#1 - Influence of early use of antimicrobial on the health and performance of Holstein calves in the first month of life

Abstract in English:

The early use of antimicrobial therapy has been introduced in many farms to prevent diarrhea and respiratory disease in young calves; however, there is controversy about whether this practice has a beneficial effect on the health of these animals. This study evaluated the influence of the early use of antimicrobials on the health and performance of neonatal Holstein calves. Twenty-six Holstein calves were screened and divided into two groups, according to the administration (ATB+), or not (ATB-) of tulathromycin (2.5mg/kg, subcutaneously) within the first 12 hours of life. Calves were evaluated by general clinical examination, fecal score, respiratory score, and external palpation of the umbilical region, besides fecal output of dry matter. Anemia was determined by using an automatic system and, also, using a commercial kit for iron dosage. Diarrhea was diagnosed by a centrifuge‑flotation technique using a sugar solution (Cryptosporidium) and multiplex semi-nested RT-PCR (rotavirus/coronavirus). The performance of the calves was estimated by Daily Weight Gain (DWG). The young dairy calves were evaluated within 12 hours of birth (≤12h) and at 3-5th (D3-5), 7-9th (D7-9), 13-15th (D13-15), 20-23rd (D20-23), and 27-30th (D27‑30) days of life. No difference was noted between the ATB+ and ATB- groups concerning heart rate, respiratory frequency, and rectal temperature. Erythrogram showed a higher frequency of anemia in ATB- group (P=0.016) at the D3-5 check-up; lower values of serum iron were also observed simultaneously (P=0.051). Thirteen cases of respiratory disease were detected during this study; however, no significant difference was observed between the groups in this regard. The frequency of diarrhea (fecal score 2-3) was high in both groups, peaking at D13-D15. No differences were noted between the groups regarding the frequency of diarrhea when considering the dry fecal matter. The predominant etiological agent for diarrhea was Cryptosporidium spp.. The DWG was similar between groups, with maximum weight reduction on D13-15. The administration of tulathromycin in prophylactic dose (2.5mg/kg) at birth decreased the frequency of anemia but did not influence weight gain or the prevalence of diarrhea.

Specific immune response in neonate Holstein heifer calves fed fresh or frozen colostrum. Pesquisa Veterinária Brasileira 37(12):1385-1394. Departamento de Medicina Interna, Faculdade de Medicina Veterinária e Zootecnia, Universidade de São Paulo, Av. Prof. Dr. Orlando Marques de Paiva 87, Cidade Universitária, Butantã, São Paulo, SP 05508-270. E-mail: viviani.gomes@usp.br The objective of this study was to evaluate the influence of viable cells from colostrum on immune development in dairy heifer calves during the first 28 days of life. The animals were distributed between 2 groups: COL+ (n=9) receiving fresh whole colostrum from their own damns; and COL- (n=10) receiving pooled frozen colostrum, containing no viable cells, from a pool of donor cows. These calves were assessed before colostrum intake (D0), 48 hours of age (D2), and weekly from D7 to D28. The development of immunity was evaluated by assessment of the phenotype of blood leukocyte subsets, and induced cytokine production after 72 hours of stimulation in culture with concanavalin A (ConA), killed Staphylococcus aureus (S.aureus) and killed Escherichia coli (E. coli) by peripheral blood mononuclear cell (PBMC). The clinical history of these calves was marked by a high frequency of diarrhea in both groups. However, COL- had greater diarrhea intensity scores (fecal score~3 of 4), and rectal temperature on D7 than COL+ calves. Moreover, bronchopneumonia (n=1) and navel inflammation were observed only in COL- calves. COL- had a lower concentration of serum iron, and a higher absolute number of lymphocytes on D7 than COL+. COL- also had a higher percentage of anemic calves than the COL+ calves on D21 and D28. In general, the percent of cells within each subset of leukocytes was similar between the groups over the experiment, except on week 1 when COL- calves had a higher percentage of lymphocytes expressing CD45RO+ (P=0.07). A steady increase in CD45RO+ and concomitant decline in CD45RO- leukocytes was observed over the course of the study, indicating the development of immune memory. The proportion of CD14MHCII+ leukocytes increased with age (P≤0.05). The median background cytokine production by PBMC that were not stimulated was below the level of detection of the assays used for both groups. The PBMC from COL+ calves stimulated with ConA secreted a larger quantity of IL-17 week 2 (COL+=2060.0pg/mL and COL-=0.0pg/mL, P=0.00). PBMC from COL+ calves stimulated with killed S. aureus whole cell antigen (P=0.05) and killed E. coli whole cell antigen (P=0.05) also secreted higher levels of IL17 than COL- calves at week 4. Clear production of IL17 was observed in PBML from COL+ calves at week 2, but the difference was not statistical different between groups. In conclusion, calves fed fresh and frozen colostrum showed no difference in cells subset profile overall. The increased percentage of leukocytes expressing the memory CD45RO+ or CD14MHCII+ over the course of the experiment indicated a maturation of the adaptive immune response after natural exposure to pathogens in the environment of the calf. The enhanced IL17 secretion by COL+ calves indicated that viable maternal cells modulated T-cell Th17 production that was primed by bacterial antigens. This mechanism could be responsible for quick and efficient activation of neutrophils for bacterial clearance. The differences in cytokine production observed between groups may help to explain the different clinical pictures observed for calves COL+ and COL- calves.

E-mail: malafaia_ufrrj@yahoo.com.br The daily weight gain, behavioral activities (grazing, ruminating and water consumption) and the number of rumen protozoa, pH, NH3, and the osmolarity of rumen fluid was evaluated for four groups of six calves HPB/Zebu submitted to daily intake of 15, 30, 60 and 90g of NaCl during 135 days (9th Jan. to 24th May 2008).Throughout the experimental period the calves were in perfect health condition and did not show any signs suggestive of chronic sodium poisoning. There was no significant difference in average daily gain of the calves as the intake of NaCl increased. No significant behavioral changes were observed in the activities of grazing and rumination. However, there was a noticeable change in the frequency of water consumption in the calves that ingested 90g/d of NaCl; they went more often to the trough and drank more water than the group that ingested only 15g sodium chloride/d. The osmolarity of ruminal liquid was higher in the group of calves fed 90g of NaCl/d. The pH, NH3 concentration and number of rumen protozoa was within the normal range.

#5 - Genital immunization of heifers with a glycoprotein E-deleted, recombinant bovine herpesvirus 1 strain confers protection upon challenge with a virulent isolate, 30(1):42-50

Abstract in English:

ABSTRACT.- Weiss M., Vogel F.S.F., Martins M., Weiblen R., Roehe P.M., Franco A.C. & Flores E.F. 2010. Genital immunization of heifers with a glycoprotein E-deleted, recombinant bovine herpesvirus 1 strain confers protection upon challenge with a virulent isolate. Pesquisa Veterinária Brasileira 30(1):42-50. Departamento de Medicina Veterinária Preventiva, Universidade Federal de Santa Maria, 97105-900 Santa Maria, RS, Brazil. E-mail: eduardofurtadoflores@gmail.com Venereal infection of seronegative heifers and cows with bovine herpesvirus type 1.2 (BoHV-1.2) frequently results in vulvovaginitis and transient infertility. Parenteral immunization with inactivated or modified live BoHV-1 vaccines often fails in conferring protection upon genital challenge. We herein report an evaluation of the immune response and protection conferred by genital vaccination of heifers with a glycoprotein E-deleted recombinant virus (SV265gE-). A group of six seronegative heifers was vaccinated with SV265gE- (0,2mL containing 106.9TCID50) in the vulva submucosa (group IV); four heifers were vaccinated intramuscularly (group IM, 1mL containing 107.6TCID50) and four heifers remained as non-vaccinated controls. Heifers vaccinated IV developed mild, transient local edema and hyperemia and shed low amounts of virus for a few days after vaccination, yet a sentinel heifer maintained in close contact did not seroconvert. Attempts to reactivate the vaccine virus in two IV vaccinated heifers by intravenous administration of dexamethasone (0.5mg/kg) at day 70 pv failed since no virus shedding, recrudescence of genital signs or seroconversion were observed. At day 70 pv, all vaccinated and control heifers were challenged by genital inoculation of a highly virulent BoHV-1.2 isolate (SV-56/90, 107.1TCID50/animal). After challenge, virus shedding was detected in genital secretions of control animals for 8.2 days (8-9); in the IM group for 6.2 days (4-8 days) and during 5.2 days (5-6 days) in the IV group. Control non-vaccinated heifers developed moderate (2/4) or severe (2/4) vulvovaginitis lasting 9 to 13 days (x: 10.7 days). The disease was characterized by vulvar edema, vulvo-vestibular congestion, vesicles progressing to coalescence and erosions, fibrino-necrotic plaques and fibrinopurulent exudate. IM vaccinated heifers developed mild (1/3) or moderate (3/4) genital lesions, lasting 10 to 12 days (x: 10.7 days); and IV vaccinated heifers developed mild and transient vulvovaginitis (3/4) or mild to moderate genital lesions (1/4). In the IV group, the clinical signs lasted 4 to 8 days (x: 5.5 days). Clinical examination of the animals after challenge revealed that vaccination by both routes conferred some degree of protection, yet IV vaccination was clearly more effective in reducing the severity and duration of clinical disease. Furthermore, IV vaccination reduced the period of virus shedding in comparison with both groups. Taken together, these results demonstrate that SV265gE- is sufficiently attenuated upon IV vaccination in a low-titer dosis, is not readily reactivated after corticosteroid treatment and lastly, and more importantly, confers local protection upon challenge with a high titer of a virulent heterologous BoHV-1 isolate. Therefore, the use of this recombinant for genital immunization may be considered for prevention of BoHV-1-associated genital disease in the field.

RESUMO.- Weiss M., Vogel F.S.F., Martins M., Weiblen R., Roehe P.M., Franco A.C. & Flores E.F. 2010. Genital immunization of heifers with a glycoprotein E-deleted, recombinant bovine herpesvirus 1 strain confers protection upon challenge with a virulent isolate.

Abstract in English:

ABSTRACT.- Henzel A., Diel D.G., Arenhart S., Vogel F.S.F., Weiblen R. & Flores E.F. 2008. [Virological and clinico-pathological features of acute vulvovaginitis and latent infection by bovine herpesvirus 1.2 in heifers experimentally infected.] E-mail: flores@ccr.ufsm.br Venereal infection of heifers and cows with bovine herpesvirus type 1.2 (BoHV-1.2) may result in vulvovaginitis and transient infertility. The acute infection is followed by the establishment of latent infection which can be periodically reactivated. We herein describe the virology and clinico-pathological aspects of acute and recrudescent vulvovaginitis in heifers inoculated with a Brazilian BoHV-1.2 isolate recovered from an outbreak of balanoposthitis. Genital inoculation of isolate SV-56/90 (108.1TCID50/animal) in four eight-months-old heifers resulted in efficient virus replication in the genital mucosa and the development of moderate to severe vulvovaginitis. The inoculated heifers shed virus in genital secretions in titers up to 107.3TCID50/mL until day 10 pi and developed genital congestion, swelling, vesicles and pustules. The vesicles and pustules increased in size eventually coalesced and became covered with a yellowish exsudate. These signs appeared at day 2 pi, increased in severity up to days 5 - 8 pi and progressively subsided thereafter. Dexamethasone administration at day 55 pi resulted in virus shedding in vaginal secretions for up to 10 days. Virus reactivation in all animals was accompanied by clinical recrudescence of the disease, yet less severe than during acute infection. Examination of sacral ganglia and lymph nodes by PCR at day 36 post-reactivation revealed the presence of latent viral DNA in the pudendal (4/4), genito-femoral, sciatic and rectal caudal (3/4) and obturator nerve ganglia (1/4); in addition to several regional lymph nodes. These results demonstrate the virulence of isolate SV-56/90 for heifers and pave the way for its use in further pathogenesis studies and vaccine-challenge trials.

[Evolution of serological antibody titres by the plate agglutination, rose Bengal and complement fixation tests in Nelore heifers (Bos indicus) vaccinated at 18 months of age with Brucella abortus strain 19] Evolução de títulos sorológicos, nas provas de soroaglutinação em placa, antígeno acidificado tamponado e fixação de complemento, em bezerras Nelore vacinadas aos 18 meses de idade com Brucella abortus amostra B 19. One hundred and eight Nelore heifers (Bos indicus) about 18 months of age were vaccinated with a standard dosage (60-120 x 109 viable organisms) of Brucella abortus strain 19 vaccine, airning at to study the persistence of serological antibody titres by the plate agglutination, rase Bengal and cornplement fixation tests. Serum samples were collected just before vaccination and after 45 days, 6, 12 and 18 months. Before the vaccination, all the heifers showed negative results by the three tests. After 45 days, all the heifers had titres higher than or equal to 1/100 by the plate agglutination test and all of them showed positive results by the rose Bengal plate test, while 2 animals had titre 1/2 and 106 animals had titres higher than or equal to 1/4 by the complement fixation test. One year after the vaccination, most of the heifers did not show significant serological antibody titres. Considering the risk to which animals in endemic areas and infected herds are subjected, and considering the clear reduction of the serological antibody titres shown by most of the vaccinated heifers, one rnay assume that, in case of Nelore heifers in endemic areas and that were not vaccinated between 3 and 8 months of age, it would be more reasonable to vaccinate them at 18 months of age than to expose them to a high risk of Brucella infection.

RESUMO.- Mathias, L.A., Chaves, L.F., Chen, A.A., Girio, R.J.S.

ABSTRACT.- Melo V.S.P., Passos L.M.F, Facuri-Filho E.J., Saturnino H.M. & Ribeiro M.F.B. 2001. Natural infection of calves by Anaplasma marginale in dairy herds of the Metalúrgica Region, Minas Gerais. Postal 486, Belo Horizonte, MG 31270-910, Brazil. The dynamic of natural infections by Anaplasma marginale in calves was evaluated during a period of one year on two farms located in the Metalúrgica Region, State of Minas Gerais, Brazil. Blood samples were collected weekly for rickettsemia and packed cell volume (PCV) determination. The animals born from March to July suffered the infection in October and November, independently of their age, whilst calves born from September to December acquired the infection during the first days of life. These animals presented patent rickettsemia from 30 days of life. During the patent period PCV decreased after one week of infection, ranging from 20 to 23%. It was concluded, that in the region studied, the transmission of A. marginale is influenced by climatic conditions, and that calves born during the dry season are more likely to acquire the infection when they are exposed to high transmission leveis during the subsequent raining season.

Depto Parasitologia, Universidade Federal de Minas Gerais, Cx. Postal 486, Belo Horizonte, MG 31270-910, Brazil.
