Cutaneous pythiosis in horses from Rio Grande do Sul, Brazil

Abstract in English:

Seventeen cases of Pythiosis in horses from Rio Grande do Sul, Brazil, are described. The lesions consisted of subcutaneous granulomas in the limbs, ventral thoracoabdominal wall and face. Specimens from the lesions were obtained through biopsy or at post-mortem examination. The disease is caused by the Pythium sp., and yeast-like Protista organism and the lesions are similar to the cutaneous Habronimiasis. Most cases of Pythiosis were observed in summer and the characteristic lesions consisted of an exuberant chronic inflammatory reaction crisscrossed by numerous irregular channels containing a central yellow and firm necrotic core (kunker). A presumptive diagnosis of Pythiosis was made in H & E preparations based on the findings of linear or round spaces in the necrotic areas which represented ghost images of the organism. Identification of the fungal elements was achieved with the use of silver stains (all cases), direct examination (2 cases) and cultures (2 cases). The morphology of the agent and histopathological lesions in all cases are identical and permit the etiologic diagnosis even in non-cultured lesions.

Foram diagnosticados 17 casos de pitiose em lesões granulomatosas dos membros, região toracoabdominal ventral e face, obtidas por biópsia e necropsia de eqüinos do Rio Grande do Sul, Brasil. A doença é causada pelo organismo Pythium sp., do reino protista, que provoca lesões semelhantes àquelas da habronemíase cutânea e ocorre, principalmente, nos meses de verão. As lesões caracterizam-se por proliferações inflamatórias exuberantes, com fistulação de onde sai material necrótico amarelado, de consistência firme. O diagnóstico foi feito através da verificação da imagem negativa de elementos filamentosos tubuliformes nas áreas necróticas, vistas nas preparações coradas pela hematoxilina e eosina, pela sua evidenciação nas impregnações pela prata, visualização em exame direto e por cultura do agente de material de dois animais. A semelhança das lesões em todos os casos assegurou o diagnóstico de pitiose.


Neutralizing antibody levels in cattle vaccinated against rabies

Abstract in English:

Three modified live virus vaccines and one inactivated vaccine against rabies were purchased in the market and applied into cattle, aiming the determination of neutralizing antibody levels induced. Each vaccine was injected as a single dose into groups of 10 animals/vaccine, according to manufacturer's recommendations, plus 10 uninoculated controls. Cattle serum samples were collected on days 0, 30 and 60 post-vaccination. The samples were tested against the strain CVS in the mouse neutralization test, and against CVS and ERA strains in the cell culture virus-neutralization test. All the four vaccines were shown not to be good rabies neutralizing antibody inducers. The results obtained with the two tests, despite the low titers observed in both, were not comparable, probably because of the differences in the amount of virus. used for challenge.

Três vacinas antirábicas com vírus modificado e uma vacina inativada foram adquiridas no comércio e aplicadas em bovinos, visando a determinação dos níveis de anticorpos neutralizantes induzidos. Cada vacina foi injetada em dose única em grupos de 10 animais/vacina, de acordo com as recomendações dos fabricantes, além de 10 controles não inoculados. Amostras de soro foram coletadas nos dias 0, 30 e 60 pós-vacinação. As amostras foram testadas contra a cepa CVS no teste de vírus-neutralização em camundongos, e contra as cepas CVS e ERA no teste de vírus-neutralização em cultivos celulares. Todas as quatro vacinas demonstraram não ser boas indutoras de anticorpos neutralizantes. Os resultados obtidos com os dois testes, apesar dos baixos títulos observados em ambos, não foram comparáveis, provavelmente devido à diferença na quantidade de vírus utilizada no desafio.


Experimental poisoning of goats by Vernonia mollissima (Compositae)

Abstract in English:

The aerial parts of Vernonia mollissima, a plant of the family Compositae, were given orally to 17 goats. An acute lethal poisoning was reproduced by the administration of single doses from 2.5 to 10 g of the plant material per kg body weight. The clinical manifestations of the poisoning were: anorexia, apathy, rumen atony, staggering gait, sternal-abdominal decubitus, hepatic sensibility, sometimes violent muscular contractions, hypothermia and death. The main post-mortem findings were observed in the liver, characterized by severe hepatic distrophy. Histological examinations revealed massive necrosis of the liver parenchyma in the animals with a clinical course of less than 24 hours. In the animals with a longer course (about 48 hours), disappearance of the hepatic cells and congestion in the center of the lobule were observed; the midlobular and peripheral parenchymal cells were swollen and vacuolized. Additionally, there were lesions in the kidneys, which consisted of regressive changes of the epithelial cells of the uriniferous tubules; they were slight in the animals with a clinical course of less than 24 hours, pronounced in the animals with a longer course. In the present investigation also data were obtained concerning the elevation of serie activity of alanine amino transferase (ALT), as parta te amino transferase (AST), lactic dehydrogenase (LDH), gamma-glutamyl transferase (GGT) and alkaline phosphatase (SAP), as well as those regarding the bilirubin and protein levels.

Descreve-se em caprinos um quadro clínico-patológico de evolução aguda, letal, pela administração de Vernonia mollissima Don, planta pertencente à família das compostas. Esta planta mostrou-se letal para 100% dos animais que receberam 10 g/kg e para 50% dos que receberam 5 e 2,5 g/kg. A dose de 1,25 g/kg não se mostrou tóxica para a espécie caprina. As doses únicas de 5 e 10 g/kg produziram intoxicação com evolução clínica inferior a 24 horas, enquanto com 2,5 g/kg a evolução foi superior a este período (de aprox. 48 horas). O quadro clínico caracterizou-se por anorexia, prostração, sensibilidade hepática, atonia de rúmen, emissão constante de berros e gemidos, decúbito esterno-abdominal e lateral, movimentos de pedalagem, às vezes contrações musculares violentas, hipotermia, estado de coma e morte. As principais lesões macroscópicas foram observadas a nível hepático, caracterizando uma grave distrofia hepática. Microscopicamente observou-se difusa necrose de coagulação no parênquima hepático, nos animais em que a evolução foi inferior a 24 horas, e desaparecimento das células centrolobulares e congestão centrolobular, além de tumefação e vacuolização dos hepatócitos nas zonas intermediária e periférica, nos casos de evolução mais prolongada. A nível renal observaram-se, na maioria dos animais, processos regressivos nas células epiteliais dos túbulos uriníferos, leves nos casos com evolução inferior a 24 horas e acentuados nos casos com evolução mais longa. O comportamento sérico das enzimas aspartato amino transferase (AST), alanina amino transferase (ALT), lactato desidrogenase (LDH), gama glutamil transferase (GGT) e fosfatase alcalina (SAP) foi de aumento progressivo e constante até o óbito. As bilirrubinas apresentaram níveis séricos variáveis individualmente e com o tempo de evolução clínica. O quociente GGT/AST foi inversamente proporcional ao grau de necrose hepática; o quociente AST/ALT foi diretamente proporcional ao grau de necrose hepática, atingindo índices elevados.


Serotypes of Haemophilus (Actinobacillus) pleuropneumoniae isolated from swine in Brazil

Abstract in English:

Thirty-three strains of Haemophilus (Actinobacillus) pleuropneumoniae biovar 1 were isolated from swine lungs with acute or chronic lesions of swine pleuropneumonia, with one exception, which was isolated from the nasal cavity of a pig from a problem herd. These strains originated from 14 herds, ten located in the State of Santa Catarina, three in Rio Grande do Sul and one in São Paulo. Serotyping was done by rapid slide agglutination (RSA), immunofluorescence (IF), counterimmunoelectrophoresis (CIE) and immunodiffusion (ID). Twenty-two (66%) of the 33 strains could be serotyped, 19 belonged to serotype 5 and three to serotype 3. All serotype 3 strains together with those not possible to be serotyped originated from swine of the sarne herd, Santa Catarina, one in which serotype 5 was also isolated. The IF and CIE were the most sensitive tests, while the ID was the least sensitive. Three strains could not be serotyped by the RSA because they auto-agglutinated.

Isolaram-se 33 amostras de Haemophilus (Actinobacillus) pleuropneumoniae, biovar 1, sendo que 32 originaram-se de pulmões de suínos com lesões macroscópicas agudas ou crônicas de Pleuropneumonia Suína (PPS) e uma amostra da cavidade nasal de um suíno de granja problema. Estas amostras foram coletadas em 14 propriedades, sendo dez localizadas no Estado de Santa Catarina, três no Rio Grande do Sul e uma em São Paulo. Para a sorotipagem utilizaram-se as técnicas de soroaglutinação rápida (SAR), imunofluorescência (IF), imunoeletroosmoforese (IEOF) e imunodifusão (ID). Das 33 amostras, 22 (66%) foram sorotipadas e, entre estas, 19 pertenciam ao sorotipo 5 e três ao sorotipo 3. As amostras do sorotipo 3 e aquelas não sorotipadas originaram-se de suínos de uma mesma propriedade do Estado de Santa Catarina, onde também isolou-se o sorotipo 5. Os testes de IF e IEOF foram os mais sensíveis e o de ID apresentou a menor sensibilidade. Três amostras não puderam ser sorotipadas pelo teste de SAR porque auto-aglutinavam.


Enzootic calcinosis in sheep caused by the ingestion of Nierembergia veitchii (Solanaceae)

Abstract in English:

A chronic debilitating disease with hypercalcemia, parathyroid cell atrophy, C cell hyperplasia, osteopetrosis, osteonecrosis, and soft tissue calcinosis is described in sheep in southeastern Rio Grande do Sul, Brazil. Sheep of both sexes and all ages, except suckling lambs, are affected. Hypercalcemia occurs from October through February during which time sheep on pasture have access to the calcinogenic plant Nierembergia veitchii. The incidence of the disease varies between years and farms with apparent correlation to the availability of the plant. Experiments in rachitic chicks demonstrated a vitamin D-like activity in N. veitchii since the addition of the plant to the diet stimulated calcium absorption, uptake of calcium by the tibia and also increased the amount of calbindin-D (the vitamin D induced protein sinthesized by the intestinal mucosa) and the calcium and phosphorus plasma levels. Clicks were fed a diet containing a high concentration of SrCl2 which inhlbits the formation of 1,25(OH)2D3 by the renal hydroxylase system. Adding a dried, ground preparation of one batch of N. veitchii to the SrC12 diet stimulated the synthesis of small quantities of intestinal calbindin-D (CaBP), but did not cause an increase in calcium absorption. Another batch of the plant was totally inactive. These data suggest that N. veitchii does contain a l,25(OH)2D3-like compound. The variable results between the two batches might be explained on the basis of lability of the substance in these plants, differences in preparation or other unknown reasons. The failure to demonstrate the presence of 1;25(OH)2D3-like activity in N. veitchii in a clear and constant way may be that an insuficiente amount of the plant was fed. Whether, in the field, N. veitchii contains sufficient amounts of the 1,25(OH)2D3 - like substance to cause calcinosis or that calcinosis is due to other factors in the plant, requires further attention.

Descreve-se em ovinos, no Rio Grande do Sul, uma doença caracterizada por emagrecimento progressivo, lúpercalcemia, atrofia das células das paratireóides, hiperplasia das células C da tireóide, osteopetrose, osteonecrose e calcificação dos tecidos moles. Ovinos de ambos sexos e todas as idades exceto lactentes são afetados. A hipercalcemia ocorre desde outubro a fevereiro, época em que os ovinos consomem a planta calcinogênica Nierembergia veitchii. A incidência da doença é variável entre anos e fazendas, com aparente correlação com a disponibilidade da planta nas pastagens. Experimentos em pintos raquíticos demonstraram que N. veitchii possui ação biológica similar à vitamina D, já que a adição da planta na dieta estimulou a absorção de cálcio, a deposição de cálcio na tíbia, e também aumentou a síntese de calbindin-D (proteína complexante do cálcio induzida pela vitamina D sintetizada no intestino) e os níveis plasmáticos de cálcio e fósforo. Pintos foram alimentados com dietas contendo altas concentrações de SrCl2 que inibe o sistema de hidrolase renal. Uma partida de N. veitchii secada e moída, agregada à dieta com SrCl2, estimulou a síntese de pequenas quantidades de calbindin-D, mas não causou aumento na absorção de cálcio. Outra partida de planta foi totalmente inativa. Esses dados sugerem que N. veitchii contém um composto similar a 1,25 (OH)2D3. Os resultados variáveis entre as duas partidas poderiam ser devidas a instabilidade da substância na planta, diferenças na sua preparação ou mitro fator desconhecido. As dificuldades em demonstrar em forma clara e constante a presença de uma substância de ação biológica similar a 1,25 (OH)2D3 em N. veitchii, poderia ser devido à administração de doses insuficientes da planta. As possibilidades de que a planta no campo contenha suficientes quantidades de um composto similar a 1,25 (OH)2D3 para causar calcinose, ou de que a calcinose seja causada por outro fator da planta requerem posterior atenção.


Influence of the milk on periodontal lesions of "cara inchada" in calves

Abstract in English:

Five calves of mixed zebu breed, 3 to 4 months old, with active periodontal lesions of "cara inchada" (CI) were confined, during 4 and a half months, to a covered corral on a farm where the disease has been a problem, in the Rondonópolis area of northem Mato Grosso, Brazil. The calves were milk-fed on cows which were kept graúng on Panicum maximum pasture known to induce a relatively high prevalence of CI (more than 30 per cent). As roughage the calves received native grass cut daily in a near "cerrado" (savanna) area. On this type of natural pasture the disease has never been observed. The periodontal lesions of the five animals persisted or worsened, and two of them died. These observations, toghether with data from former studies, suggest that a determinating factor of CI may exist also in the milk of cows kept on pastures where the disease occurs.

Cinco bezerros, mestiços zebu, de 3 e 4 meses de idade, com lesões peridentárias ativas da "cara inchada" (CI), foram confinados num curral de fazenda com o problema da doença, na região de Rondonópolis, Mato Grosso, durante 4 meses e meio. Os bezerros foram amamentados pelas respectivas mães mantidas em pastos com prevalência realtivamente alta de CI (mais que 30%), as quais eram levadas ao curral duas vezes ao dia; como volumoso os bezerros receberam, no cocho, capim nativo recentemente cortado em área sob vegetação de cerrado, dentro da própria fazenda. A CI nunca foi observada em bezerros criados neste tipo de pastagem. As lesões peridentárias destes bezerros persistiram ou se agravaram e dois dos animais morreram. Estas observações, em conjunto com os dados obtidos em outros estudos anteriores, sugerem que o fator determinante da CI existia também no leite de vacas mantidas em pasto positivo para a doença.
