#1 - Detection of virulence genes, antibiotic resistance genes and antibiotic resistance of Escherichia coli isolated from diarrheic lambs in Anhui Province, China

Lamb diarrhea seriously restricts the development of the sheep industry. Infectious pathogens often cause diarrhea, and E. coli isolates are highly distributed in infectious diarrhea. The study aimed to investigate the prevalence of pathogenic E. coli in diarrhea lambs and determine the distribution of virulence genes, antibiotic resistance genes, and antibiotic resistance. One hundred seventy-eight E. coli isolates were isolated from the feces of 204 diarrhea lambs. The virulence genes mdh, hlyF, iss, ompA, fimC, iucD, st, lt, stx1, stx2, and antibiotic resistance genes including tetA, tetB, aac (6′)-II, blaCMY-2, qnr, aadA1, sul1, blaTEM, blaCTX-M were detected by polymerase chain reaction (PCR). Antibiotic resistance was determined by Kirby-Bauer Disc Diffusion method. There were 109 (61.24%, 109/178) enterotoxigenic E. coli (ETEC), 119 (66.85%, 119/178) Shiga toxin-producing E. coli (STEC), and 95 (53.37%, 95/178) hybrid STEC/ETEC isolates. The highest prevalent virulence genes were ompA (80.90%, 144/178) and fimC (67.98%, 121/178), and the lowest was iucD (8.99%, 16/178). The most commonly detected antibiotic resistance genes were tetA/tetB (92.70%, 165/178), qnr (75.84%, 135/178), and sul1 (62.92%, 112/178), no blaTEM or blaCTX-M genes were detected. All isolates had high antibiotic resistance to lincomycin (96.63%, 172/178), tetracycline (88.76%, 158/178), and co-trimoxazole (80.34%, 143/178), and the multidrug resistant (MDR) rate reached 93.82% (167/178). The high prevalence of ETEC and STEC indicates that E. coli is one of the critical pathogenic agents leading to diarrhea in lambs in the region, and its high antibiotic resistance, especially MDR, should be brought to our attention.

#2 - Monitoring periodontal lesions and their effects during pregnancy: microbiological aspects of the oral cavity and amniotic fluid in pregnant ewes

Abstract in English:

Periodontitis affects the teeth supporting tissues, leading to tooth loss and damage to animal health. Evidence in humans suggests that oral microorganisms spread systemically, increasing the risk of pregnancy disorders such as miscarriage, prematurity, and low birth weight. This study aimed to verify whether periodontopathogenic microorganisms reach the transplacental unit, culminating in problems in pregnant ewes. After analyzing the oral cavity, 10 clinically healthy pregnant ewes (OGCH group) and 10 pregnant ewes with periodontitis (OGP group) were selected. The subgingival biofilm was collected for the polymerase chain reaction (PCR) test and amniotic fluid for both the PCR and interleukin (IL) analysis. Peripheral blood was collected for complete blood count, and analyses of IL-6, IL1-β, and tumor necrosis factor-α were performed. Placental fragments were collected to assess the inflammatory changes using optical microscopy. After giving birth, both the ewes and their lambs were weighed. On clinical examination, a positive correlation between bleeding and suppuration (correlation index – CI=0.54), suppuration and marginal gingivitis (CI=0.34), and marginal gingivitis and edema (CI=0.54) was observed. The weights of the ewes (p=0.013) and their respective lambs (p=0.04) in the OGP group were lower than those of their OGCH group counterparts. The hematological analysis revealed that the OGP group ewes showed a slight increase in the mean corpuscular volume (p=0.2447), segmented cells (p=0.3375), and eosinophils (p=0.3823) when compared with the OGCH group ewes, without a statistical difference. Regarding the microorganisms detected in the oral cavity, there was a significant difference between the occurrence of periodontal pockets and the presence of Fusobacterium necrophorum (p=0.0328), Porphyromonas asaccharolytica (p=0.0392), and the Mollicutes class (p=0.0352). Staphylococcus genus (p=0.9107) and Archaea domain (p=0.7245) were detected in the amniotic samples of both groups, without a significant difference, whereas P. asaccharolytica (p=0.2685) was only detected in one sample in the OGCH group. The expression of cytokine IL-6 in the OGP group differed significantly between the prepartum and postpartum periods (p=0.0039); moreover, it differed significantly in the postpartum period between the OGCH and OGP groups (p=0.0198). Histological examination showed a higher percentage of placental changes in the OGP group (70%) than in the OGCH group, such as the presence of macrophages, neutrophils, plasma cells, and multifocal areas of calcification. These results do not corroborate the hypothesis of dissemination of oral microorganisms to the placental unit, suggesting that it constitutes placental isolation in sheep.

A periodontite afeta os tecidos de suporte dos dentes levando à perda dentária e danos à saúde do animal. Evidências em humanos sugerem que os microrganismos orais se espalham sistemicamente, aumentando o risco de distúrbios da gravidez, como aborto espontâneo, prematuridade e baixo peso ao nascer. Este estudo teve como objetivo verificar se microrganismos periodontopatogênicos atingem a unidade transplacentária, culminando em problemas em ovelhas gestantes. Após análise da cavidade oral, foram selecionadas 10 ovelhas gestantes clinicamente saudáveis (grupo OGCH) e 10 ovelhas gestantes com periodontite (grupo OGP). O biofilme subgengival foi coletado para o teste de reação em cadeia da polimerase (PCR) e o líquido amniótico para teste de PCR e análise de interleucina (IL). Sangue periférico foi coletado para hemograma completo e análises de IL-6, IL1-β e fator de necrose tumoral-α foram realizadas. Fragmentos de placenta foram coletados para avaliação das alterações inflamatórias por meio de microscopia óptica. Após o parto, as ovelhas e seus cordeiros foram pesados. Ao exame clínico, observou-se correlação positiva entre sangramento e supuração (índice de correlação – IC=0,54), supuração e gengivite marginal (IC=0,34) e gengivite marginal e edema (IC=0,54). Os pesos das ovelhas (p=0,013) e de seus respectivos cordeiros (p=0,04) do grupo OGP foram inferiores aos do grupo OGCH. A análise hematológica revelou que as ovelhas do grupo OGP apresentaram discreto aumento no volume corpuscular médio (p=0,2447), células segmentadas (p=0,3375) e eosinófilos (p=0,3823) quando comparadas com as ovelhas do grupo OGCH, sem diferença estatística diferença. Em relação aos microrganismos detectados na cavidade oral, houve diferença significativa entre a ocorrência de bolsas periodontais e a presença de Fusobacterium necrophorum (p=0,0328), Porphyromonas asaccharolytica (p=0,0392) e da classe Mollicutes (p=0,0352). O gênero Staphylococcus (p=0,9107) e o domínio Archaea (p=0,7245) foram detectados nas amostras amnióticas de ambos os grupos, sem diferença significativa, enquanto P. asaccharolytica (p=0,2685) foi detectado apenas em uma amostra do grupo OGCH. A expressão da citocina IL-6 no grupo OGP diferiu significativamente entre os períodos pré e pós-parto (p=0,0039); além disso, diferiu significativamente no período pós-parto entre os grupos OGCH e OGP (p=0,0198). O exame histológico mostrou maior porcentagem de alterações placentárias no grupo OGP (70%) do que no grupo OGCH, como a presença de macrófagos, neutrófilos, plasmócitos e áreas multifocais de calcificação. Esses resultados não corroboram a hipótese de disseminação de microrganismos orais para a unidade placentária, sugerindo que se trata de um isolamento placentário em ovinos.


#3 - Effect of flunixin or ketoprofen in caudectomy by elastration in lambs: pain and neutrophil function

Abstract in English:

Painful procedures can affect the function of innate immune cells, such as neutrophils and macrophages, increasing the risk of infectious diseases. The present work aimed to verify if the analgesics flunixin meglumine or ketoprofen can attenuate the pain/discomfort of newborn lambs submitted by elastration tail docking and thereby avoid the impairment of blood granulocytes function. Twenty-one neonate lambs were divided into three treatments: the control group (n=7), not subjected to caudectomy; the flunixin group (n=7), subjected to caudectomy under local anesthesia and analgesia with two doses of flunixin meglumine; and the ketoprofen group (n=7), subjected to caudectomy under local anesthesia and two doses of ketoprofen. Pain indicators were observed by pain posture score (PS), the number of vocalizations (V), frequency of the movement of the ears (EF), and respiratory rates (RR), observed by a 10 minutes videos for each time points: -15min, 6h, 48h, and 144h. At the same time points, the reactive oxygen species (ROS) production and phagocytosis of blood granulocytes were measured by flow cytometry. At 6h after caudectomy, there was a pain indicator increase (RR, V, and PS), a blood granulocyte percentage increase, and a granulocytes phagocytosis reduction for both groups. At 48h, the ketoprofen group spend more time in pain posture and, at 144h, they exhibited a ROS production granulocyte reduction without signs of pain. We conclude the flunixin meglumine and ketoprofen did not prevent the acute pain/discomfort caused by caudectomy, because the groups showed a pain behavior and impaired of the innate immune response however, the flunixin meglumine was effective in controlling the chronic pain and their effects on blood granulocytes function in compare ketoprofen.

Procedimentos dolorosos podem afetar a função das células imunes inatas como neutrófilos e macrófagos, aumentando o risco de ocorrer doenças infeciosas. Desta maneira, o presente trabalho pretendeu verificar se os analgésicos flunixin meglumine ou cetoprofeno conseguem atenuar a dor/desconforto de cordeiros neonatos submetidos a caudectomia por elastração, evitando assim o comprometimento da função dos granulócitos sanguíneos. Para tanto 21 cordeiras foram aleatoriamente divididas em três tratamentos: grupo controle (n=7) não submetido a caudectomia, grupo flunixin (n=7) submetido a caudectomia precedida por anestesia local e duas doses de flunixin meglumine, e cetoprofeno (n=7) submetido a caudectomia precedida por anestesia local e duas doses de cetoprofeno. Mensurou-se os indicadores de dor/desconforto após caudectomia por observações de escore de postura de dor (EP), número de vocalizações (V), frequência de movimentar as orelhas (FO) e frequência respiratória (FR), observados em vídeos de 10 minutos nos momentos -15 min e 6, 48 e 144h. Nos mesmos momentos, avaliou-se a as funções de produção de espécies reativas de oxigênio (ERO) e de fagocitose por granulócitos sanguineos em citometria de fluxo. Notou-se aumento dos indicadores de dor (FR, de V e de EP), da porcentagem dos granulócitos sanguíneos e redução da eficiência de fagocitose em ambos os grupos as 6h. As 48h, os animais do grupo cetoprofeno ainda apresentava mais tempo em postura de dor que os demais grupos e as 144h, apresentou redução da produção ERO por granulócitos. Tais achados permitem concluir que tanto o flunexin meglumine como o cetoprofeno não preveniram dor/desconforto agudo promovido pela elastração, pois os dois grupos manifestaram comportamento de dor e redução da resposta imune inata. Ainda, o analgésico flunixin meglumine foi efetivo em controlar a dor mais tardia e seus efeitos na função de granulócitos sanguíneos em comparação ao cetoprofeno.


#4 - Eimeria spp. infection in lambs from southern Brazil

Abstract in English:

Eimeria infections are common in the sheep industry worldwide. Lambs are more susceptible to coccidiosis, especially in stressful conditions, being infected by different species of the parasite. Eimeria crandallis and Eimeria ovinoidalis are considered the most pathogenic, causing reduced growth, dehydration, anorexia, and death. In this study, the frequency of Eimeria species was evaluated in lambs from the southern region of the Rio Grande do Sul state, Brazil. Fecal samples from 248 lambs, from 19 farms, were tested for the presence of oocysts. The positive samples were re-examined and the sporulated oocysts analyzed morphometrically to identify the presence of Eimeria species. In 100% of the evaluated farms, there were animals positive for the protozoan. The frequency of Eimeria species was: E. ovinoidalis (94.74%), E. crandallis (89.47%), E. granulosa (78.95%), E. parva (68.42%), E. ahsata (63.13%), E. punctata (42.11%), E. bakuensis (36.84%), E. faurei (10.53%), and E. pallida (5.26%). Mixed infection was found in 94.74% of the samples. This research describes, for the first time, the occurrence of E. crandallis and E. ovinoidalis infecting lambs in the study area. The wide distribution of this protozoan and the high frequency of pathogenic species show the importance and potential damage of sheep coccidiosis in herds from Rio Grande do Sul.

As infecções por Eimeria são comuns na ovinocultura mundial. Cordeiros são mais suscetíveis a coccidiose, especialmente em condições estressantes, sendo infectados por diferentes espécies do parasito. Eimeria crandallis e Eimeria ovinoidalis são consideradas as mais patogênicas, causando redução do crescimento, desidratação, anorexia e morte. Neste estudo, a prevalência de Eimeria spp. foi avaliada em cordeiros da região sul do Estado do Rio Grande do Sul, Brasil. Amostras fecais de 248 cordeiros, provenientes de 19 fazendas, foram testadas quanto à presença de oocistos. As amostras positivas foram reexaminadas e os oocistos esporulados analisados morfometricamente para identificação das espécies de Eimeria presentes. Em 100% das fazendas avaliadas houve animais positivos para o protozoário. A frequência das espécies de Eimeria foi: E. ovinoidalis (94.74%), E. crandallis (89.47%), E. granulosa (78.95%), E. parva (68.42%), E. ahsata (63.13%), E. punctata (42.11%), E. bakuensis (36.84%), E. faurei (10.53%) e E. pallida (5.26). Infecção mista foi encontrada em 94.74% das amostras. Este trabalho descreve pela primeira vez a ocorrência de E. crandallis e E. ovinoidalis infectando cordeiros na área de estudo. Este trabalho descreve pela primeira vez a ocorrência de E. crandallis e E. ovinoidalis infectando cordeiros na área de estudo. A ampla distribuição desse protozoário e a alta frequência das espécies patogênicas evidenciam a importância da coccidiose ovina e os danos potenciais nos rebanhos do Rio Grande do Sul.


#5 - Haematological variables of Santa Ines and Ile de France suckling lambs: Influence of Haemonchus contortus infection

Abstract in English:

Haemonchus contortus is the major gastrointestinal parasite of sheep raised in tropical and subtropical areas worldwide. This trial aimed to evaluate the influence of H. contortus infection on the bone marrow response of Santa Ines (SI) and Ile de France (IF) suckling lambs experimentally infected with H. contortus. Fourteen SI lambs and 12 IF lambs were randomized in four groups: infected SI (n=8), non-infected SI (n=6), infected IF (n=8) and non-infected IF (n=4). Lambs of infected groups were submitted to 27 infections, conducted every two days, from 14 to 68 days of age, and each lamb received a total of 5400 H. contortus infective larvae (L3). Ten blood samples were obtained during the experimental period to be used for erythrocyte and leukocyte counts, packed cell volume and total plasma protein estimation. Additionally, it was carried out a differential leukocyte count. Lambs from control groups did not shed eggs in faeces all over the experiment, while infected Santa Ines and Ile de France lambs presented means of 2963 EPG (Eggs Per Gram of faeces) and 8175 EPG in the last sampling (P<0.05), respectively. Infected Santa Ines lambs had an increase in eosinophil release, however differences (P<0.05) on circulation number in comparison with infected Ile de France lambs were identified only in the last sampling (54 days post first infection). The mild H. cortortus infection did not produce significant changes in the blood variables of the Ile de France and Santa Ines suckling lambs.

Haemonchus contortus é o principal parasita gastrintestinal de ovinos criados em áreas tropicais e subtropicais. Este estudo objetivou avaliar a influência da infecção por H. contortus na resposta medular de cordeiros lactentes das raças Santa Inês (SI) e Ile de France (IF) infectados experimentalmente com H. contortus. Quatorze cordeiros SI e 12 cordeiros IF, foram distribuídos em quatro grupos: SI infectado (n=8), SI não infectado (n=6), IF infectado (n=8) e IF não infectado (n=4). Cordeiros dos grupos infectados foram submetidos a 27 infecções, realizadas a cada dois dias, do 14º até 68º dia de vida, com um total de 5400 larvas infectantes (L3) de H. contortus por animal. Foram realizadas 10 coletas de sangue ao longo do período experimental para determinação do volume globular, proteínas plasmáticas totais, contagem de eritrócitos e leucócitos, além do diferencial de leucócitos. Cordeiros controles não infectados não eliminaram ovos nas fezes durante todo o experimento, enquanto que os cordeiros Santa Ines e Ile de France infectados apresentaram em média 2963 ovos por grama de fezes (OPG) e 8175 OPG na última coleta (P<0,05), respectivamente. Cordeiros Santa Inês infectados tiveram um aumento na produção de eosinófilos, mas diferenças (P<0.05) no número de eosinófilos sanguíneos em comparação a cordeiros Ile de France infectados foram detectadas somente na última coleta (54 dias após a primeira infecção). A infecção leve por H. cortortus não induziu alterações significativas nas variáveis sanguíneas de cordeiros lactentes Ile de France e Santa Inês.


#6 - Response of vital functions, Apgar and cortisol in the prognosis of vigor against neonatal factors of lambs

Abstract in English:

The management of lambs during the neonatal period has been studied in several researches due to the vital and hormonal organic adaptations undergone by the calf after birth. However, gender, number of pups and type of delivery play an important role in understanding neonatal vigor. The study of these groups with the monitoring of clinical evolution and cortisol metabolism becomes an indispensable subsidy for a better understanding of this neonatal phase, aiming to minimize the losses generated. The objective of this study was to evaluate the influence of gender, number of pups and type of delivery in the prognosis of neonatal vigor of lambs through clinical and cortisol diagnosis. Thirty crossbred Santa Inês lambs with Dorper in the neonatal phase were divided into three groups: male and female, number of pups (single and twin) and type of delivery (eutocic and dystocic). In each group, clinical evaluation of heart and respiratory rate, rectal temperature, Apgar score and weight were performed; and with the exception of cortisol, all evaluations were performed at fifteen and sixty minutes, as well as at twelve and twenty-four hours. In addition, blood samples were collected for cortisol dosage obtained in two moments at fifteen and sixty minutes using the radioimmunoassay technique. Among the three experimental groups related to lamb vigor, the heart rate was the only one that showed lower mean values ​​(P<0.05) at twenty-four hours in the male group 90.00±20.20bpm, twins 96.44±20.02bpm and eutocic 93.25±18.11bpm. Differences in respiratory rate values ​​were observed in the eutocic group (64.00±14.75mpm) at twenty‑four hours. In the group of males there was a significant reduction in body temperature during the evaluation moments (P<0.05). Lambs from the group of twins showed lower body weight during the evaluations. At both times the analysis of serum cortisol was less than at sixty minutes. It was concluded that soon after the birth there were marked changes in the physiological parameters and weight of Santa Inês lambs, but were not enough to cause negative effects on the vigor of the neonates, indicating the occurrence of effective neonatal adaptation capacity in this species.

O manejo dos cordeiros durante o período neonatal tem sido objeto de estudo em diversas pesquisas devido às adaptações orgânicas vitais e hormonais sofridas pela cria após o parto. Todavia, o gênero, número de filhotes e o tipo de parto parecem desempenhar um papel importante para melhor compreensão do vigor neonatal. Além disso, o estudo destes grupos com o acompanhamento da evolução clínica e do metabolismo do cortisol torna-se um subsidio indispensável para melhor compreensão dessa fase neonatal, visando minimizar as perdas geradas. Dessa forma, o objetivo do presente trabalho foi avaliar a influência do gênero, número de filhotes e tipo de parto na apresentação do vigor neonatal dos cordeiros através do diagnostico clinico e de cortisol. Foram utilizados trinta cordeiros mestiços da raça Santa Inês com Dorper em fase neonatal divididos em três grupos: gênero (macho e fêmea), número de filhotes (único e gemelar) e tipo de parto (eutócico e distócico). Em cada grupo, foi realizada a avaliação clínica da frequência cardíaca e respiratória, temperatura retal, escore Apgar e peso; e com a exceção do cortisol, todas as avaliações foram realizadas aos quinze e sessenta minutos, como também às doze e vinte e quatro horas. Adicionalmente, procedeu-se com a coleta de amostras de sangue total para dosagem de cortisol obtida em dois momentos aos quinze e sessenta minutos através da técnica de radioimunoensaio. Dentre os três grupos experimentais relacionados com vigor dos cordeiros, a frequência cardíaca foi a única que evidenciou menores médias (P<0,05) às vinte e quatro horas no grupo dos machos 90,00±20,20bpm, gêmeos 96,44±20,02bpm e eutócicos 93,25±18,11bpm. Observou‑se no grupo eutócico diferenças nos valores da frequência respiratória de 64,00±14,75mpm às vinte e quatro horas. No grupo dos machos houve redução significativa na temperatura corpórea durante os momentos de avaliação (P<0,05). Cordeiros do grupo de gêmeos demonstraram menor peso corpóreo durante as avaliações. Em ambos momentos a análise do cortisol sérico demonstrou se menor aos sessenta minutos. Pôde se concluir que logo após o parto ocorreram alterações marcantes nos parâmetros fisiológicos e peso de cordeiros Santa Inês, porém não foram suficientes para causar efeitos negativos sobre o vigor dos neonatos, indicando a ocorrência de efetiva capacidade de adaptação neonatal nesta espécie.

E-mail: mege@fmvz.unesp.br Devido à imaturidade cardíaca, os recém-nascidos apresentam uma morfologia e débito cardíacos diferenciados quando comparado aos adultos. O objetivo deste estudo foi descrever as alterações cardíacas ecocardiográficas e silhueta cardíaca ao exame radiográfico a partir do método VHS (vertebral heart size) em cordeiros recém-nascidos durante o período neonatal. Vinte e oito cordeiros saudáveis da raça Ile de France foram avaliados desde o nascimento até os 35 dias de idade. Ao exame ecocardiográfico avaliou-se em sístole e diástole, os índices relacionados aos ventrículos direito (VD) e esquerdo (VE), a parede livre do ventrículo esquerdo, aorta (Ao), átrio esquerdo (Ae), septo e ponto E, e diâmetros finais. Foi feita a relação entre o átrio esquerdo e aorta, além da fração de ejeção e de encurtamento. O VHS foi mensurado a partir do tamanho dos eixos longo e curto do coração em relação às vértebras torácicas. Os resultados obtidos demonstraram aumento do VE em relação ao VD no decorrer dos 35 dias (p<0,05). Os índices calculados e a relação AE:Ao não tiveram diferença. Os valores de VHS tiveram distribuição normal com média de 9,67 vértebras em todo o período. Os parâmetros radiográficos e ecocardiográficos mostraram diferenças significativas entre os momentos analisados. Os valores VHS variaram de 8.4 a 11.2 vertebras. Para evitar que essas alterações sejam erroneamente interpretadas como cardiopatias na espécie, este estudo demonstra às peculiaridades do desenvolvimento cardíaco neonatal em cordeiros, além de descrever parâmetros para esta faixa etária em cordeiros da raça Ilê de France.

E-mail: mege@fmvz.unesp.br Due to cardiac immaturity, newborns have a different morphology and cardiac output when compared with adults. The purpose of this study was to describe the cardiac changes in echocardiographic characteristics and vertebral heart size (VHS) in newborn lambs during the neonatal period. Twenty-eight healthy Ile de France lambs were evaluated from birth to 35 days old. With the assistance of echocardiography, this study evaluated, both in systole and diastole, indexes related to the right and left ventricles, the aorta (Ao), the left atrium (LA), the E point to septal separation (EPSS) and the thickness of the interventricular septum (VSd, VSs). The left atrium-to-aorta ratio (LA:Ao) was analyzed and both the shortening (SF%) and ejection fractions were calculated (EF%). The VHS was obtained by measuring the heart’s long and short axes and comparing it to the thoracic vertebrae. Throughout the first 35 days, the results obtained demonstrated an increased LV:RV ratio when compared to adult lambs. The calculated indexes and the LA:Ao ratio did not present statistically significant differences. The VHS values were normally distributed, presenting a mean of 9.67 vertebrae (v) during the entire period. Both radiographic and echocardiographic parameters showed significant differences between the analyzed moments. The VHS values ranged from 8.4v at the minimum end to 11.2v at the maximum. In order to prevent these changes from being misdiagnosed as heart diseases, this study contributed to take note of neonatal physiological anatomy peculiarities and in addition, it describes the parameters for this age in Ilê de France lambs.

E-mail: mege@fmvz.unesp.br Newborn animals, in the fetal-to-neonatal transition, usually face several challenges in their first 24 hours, including issues with acid-base balance, glycemic levels and oxygenation. Difficulties to overcome such issues have caused several deaths among newborns. Therefore, studies have been carried out in order to evaluate them. The main purpose of this study is to evaluate the correlation between the serum lactate level in the mother, in the placenta and in the newborn. Moreover, the study measured the lactate level, blood glucose level and blood gas level in the first 24 hours. Tests were carried out right after birth, and at 4, 8, 12 and 24 hours after. Lactate levels were quite similar to the placental levels (p=0.991) which, in turn, were significantly different from the mother’s (p=0.011). Results showed that, shortly after birth, the production of lactate in the placenta is part of the issue. Along the first 24 hours, the study observed a reduction of the levels of lactate in newborns; the levels were closer to the normal index levels for the species. Regarding the blood gas test results, we observed mild metabolic acidosis at birth; acid-base balance was completely stable at the end of the period.

Falhas nesse sistema são a maior causa de morte nesse período e os estudos de viabilidade neonatal tem se concentrado nas avaliações destas variáveis. O objetivo do presente estudo foi avaliar a comparação da lactatemia materna, placentária e neonatal ao nascimento e, ainda, observar o comportamento do lactato, glicemia e hemogasometria nas primeiras 24 horas de vida do neonato ovino. Para tanto, as avaliações foram realizadas ao nascimento, 4, 8, 12 e 24 horas após. A lactatemia neonatal se mostrou próxima à placentária (p=0,991) e significativamente diferente da materna (p=0,011), o que sugere uma influência da produção de lactato pela placenta na lactatemia neonatal ao nascimento. Ao longo das 24 horas de vida foi possível observar a depuração de lactato pelo organismo do neonato, aproximando assim dos valores de referência para a espécie. Na hemogasometria foi possível observar discreta acidose metabólica, com normalização já nas primeiras 4 horas e estabilização completa do equilíbrio ácido-básico ao final das 24 horas de vida.

E-mail: martalizandra@gmail.com To evaluate the effect of supplementation of iron dextran on blood variables and iron metabolism in lambs experimentally infected by Haemonchus contortus, four experimental groups were used: uninfected and non-supplemented animals (GI); infected animals supplemented with iron (GII); uninfected animals supplemented with iron (GIII); and infected non-supplemented animals (GIV). Groups II and IV received 10,000 larvae (L3) of Haemonchus contortus, and groups II and III received three doses of iron dextran (20mg/kg) intramuscularly with seven days of interval. Blood and faeces samples were collected on days 10 (D10), 17 (D17), 24 (D24), and 31 (D31), in order to determine red blood cell counts, iron metabolism, and EPG. Infected animals developed anemia from D24 and anemia was more severe on D31. Animals from GII had higher hematocrit and hemoglobin concentration compared to animals of GIV on D31. Iron stores in the bone marrow were higher in GII and GIII compared to GI and GIV. The GIV showed lower seric levels of iron on D24 compared to the other groups. The iron supplementation reduces the severity of the anemia caused by infection with Haemonchus contortus in lambs, improving erythropoietic response after blood loss.

#10 - Urinary fractional excretion of sodium, potassium and chloride in lambs supplemented with ammonium chloride to prevent urolithiasis

Abstract in English:

Urolithiasis is an important disease of lambs confined. The urine acidification, by ammonium chloride intake, is the preventive method most frequently employed. Due to the lack of specific information in sheep, this study was performed to evaluate the electrolyte changes that occur in the urine of lambs receiving ammonium chloride in the diet. One hundred male lambs, 3 months old, were kept in a feedlot during 56 days, and distributed in 3 groups: G1 (n=40) receiving 400mg/kg BW of ammonium chloride/day during 21 days; G2 (n=40) receiving 400mg/kg BW of ammonium chloride/day during 42 days; and G3 (n=20) that did not receive ammonium chloride. The lambs were examined and blood and urine samples were collected every 7 days: 0 (the beginning of ammonium chloride intake), 7, 14, 21, 28, 35, and 42 days. Serum and urine sodium (Na+), potassium (K+), chloride (Cl-), and creatinine concentrations were measured. The urinary fractional excretion (FE) of electrolytes and the urine strong ion difference [(Na+ + K+) - Cl-] were calculated. FEs of Na+, K+, and Cl- did not vary over time in G3, proving that the feedlot diet, by itself, did not influence the urinary excretion of these electrolytes. The ingestion of ammonium chloride, instead, influenced FEs over the time of feedlot. The urinary SID was more accurate than the FE of Cl- to demonstrate that the concentration of Cl- increased in the urine. It highlights the relevance of this variable.

A urolitíase é uma doença importante de cordeiros confinados. A acidificação da urina, pela ingestão de cloreto de amônio, é o método preventivo mais frequentemente empregado. Devido à falta de informação específica em ovinos, este estudo foi realizado para avaliar as alterações que ocorrem nos eletrólitos urinários de cordeiros, que receberam cloreto de amônio na dieta. Foram utilizados 100 cordeiros, com 3 meses de idade, que foram mantidos em confinamento durante 56 dias, e distribuídos em 3 grupos: G1 (n=40) que receberam 400mg/kg de peso vivo (PV) de cloreto de amônio/dia, durante 21 dias; G2 (n=40) que receberam 400mg/kg de PV de cloreto de amônio/dia durante 42 dias; e G3 (n=20) que não receberam cloreto de amônio. Os cordeiros foram examinados e as amostras de sangue e urina foram colhidas a cada 7 dias: 0 (antes do início da ingestão de cloreto de amônio), 7, 14, 21, 28, 35, e 42 dias. As concentrações séricas e urinárias de sódio (Na+), potássio (K+), cloreto (Cl-), e de creatinina foram mensuradas em todos os momentos de colheita. A excreção fracionada urinária (EFu) de eletrólitos e a diferença de íons fortes (SID) na urina [(Na+ + K+) - Cl] foram calculadas. A EFu de Na+, K+ e Cl- não variou ao longo do tempo em G3, provando que a dieta de confinamento, por si só, não influenciou a excreção urinária destes eletrólitos. A ingestão de cloreto de amônio, pelo grupo G1 e G2, influenciou a EFu sobre o tempo de confinamento. A SID urinária foi mais precisa do que a EFu de Cl- para demonstrar que a concentração de Cl- aumentou na urina, o que destacou a relevância desta variável.
