#1 - Ultrastructural lesions in the myocardium and kidneys of rabbits in experimental acute Amorimia exotropica poisoning, 36(3):161-166

E-mail: gmpsoares@gmail.com Amorimia exotropica is an important plant associated with sudden death in cattle in Southern Brazil. In order to understand the mechanisms by which A. exotropica causes acute lesions in the heart and kidney of intoxicated animals, an experiment was conducted to determine the histopathology and ultrastructure of myocardial and renal lesions of intoxicated rabbits. After receiving 18g/kg of dried plant, six rabbits died suddenly. At necropsy, the liver was swollen and no other macroscopic lesions were observed. Histologically, centrolobular and midzonal hepatocytes were vacuolated. These vacuoles were strong PAS stained positive, suggesting that they corresponded to glycogen accumulations. In some regions of the ventricular septum and ventricles were found vacuoles of different sizes and the kidneys of two rabbits showed vacuolar degeneration on distal convoluted tubules. Ultrastructurally, the myocardium had cardiomyocytes swelling with separation of myofibrils bundles and rupture and disorganization of the sarcomeres. The mitochondria displayed swelling, disorganization, disruption of the mitochondrial cristae, and electron-dense matrix. Some mitochondria exhibited eccentric projections of their membranes with disruption of both outer and inner membranes. The sarcoplasmic reticulum had no alterations, whereas the T-tubule system was occasionally dilated and ruptured. The kidneys had mitochondrial swelling with disorganization and disruption of the mitochondrial cristae. The vacuoles result from the swelling of the endoplasmatic reticulum and usually were located between two basolateral infoldings and mitochondria, occurring preferentially around the nucleus. The myocytes and T system damages induced by A. exotropica result in acute heart failure and death. Furthermore, this mechanism of cardiotoxicity may be common to all plant containing monofluoroacetate.

Visando compreender os mecanismos pelos quais a A. exotropica provoca lesões agudas no coração e rins de animais intoxicados, foi conduzido uma intoxicação experimental em coelhos para determinar a histopatologia e ultraestrutura da lesão miocárdica e renal. Depois de receber 18g/kg de planta seca, seis coelhos morreram subitamente. Na necropsia, o fígado apresentava acentuação do padrão lobular. Os demais órgãos não apresentaram alterações macroscópicas. Histologicamente, os hepatócitos centrolobulares e mediozonais estavam vacuolizados e coraram-se fortemente com PAS. Em algumas regiões foram observados vacúolos de diferentes tamanhos no septo ventricular e ventrículos e os rins de dois coelhos mostraram degeneração vacuolar nos túbulos contorcidos distais. Ultraestruturalmente, o miocárdio apresentou cardiomiócitos tumefeitos com separação das bandas de miofibrilas e ruptura e desorganização dos sarcômeros. As mitocôndrias estavam tumefeitas exibindo desorganização das cristas mitocondriais, e a matriz estava eletrodensa. Algumas mitocôndrias exibiam projecções excêntricas das suas membranas com ruptura das membranas externas e internas. O retículo sarcoplasmático não tinha alterações, e os túbulos T estavam ocasionalmente dilatados e rompidos. Os rins apresentavam tumefação mitocondrial com desorganização e ruptura das cristas mitocondriais. Os vacúolos resultam da expansão do retículo endoplasmático e foram localizados geralmente entre duas invaginações basolaterais e as mitocôndrias, ocorrendo preferencialmente ao redor do núcleo. A lesão nos miócitos e o dano no sistema T induzido pela A. exotropica resultam na insuficiência cardíaca aguda e morte. Este mecanismo de cardiotoxicidade pode ser comum a todas as plantas contendo monofluoroacetato.

E-mail: jrobertok@usp.br The umbilical cord blood (UCB) is an important source of pluripotent stem cells, which motivated researches on ontogeny and transplantation. The morphological characterization of umbilical cord cells is the first step to establish subsequent experiments on these areas. Although some information on humans can be found, no data on UCB is available for bovines. Therefore, this work is the first attempt to conduct an ultrastructural characterization of bovine umbilical cord blood. Blood was collected from the umbilical cord of twenty fetuses by punction of the umbilical vein. Samples were processed for whole leucocytes observation by centrifugation and the buffy coat was collected. Cells were washed and pelleted and prepared according to the standard protocol of the transmission electron microscopy. The presence of cells with morphologic characteristics compatible with the precursors from the erythrocytic, neutrophilic, eosinophilic, basophilic, and lymphocytic lineages was observed. Atypical cells with peculiar morphological features, strongly similar to apoptotic cells, were seen. Bovine neutrophils with three types of cytoplasmic granules were also found in the blood. The ultrastructural characteristics of observed bovine UCB cells where similar to those found in other species, suggesting that bovines could possibly constitute an experimental model for approaches on UCB cells research.

E-mail: jrobertok@usp.br O sangue de cordão umbilical (SCU) é uma importante fonte de células progenitoras pluripotentes, que motiva pesquisas em ontogenia e transplantes. A caracterização morfológica das células de cordão umbilical é o primeiro passo para se estabelecer experimentos subsequentes nessas áreas. Embora algumas informações sobre SCU em humanos possam ser encontradas, não existe nenhuma informação disponível sobre elas em bovinos. Portanto, este trabalho é a primeira tentativa de se conduzir uma caracterização ultra-estrutural do sangue de cordão umbilical bovino. O sangue foi coletado do cordão umbilical de 20 fetos por punção da veia umbilical. As amostras foram processadas para observação dos leucócitos totais por centrifugação pela coleta do botão leucocitário. As células foram lavadas, peletizadas e preparadas de acordo com protocolo padrão para microscopia eletrônica de transmissão. A presença de células com características morfológicas compatíveis com precursores das linhagens eritrocítica, neutrofílica, eosinofílica, basofílica e linfocítica foram observadas. Células atípicas com características morfológicas peculiares muito semelhantes a células em apoptose foram observadas. No sangue do cordão umbilical também foi encontrado neutrófilos bovinos apresentando três tipos de grânulos citoplasmáticos. As características ultra-estruturais do SCU bovino foram semelhantes às encontradas em outras espécies, sugerindo que esta espécie possa servir como modelo experimental para abordagens em pesquisas sobre sangue de cordão umbilical.

However this model was not sufficient to induce evident histophatological changes in the mucosal layer. The aim of this study was to evaluate the mucosa subjected to that model of small colon distention by histomorphometry and scan electronic microscopy (SEM). Sixteen horses were used. In the distended group (DG), nine of them were subjected to distention of the small colon by a surgically implanted intraluminal balloon that was inflated with a pressure of 40mm Hg during 4 hours. In the sham-operated group (SG), the balloon was implanted but not inflated. Full-thickness intestinal samples were collected before and after obstruction and after 1.5 and 12 hours of decom-pression. By SEM, it was observed that the mucosa turned flat and smooth after distention and returned to the wrinkled original appearance after decompression. Twelve hours after decompression the mucosa had a more irregular appearance with points of fragmentation. There was a reduction in mucosa thickness after distention, returning to basal values after decompression. Instead of the fact that there were changes in appearance and thickness, it was concluded that the mucosa could borne up the compression caused by distention returning to the original characteristics without major lesions.

These cells can be differentiated into olfactory receptors. An immunolocalization study and ultrastructural analysis by transmission electron microscopy of olfactory epithelium of mongrel dogs were made using 3 males (one year old) and 2 females (three years old). Labeled cells with positive staining by Proliferating cell nuclear antigen (PCNA) were observed in specific areas of the olfactory epithelium, especially above the basal membrane. The ultrastructure revealed cells adjacent to the basal membrane with morphology resembling sustentacular cells, supporting the idea of renewal of sustentacular and olfactory sensorial cells. Olfactory epithelium contains basal cells committed to self-renewal, characterized by high metabolic activity, identified by positive reaction to PCNA. These results suggested the renewal of sustentacular and sensorial olfactory cells through the same pathway.

The mushroom was given to three 9 to10-month-old Jersey calves immediately after harvesting. Daily doses were around 20g/kg of body weight during 7 (Calf 1) or 13 days (Calves 2-3), and the total doses of mushroom given were 140, 268, and 261g/kg of body weight, respectively. One calf (Calf 4) with same age and breed was used as control. Clinical signs were characterized by prostration, anorexia, hyperemia of oral mucosa, and loosening of long hairs of the tail tip under mild traction. The calves were submitted to euthanasia and necropsied on days 8 (Calf 1) and 15 (Calves 2-4) after the beginning of the experiment. Microscopically, there was smoothness of dorsal epithelium of tongue with absence of filiform papillae, vacuolation of keratinocytes, and loosening of the keratin layer. In the hooves, there was vacuolation and irregular keratinization of the laminar epidermis and hyperplasia of keratinocytes. Hyperkeratosis, vacuolation of the external root sheath, thickening of tricholemal keratin, and inflammatory infiltration around hair follicles were observed on the skin of the tail tip. Immunohistochemical results with anti-pancytoceratin and anti-Ki67 (cell proliferation marker) antibodies showed no differences between the tongue dorsal epithelium of the control and experimental calves. Ultrastructural study demonstrated decrease in tonofilaments and increased intercellular spaces of the spinous layer of the tongue dorsal epithelium. The results of this study favor the hypothesis of an interference with the epithelial keratinization mechanisms by the toxic principles of Ramaria flavo-brunnescens.

E-mail: bonel-raposo@brturbo.com Green leaves of Myoporum laetum were collected during spring and summer, and administered to five sheep and six steers at dosages of 20 and 30 g/kg. Liver biopsies were taken before (Controls) and 1, 3 and 7 days after dosage. ln sheep, the clinicai signs were depression, rumen hypomotility, dried feces, tenesmus, teeth grinding, dyspnea and typical lesions of photosensitization. ln cattle, the clinicai picture was much less pronounced. The main histological findings in sheep were vacuolization of hepatocytes, portal ftbrosis, bile duct proliferation and necrosis of periportal hepatocytes; the ultrastructural examination revealed hyperplasia of the smooth endoplasmic reticulum, hepatocellular hydropic degeneration, presence of crystals and severa! other degenerative changes. ln cattle both, the histological and the ultrastructural findings, were less evident.

E-mail: bonel-raposo@brturbo.com Amostras de Myoporum laetum foram colhidas durante a primavera e verão e administradas a cinco ovinos e seis bovinos em doses únicas de 20 e 30 g/kg. Biópsias hepáticas foram colhidas antes (controles) e 1, 3 e 7 dias após a dosagem da planta. Estas biópsias foram analisadas histológica e ultraestruturalmente. Os sinais clínicos, em ovinos, caracterizaramse, especialmente; pordepressão, diminuição dos movimentos ruminais, fezes ressequidas, tenesmo, ranger de dentes, dispnéia e lesões típicas de fotossensibilização. Em bovinos, o quadro clínico foi discreto. Os principais achados histológicos, em ovinos, incluíram vacuolização de hepatócitos, fibrose portal, proliferação de duetos biliares e necrose de hepatócitos periportais. Os estudos ultra-estruturais, em ovinos, revelaram hiperplasia do retículo endoplasmático liso, tumefação de hepatócitos, degranulação e vesiculação do retículo endoplasmático rugoso, presença de cristais aciculares, retenção biliar, tumefação de mitocôndrias e várias outras alterações degenerativas. Em bovinos, tanto os achados histológicos, quanto os ultra-estruturais foram menos evidentes.

E-mail: amg@ufla.br A transmission electron microscope study was carried out on Babesia equi obtained from a splenectomized horse, from the municipality of Santa Luzia, Minas Gerais, Brazil. The isolate was inoculated into two splenectomized foals (1.05 x 1010 parasitized erythrocytes by B. equi). Trophozoites have a single membrane in direct contact with the cytoplasm of the red blood cells, a prominent nucleus, well-developed rough and smooth endoplasmic reticulum, numerous free ribosomes and small food vacuóles. B. equi trophozoites have a cytostome and a long tubular feeding structure in direct contact with the blood plasmá.

E-mail: amg@ufla.br Neste estudo de microsco-pia eletrônica de transmissão utilizou-se um isolado de Babesia equi obtido a partir de um eqüino esplenectomizado, oriundo do município de Santa Luzia, Minas Gerais, Brasil. O isolado foi inoculado em dois potros esplenectomizados (1,05 x 1010 eritrócitos parasitados com B. equi). Os trofozoítos apresentaram uma membrana simples em· contato direto com o citoplasma das· hemácias, núcleo proeminente, retículo endoplasmático liso e rugoso bem desenvolvidos, numerosos ribosomos livres e pequenos vacúolos alimentares. Em trofozoítos de B. equi foi observado citostoma e uma longa estrutura tubular de alimentação em contato direto com o plasma sangüíneo.

[Observação ultra-estrutural das vias aéreas de suínos recuperados e suscetíveis após inoculação com Mycoplasma hyopneumoniae.] Pesquisa Veterinária Brasileira 18(1):1-7. To determine the morphological differences in the epithelium of the airways of recovered and susceptible pigs after Mycoplasma hyopneumoniae challenge, twenty-four 4-week-old M. hyopneumoniae-free pigs were intratracheally inoculated with 107ccu/ml of a pure low-passaged culture of the P5722-3 strain of M. hyopneumoniae challenge material. Eight pigs (group I) were challenged at the beginning of the experiment and rechallenged 3 months late 1: Group II pigs were also challenged at the beginning of the experiment and necropsied 3 months later: Group III pigs were challenged at the sarne time as the rechallenge of group I pigs. Eight nonchallenged pigs served as contrais (group IV). Three days after the second challenge of group I and the first challenge of group III, and every 3 and 4 days thereafter, two pigs from each group were euthanatized by electrocution and necropsied. Samples of bronchi and lung tissue were examined using light and electron microscopy (SEM and TEM). Macroscopic lesions were observed in the lungs of all group III pigs (average = 4.74%) and were characterized by purple-red areas of discoloration and increased firmness affecting the cranioventral aspect of the lungs. Macroscopic lesions of pneumonia in groups I and II were minimal (less than 1%). There were no gross lesions of pneumonia in contrai (group IV) pigs. Microscopic lesions were characterized by hyperplasia of the peribronchial lymphoid tissue and mild neutrophilic infiltrates in alveoli. Electron microscopy showed patchy areas with loss of cilia and presence of leukocytes and mycoplasmas in bronchi of susceptible pigs (group III). The bronchial epithelium of rechallenged (group I), recovered (group II), and control (group IV) pigs was ultrastructurally similar indicating recovery of the former two groups. Although mycoplasmas were seen among cilia, a second challenge on pigs of group I did not produce another episode of the disease nor did it enhance morphological changes, suggesting that those pigs could become carriers of M. hyopneumoniae.

Para determinar as diferenças morfológicas no epitélio das vias aéreas de suínos suscetíveis e recuperados da infecção por Mycoplasma hyopneumoniae, 24 leitões com 4 semanas de idade, livres de infecção por M. hyopneumoniae, foram inoculados intratraquealmente com material contendo 107ccu/ml de cultura pura da cepa P5722- 3 de M. hyopneumoniae. Oito leitões (grupo I) foram inoculados no início do experimento e reinoculados 3 meses mais tarde, após apresentarem a doença e terem se recuperado. Os oito leitões do grupo II foram também inoculados no início do experimento e necropsiados 3 meses depois. Os leitões do grupo III foram inoculados ao mesmo tempo da reinoculação dos leitões do grupo I. Oito leitões não inoculados serviram como controles (grupo IV). Três dias após a reinoculação dos leitões do grupo I e a inoculação do grupo III e, a partir disso, a cada 2 dias, dois leitões de cada grupo foram sacrificados e necropsiados. Segmentos de brônquios e pulmão foram examinados em microscopia ótica e microscopia eletrônica de transmissão e de varredura. Lesões macroscópicas foram observadas nos pulmões de todos os leitões do grupo III (média: 4.74% da superfície) e eram caracterizadas por áreas firmes e avermelhadas nas porções cranioventrais. Lesões macroscópicas semelhantes foram mínimas em pulmões de leitões dos grupos I e II (menos de 1%) e ausentes.no grupo IV. As lesões microscópicas consistiam de hiperplasia do tecido linfóide peribronquial e infiltrado neutrofilico leve nos alvéolos. Na microscopia eletrônica havia lesões focais no epitélio ciliado, caracterizadas por perda de cílios e descamação de células, e presença de leucócitos e micoplasmas nos brônquios de leitões suscetíveis (grupo III). O epitélio bronquial de leitões reinoculados (grupo I), recuperados (grupo II) e controles (grupo IV) era ultraestruturalmente similar, indicando recuperação nos primeiros dois grupos. Embora micoplasmas tenham sido observados entre cilios, uma segunda inoculação em leitões do grupo I não causou outro episódio da doença nem aumentou as alterações morfológicas nos brônquios, sugerindo que esses leitões podem se tornar portadores de micoplasmas.

Estudos histológico, histoquímico e ultra-estrutural de fígados e linfonodos de bovinos com presença de macrófagos espumosos ("foam cells"). Pesquisa Veterinária Brasileira 18(1):29-34. Depto Patologia Clínica Veterinária, Faculdade de Veterinária, UFRGS, Cx. Postal 15094, Porto Alegre, RS 91540-000, Brazil. Samples of liver, kidney, spleen, and hepatic, mesenteric, retromandibular, prescapular and mediastinic lymph nades, collected from 12 groups of cattle of different ages from the State of Mato Grosso, were studied. A total of 120 bovines was examined. According to their history, the animals were kept in pastures where Brachiaria decumbens and Brachiaria brizantha were the predominant plants. Grossly there was a yellowish liver which was more evidente after 24 hr. fixation with formalin. Hepatic and mesenteric lymph nades had normal size, howeve1; on the cut surface there were whitish grooves, extending from the cortex to the medulal; and multiple small white nodular areas in the medulal: Beside these areas there were red, sometimes hemorrhagic, foci. Microscopically all animals had cells with foamy citoplasm in the liver and in the hepatic and mesenteric lymph nades, of ten forming multinucleated cells. In the lymph nodes these infiltrates were adjacent to necrotic, hemorrhagic areas. In the liver the foamy cells were irregularly disseminated throughout the parenchyma or formed nodules around the central vein. These cells did not stain with PAS but weakly with oil red. Ultrastructurally foamy cells and hepatocytes showed negative images of crystals involved partially or totally by membranes. Mediastinic, retromandibular and prescapular lymph nades of the sarne animals showed no gross and microscopic alterations.

Foram examinadas amostras de fígado, rins, baço e linfonodos hepáticos, mesentéricos, retro-mandibulares, pré-escapulares e mediastínicos de 12 lotes em um total de 120 animais, envolvendo diferentes faixas etárias, todos oriundos do Estado de Mato Grosso. Os animais haviam sido mantidos em pastos onde Brachiaria decumbens e Brachiaria brizantha eram as forrageiras predominantes. Macroscopicamente o fígado desses animais mostrava coloração amarelada, mais evidente após 24 horas de fixação em formol a 10%. Nos linfonodos hepáticos e mesentéricos foram evidenciadas, na superfície de corte, estriações esbranquiçadas de forma radiada na cortical e medular com pequenas áreas brancas nodulares multifocais principalmente na medular. Em muitos casos, associado com essas áreas foram vistos focos de aspecto hemorrágico. Microscopicamente foram encontradas, no fígado, linfonodos hepáticos e mesentéricos, células com citoplasma espumoso, muitas das quais multinucleadas. Nos linfonodos hepáticos e mesentéricos estes infiltrados estavam associados a áreas de necrose e hemorragia. No fígado, as células de citoplasma espumoso estavam presentes em todo parênquima, de forma irregular, geralmente formando nódulos ao redor da veia centrolobula1: Estas células não se coravam pelo ácido periódico de Schiff (PAS) e apenas fracamente na coloração de gordura pelo Oil Red O. Na ultraestrutura as células de citoplasma espumoso apresentavam fendas, parcial ou totalmente delimitadas por membrana, que representam a imagem negativa de cristais, presentes também no citoplasma dos hepatócitos.


#10 - Ultrastructural alterations of bone cells in the experimental Solanum malacoxylon poisoning in rabbits

Abstract in English:

The ultrastructure of bone cells of mid-diaphysis of the humerus of young rabbits that received by gastric tube a water estract of Solanum malacoxylon corresponding to 100 mg/kg of body weight is described. Atrophic, degenerated and necrotic osteocytes anel mineralization of perilacunar osteoiel were the main lesions. Osteoblasts were atrophic with diminution of synthetic organels. Osteoclasts exhibited a ruffle border in dose contact with the non eroded mineralized matrix, poorly developecl vesicular region and absent clear zone. Macrophages in fusion forming multinucleated giant cells wcre seen in some resorption cavities. It is suggested that the 1,25(OH)2D3 present in the plant is responsible for the cell changes, inhibition of osteoclastic and osteocytic resorption anel osteopetrosis.

É descrita a ultra-estrutura das células ósseas da cortical da diáfise do úmero de coelhos jovens que receberam, diariamente, via sonda gástrica, um extrato aquoso de Solanum malacoxyloncorrespondente a 100mg da planta seca por quilograma de peso corporal e foram sacrificados 96 horas após o início do experimento. Osteócitos mostravam diversos graus de atrofia e necrose. Lacunas osteocíticas com restos celulares ou vazias foram vistas com frequência. Mineralização do tecido osteóide perilacunar foi constantemente observada. Os osteoblastos mostravam atrofia, diminuição das organelas de síntese e, em algumas oportunidades, exibiam núcleos picnóticos e estavam livres nos canais de Havers. Os osteoclastos mostravam borda pregueada em íntimo contato com a matriz mineralizada não erodida, região vesicular pouco desenvolvida e ausência da zona clara na periferia da borda pregueada. Em alguns canais de Havers, foram identificados macrófagos em processo de fusão para formarem células gigantes. Sugere-se que a osteopetrose descrita na intoxicação por S. malacoxylon seja decorrente de inibição das atividades de reabsorção osteocítica e osteoblástica, por efeito direto da ação de 1,25(OH)2D3.
