Samples of the jejunum, ileum, colon, cecum and feces were collected from the piglets and analyzed to determine the presence of the following enteropathogens: enterotoxigenic Escherichia coli (ETEC), Clostridium perfringens types A and C, Clostridium difficile, rotavirus and Isospora suis. Among diarrheic piglets, 23.3% were positive for C. difficile, 70% for C. perfringens type A cpb2+, 14.3% for rotavirus and 10% for ETEC. Among non-diarrheic control piglets, 10% were positive for C. difficile, 76.7% for C. perfringens type A cpb2+, 0% for rotavirus, 3.3% for ETEC and 3.3% for I. suis. C. perfringens type C was not detected in any of the animals. Histological lesions characteristic of C. difficile, E. coli and rotavirus were observed. However, no C. perfringens type A suggestive lesions were detected. There was a positive correlation between mesocolon edema and the presence of C. difficile toxins. Although C. perfringens type A cpb2+ was the most frequently detected enteropathogen, there was no association between its presence and diarrhea or macro or microscopic changes. C. difficile and Rotavirus were the most relevant pathogens involved with neonatal diarrhea in this study, and histopathology associated with microbiological test proved to be the key to reach a final diagnosis.

Entre os animais diarréicos, 23.3% foram positivos para C. difficile, 70% para C. perfringens tipo A cpb2+, 14.3% para rotavirus e 10% para ETEC. Entre os leitões não-diarréicos, 10% foram positivos para C. difficile, 76.7% para C. perfringens tipo A cpb2+, 3.3% para ETEC e 3.3% for I. suis. For enteropathogenic bacteria of the Enterobacteriaceae family, 1ml samples were transferred for enrichment in Rappaport- Vassiladis broth and Kauffmann Tetrathionate Broth followed by streak onto Hektoen Enteric Agar and Salmonella-Shigella Agar. Simultaneously at each visit samples of water from fish and macrophyte tanks were collected for monitoring faecal coliforms (MPN) using A1 medium. Among the 116 isolates, Aeromonas spp. were the most frequent (67.2%) with 9 species (A. veronii, biogroup sobria, A. hidrophila, A. sobria, A. trota, A. eucrenophila, A. veronii biog. veronii, A. media, A. cavia and A. jandaei), followed by Edwardsiella tarda (16.4%), Plesiomonas shigelloides (12.9%) and Salmonella spp. (3.4%). The NMP of fecal coliforms showed higher values in the fish tanks (>1800/100ml).
