E-mail: hill@vm.uff.br O objetivo deste trabalho foi de analisar morfometricamente fragmentos histológicos do baço de animais normais e diabéticos, comparando os resultados encontrados e relacionando-os ao sexo e a suplementação da vitamina C. Foram utilizados 32 ratos Wistar, os quais foram analisados número de vasos, o número de folículos germinativos (polpa branca) e o diâmetro dos vasos de cada animal. As análises histológicas e morfometricas foram feitas em amostras de 5µm de espessura demonstrando que: na quantidade de folículos germinativos, observamos resultados comparando, independente do sexo, animais controles suplementados com vitamina C e controles não suplementados (p£”0,05; F=0,1452); na quantidade de vasos, observamos resultados comparando, fêmeas diabéticas suplementadas pela vitamina C e fêmeas diabéticas não suplementadas (pd”0,05; F=6.8893); e no diâmetro dos vasos, observamos resultados comparando fêmeas, tanto no grupo controle quanto ao grupo diabético, suplementadas pela vitamina C quando comparadas às fêmeas não suplementadas pela vitamina C (p<0,05; U=121.50; Z(U)=2.1234) e (p<0,05; F=4.8134). De um modo geral, a indução de diabetess modifica o diâmetro vascular nas fêmeas e que a administração de vitamina C interfere nos dados métricos relativos ao diâmetro vascular somente nas fêmeas.

Had been used 32 Wistar rats, which had been analyzed: number of vases; the number of germinative folicules (white pulp); and the diameter of the vases of each animal. Histological and morphometric analyses were applied on 5µm thick samples and showed that: in the amount of germinative folicules, we observe resulted comparing, independent of the sex, animals controls supplemented with vitamin C and controls not supplemented (p£0.05; F=0.1452); in the amount of vases, we observe resulted comparing, of diabetic females supplemented by vitamin C when compared with not supplemented (pd”0.05; F=6.8893); and in the diameter of the vases, we observe resulted comparing females, as much in the group has controlled how much to the diabetic group, supplemented with vitamin C when compared with the females not supplemented with vitamin C (p<0.05; U=121.50; Z(U)=2.1234) and (p<0,05; F=4.8134). In a general way, the induction of diabetes modifies the vascular diameter in the females and that the vitamin C administration only intervenes with relative metric data to the vascular diameter in the females.


#112 - Intoxicações natural e experimental por salinomicina em coelhos, 29(9):695-699

ABSTRACT.- Peixoto P.V., Nogueira V.A., Gonzaléz A.P., Tokarnia C.H. & França T.N. 2009. Accidental and experimental salinomycin poisoning in rabbits. Pesquisa Veterinária Brasileira 29(9):695-699. Projeto Sanidade Animal Embrapa/UFRRJ, Seropédica, RJ 23890-000, Brazil. E-mail: pfpeixoto@terra.com.br An outbreak of salinomycin poisoning in rabbits is described. At least 27 out of 2,000 rabbits reared on a farm died after the coccidiostatic drug sulfaquinoxaline was substituted by salinomycin in the feed. An average of 26.9ppm salinomycin was detected in the ration given to the rabbits. Clinical signs included anorexia, apathy and bradykinesia, which progressed to incoordination and recumbency. Gross lesions consisted of pale areas in the skeletal muscles. The histopathological findings showed severe necrotic degenerative myopathy in association with infiltration of neutrophils and macrophages. One rabbit exhibited similar alterations in the myocardium. Mineralization was observed in the affected skeletal muscles in some cases. In order to verify if the poisoning was due to salinomycin, 20 rabbits were divided into five groups and a ration containing the drug at doses of 10, 25, 50, 75 and 100ppm was given. The administration of doses higher than 50ppm resulted in manifestation of the clinical signs seen in the outbreak of poisoning. It was concluded, that probably an error related to the mixture of salinomycin in the feed was the cause of deaths in the spontaneous outbreak of poisoning on the rabbit farm.

E-mail: pfpeixoto@terra.com.br Relata-se, pela primeira vez, um surto de intoxicação por salinomicina em coelhos. De 2000 animais, no mínimo 27 morreram após troca do coccidiostático sulfaquinoxalina pela salinomicina. A análise de parte da ração detectou 26,9ppm de salinomicina. Os sinais clínicos observados foram anorexia, apatia e lentidão com evolução para incoordenação dos movimentos e decúbito. As lesões macroscópicas consistiram de áreas pálidas na musculatura esquelética. O exame histopatológico evidenciou miopatia degenerativo-necrótica. Adicionalmente, verificou-se reação inflamatória constituída por neutrófilos e macrófagos. Um coelho apresentou lesões similares no miocárdio. Em alguns casos, mineralização estava presente nos músculos esqueléticos afetados. Vinte coelhos experimentais foram divididos em 5 grupos que receberam 10, 25, 50, 75 e 100ppm de salinomicina por via oral, com a finalidade de reproduzir a intoxicação. Os animais que receberam a partir de 50ppm de salinomicina apresentaram sinais clínicos semelhantes aos observados no surto espontâneo. Nossos resultados indicam que, provavelmente, erro na mistura da substância à ração causou a morte dos coelhos.

E-mail: lucianelaskoski@hotmail.com The gastrointestinal diseases of horses are aggravated by complications such as laminitis. The laminitis etiopathogeny are connected with lamellar basement membrane degradation by matrix metalloproteinases (MMPs). Inflammatory cytokines and leukocytes enzymes can active MMPs. The object of this study was to evaluate morphological changes on lamellar tissue of horses with colic syndrome and its association with clinical and laboratorial parameters. It was observed intensive destruction of lamellar architecture, mainly on animals with severe physical and laboratorial alterations, such as delayed capillary refill time, congested mucous membrane, tachycardia, hemoconcentration and low count of platelet and leukocytes. The results sign to the most likely moment of development of lamellar tissue injuries in horses with colic syndrome, which can be adopted preventive measures against laminitis.

A ativação das MMPs pode ocorrer devido à liberação local de citocinas inflamatórias ou enzimas provenientes de leucócitos infiltrados no tecido laminar. O objetivo deste trabalho foi avaliar as alterações morfológicas do tecido laminar de equinos com síndrome cólica letal e sua provável associação com parâmetros clínicos e laboratoriais. Observou-se intensa destruição da arquitetura laminar, principalmente nos animais com alterações físicas e laboratoriais mais acentuadas, como tempo de preenchimento capilar prolongado (TPC), membranas mucosas congestas, taquicardia, hemoconcentração e redução nas contagens de plaquetas e leucócitos. Os resultados sinalizam o provável momento do desenvolvimento de lesões do tecido laminar em equinos com síndrome cólica, no qual é possível adotar medidas preventivas contra a laminite.

E-mail: saulo-andrade@bol.com.br The aim of this study was to determine the clinical-pathological alterations and laboratory findings in cattle inoculated with Bothrops alternatus venom, with the intention of providing information for the establishment of diagnosis and differential diagnosis procedures, as well as to elucidate some obscurities observed in the pertinent literature. The lyophilized venom was diluted in 1 ml of physiologic solution. It was administered to 5 bovines by the subcutaneous route at doses of 0.0625, 0.125 and 0.25mg/kg body weight, and to 2 bovines by the intramuscular route at doses of 0.25 e 0.45mg/kg. Six bovines died and the only animal that survived, who had subcutaneously received the venom at a dose of 0.0625mg/kg, recovered. The first clinical signs were observed from 25min to 5h30min after the inoculation. The clinical evolution time varied from 7 hours 18 minutes to 92 hours. Regardless of the dose, the clinical picture was characterized by swelling (hemorrhage/hematoma) at the site of inoculation, increase in bleeding time and capillary refill time, paleness of mucous membranes and apathy. The laboratory exams revealed progressive normocytic normochromic anemia, thrombocytopenia, reduction in fibrinogen and total plasma proteins, decreased hematocrit and hemoglobin, and slight increase in creatine kinase and lactate dehydrogenase. When subjected to necropsy, the animals that received the venom through the subcutaneous route showed large hematomas and hemorrhagic areas in the subcutaneous tissue, extending from the site of inoculation. The animals in which inoculation was performed intramuscularly exhibited, in addition, intramuscular hemorrhage. The left endocardium showed extensive hemorrhagic lesions, and petechiae were found on the serosae of rumen, omasum, abomasum and gall bladder. The colon, rectum and perirenal areas were surrounded by clotted blood in three animals. Besides hemorrhage, the histological examination revealed hemorrhage and coagulative muscle necrosis in the vicinities of the inoculation site in the animals that received the venom by the intramuscular route. These lesions were mild in the muscles close to the site of inoculation in the animals inoculated by the subcutaneous route. The swelling on the site of inoculation and its surroundings was a consequence of blood accumulation and not merely edema. Myoglobinuria and gross or microscopic lesions were not observed in the kidneys. This study indicates that B. alternatus can lead adult bovines to death if they release all their venom during a bite. #115 - Susceptibility of laboratory animals to the intoxication by Ateleia glazioviana (Leg. Papilionoideae), 22(2):73-78

ABSTRACT.- Leite L.G., Riet-Correa F., Medeiros R.M.T., Piacenti A., Aragão M. & Schons S.V. 2002. Susceptibility of laboratory animals to the intoxication by Ateleia glazioviana (Leg. Papilionoideae).] Susceptibilidade de animais de laboratório à intoxicação por Ateleia glazioviana (Leg. Papilionoideae). Pesquisa Veterinária Brasileira 22(2):73-78. Laboratório Regional de Diagnóstico, Faculdade de Veterinária, Universidade Federal de Pelotas, 96010-900 Pelotas, RS, Brazil. E-mail: lgmv@alternet.com.br Rabbits, guinea pigs, rats and mice were used to reproduce the cardiac lesions observed in ruminants in the intoxication by Ateleia glazioviana. Four animals of each species were fed during four months with a ration containing 10% of dry A glazioviana. The pellets were made by mixing 700 g of commercial ration, 200 g of cornstarch, 1000 mi of distilled water and 100g of the dry plant. Later this mixture was dried at 100ºC during 16-20 hours. Four control animals of each species were given similar food, but containing dry ryegrass (Lolium multiflorum) instead of A glaziovidna. Water was offered ad libitum and a daily amount of green oats (Avena sativa) was given to rabbits and guinea pigs. No clinical signs of disease were observed during the experiment. Ali animals were weighted weekly in the last 5 weeks of the experiment and no differences were observed in weight gains between treated and control groups. At the end of dosing ali animals were euthanatized and necropsied. No macroscopic or histologic lesions were observed in the heart or other organs. The abortive properties of A glazioviana were studied in rats. One group of 11 pregnant rats was fed, from day 7 to day 21 of gestation, ration containing 10% of A. glazioviana, prepared in the sarne way than in the previous experiment. A control group received the ration with 10% ryegrass. Ali rats delivered normal litters. To determine a possible loss of toxicity of the plant during the pellet preparation, dry A. glazioviana was heated for 16-20 hours at 100ºC and administered orally to one ewe. This sheep ingested daily 2.65 g/kg bw of dry A. glazioviana for 26 days. During the experimental period the animal had tachycardia and cardiac arrhythmia. At the end of the experiment bradycardia and severe depression were observed. It was euthanatized 7 days after the last plant administration. The sheep was 2.5-3 months pregnant and the fetus was apparently normal with no autolysis. Ascites and hydrothorax were observed at necropsy. Whitish areas were observed in the myocardium and the liver was pale. Degeneration and necrosis of cardiac fibers and proliferation of fibrous tissue were observed histologically in the heart. The liver had centrilobular degeneration of hepatocytes and congestion. These experiments indicate that A. glazioviana is not cardiotoxic for laboratory animals and suggest that the plant has a heat-stable active principie similar to the active principie of plants causing cardiac fibrosis (gousiekte) in Southern Africa.

E-mail: lgmv@alternet.com.br Coelhos, cobaias, ratos e camundongos foram utilizados com a finalidade de reproduzir a forma cardíaca da intoxicação por Ateleia glazioviana. Quatro animais de cada espécie receberam por 4 meses a planta, secada à sombra, na concentração de 10% na ração, fornecida na forma de pelets. Os pelets foram preparados misturando 700 g de ração comercial, 200 g de amido de milho, 1000 mi de água destilada e 100g de planta seca e, posteriormente, secados em estufa a 100°C, durante 16 a 20 horas. Quatro animais de cada espécie serviram como testemunhas, recebendo a ração preparada da mesma forma, porém com a utilização de azevém (Lolium multijlorum) em lugar de A glazioviana. Era fornecida água à vontade e, para os coelhos e cobaias, também aveia (Avena sativa) verde diariamente. Esses animais não apresentaram nenhuma alteração clínica, e foram pesados semanalmente nas últimas 5 semanas do período experimental, não apresentando diferença significativa no ganho de peso. À necropsia não foram observadas alterações macroscópicas e pelo exame histológico também não foram detectadas lesões significativas no coração e em outros órgãos. Para testar a atividade abortiva de A glazioviana, ração contendo 10% de planta seca, preparada da mesma forma que no experimento anterior, foi administrada a 11 outros ratos, fêmeas, prenhes, nos dois últimos terços da gestação. Um grupo controle de 11 fêmeas recebeu a ração com azevém a 10%, durante o mesmo período. Esses animais tiveram filhotes normais e no prazo correto. Para determinar a possível perda de toxidez da planta durante a preparação dos pelets, A glazioviana foi aquecida por 16 a 20 horas a 100ºC e, posteriormente, administrada a um ovino em 26 doses diárias de 2,65 g/kg de planta seca. Durante o período experimental, a ovelha apresentou batimentos cardíacos mais fortes, taquicardia e arritmia cardíacas, depois bradicardia e, no período final, apatia acentuada. Foi sacrificada 7 dias após a última administração da planta. Esse animal apresentava gestação de 2,5 a 3 meses e o feto não apresentava sinais de autólise. As alterações macroscópicas mais evidentes à necropsia foram ascite, hidrotórax, áreas esbranquiçadas no músculo cardíaco e fígado de coloração clara. Na histologia do coração foram observadas degeneração e necrose de fibras cardíacas e proliferação de tecido conjuntivo fibroso. O fígado apresentava congestão e degeneração de hepatócitos nas áreas centrolobulares. Os resultados indicam que esses animais de laboratório não foram susceptíveis à ação cardiotóxica de A. glazioviana, por via oral, e sugerem a possibilidade de que o princípio ativo da planta, que é resistente ao calor, seja semelhante ao princípio ativo das plantas do sul da África que causam fibrose cardíaca.


#116 - Clinical and laboratorial evaluation of Nellore cattle experimentally infected with Trypanosoma vivax, 21(4):157-161

[Clinical and laboratorial evaluation of Nellore cattle experimentally infected with Trypanosoma vivax] Avaliação clínico and laboratorial de bovinos nelore infectados experimentalmente com Trypanosoma vivax. Pesquisa Veterinária Brasileira 21(4):157-161. Embrapa Gado de Corte, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil. In arder to evaluate the clinical-laboratorial alterations, six Neil ore calves were inoculated with 107 Trypanosoma vivax isolated from Poconé region, Mato Grosso, Brazil. The animals were evaluated daily for rectal temperature, packed cell volume (PCV), parasitemia, antibody production, calor of mucous membranes, behavior and appetite. Blood and serum samples for biochemical evaluation for aspartate aminotransferase (AST), alkaline phbsphatase (AF), gamma glutamyltransferase (GGT), cholesterol, urea, creatinine, creatine kinase (CK), calcium, phosphorus and proteinogram were collected on days 4, 8, 12, 16, 23 and 30 post inoculation (DPI). During the following 6 months rectal temperature, PCV and parasitemia were evaluated weekly. T. vivax was evidenced from 1 DPI in all calves and persisted until day 30 in five of six animals. A remarkable decrease (p<0.05) of PCV mean value (25%) was observed on 10 DPI. The animals presented no alterations in their clinical or serum biochemical state during the trial. Seroconversion took place 6 and 8 DPI, and all the animals remained seropositive during the 30 days of experiment. In all the experimental animals the occurrence of T. vivax infection was verified, characterized by the increase of corporal temperature, presente of the blood protozoa and reduction of the globular volume, without altéations in the other variables analyzed. Nellore calves, when experimentally inoculated with T. vivax, are able to establish a balance between host-parasite relationship.

RESUMO.- Schenk M.A.M., Mendonça C.L., Madruga C.R., Kohayagawa A. & Araújo F.R. 2001. [Clinical and laboratorial evaluation of Nellore cattle experimentally infected with Trypanosoma vivax] Avaliação clínico and laboratorial de bovinos nelore infectados experimentalmente com Trypanosoma vivax. Pesquisa Veterinária Brasileira 21(4):157-161. Embrapa Gado de Corte, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil. Avaliaram-se as alterações clínico-laboratoriais de seis bezerros Nelore, de ambos os sexos, inoculados experimentalmente com 107 organismos viáveis de Trypanosoma vivax, isolados de bovinos da região de Poconé, Estado de Mato Grosso. Os animais foram observados diariamente, durante 30 dias, quanto aos parâmetros de temperatura retal, volume globular (VG), parasitemia, produção de anticorpos, coloração de mucosas, comportamento e apetite. Deteminaram-se os níveis séricos de aspartato aminotransferase (AST), fosfatase alcalina (FA), gama glutamiltransferase (GGT), creatina kinase (CK), colesterol, uréia, creatinina, cálcio, fósforo e o perfil eletroforético das proteínas séricas aos 4, 8, 12, 16, 23 e 30 dias pós-inoculação (DPI). Durante os 6 meses seguintes, os animais foram observados semanalmente, avaliando-se a temperatura retal, o VG e a parasitemia. T. vivax foi evidenciado a partir do terceiro e quarto DPI em todos os bezerros e persistiu até o 30º DPI em cinco dos seis animais em estudo. Ocorreu um decréscimo significativo (p<0,05) do valor médio do VG (25%) aos dez DPI. Os animais não apresentaram qualquer alteração no quadro clínico, bem como na avaliação da bioquímica sérica durante o período experimental. A soroconversão ocorreu aos 6 e 8 DPI, permanecendo todos os animais soropositivos nos 30 dias experimentais. Bovinos nelores jovens, infectados experimentalmente com T. vivax, foram capazes de estabelecer um equilíbrio na relação hospedeiro-parasita.

Papilionoideae) poisoning in cattle and laboratory animals. Pesquisa Veterinária Brasileira 21 (1):5-7. [Alterações histológicas na intoxicação experimental por Riedeliella graciliflora (Leg. Papilionoideae) em bovinos e animais de laboratório.] Laboratório Regional de Diagnóstico, Faculdade de Veterinária, Universidade Federal de Pelotas, 96010-900 Pelotas, RS, Brazil. Tissues from cattle, mice, rats and guinea pigs experimentally intoxicated by Riedeliella graciliflora were studied histologically. Cattle lymph nodes, spleen, Peyer patches and peribronchial lymphoid tissues had diffuse necrosis of lymphocytes, mainly in the germinal centers of the follicles. This lesion was less severe in laboratory animals. All species had severe enteritis with infiltration of the lamina propria by mononuclear cells. Some cells in this infiltrate were necrotic. Degeneration and necrosis of the epithelial cells, mainly in the tip of the villi, and detachment of the epithelial lining from the lamina propria were also observed. In the liver the trabecular structure was disrupted and the hepatocytes had some degree of individual necrosis and degeneration. A tubular nephrosis was observed in the kidneys. Liver, lung, kidney, intestine and lymph nodes had different degrees of congestion. Those lesions are similar than those caused by Polygala klotzscllii, a plant that contains 5-metoxi-podophyllotoxin.

Foi realizado um estudo histológico de bovinos, camundongos, ratos e coelhos intoxicados experimentalmente por Riedeliella graciliflora. Em bovinos os gânglios linfáticos, baço, placas de Peyer e tecido peribronquial apresentavam necrose do tecido linfático, afetando, principalmente, os centros germinativos dos folículos. Esta lesão foi menos severa em animais de laboratório. Em todas as espécies havia severa enterite e infiltração da lâmina propria do intestino por células mononucleares. Algumas destas células estavam necróticas. Degeneração e necrose do epitélio, principalmente na superfície das vilosidades, e descamação das células epiteliais foram, também, observadas. No fígado havia desorganização da estrutura trabecular e alguns hepatócitos apresentavam-se degenerados ou necróticos. Os rins apresentavam nefrose tubular. Diferentes graus de congestão foram observados no fígado, pulmão, rins, intestinos e linfonodos. As lesões observadas são similares às causadas por Polygala klotzschii, uma planta que contém 5-metoxi-podofilotoxina.


#118 - Experimental type C botulism in goats

Abstract in English:

The present paper describes the subcutaneous inoculation of goats with botulinum toxin type C to determine the doses required to cause various clinical signs, and to evaluate the bioassay as a means of laboratory confirmation of the diagnosis of botulism. Serial double dilutions in doses ranging from 15.6 to 500 DL50/kg were administered to 6 goats. The animals were checked daily to observe development of characteristic signs. Blood and liver samples were collected to detect the toxin by bioassay in mice. Doses of 500 and 250 DL50/kg induced acute botulism, death occurring between 42 and 46 hours post-inoculation, but the toxin was only detected in serum samples taken from the goat which received the larger dose. Animals inoculated with doses of 125, 62.5 and 31.3 DL50/kg developed the sub-acute forro, but the toxin could not be detected in their blood serum. The chronic forro of botulism was observed in those which received 15.6 DL50/kg doses and the toxin could not be demonstrated either in their serum samples. The results confinn that goats are highly susceptible to botulinum type C toxin and that these animals develop the sarne clinical signs as seen in bovines.

O objetivo do presente trabalho foi reproduzir o botulismo em caprinos, induzido pela toxina tipo C e determinar as doses para desenvolver as diferentes formas clínicas, bem como para avaliar a eficácia do bioensaio em camundongos. Foram utilizados 6 caprinos, inoculados por via subcutânea com a toxina tipo C, em doses de 500 a 15,6 DL50/kg de peso vivo, em diluições duplas seriadas. Os animais foram observados quanto ao desenvolvimento de sintomatologia característica de botulismo e amostras de soro sanguíneo e fígado foram coletadas para pesquisa da toxina pelo bioensaio em camundongos. As doses de 500 e 250 DL50/kg induziram quadro agudo de botulismo evoluindo para a morte entre 42 e 46 horas pós-inoculação. A toxina foi detectada somente no soro do animal que recebeu a dose de 500 DL50/kg. Os animais que receberam as doses de 125, 62,5 e 31,3 DL50/kg desenvolveram quadro subagudo da doença, não sendo detectado a toxina nas amostras analisadas. Observou-se quadro crônico de botulismo no animal inoculado com a dose de 15,6 DL50/kg, não se constatando a presença da toxina no soro. Os resultados confirmaram a alta susceptibilidade dos caprinos à toxina botulínica tipo C, que apresentaram quadros clínicos semelhantes aos observados em bovinos.


#119 - Diagnosis of botulismo of cattle in Brazil by micro-complement fixation

Abstract in English:

The heat induced micro-complement-fixation test was used for the laboratory diagnosis of botulism in cattle. The following specimens were collected in southeastem and central-westem Brazil: Thirty six samples of blood serum, liver and rumen liquid from 23 bovines with clinical signs of botulism (a condition commonly called "doença da vaca caída" - fallen cow disease) and from 1 bovine inoculated subcutaneously with botulinum type C toxin. Samples of blood serum, liver and rumen liquid collected from healthy bovines were used as negative controls. Sixteen out of 17 serum samples were positive for botulinum toxin in cattle with clinical signs of acute, subacute and chronic botulism. All 9 liver samples were positive including those from chronic cases. Two samples of rumen liquid from bovines with the acute disease gave a positive test, but 2 other chronic cases were negative. All 3 samples collected from the bovine with subacute clinical sigos gave positive results. From 36 samples examined, 31 were positive; of these 2 were of type C, 5 type D and 24 samples had to be grouped within the CD complex. All samples gave negative results with the mouse bioassay, including the material collected from the experimental case. In conclusion at least one sample of each of the 24 cases examined gave a positive result for botulinum toxin; thus this highly sensitive test confirmed the diagnosis that the "doença da vaca caída" is indeed botulism, caused by type C and D toxins.

Foi utilizada a técnica de Microfixação de Complemento Induzida pelo Aquecimento para o diagnóstico laboratorial do botulismo em bovinos. Neste sentido foram colhidas, nas regiões Sudeste e Centro-Oeste do Brasil, 36 amostras constituídas por soro sangüíneo, fígado e líquido ruminal de 23 bovinos com quadro clínico de botulismo, vulgarmente conhecido como “doença da vaca caída”, e de 1 bovino intoxicado experimentalmente pela toxina botulínica tipo C. Como controle negativo foram utilizados soro sangüíneo, fígado e líquido ruminal de bovinos sadios. De 17 soros examinados de bovinos com quadros clínicos agudo, subagudo e crônico, 16 foram positivos para a presença de toxina; todas as 9 amostras de fígado também foram, positivas, mesmo de animais com intoxicação crônica. Duas amostras de líquido ruminal de bovinos com doença aguda foram, positivas no teste, enquanto 2 amostras colhidas de animais com evolução crônica foram negativas. As 3 amostras oriundas do bovino intoxicado experimentalmente e apresentando evolução subaguda, foram positivas na Microfixação. Das 36 amostras examinadas, 31 foram positivas no teste, sendo 2 pertencentes ao tipo C, 5 ao tipo D, e 24 agrupadas dentro do complexo CD. Todas essas amostras foram negativas no Bioensaio em camundongo, inclusive o material colhido do bovino intoxicado experimentalmente. Com base nos resultados, e tendo como referência o Bioensaio em camundongo, a Microfixação de Complemento demonstrou um excelente desempenho, permitindo o diagnóstico laboratorial de botulismo em pelo menos um material de cada um dos 24 casos examinados, confirmando assim o diagnóstico de que a "doença da vaca caída" é de fato botulismo causado pelas toxinas C e D.


#120 - Outbreak of mycobacteriosis in a froggery caused by Mycobacterium marinum

Abstract in English:

An outbreak of mycobacteriosis (tuberculosis) caused by Mycobacterium marinum in a colony of Rana catesbeiana in the State of Rio de Janeiro is described. The disease was introduced in the froggery by infected animals and, in 3 months, about 50% of 160 animals became contaminated and died. The sick frogs had shown clinically progressive anorexia, apathy, adynamia, having the head lowered and lost weight with the evolution of the disease. The necropsy had shown nodular whitish caseous lesions in livers, spleens, kidneys, rarely in lungs, with variable number and size of the nodules. Microscopically, the lesions were characterized as granulomas in different evolutioning stages. These had origin in epithelioid cell agglomerations which became caseous necrosis later. The so initiated nodules were surrounded by some macrophage cells and by concentric connective tissue layers, limiting the nodules themselves and from the normal adjacent organ tissues. A great number of isolated or aggregated acid-fast bacteria was seen scattered in the nodule structure. The identification of M. marinum was based on the cultural and biochemical behaviour, with special regard to the photochromogenicity.

Foi descrito um surto de micobacteriose causado por Mycobacterium marinum numa criação de rãs da espécie Rana catesbeiana no Estado do Rio de Janeiro. A doença foi introduzida no ranário através da compra de rãs infectadas e propagou-se em 3 meses a cerca de 50% das rãs de um lote de 160 animais que adoeceram e morreram. Clinicamente as rãs doentes demonstravam progressiva anorexia, apatia, adinamia e postura cabisbaixa, perdendo peso a medida que a doença evoluia. A necrópsia revelou lesões nodulares caseosas de cor esbranquiçada no fígado, baço e rins, raramente nos pulmões, variando o número e tamanho dos nódulos. Microscopicamente as lesões eram constituídas por granulomas, em vários estágios de evolução. Estes iniciavam-se por aglomerados de células epitelióides que posteriormente eram levados a necrose por caseificação. Os nódulos assim formados eram envoltos por alguns macrófagos e por camadas concêntricas de tecido conjuntivo que delimitavam os nódulos entre si e do tecido normal adjacente do órgão. Grande número de bastonetes isolados ou formando agregados bacterianos, álcool-ácido resistentes, encontravam-se entre as estruturas do nódulo. A identificação de M. marinum baseou-se no comportamento cultural e bioquímico, destacando-se a fotocromogenicidade.
