#1 - Characterization of nonpathological intrascleral cartilage in the domestic sheep (Ovis aries)

Abstract in English:

Birds, cartilaginous and teleost fish, reptiles, and some amphibians have intrascleral cartilage and/or bone; however, these are rarely reported in therian mammals. This study aimed to investigate and characterize a nonpathological formation of cartilage in the posterior sclera of sheep macroscopically, histologically, and by immunohistochemical exam (IHC). Ninety eyes from 45 domestic sheep were collected, underwent gross examination, fixed in formalin, and embedded in paraffin for the microscopical assessment. Sections with histological shreds of cartilage were selected to perform IHC to confirm the presence of cartilage. Intrascleral cartilage was detected in 60 eyeballs (66.66%) from 37 sheep (82.22%). A slight whitish thickening was grossly seen in the posterior sclera. The histologic exam revealed a few scattered, isolated chondrocytes to larger aggregates of cartilaginous islands in the posterior sclera. Eighteen (30%) of 60 eyeballs revealed marked anti-collagen type II immunolabeling. The development of cartilaginous structures in the eyes is considered rare in mammalian animals. The high occurrence of intrascleral cartilage in the examined sheep eyes suggests that this finding corresponds to an anatomical component of sheep sclera, despite the age, breed, or body condition.

Aves, peixes cartilaginosos e teleósteos e répteis, bem como alguns anfíbios possuem cartilagem e/ou osso intraescleral, contudo, isso é raramente descrito em marsupiais. O objetivo deste trabalho foi investigar e caracterizar a formação não patológica de cartilagem hialina na esclera posterior de ovinos, por exame macroscópico, histológico e imuno-histoquímico. Olhos de 45 ovinos foram coletados, submetidos a exame macroscópico, fixados em formalina a 10% tamponada e incluídos em parafina para o exame histológico. Amostras com evidência de formação cartilaginosa ao exame histológico foram selecionadas para a realização da técnica de imuno-histoquímica para confirmação. Cartilagem intraescleral foi detectada em 60 amostras oculares (66,66%) de 37 ovinos (82,22%), caracterizada na avaliação macroscópica como discretos espessamentos esbranquiçados na esclera posterior. O exame histológico revelou formações que variaram de condrócitos isolados e dispersos a grandes agregados, formando ilhas cartilaginosas na esclera. Dezoito (30%) de 60 bulbos oculares mostraram imunomarcação anti-colágeno tipo II. O desenvolvimento de estruturas cartilaginosas nos bulbos oculares é considerado raro em mamíferos. A identificação de cartilagem intraescleral em 66% dos olhos ovinos avaliados no estudo indica que a formação de cartilagem corresponde a um componente anatômico normal na esclera de ovinos, independente de idade, raça ou condição corporal.


#2 - Ultrastructural and morphometric description of the ear skin and cartilage of two South American wild histricognate rodents (Dasyprocta leporina and Galea spixii)

Abstract in English:

Skin and cartilage have been the main source for the recovery of somatic cells to be used in conservation strategies in wild mammals. In this sense, an important step for the cryopreservation of these samples is to recognize the properties of the skin and cartilage. Thus, knowing that the skin may differ among species and aiming to contribute to the establishment of cryobanks, the study examined the differences in the ear skin and cartilage of wild rodents from South America, agouti (Dasyprocta leporina) and spix’s yellow-toothed cavy (Galea spixii). Ultrastructural and quantitative methods were used to measure skin and cartilage thickness, density of collagen and elastic fibers, cell type number and distribution, and proliferative activity. Although ultrastructural analysis revealed a similar pattern between species, morphometric analysis of the skin and cartilage showed differences between agoutis and cavies regarding thickness of epidermis layers (corneum: 5.3±2.5µm vs. 3.9±0.6µm; intermediate: 16.4±6.2µm vs. 23.4±8.1µm; basal: 9.9±2.1µm vs. 4.8±0.5µm), dermis (183.1±44.0µm vs. 258.2±22.9µm), total skin (211.8±46.0µm vs. 290.3±23.7µm) and perichondrium (27.6±6.1µm vs. 10.5±1.8µm). A greater number of epidermal cells (61.7±15.2 vs. 24.8±7.6) and chondrocytes (32.7±9.0 vs. 27.5±4.7) were observed in agouti, while the cavy presented a greater number of melanocytes (12.6±4.7 vs. 29.9±6.2), keratinocytes (14.7±4.2 vs. 29.8±7.6), and fibroblasts (103.6±24.7 vs. 112.2±11.3). Moreover, a higher percentage of collagen fibers and proliferative activity was observed in the skin of cavies, when compared to the skin of agoutis. Therefore, there are differences between agouti and cavy for ear skin and cartilage, requiring the establishment of species-specific cryopreservation protocols.

A pele e cartilagem têm sido uma importante fonte de recuperação de células somáticas a serem utilizadas em estratégias de conservação em mamíferos silvestres. Nesse contexto, uma importante etapa para criopreservação é conhecer, inicialmente, as propriedades que compõem a pele e cartilagem. Sabendo, então, que a pele pode diferir-se entre espécies e com o objetivo de contribuir para o estabelecimento de criobancos, o estudo evidenciou as diferenças da pele e da cartilagem do pavilhão auricular apical de cutias (Dasyprocta leporina) e preás (Galea spixii) que são roedores silvestres presentes na América do Sul. Para tanto, métodos ultraestruturais e quantitativos foram utilizados para mensurar a espessura da pele e da cartilagem, densidade de fibras colágenas e elásticas, número e distribuição dos tipos celulares e atividade proliferativa. Embora as propriedades ultraestruturais em cutias e preás tenham se mostrado semelhantes, avaliações acerca da morfometria da pele e da cartilagem demonstrou diferenças, especialmente nas camadas epidérmicas (córnea: 5,3±2,5µm vs. 3,9±0,6µm; espinhosa: 16,4±6,2µm vs. 23,4±8,1µm; basal: 9,9±2,1µm vs. 4,8±0,5µm), derme (183,1±44,0µm vs. 258,2±22,9µm), pele total (211,8±46,0µm vs. 290,3±23,7µm) e pericôndrio (27,6±6,1µm vs. 10,5±1,8µm). Além disso, um número maior de células epidérmicas (61,7±15,2 vs. 24,8±7,6) e condrócitos (32,7±9,0 vs. 27,5±4,7) foram observados em cutias, enquanto em preás um maior número de melanócitos (12,6±4,7 vs. 29,9±6,2), queratinócitos (14,7±4,2 vs. 29,8±7,6) e fibroblastos (103,6±24,7 vs. 112,2±11,3) foram evidenciados. Ainda, em preás, uma maior porcentagem de fibras colágenas e da atividade proliferativa foram observadas quando comparadas a pele de cutias. Portanto, existem diferenças entre cutias e preás para pele e cartilagem do pavilhão auricular, exigindo desta forma um estabelecimento de protocolos de criopreservação específica para cada uma destas espécies.

E-mail: fe.kawamoto@gmail.com This study aimed to determine the characteristics and applicability of magnetic resonance imaging (MRI) in the evaluation of autogenous osteochondral graft in intact or macerated format, with or without insulin-like growth factor type 1 (IGF-1) used in repair of cartilage lesions induced in rabbits. Nine New Zealand rabbits were used, in which 18 stifle joints underwent grafting procedure in the femoral trochlear groove. These were divided into four groups, referred as intact osteochondral graft + IGF-1 (n=5), intact osteochondral graft + saline solution (n=4), macerated osteochondral graft + IGF-1 (n=5) and macerated osteochondral graft + saline solution (n=4). Animals were euthanized 12 weeks after surgery and the joints were subjected to MRI using a high magnetic field scanner of 1.5 Tesla. In addition, samples of grafting sites were subjected to anatomopathological examination. The MRI was effective as a noninvasive method to evaluate the repair tissue in osteochondral grafts in articular cartilage of the femur of rabbits by providing complementary data to macroscopic and histological examinations. Through these images and anatomopathological examinations satisfactory results were observed in relation to the repair process of autogenous osteochondral grafts in cartilage of rabbits, regardless of its format or the addition of IGF-1.

Foram utilizados 9 coelhos da linhagem Nova Zelândia, em que as 18 articulações fêmoro-tíbio-patelares foram submetidas à enxertia osteocondral autógena no sulco troclear femoral. Estas foram divididas em quatro grupos, denominados como enxerto osteocondral íntegro + IGF-1 (n=5), enxerto osteocondral íntegro + solução fisiológica (n=4), enxerto osteocondral macerado + IGF-1 (n=5) e enxerto osteocondral macerado + solução fisiológica (n=4). Os animais foram eutanasiados em 12 semanas após a cirurgia e as articulações foram submetidas ao exame de ressonância magnética utilizando um aparelho scanner de 1,5 Tesla de alto campo magnético. Além disso, amostras dos locais de enxertia foram submetidas aos exames anatomopatológicos. O exame de ressonância magnética mostrou-se eficaz como um método não invasivo para avaliação do tecido de reparação em enxertos osteocondrais na cartilagem articular do fêmur de coelhos, fornecendo dados complementares aos exames macroscópicos e histológicos. Por meio destas imagens e dos exames anatomopatológicos, foram observados resultados satisfatórios em relação ao processo de reparação dos enxertos osteocondrais autógenos na cartilagem de coelhos, independentemente de seu formato ou da adição de IGF-1.

E-mail: faleirosufmg@gmail.com The aim of this study was to evaluate the joint cartilage repair by macroscopic (via arthroscopy) and histological (biopsy fragments) analyses in chondral defects induced into equine femoral trochlea treated by microperforation associated with or without intra-articular administration of kartogenin. Six horses weighing 342±1.58 kg (mean ± SD), aged approximately 7.2±1.30 years and with a body condition score of 7.1±0.75, were used. The horses underwent arthroscopy for induction of 1-cm2 chondral lesions in lateral femoral trochlea immediately treated by microperforation of the subchondral bone of both knees. Four weekly intra-articular injections of kartogenin (20µM) in one knee (treated group) and Ringer lactate solution in the contralateral joint (control group) were performed during the postoperative period. After 60 days, macroscopic evaluations were performed by video-arthroscopy, and biopsy samples of the repair tissue were taken for histopathological healing evaluation. No significant change was observed in macroscopic and histological scores for chondral healing between treated and untreated groups (P>0.05). The overall mean percentage of hyaline cartilage in both groups (17.5%) was consistent with other international studies using other types of chondral microperforation; however, no statistical differences were observed between groups (P>0.05). In conclusion, the therapy with kartogenin, according to the used protocol, did not produce any macroscopic and histological healing improvement in induced chondral lesions treated with microperforations in equine femoral trochlea.

Foram realizadas quatro aplicações semanais com 20 µM de cartogenina intra-articulares em um dos joelhos (grupo tratado) e solução de ringer com lactato na articulação contralateral (grupo controle). Após o período de 60 dias, foram feitas as avaliações macroscópicas, através de videoartroscopias, e histológicas, através de biopsia. Não foram observadas diferenças significativas nos escores macroscópicos e histológicos para reparação condral entre animais dos grupos tratados e não tratados (P>0,05). De modo geral, a porcentagem média de cartilagem hialina no tecido de reparo (17,5%) foi condizente com a literatura internacional usando outros tipos de perfuração condral. Entretanto, não se observaram diferenças estatísticas entre grupos (P>0,05). A terapia com cartogenina, segundo protocolo utilizado, não produziu melhora do processo cicatricial em lesões condrais induzidas e tratadas com microperfurações na tróclea lateral do fêmur em equinos.

E-mail: ricarlo@ufv.br In the present study was evaluated the tissue architecture, the percentage of cellular population, as well as viability and distribution of cells in fresh menisci of rabbits and preserved in 98% glycerin. Were analyzed medial menisci of rabbits freshly slaughtered, which were distributed into three groups: the MF group (n=7), composed of fresh menisci, corresponded to the control group; the MG group (n=7), composed by menisci preserved in 98% glycerin, for 30 days, and the MR group (n=7) by menisci preserved in 98% glycerin and rehydrated in NaCl 0.9% for 12 hours. In all menisci were identified and quantified the different cell types: fibroblasts/fibrocytes and condrocytes. The cell population percentage was statistically similar in all groups. All menisci preserved in the MG and MR groups showed a lower intensity of color and shrinkage of collagen fibers, reduced volume and higher intensity of staining of nucleus (chromatin condensation), as compared to fresh menisci (MF), featuring the phenomenon of cell lysis. The cartilaginous matrix of preserved menisci proved to be well preserved because the tissue architecture was maintained. It was concluded that 98% glycerin is an optional preservation mean for meniscal allografts with a devitalized collagenous matrix.

Em todos os meniscos foram identificados e quantificados os diferentes tipos celulares: fibroblastos/fibrócitos e condrócitos. A população celular foi estatisticamente semelhante nos três grupos de meniscos, sendo que os meniscos preservados, grupos MG e MR, apresentaram menor intensidade de coloração e retração das fibras colágenas, diminuição de volume e maior intensidade de coloração dos núcleos (condensação da cromatina), em relação aos meniscos frescos (MF), caracterizando o fenômeno de lise celular. A matriz fibrocartilaginosa dos meniscos preservados revelou-se bem preservada mantendo a arquitetura tecidual dos meniscos. Conclui-se que a glicerina 98% é uma opção de meio de preservação para meniscos objetivando aloenxerto, com matriz colágena desvitalizada.

E-mail: ricarlo@ufv.br In the present study was evaluated the effect of 98% glycerin on the collagen fibers, tissue architecture and size of medial meniscuses of New Zealand rabbits. The animals were separated into three groups: (1) Group MF of fresh meniscus, (2) Group MG of meniscus preserved in glycerin for 30 days, and (3) Group MR of meniscus preserved in glycerin for 30 days and rehydrated in NaCl 0.9% for 12 hours. Histological sections were stained with sirius red for identification of collagen types, which were examined with a polarized light microscope. The total collagen concentration and the fiber arrangement were evaluated. Group MF presented higher Type I collagen concentration and lower Type III collagen concentration when compared with Group MG and MR. This fact is due to the water loss and consequent reduction in size and subsequent retraction of the collagen fibers of the meniscus from these groups, caused by dehydration. This may have occurred because those Type I fibers, thicker and in larger quantities, become more evident than Type III collagen fibers, which are more slender and fragile (fibrils). In the three groups studied, the collagen fibers presented themselves in a circumference form, interposed by radially oriented fibers. All of the meniscuses from Group MF presented fibers arranged obliquely, while in the treated groups, a slight disorganization of collagen fibers could be observed in some areas, what corresponds to 42.8% and 14.3% of the meniscuses, respectively. The MG group presented a significant decrease (p<0.05) in size if compared with the MF group. In the MR group, 85.7% of the meniscuses went back to the original size after rehydratation. The 98% glycerin is effective in preserving the meniscus, following rehydratation, maintaining size, structural architecture, integrity and percentage of collagen of the meniscus preserved similar to the fresh one.

Os meniscos frescos apresentaram significativamente maior concentração de fibras colágenas Tipo I e menor concentração de fibras colágenas Tipo III que os meniscos preservados (MG e MR); isto ocorreu devido à perda de água e conseqüente redução do tamanho dos meniscos e retração das fibras colágenas dos meniscos dos Grupos MG e MR; isto pode ter feito com que as fibras Tipo I, mais espessas e em maior quantidade, se tornassem mais evidentes do que as fibras colágenas Tipo III, que são mais delgadas e frágeis (fibrilas). Nos três grupos estudados, as fibras colágenas apresentaram-se de forma circunferencial, interpostas por fibras orientadas radialmente. Entretanto, nos grupos tratados (MG e MR) foi observado, em pequenas áreas, leve desorganização das fibras colágenas, o que correspondeu a 42,8% e 14,3% dos meniscos, respectivamente. O grupo de meniscos em glicerina apresentou redução significativa (p<0,05) no tamanho em relação ao Grupo MF. No Grupo MR, 85,7% dos meniscos retornaram ao seu tamanho inicial após a reidratação. [Implantation of the autologous auricular cartilagem in the repair of penile deviation in cattle.] Implante de cartilagem auricular autóloga no reparo de desvio de pênis de bovinos. Pesquisa Veterinária Brasileira 29(3):258-262. Faculdade de Medicina Veterinária, Universidade Federal de Uberlândia, Av. Pará 1720, Campus Umuarama, Uberlândia, MG 38400-902, Brazil. E-mail: duvaldo@ufu.br Eighteen 18 to 24-month-old mixed breed bullocks were submitted to removal of the apical ligament of the penis to induce penile deviation. After 60 days from the surgery the steers were put together with cows in heat. A ventral and right lateral penis deviation leading to incapacity to copulate was observed. The bullocks were then submitted to an autolgous transplant of a cartilage segment from the ear pinna placed on top of the tunica albuginea, replacing the penile ligament apical tha had been removed. The implanted material was well tolerated; histologically with only infiltration few mononuclear cells, polymorpho nucleated cells and giant cells, and fibrosis, which caused strong adherence and provided ability to sustain the free penile extremity. The bullocks that were cows in heat did not show any degree of penile deviation nor any change in sexual behavior. The surgical method described here was shown to be satisfactory as to be indicated for correction of ventral and right lateral penile deviation in bullocks.
