#541 - Aspectos epidemiológicos da seneciose na região sul do Rio Grande do Sul, Brasil

E-mail: fernando@alternet.com.br Seneciosis is the main cause of livestock mortality due to poisonous plants in the State of Rio Grande do Sul, Brazil. This paper presents epidemiological data of 24 outbreaks in cattle and one in horses, diagnosed by the Regional Diagnostic Laboratory at Pelotas University in southern Rio Grande do Sul, from 1998 to 2000. Additionally, data of 54 outbreaks which occurred in 1978-1997 were analyzed. The farms where outbreaks occurred in 1998-2000 were visited to obtain clinical and epidemiological data and to verify the presence of Senecio spp. Eleven outbreaks (45.83%) affected cattle up to 3 years of age, and 13 (54.16%) cattle over 3 years. Nine (37.5%) outbreaks affected females and 15 (62.5%) males. Ten (41.66%) outbreaks occurred in spring, 4 (16.66%) in summer, 5 (20.83%) in autumn, and 5 (20.83%) in winter. Morbidity rate was estimated with 4.92% and case fatality with 95.59%. The predomionating Senecio species were S. brasiliensis on 12 farms (57.14%), S. selloi on 10 (47.61%), S. oxyphyllus on 6 (28.57%), S. heterotrichius on 3 (14.28%), and S. leptolobus on 1 farm (4.76%). The main clinical signs were progressive emaciation, incoordination, diarrhea, tenesmus, rectal prolapse and aggressiveness. The clinical manifestation periods of affected cattle observed during the farm visits, or of cattle sent for post-mortem examination,were 24-96 hours in 4 outbreaks (16.66%), 4-7 days in 7 (29.16%), 1-2 weeks in 4 (16.66%), 2-3 weeks in 2 (8.33%), 1-2 months in 2 (8.33%), and 2-3 months in 1 outbreak (4.16%). In 4 outbreaks (16.66%) the clinical manifestation period was not established. In outbreaks with longer clinical manifestation periods some animals showed photosensitization. The main necropsy findings were a hard and enlarged liver, distended gall bladder, edema of the mesenterium and abomasum, and increased amount of liquid in the cavities. Histopathological findings were fibroplasia, megalocytosis and biliary ductal proliferation of the liver, and spongy degeneration of the cerebral white matter. Samples of different Senecio species, in different seasons, were analyzed for detection of pyrrolizidine alkaloids (PAs) by thin layer chromatography. Retrorsine was found in Senecio brasiliensis, S. heterotrichius, S. selloi and S. oxyphyllus. In S. brasiliensis and S. heterotrichius one and two more non-identified PAs were detected, respectively. The total PAs concentration by spectrophotometric method was 0.25% for S. brasiliensis, 0.19% for S. heterotrichius, 0.03% for S. oxyphyllus, and 0.03% for S. selloi. The highest PAs concentration occurred in winter (June/July). No alkaloids were found in samples of S. leptolobus. These results show that S. brasiliensis is the most important cause of seneciosis in southern Rio Grande do Sul. Additional data obtained dealt with 54 outbreaks of PAs poisoning in 1978-1997. During this period, 7 outbreaks (12.96%) affected cattle up to 3 years of age, 39 (72.22%) cattle over 3 years, and 3 outbreaks (5.55%) affected cattle of different ages. In 5 outbreaks (9.25%) the age was not informed. Seven outbreaks (12.96%) affected males, 39 (72.22%) females, 3 (5.55%) both sexes, and in 5 outbreaks (9.25%) the sex was not informed. Twenty-three outbreaks (42.59%) occurred in spring, 9 (16.66%) in summer, 9 (16.66%) in autumn, and 13 (24.07%) in winter. The greater number of outbreaks during 1998-2000 (24 outbreaks in 3 years) in regard to 1978-1997 (54 outbreaks in 20 years) is probably due to a decrease of more than 50% in the number of sheep in the region.

E-mail: lafranco@vet.ufg.br With the objective to investigate a possible association between foot diseases, clinical mastitis and/or metritis and predisposing factors for foot diseases, 5300 dairy cows from 80 intensive and semi-intensive farms were used. In 325 (6.13%) cows only foot disease was diagnosed, in 35 (0.66%) foot disease and clinical mastitis, in 52 (0.98%) foot disease and metritis, in 28 (0.53%) foot disease, clinical mastitis and metritis, in 128 (2.42%) only metritis, in 165 (3.11%) only clinical mastitis, and in 89 (1.68%) cows metritis and clinical mastitis. Rapid changes in the diet, high exposure time of hoof horn to slurry and wet conditions, concrete floors, use of footbaths, low frequency of claw trimming, irregular quarantine, and no attention to health aspects during acquisition of the animals were considered as major risk factors for the occurrence of those diseases. The McNemar test for dependent samples showed significant statistical differences between the occurrence of foot disease, clinical mastitis and metritis, in spite of the poor association between those diseases. It was concluded that there was no expressive relationship between foot diseases, clinical mastitis and metritis in lactent cows.

E-mail: claudioslbarros@uol.com.br Granulomatous meningoencephalitis was observed in 7 out of 8 adult dairy cows affected by vetch-associated systemic granulomatous disease, although there was no neurological signs associated with the condition. The cellular inflammatory infiltrates were located in the leptomeninges and as perivascular cuffings within the brain and consisted of epithelioid macrophages, lymphocytes, plasm cells and eosinophils. These inflammatory changes varied from mild to severe from animal to animal and among different brain regions of the same animal. Perivascular cuffings were usually more marked than the leptomeningeal infiltrates. Affected brain regions, in decreasing order of intensity, included diencephalon through the level of massa intermedia, mesencephalon at the level of rostral colliculi, pons and cerebellar peduncles, medulla at the level of obex, frontal lobe at the level of the genu of the corpus callosum, and cerebellum. The character and the distribution of the inflammatory changes are emphasized regarding the differential diagnosis with other diseases and lesions of the bovine central nervous system in the context of the Brazilian surveillance program for bovine spongiform encephalopath.

Since little is known about the ultrastructural and morphometrical aspects of this microorganism, the present work aimed to deal with these issues. The ultrastructural study revealed the presence of structures corresponding to tubules disseminated throughout the soma of M. suis. A variable separation between the microorganism membrane and that of the erythrocyte was also observed. The structural and positional attitude of M. suis could allow speculation about its mechanism of action.

[Histological and ultrastructural evolution of liver lesions in experimental Myoporum laetum poisoning of sheep and cattle.] Evolução das lesões histológicas e ultra-estruturais no fígado de ovinos e bovinos experimentalmente intoxicados por Myoporum laetum. E-mail: bonel-raposo@brturbo.com Green leaves of Myoporum laetum were collected during spring and summer, and administered to five sheep and six steers at dosages of 20 and 30 g/kg. Liver biopsies were taken before (Controls) and 1, 3 and 7 days after dosage. ln sheep, the clinicai signs were depression, rumen hypomotility, dried feces, tenesmus, teeth grinding, dyspnea and typical lesions of photosensitization. ln cattle, the clinicai picture was much less pronounced. The main histological findings in sheep were vacuolization of hepatocytes, portal ftbrosis, bile duct proliferation and necrosis of periportal hepatocytes; the ultrastructural examination revealed hyperplasia of the smooth endoplasmic reticulum, hepatocellular hydropic degeneration, presence of crystals and severa! other degenerative changes. ln cattle both, the histological and the ultrastructural findings, were less evident.

Biópsias hepáticas foram colhidas antes (controles) e 1, 3 e 7 dias após a dosagem da planta. Estas biópsias foram analisadas histológica e ultraestruturalmente. Os sinais clínicos, em ovinos, caracterizaramse, especialmente; pordepressão, diminuição dos movimentos ruminais, fezes ressequidas, tenesmo, ranger de dentes, dispnéia e lesões típicas de fotossensibilização. Em bovinos, o quadro clínico foi discreto. Os principais achados histológicos, em ovinos, incluíram vacuolização de hepatócitos, fibrose portal, proliferação de duetos biliares e necrose de hepatócitos periportais. Os estudos ultra-estruturais, em ovinos, revelaram hiperplasia do retículo endoplasmático liso, tumefação de hepatócitos, degranulação e vesiculação do retículo endoplasmático rugoso, presença de cristais aciculares, retenção biliar, tumefação de mitocôndrias e várias outras alterações degenerativas. Em bovinos, tanto os achados histológicos, quanto os ultra-estruturais foram menos evidentes.

Postal 486, Belo Horizonte, MG 31270-901, Brazil.E-mail:mmot@icb.ufmg.br Biological control is a non-chemical alternative method with its main goal to reduce the amount of parasite population using natural antagonists. The administration of nematophagous fungi to animais has been considered an alternative in gastrointestinal helminthiasis prophylaxis. The nematophagous fungi produce trap-shaped structures, which are responsible for capturing and destroying the free-living stages of nematodes. The genera Arthrobotrys, Duddingtonia and Monacrosporium has been shown efficacy in laboratory and field experiments against cattle, equine, ovine and swine parasites. Severa) fungi formulations have been evaluated, but there is so far no commercial product available. The association of research groups with industry could improve the successful implementation of this control method.

A administração de fungos nematófagos aos animais domésticos é considerada uma promissora alternativa na profilaxia das helmintíases gastrintestinais parasitárias. Os fungos nematófagos desenvolvem estruturas em forma de armadilhas, responsáveis pela captura e destruição dos estágios infectantes dos nematóides. Os fungos dos gêneros Arthrobotrys, Duddingtonia e Monacrosporium têm demonstrado eficácia em experimentos laboratoriais e no campo no controle de parasitos de bovinos, eqüinos, ovinos e suínos. Diversas formulações fúngicas têm sido avaliadas, no entanto, ainda não há nenhum produto comercial disponível. A associação dos grupos de pesquisa e o envolvimento das indústrias poderão colaborar para o sucesso na implementação desta forma de controle.

E-mail: amg@ufla.br Neste estudo de microsco-pia eletrônica de transmissão utilizou-se um isolado de Babesia equi obtido a partir de um eqüino esplenectomizado, oriundo do município de Santa Luzia, Minas Gerais, Brasil. O isolado foi inoculado em dois potros esplenectomizados (1,05 x 1010 eritrócitos parasitados com B. equi). Os trofozoítos apresentaram uma membrana simples em· contato direto com o citoplasma das· hemácias, núcleo proeminente, retículo endoplasmático liso e rugoso bem desenvolvidos, numerosos ribosomos livres e pequenos vacúolos alimentares. Em trofozoítos de B. equi foi observado citostoma e uma longa estrutura tubular de alimentação em contato direto com o plasma sangüíneo.

E-mail: pvpeixoto@uol.com.br The reproduction performance of about 1,200 beef cows, raised on pasture and given three different mineral supplements, was evaluated during 5 years in Cássia county, Minas Gerais, Brazil. The cows were divided into three groups: For one group (A) a comercial mineral mixture was used in 1997 and 1998; for the second group (B) common salt and dicalcium phosphate 1:1 was offered in 1999; and the third group (C) received only common salt during the sarne year. ln 2000 and 2001, ali the cows received exclusively common salt. There was no decrease in i:eproduction efficiency when only common salt was given; on the contrary, the reproduction performance of the inseminated e pregnant cows, thát was respectively 92.5 and 78.2 % (1997) and 92.2 and 80.5 % (1998), increased to 94.5 and 85.7 % (2000) and 96.7 and 89.7 % (2001). The decrease of reproduction performance of the cattle in 1999 was probably due to the severe drought during this year. The authors emphasize the importance of a selective mineral supplementation, i.e. to supplement cattle with mineral mixtures including only the elements which the animais can not obtain from the pasture. Theselective mineral supplementation requires the assistance of professionals with training and experience in clinics, pathology and nutrition of ruminants. These professionals should be able to accompany and evaluate the nutritional condition of a herd when a selective mineral mixture is used. Selective mineral supplementation could represent a two or threefold economy of the total cost of conventional supplementation with "complete" mineral mixtures. The authors discuss ifthe improvement of the reproduction performance of the cattle used in this fiel d experiment was due to a better management of the herd or to possible antagonism between the minerais.

Enquanto que a mistura A (sal mineral comercial) foi oferecida nos anos de 1997 e 1998, a mistura B (cloreto de sódio 50% e fosfato bicálcico 50%) foi disponibilizada, em 1999, para as matrizes mantidas em um dos setores da fazenda e a mistura C (apenas cloreto de sódio), foi oferecida, neste mesmo ano, para os lotes cria dos em outros dois setores. Nos anos de 2000 e 2001, a mistura C foi ofertada a todas matrizes. Não houve queda na eficiência reprodutiva pela suplementação exclusiva com cloreto de sódio, pelo contrário, as percentagens de matrizes inseminadas e matrizes prenhes, que eram, respectivamente, de 92,5 e 78,2 (1997) e 92,2 e 80,5 (1998) alcançaram níveis de 94,5 e 85,7 (2000) e 96,7 e 89,7 (2001). A redução nos índices de matrizes inseminadas e de prenhez verificadas no ano de 1999 foi atribuída à baixa precipitação pluviométrica registrada naquele ano. Os autores ressalvam, porém, que a suplementação seletiva, isto é, aquela baseada na exclusiva administração do (s) mineral (is) que está (ão) faltando em uma determinada fazenda, só deve ser implantada mediante avaliação e acompanhamento clínico-nutricional do rebanho, o que exige apoio de profissional com adequados conhecimentos sobre nutrição, deficiências minerais e clínica de ruminantes. Por outro lado, a suplementação seletiva, pode representar uma despesa 2 a 3 vezes menor do que a verificada com a "mineralização" convencional do rebanho. Discute-se se a melhoria dos índices reprodutivos deveu-se à adequação do manejo ou à cessação de possível antagonismo entre os minerais.

The susceptibility of antimicrobials was studied in Gram positive and catalase negative cocci (21 samples of Lactococcus garvieae and 6 Enterococcus gallinarum), isolated from the milk of cows with subclinical mastitis, belonging to six buffalo herds in the State of Rio de Janeiro. The test used was diffusion of disks in agar Müller Hinton, according to recommendations of the National Committee for Clinicai Laboratory Standards - NCCLS. There were tested disks with ampicillin (10mg), cefalotin (30mg), cefotaxime (30mg), cefoxitin (30mg), doranfenicol (30mg), eritromycin (15mg), gentamycin (10mg), nitrofurantoin (300mg), norfloxacin (10mg), penicillin (1 O IU), tetracydin (30mg) and vancomycin (30mg). The results showed that with Lactococcus garvieae, the most efficient antimicrobial was nitrofurantoin, revealing 85.71% sensibility, followed by cefotaxime (61.90%), vancomycin (52.38%), norfloxacin (47.62%) and cefalotin (47.62%). The highest resistance was developed against penicillin and ampicillin, with 95.24% resistance for the two antimicrobials. The susceptibility profile developed by the strains of Enterococcus gallinarum showed low sensibility against the tested antimicrobials; the highest resistance observed was against eritromycin and gentamycin, with 33.34% sensibility for both. The antimicrobial evaluation showed 100% resistance against vancomycin and tetracyclin, followed by cloranfenicol, penicillin, ampicillin, cefoxitin, cefotaxim, norfloxacin and nitrofurantoin; all of them showed a resistance of 83.33% with the samples tested.

Estudou-se o perfil de susceptibilidade a antimicrobianos em cocos Gram-positivos catalase negativos (21 amostras de Lactococcus garvieae e 6 de Enterococcus gallinarum), isoladas do leite de fêmeas com mastite subdínica e pertencentes a uma população composta por seis rebanhos bubalinos localizados no Estado do Rio de Janeiro. O teste utilizado foi o da difusão de discos em agar Müller Hinton, segundo recomendações do National Committee for Clinicai Laboratory Standards - NCCLS, tendo sido testados discos com ampicilina (10mg), cefalotina (30mg), cefotaxima (30mg), cefoxitina (30mg), cloranfenicol (30mg), eritromicina (15mg), gentamicina (10mg), nitrofurantoína (300mg), norfloxacina (10mg), penicilina (10 UI), tetracidina (30mg) e vancomicina (30mg). Os resultados evidenciaram que em se tratando de Lactococcus garvieae, o antimicrobiano mais eficiente foi o nitrofurantoína com 85,71% de sensibilidade, seguido da cefotaxima (61,90%), vancomicina (52,38%), norfloxacina (47,62%) e cefalotina (47,62%). A maior resistência foi desenvolvida frente a penicilina e ampicilina, com 95,24% de resistência para os dois antimicrobianos testados. O perfil de susceptibilidade desenvolvido pelas amostras de Enterococcus gallinarum, mostrou baixa sensibilidade frente aos antimicrobianos testados, onde os maiores índices foram observados frente eritromicina e gentamicina, com 33,34% de sensibilidade para ambos; quanto à resistência desenvolvida, foi possível observar 100% de resistência com relação a vancomicina e tetraciclina, seguindo-se cloranfenicol, penicilina, ampicilina, cefoxitina, cefalotina, cefotaxima, norfloxacina e nitrofurantoína, todas evidenciando uma resistência de 83,33% das amostras testadas.

[Clinical and hematological evaluation of Nelore calves experimentally infected with isolates of Babesia bigemina from the Southeastern, Northeastern and Northern regions of Brazil.] Avaliação clínica e hematológica em bezerros Nelore infectados experimentalmente com isolados de Babesia bigemina das regiões Sudeste, Nordeste e Norte do Brasil. Pesquisa Veterinária Brasileira 23(2):52-60. Clínica de Bovinos, Campus Garanhuns, Univ. Fed. Rural de Pernambuco, Cx. Postal 152, Garanhuns, PE 55292-901, Brazil. A comparative study was made regarding the clinical and hematological alterations caused by isolates of Babesia bigemina from southeastern, northeastern and northern Brazil in experimentally infected Nelore calves. Eighteen calves between 7 and 9 months of age, without antibodies against Babesia sp and raised free from ticks, were used. Three animais were previously inoculated with 2.0x109 parasitic erythrocytes (PE) for each stabilate. The other 15 calves were subdivided into three groups, with five animais each, that were subinoculated with 1.0x1010 PE of the respective isolates. The clinical and hematological alterations were evaluated by the determination of parasitaemia, haemogram, plasmatic fibrinogen,reticulocyte count, descriptive analysis of the bone marrow and erythrocytic osmotic fragility, for 30 days, totalizing seven moments of observation. The follow-up of the immunological response by the indirect fluorescent antibody test was carried out daily until the 10th day after inoculation (DAI) and after that, on the 151 20th, 25th and 30th DAI. A mild clinical manifestation of the disease was observed. The laboratory findings revealed low leveis of parasitaemia; decrease of the erythrogram values; absence of reticulocytes, initial decrease in the total count of leukocytes, neutrophils and lymphocytes with a posterior elevation of the number of these cells; hypercellularity of the erythrocytic series and decrease of the myeloid: erythroid relation which was more accentuated between the 8th and 12th DAI, and an increase of the erythrocytic osmotic fragility in the groups inoculated with the Southeast and Northeast isolates. None of the three isolates of B. bigemina gave rise to the clinical characteristic form of the disease, although they induced an humoral immune response.

O presente trabalho teve por objetivo estudar comparativamente as alterações clínicas e hematológicas desencadeadas por isolados de Babesia bigemina das regiões Sudeste, Nordeste e Norte do Brasil em bezerros Nelore infectados experimentalmente. Foram utilizados 18 bezerros com idade entre sete e nove meses, isentos de anticorpos contra Babesia sp. e criados livres de carrapatos. Três animais foram previamente inoculados com 2,0x109 eritrócitos parasitados (EP) para cada isolado. Os outros 15 bezerros foram subdivididos em três grupos de cinco animais, que foram subinoculados com 1,0x1010 EP dos respectivos isolados. Foram avaliadas as alterações clínicas e hematológica por meio da determinação da parasitemia, do hemograma, do fibrinogênio plasmático, da contagem de reticulócitos, da análise descritiva da medula óssea e da fragilidade osmótica eritrocitária, no decorrer de 30 dias, perfazendo um total de sete momentos de observação. O acompanhamento da resposta imunológica pelo teste de imunofluorescência indireta foi realizado diariamente até o 10º dia pós-inoculação (DPI) e posteriormente no 15º, 20º, 25º e 30º DPI. Clinicamente, observou-se uma manifestação muito branda da doença. Os achados laboratoriais revelaram baixos níveis de parasitemia; decréscimo nos valores do eritrograma; ausência de reticulócitos; diminuição inicial na contagem total dos leucócitos, neutrófilos e linfócitos com posterior elevação do número destas células; hipercelularidade da série eritrocítica e decréscimo da relação mielóide:eritróide mais acentuada entre o 8º e 12º DPI e um aumento da fragilidade osmótica eritrocitária nos grupos inoculados com os isolados sudeste e nordeste. Nenhum dos três isolados de B. bigemina desencadeou a forma clínica característica da enfermidade, apesar de induzirem uma resposta imune humoral.
