#1 - Mast cells in the pathophysiology of Duchenne muscular dystrophy in Golden Retriever dogs

Abstract in English:

The Golden Retriever muscular dystrophy (GRMD) is one of the best models of Duchenne muscular dystrophy (DMD), with similar genotypic and phenotypic manifestations. Progressive proliferation of connective tissue in the endomysium of the muscle fibers occurs in parallel with the clinical course of the disease in GRMD animals. Previous studies suggest a relationship between mast cells and the deposition of fibrous tissue due to the release of mediators that recruit fibroblasts. The aim of this study was to evaluate the presence of mast cells and their relationship with muscle injury and fibrosis in GRMD dogs of different ages. Samples of muscle groups from six GRMD and four control dogs, aged 2 to 8 months, were collected and analyzed. The samples were processed and stained with HE, toluidine blue, and Azan trichrome. Our results showed that there was a significant increase in infiltration of mast cells in all muscle groups of GRMD dogs compared to the control group. The average number of mast cells, as well as the deposition of fibrous tissue, decreased with age in GRMD dogs. In the control group, all muscle types showed a significant increase in the amount of collagenous tissue. This suggests increased mast cell degranulation occurred in younger GRMD dogs, resulting in increased interstitial space and fibrous tissue in muscle, which then gradually decreased over time as the dogs aged. However, further studies are needed to clarify the role of mast cells in the pathogenesis of fibrosis.

O cão Golden Retriever distrófico (Golden Retriever muscular dystrophy - GRMD) é um dos melhores modelos da distrofia muscular de Duchenne (DMD), com manifestações genotípicas e fenotípicas similares. A proliferação progressiva de tecido conjuntivo no endomísio das fibras musculares ocorre paralelamente ao curso clínico da doença em animais GRMD. Estudos anteriores sugerem uma relação entre os mastócitos e a deposição de tecido fibroso devido à liberação de mediadores que recrutam fibroblastos. O objetivo deste estudo foi avaliar a presença de mastócitos e sua relação com a lesão muscular e fibrose em cães GRMD de diferentes idades. Amostras de grupos musculares de seis GRMD e quatro controles, com idade entre 2 a 8 meses, foram coletadas e analisadas. As amostras foram processadas e coradas com HE, azul de toluidina e tricrômico de Azan. Nossos resultados mostraram que houve um aumento significativo na infiltração de mastócitos em todos os grupos musculares de cães GRMD em comparação com o grupo controle. O número médio de mastócitos, assim como a deposição de tecido fibroso, diminuiu com a idade em cães GRMD. No grupo controle, todos os tipos musculares mostraram um aumento significativo na quantidade de tecido colágeno. Isto sugere o aumento da degranulação de mastócitos em cães GRMD mais jovens, resultando em aumento do espaço intersticial e tecido fibroso no músculo, que então gradualmente diminuiu com o tempo à medida que os cães envelheceram. No entanto, mais estudos são necessários para esclarecer o papel dos mastócitos na patogênese da fibrose.


#2 - Pathogenesis of Bovine alphaherpesvirus 2 in calves following different routes of inoculation

Abstract in English:

Bovine alphaherpesvirus 2 (BoHV-2) is the agent of herpetic mammilitis (BHM), a cutaneous and self-limiting disease affecting the udder and teats of cows. The pathogenesis of BoHV-2 is pourly understood, hampering the development of therapeutic drugs, vaccines and other control measures. This study investigated the pathogenesis of BoHV-2 in calves after inoculation through different routes. Three- to four-months seronegative calves were inoculated with BoHV-2 (107TCID50.mL-1) intramuscular (IM, n=4), intravenous (IV, n=4) or transdermal (TD) after mild scarification (n=4) and submitted to virological, clinical and serological monitoring. Calves inoculated by the IV route presented as light increase in body temperature between days 6 to 9 post-inoculation (pi). Virus inoculation by the TD route resulted in mild inflammatory lesions at the sites of inoculation, characterized by hyperemia, small vesicles, mild exudation and scab formation, between days 2 and 8pi. Virus or viral DNA was detected by PCR in the crusts/swabs collected from lesions of 3 out of 4 animals inoculated TD from day 2 to 8pi. Viremia was detected in 3/4 animals of the IM group (from day 4 to 8pi); in 2/4 animals of the IV group (days 6 and 8pi) but not in the TD group. Calves from all inoculated groups seroconverted to BoHV-2 in titers from 4 to 64, as indicated by virus-neutralizing (VN) assays performed in sera collected at day 15pi. Administration of dexamethasone (Dex) to the inoculated calves at day 48pi, did not result in virus reactivation as indicated by lack of virus detection in the blood and/or in inoculation sites and no increase in VN antibody titers. These results demonstrated that BoHV-2 was able to replicate efficiently in calves following different routes of exposure, produced viremia after IM and IV inoculation and was not reactivated by Dex treatment.

O alfaherpesvírus bovino 2 (BoHV-2) é um agente etiológico da mamilite herpética (BHM), uma doença cutânea e autolimitante do úbere e tetos de vacas. Pouco se sabe sobre a patogênese do BoHV-2, dificultando o desenvolvimento de medicamentos terapêuticos e vacinas. Este estudo investigou a patogênese do BoHV-2 em bezerros após a inoculação por diferentes vias. Bezerros soronegativos de três a quatro meses foram inoculados com BoHV-2 (107TCID50.mL-1) por via intramuscular (IM, n=4), por via intravenosa (IV, n=4) ou transdérmica (TD, n=4) após escarificação leve e submetidos a monitoramento virológico, clínico e sorológico. Os bezerros inoculados pela via IV apresentaram aumento leve da temperatura corporal entre os dias 6 a 9 pós-inoculação (pi). A inoculação do vírus pela via TD resultou em lesões inflamatórias leves nos locais de inoculação, caracterizadas por hiperemia, pequenas vesículas, exsudação leve e formação de crostas, entre os dias 2 e 8pi. O vírus ou DNA viral foi detectado por PCR nas crostas/swabs coletados de lesões de 3 de 4 animais inoculados TD do dia 2 ao 8pi. Viremia foi detectada em 3/4 dos animais do grupo IM (do dia 4 ao 8pi); em 2/4 animais do grupo IV (dias 6 e 8pi), mas não no grupo TD. Bezerros de todos os grupos inoculados soroconverteram o BoHV-2 em títulos de 4 a 64, conforme indicado por ensaios de vírus-neutralização (VN) realizados em soro coletado no dia 15pi. Administração de dexametasona (Dex) nos bezerros inoculados no dia 48pi, não resultou em reativação do vírus, como indicado pela falta de detecção de vírus no sangue e/ou nos locais de inoculação e pela ausência de aumento nos títulos de anticorpos. Estes resultados demonstraram que o BoHV-2 foi capaz de replicar eficientemente em bezerros seguindo diferentes vias de inoculação, produziu viremia após a inoculação IM e IV e não foi reativado pelo tratamento com Dex.

E-mail: prof_ufg.dmv@hotmail.com Morfometric studies of bovine and buffalo digits can help to understand the etiopathogeny of digital diseases. This study described morphometric characteristics of digits of Curraleira (Bos taurus), Pantaneira (Bos taurus) Nelore (Bos indicus) breeds of cattle and Murrah buffalo (Bubalus bubalis) and stablish possible relation among the parameters and digital infirmities. Were used ten animals of each breed and specie. Two limbs were evaluated, a toracic and a pelvic, in a total of 80 distal limbs. Morphometric measurements were obtained using a graduated paquimeter and angles using a metallic protactor. The main parameters evaluated were hoof dorsal angle (A), dorsal wall length (B), heel height (C), toe height (D), hoof length (E), hoof diagonal hoof (F), lateral digit width (H), medial digit width (I) and medial digit length (J). For results averages comparison among breeds were used Tukey test (p<0,05). Multivariate analysys for graphic representations of canonic variables was used to express similarity of measures studied among groups, using R software. Results shows tha bubaline present higher morphometric measures for the variables B, C, D, E, F, G, H, I and J, only for variable A presented lower measures comparing the bovine breeds studied. There is similarity between the three breeds of cattle studied about toe height (D), lateral digit width (G) and medial digit width (H), which differed of values observed in buffalo. It was concluded that digital morphometry can influence the occurence of digital infirmities, but doesn´t act as an isolated factor, needing interaction of other structural, environmental and management factors for its occurence.

Este estudo objetivou descrever as características morfométricas dos dígitos de bovinos das raças Curraleira (Bos taurus), Pantaneira (Bos taurus), Nelore (Bos indicus) e de bubalinos (Bubalus bubalis) da raça Murrah e estabelecer possível relação entre tais medidas e a ocorrência de enfermidades digitais. Na pesquisa foram utilizados dez animais, saudáveis, de cada raça e espécie. Foram avaliados dois membros de cada animal, sendo um torácico e outro pélvico, totalizando 80 extremidades distais. As medidas morfométricas foram obtidas com auxílio de um paquímetro mecânico graduado e os ângulos das pinças conferidos por meio de transferidor metálico. Os principais parâmetros digitais avaliados foram o ângulo dorsal do casco (A), comprimento da parede dorsal (B), altura do talão (C), altura da pinça (D), comprimento do casco (E), comprimento diagonal do casco (F), largura do dígito lateral (G), largura do dígito medial (H), comprimento do dígito lateral (I) e comprimento do dígito medial (J). Para a comparação de médias dos resultados obtidos entre as raças foi utilizado o teste de Tukey (p<0,05). A análise multivariada para as representações gráficas das variáveis canônicas foi empregada para expressar a similaridade das medidas estudadas entre os grupos, no qual se utilizou o software R. Os resultados revelaram que os bubalinos apresentam as maiores medidas morfométricas para as variáveis B, C, D, E, F, G, H, I e J e apenas na variável A apresentaram medidas inferiores entre as diferentes raças de bovinos estudadas. Existe similaridade entre as três raças de bovinos estudadas em relação às variáveis, altura da pinça (D), largura do dígito lateral (G) e largura do dígito medial (H) as quais se distanciam dos valores encontrados para essas variáveis nos bubalinos, Concluiu que a morfometria digital pode influenciar na ocorrência de enfermidades digitais, mas não age como fator isolado, necessitando da interação com outros fatores estruturais, ambientais e de manejo para a manifestação dessas doenças.

E-mail: anemiaveterinaria@yahoo.com.br The pathogenesis, clinical, hematological and pathological features of the natural infection by the protozoan organism Rangelia vitalii (canine rangeliosis) was studied in 35 dogs that died due this condition. The results allow for the following set of conclusions on canine rangeliosis: (1) causes an exclusively extravascular immune mediated hemolysis; (2) is invariably associated with some degree of hemorrhage observed at necropsy, but no always clinically apparent; (3) the clinical signs that are the hallmark of the disease are anemia, icterus and splenomegaly; (4) the main hematological aspect that establishes a clinical suspect is the development of anemia with signs of intense erythroid regeneration; (5) the three main differential diagnosis are leptospirosis, babesiosis and e acute monocytotropic ehrlichiosis; (6) the main observed histopathological lesion is an association of lymphoid hyperplasia with mononuclear inflammatory reaction, predominantly plasmacytic, but occasionally granulomatous; (7) other frequently found lesions are secondary to a marked regenerative anemia; (8) large numbers of the etiologic agent can be easily demonstrate in most tissues, mainly in lymph nodes, spleen, bone marrow, heart, and tonsils.

The chicken inoculated with live bacterial cultures were submitted to euthanasia 21 days later, and the cecum was examined histologically using hematoxylin-eosin, silver staining and immunohistochemistry with a polyclonal antibody anti-Brachyspira pilosicoli. With silver staining and immunohistochemistry, respectively, 21.59% and 70.96% of the chicken showed colonization of the cecal epithelium with B. pilosicoli. Differences in the type of colonization characterized by continuous adhesion, focal adhesion or free bacteria in the intestinal lumen were observed. Immunohistochemistry was more efficient to assess intestinal colonization, because 49.37% more colonized chicken could be detected with silver staining. With three strains, elongated figures inside the cytoplasm of epithelial cecal cells in the inoculated chicken were observed.

E-mail: elporti@fcv.unlp.edu.ar Swine eperythrozoonosis is a haemotrophic disease caused by Eperythrozoon suis, actually called Mycoplasma suis, an extracellular bacterial organism that apparently adheres to pig erythrocyte membrane, inducing its deformation and damage. Since little is known about the ultrastructural and morphometrical aspects of this microorganism, the present work aimed to deal with these issues. The ultrastructural study revealed the presence of structures corresponding to tubules disseminated throughout the soma of M. suis. A variable separation between the microorganism membrane and that of the erythrocyte was also observed. The structural and positional attitude of M. suis could allow speculation about its mechanism of action.

#7 - ARE ALVEOLAR BONE CHANGES A DETERMINANT FACTOR FOR "CARA INCHADA" IN CATTLE ?

Abstract in English:

In order to study possible alterations of the skeleton which might play a role in the pathogenesis of the periodontitis of "cara inchada" in young cattle, ribs from 20 affected calves, 2 to 10 months old, were examined. Electrolytically decalcified longitudinal sections of the costochondral junction and cross sections through the corpus costae, stained with Haematoxylin-Eosin, were studied. In five calves, longitudinal sections of the proximal humerus were examined as well. The status of mineralization was checked by microradiograms. Systemic alteration of the skeleton due to disturbances of mineral metabolism could not be shown in any of the animals. In seven 2 to 4 months old calves, no bone changes were found. The reduced osteogenesis in six 3 to 5 months old calves and the reduced osteogenesis and diminished chondral growth in seven 5 to 10 months old calves are therefore a consequence of the disease. The results show that the development of the alveolar bone was not defective, so this cannot be a determinant factor for the development of the periodontitis of "cara inchada" in cattle.

Para verificar possíveis alterações do esqueleto, que poderiam ter papel importante na patogênese da periodontite da "cara inchada" em bovinos jovens, costelas de 20 bezerros com 2 a 10 meses de idade e afetados pela doença foram estudadas. Examinaram-se cortes longitudinais eletroliticamante descalcificados das junções cartilaginosas e cortes transversais do corpus costae, corados pela hematoxilina-eosina. Em cinco bezerros examinaram-se também cortes longitudinais do úmero proximal. O estado de mineralização óssea foi avaliado através de microrradiogramas. Em nenhum dos animais alterações do esqueleto por distúrbios do metabolismo mineral podiam ser demonstradas. Nos bezerros com 2 a 4 meses de idade nenhuma alteração patológica do esqueleto foi encontrada. Por isso, a osteogênese reduzida observada nos bezerros com 3 a 5 meses de idade, bem como a osteogênese e o crescimento cartilaginoso reduzidos nos bezerros de 5 a 10 meses de idade, devem ser interpretados como consequências da doença. Os resultados desse estudo demonstram que não existe um desenvolvimento deficiente do osso alveolar como fator predisponente para o desenvolvimento da periodontite da "cara inchada" nos bovinos.


#8 - Reproduction of caseous lymphadenitis in goats by small numbers of Corynebacterium pseudotuberculosis

Abstract in English:

Caseous lymphadenitis was reprodued experimentally by intradennic inoculation of 5, 20 and 100 washed Corynebacterium pseudotuberculosis bacteria in 26 normal goats. Between day 2 and 9 after inoculation, there was local erythema, papula and skin ulceration and, at day 15, a small enlargement of the local lymph node. Between day 45 and 180, independent of the infective dose rate, the infection developed to an abscess in the local lymph node of about one third of the goats. A similar number showed a pronounced enlargement of the local lymph node, but these later became normal again. The remainder of the goats showed an early return to normal after the initial signs of infection. All 26 goats, except 2, presented an antitoxin titre during some phase of the infection. The experimental·infection was shown to develop as the natural. It was concluded that selfcures, demonstrated experimentally, must also occur in the field, making clinical and immunological diagnosis difficult.

Abstract in Portuguese:

A linfadenite caseosa foi reproduzida experimentalmente através da inoculação intradérmica de 5, 20 e 100 bactérias lavadas de C. pseudotuberculosis em 26 caprinos normais. A infecção manifestou-se, inicialmente, por formação de eritema, pápula, pústula e ulceração local da pele entre o 2º e o 9º dia pós-inoculação e no 15º dia por um ligeiro aumento do linfonodo satélite. Posteriormente, entre o 45º e o 180º. dia, independentemente da dose infectante usada, a infecção evoluiu para abscedação do linfonodo local em cerca de 1/3 dos caprinos. Em outro número similar, observou-se acentuado aumento do volume do linfonodo mas que depois regrediu e, finalmente, nos restantes houve rápida normalização após os sinais iniciais da infecção. Dos 26 caprinos apenas 2 não apresentaram antitoxinas no soro durante nenhuma fase da infecção. Foi demonstrado com o presente experimento que a infecção evoluiu simulando a natural e concluiu-se que as curas espontâneas observadas também devem ocorrer nas infecções de campo, dificultando o diagnóstico clínico e imunológico.
