#1 - Monitoring periodontal lesions and their effects during pregnancy: microbiological aspects of the oral cavity and amniotic fluid in pregnant ewes

Periodontitis affects the teeth supporting tissues, leading to tooth loss and damage to animal health. Evidence in humans suggests that oral microorganisms spread systemically, increasing the risk of pregnancy disorders such as miscarriage, prematurity, and low birth weight. This study aimed to verify whether periodontopathogenic microorganisms reach the transplacental unit, culminating in problems in pregnant ewes. After analyzing the oral cavity, 10 clinically healthy pregnant ewes (OGCH group) and 10 pregnant ewes with periodontitis (OGP group) were selected. The subgingival biofilm was collected for the polymerase chain reaction (PCR) test and amniotic fluid for both the PCR and interleukin (IL) analysis. Peripheral blood was collected for complete blood count, and analyses of IL-6, IL1-β, and tumor necrosis factor-α were performed. Placental fragments were collected to assess the inflammatory changes using optical microscopy. After giving birth, both the ewes and their lambs were weighed. On clinical examination, a positive correlation between bleeding and suppuration (correlation index – CI=0.54), suppuration and marginal gingivitis (CI=0.34), and marginal gingivitis and edema (CI=0.54) was observed. The weights of the ewes (p=0.013) and their respective lambs (p=0.04) in the OGP group were lower than those of their OGCH group counterparts. The hematological analysis revealed that the OGP group ewes showed a slight increase in the mean corpuscular volume (p=0.2447), segmented cells (p=0.3375), and eosinophils (p=0.3823) when compared with the OGCH group ewes, without a statistical difference. Regarding the microorganisms detected in the oral cavity, there was a significant difference between the occurrence of periodontal pockets and the presence of Fusobacterium necrophorum (p=0.0328), Porphyromonas asaccharolytica (p=0.0392), and the Mollicutes class (p=0.0352). Staphylococcus genus (p=0.9107) and Archaea domain (p=0.7245) were detected in the amniotic samples of both groups, without a significant difference, whereas P. asaccharolytica (p=0.2685) was only detected in one sample in the OGCH group. The expression of cytokine IL-6 in the OGP group differed significantly between the prepartum and postpartum periods (p=0.0039); moreover, it differed significantly in the postpartum period between the OGCH and OGP groups (p=0.0198). Histological examination showed a higher percentage of placental changes in the OGP group (70%) than in the OGCH group, such as the presence of macrophages, neutrophils, plasma cells, and multifocal areas of calcification. These results do not corroborate the hypothesis of dissemination of oral microorganisms to the placental unit, suggesting that it constitutes placental isolation in sheep.

A periodontite afeta os tecidos de suporte dos dentes levando à perda dentária e danos à saúde do animal. Evidências em humanos sugerem que os microrganismos orais se espalham sistemicamente, aumentando o risco de distúrbios da gravidez, como aborto espontâneo, prematuridade e baixo peso ao nascer. Este estudo teve como objetivo verificar se microrganismos periodontopatogênicos atingem a unidade transplacentária, culminando em problemas em ovelhas gestantes. Após análise da cavidade oral, foram selecionadas 10 ovelhas gestantes clinicamente saudáveis (grupo OGCH) e 10 ovelhas gestantes com periodontite (grupo OGP). O biofilme subgengival foi coletado para o teste de reação em cadeia da polimerase (PCR) e o líquido amniótico para teste de PCR e análise de interleucina (IL). Sangue periférico foi coletado para hemograma completo e análises de IL-6, IL1-β e fator de necrose tumoral-α foram realizadas. Fragmentos de placenta foram coletados para avaliação das alterações inflamatórias por meio de microscopia óptica. Após o parto, as ovelhas e seus cordeiros foram pesados. Ao exame clínico, observou-se correlação positiva entre sangramento e supuração (índice de correlação – IC=0,54), supuração e gengivite marginal (IC=0,34) e gengivite marginal e edema (IC=0,54). Os pesos das ovelhas (p=0,013) e de seus respectivos cordeiros (p=0,04) do grupo OGP foram inferiores aos do grupo OGCH. A análise hematológica revelou que as ovelhas do grupo OGP apresentaram discreto aumento no volume corpuscular médio (p=0,2447), células segmentadas (p=0,3375) e eosinófilos (p=0,3823) quando comparadas com as ovelhas do grupo OGCH, sem diferença estatística diferença. Em relação aos microrganismos detectados na cavidade oral, houve diferença significativa entre a ocorrência de bolsas periodontais e a presença de Fusobacterium necrophorum (p=0,0328), Porphyromonas asaccharolytica (p=0,0392) e da classe Mollicutes (p=0,0352). O gênero Staphylococcus (p=0,9107) e o domínio Archaea (p=0,7245) foram detectados nas amostras amnióticas de ambos os grupos, sem diferença significativa, enquanto P. asaccharolytica (p=0,2685) foi detectado apenas em uma amostra do grupo OGCH. A expressão da citocina IL-6 no grupo OGP diferiu significativamente entre os períodos pré e pós-parto (p=0,0039); além disso, diferiu significativamente no período pós-parto entre os grupos OGCH e OGP (p=0,0198). O exame histológico mostrou maior porcentagem de alterações placentárias no grupo OGP (70%) do que no grupo OGCH, como a presença de macrófagos, neutrófilos, plasmócitos e áreas multifocais de calcificação. Esses resultados não corroboram a hipótese de disseminação de microrganismos orais para a unidade placentária, sugerindo que se trata de um isolamento placentário em ovinos.


#2 - Periodontal diseases and tooth wear in sheep flocks in the State of Goiás, Brazil

Abstract in English:

Dental and periodontal affections consist of health concerns of great importance in sheep flocks, as they affect the health and welfare of animals and represent one of the main causes of premature slaughter. Studies on the occurrence of these conditions in sheep are scarce in Brazil. The present study aimed to evaluate the prevalence of periodontal diseases and tooth wear in sheep flocks in the State of Goiás. For this, a clinical oral examination was conducted on 325 sheep from seven farms in six municipalities of the State. Periodontal lesions, characterized by gingival recession in at least one incisor tooth, were observed in 48.3% (157/325) of the sheep, while in cheek teeth it occurred in 13.2% (43/325) of the evaluated sheep. Dental wear on cheek teeth was identified in 69.2% (255/325) of the sheep, and 10.7% (35/325) of the ovines showed dental wear on incisor teeth. Sheep older than 36 months had an increase in the frequency of tooth wear. A total of 171 (52.62%) out of the 325 examined sheep had signs of bleeding on probing, which is compatible with gingivitis, and 69 (40.36%) were older than 36 months. Thirty-eight (11.69%) out of the 325 evaluated ovines had gingivitis in all teeth, among which 21 (55.26%) were older than 36 months. Logistic regression analysis allowed the observation that age was associated with the occurrence of gingivitis. Less frequent changes such as periodontal pockets and loss of at least one tooth were also observed in 4.9% (16/325) and 17.8% (58/325) of the sheep, respectively. It indicates that the presence of periodontal diseases and tooth wear is a common occurrence in the evaluated flocks. Therefore, there is a need to include the oral health of sheep in the routine of the farm as an essential preventive measure to improve the quality of life, animal welfare, and zootechnical indices.

As afecções dentárias e periodontais constituem uma preocupação sanitária de grande importância em rebanhos ovinos, uma vez que afetam a saúde e o bem-estar dos animais, e representam uma das principais causas para o abate prematuro. No Brasil, estudos sobre a ocorrência dessas afecções em ovinos são escassos. Assim, o presente estudo teve como objetivo avaliar a prevalência de doenças periodontais e desgaste dentário em rebanhos ovinos no estado de Goiás. Para isso, realizou-se o exame clínico bucal em 325 ovinos em sete propriedades rurais de seis municípios do estado. As lesões periodontais, caracterizadas por recessão gengival em, pelo menos, um dente incisivo, foram observadas em 48,3% (157/325) dos ovinos, enquanto nos mastigatórios ocorreu em 13,2% (43/325) dos ovinos avaliados. O desgaste dentário em dentes mastigatórios foi identificado em 69,2% (255/325) dos ovinos e 10,7% (35/325) dos ovinos apresentaram desgaste dentário em dentes incisivos. Ovinos com idade superior a 36 meses tiveram um aumento na frequência do desgaste dentário. Dos 325 ovinos examinados, 171 (52,62%) apresentaram sinais de sangramento à sondagem, compatíveis com o quadro de gengivite, sendo que 69 (40,36%) apresentaram idade superior a 36 meses. Dos 325 ovinos avaliados, 38 (11,69%) apresentaram gengivite em todos os dentes, dos quais 21 (55,26%) apresentaram idade superior a 36 meses. Pela análise de regressão logística, foi possível observar que a idade mostrou associação com a ocorrência de gengivite. Alterações menos frequentes como a bolsa periodontal e a perda de, pelo menos, um elemento dentário também foram observadas em 4,9% (16/325) e 17,8% (58/325) dos ovinos, respectivamente. Os resultados do presente estudo indicam que a presença doenças periodontais e desgaste dentário é uma ocorrência comum nos rebanhos avaliados. Portanto, enfatiza-se a necessidade de incluir a saúde bucal dos ovinos na rotina da propriedade como uma medida preventiva essencial para melhorar a qualidade de vida e o bem-estar dos animais, bem como os índices zootécnicos.


#3 - Periodontitis in sheep in Pernambuco, Northeastern Brazil

Abstract in English:

Cases of periodontitis in sheep from the Agreste and Zona da Mata regions of Pernambuco, Northeastern Brazil, are described. Information on breed, age and sex of affected animals as well as clinical and pathological aspects of the disease were obtained during farm visits. Fifteen animals from a total of 450 Santa Inês sheep, aged between three and twelve years, showed low body condition score, dull coat, and difficulty in feeding followed by pain on palpation of the mandibular region. Physical examination of the oral cavity of affected animals and post mortem findings of slaughtered individuals revealed varying degrees of periodontitis, wear, fracture, mobility, and loss of teeth associated with gingival retraction, presence of biofilm and food packing between incisors, premolars and molars teeth. Increased mandibular volume was observed in eight animals and, in two of them, an abscess fistula, exclusively in mandibular teeth. The occurrence of periodontitis in sheep had not been recorded until now in Northeastern Brazil and, thus, this is the first description of cases in the region.

Descrevem-se casos de periodontite em ovinos provenientes das regiões do Agreste e Zona da Mata de Pernambuco, Nordeste do Brasil. Informações sobre raça, idade e sexo dos animais afetados bem como os aspectos clínicos e patológicos dos surtos foram obtidos a partir de visitas às propriedades e são reportados. 15 animais de um total de 450 ovinos da raça Santa Inês, com idade entre três a doze anos de idade manifestaram baixa condição corporal, pelagem sem brilho e dificuldade de alimentar-se, acompanhado de dor à palpação na região mandibular. O exame físico da cavidade oral dos animais afetados e os achados post mortem de indivíduos abatidos revelaram graus variados de periodontite, desgaste, fratura, mobilidade e perda dentária, associados a retração gengival, presença de biofilme e compactação de resíduos alimentares entre os dentes incisivos, pré-molares e molares. Em oito animais havia aumento de volume mandibular e em dois deles observaram-se abscessos fistulados que ocorreram exclusivamente nos dentes mandibulares. A ocorrência de periodontite em ovinos não têm sido registrada no Nordeste do Brasil, sendo estes os primeiros casos descritos na região.


#4 - Pathology of chronic ovine periodontitis

Periodontitis is an inflammatory process of infectious origin affecting the teeth and their supporting structures, causing significant economic losses and reducing animal welfare. Bacteria in the gingival biofilm are one of the main factors in initiating inflammatory lesions. Bacteria act directly on tissues or indirectly through substances that cause tissue damage. Studies on the etiopathogenesis of periodontitis in Brazilian sheep herds are scarce. The present study aimed to characterize histologically periodontal lesions of culled sheep from the Brazilian breed, Santa Inês. Periodontal lesions, such as periodontal pockets containing plant tissue and bacteria, replacement of the periodontal ligament by connective tissue and inflammatory cells, superficial pustules, hydropic epithelial degeneration, and epithelium hyperplasia, were observed. Submucosal changes were characterized by granulation tissue, edema, swelling of the endothelial cells, bacteria, and predominantly perivascular lymphoplasmacytic inflammatory infiltrate. In the alveolar bone, osteoclastic resorption and bone apposition were observed. This study revealed subacute to chronic inflammation, alveolar bone resorption, and cortical bone apposition in ovine periodontitis. Thus, these findings can contribute to the evolution of knowledge about the etiopathogenesis of ovine periodontitis and, possibly, the development of measures to control the disease.

A periodontite é um processo inflamatório de origem infecciosa que afeta os dentes e suas estruturas de suporte, causando perdas econômicas significativas e redução do bem-estar animal. As bactérias do biofilme gengival são um dos principais fatores envolvidos no início das lesões inflamatórias. As bactérias agem diretamente nos tecidos ou indiretamente por meio de substâncias que causam dano tecidual. Estudos sobre a etiopatogenia da periodontite em rebanhos ovinos brasileiros são escassos. O presente estudo teve como objetivo caracterizar histologicamente as lesões periodontais de ovinos de descarte da raça brasileira Santa Inês. Foram observadas lesões periodontais, como bolsas periodontais contendo tecido vegetal e bactérias, substituição do ligamento periodontal por tecido conjuntivo e células inflamatórias, pústulas superficiais, degeneração hidrópica epitelial e hiperplasia do epitélio. As alterações da submucosa foram caracterizadas por tecido de granulação, edema, tumefação das células endoteliais, bactérias e infiltrado inflamatório linfoplasmocitário predominantemente perivascular. No osso alveolar, observou-se reabsorção osteoclástica e aposição óssea. Este estudo revelou inflamação subaguda a crônica, reabsorção óssea alveolar e aposição de osso cortical na periodontite ovina. Assim, esses achados podem contribuir para a evolução do conhecimento sobre a etiopatogenia da periodontite ovina e para o desenvolvimento de medidas de controle da doença.


#5 - Periodontal disease and its complications in cats from a shelter in the Central Region of Rio Grande do Sul

Abstract in English:

Periodontal disease is the most common infectious disease that occurs in feline patients. Although it occurs in both sexes, different age groups, and any breeds, the prevalence and severity seem much higher in cats living in animal shelters. This paper aimed to describe the clinical, radiological, cytopathological, and virological aspects of periodontal disease and its complications in cats, based on these aspects and, consequently, on the importance it brings to cat feline medicine in shelter cats. For this, nine cats with periodontal disease from a single animal shelter were evaluated. These cats demonstrated a disease characterized by halitosis, excessive salivation, and oral discomfort. Lymphadenomegaly of the mandibular and retropharyngeal lymph nodes was observed in 44.4% of the cases. Oral lesions consisted of varying degrees of gingival hyperemia, complete loss of free gingival margins, and consequently gingival retraction, dental calculus deposition, dental mobility, complete exposure of the furcation of premolars and molars, and dental roots of canines and incisors, loss of bone radiopacity due to alveolar bone resorption and tooth loss. Complications included chronic ulcerative paradental stomatitis (22.2%), faucitis (22.2%), and chronic gingivostomatitis (11.1%). None of the cats affected by periodontal disease was positive for FIV or FeLV. In 33.3% of the cases, cats were carriers of feline calicivirus, but not feline herpesvirus.

Doença periodontal é a mais comum doença infecciosa que ocorre em pacientes felinos. Embora ocorra em gatos de ambos os sexos, diferentes faixas etárias e quaisquer raças, a prevalência e a gravidade parece muito maior em gatos que vivem em abrigos para animais. Baseado nesses aspectos e, consequentemente, na importância que ela traz para a medicina felina de gatos de abrigos, o objetivo desse artigo é descrever os aspectos clínicos, radiológicos, citopatológicos e virológicos da doença periodontal e suas complicações em gatos. Para isso, nove gatos com doença periodontal oriundos de um único abrigo de animais foram avaliados. Esses gatos demonstraram uma doença caracterizada por halitose, salivação excessiva e desconforto oral. Linfadenomegalia dos linfonodos mandibulares e retrofaríngeos foi observada em 44,4% dos casos. As lesões orais consistiam de graus variados de hiperemia gengival, perda completa das margens gengivais livres e, consequentemente, retração gengival, deposição de cálculo dental, mobilidade dentária, exposição completa da furca dos pré-molares e molares e das raízes dentárias dos caninos e incisivos, perda de radiopacidade óssea devido à reabsorção de osso alveolar e perda dentária. Complicações incluíram estomatite paradental ulcerativa crônica (22,2%), faucite (22,2%) e gengivoestomatite crônica (11,1%). Nenhum dos gatos afetados pela doença periodontal foi positivo para FIV ou FeLV. Em 33,3% dos casos, os gatos eram portadores do calicivírus felino, mas não do herpesvírus felino.


#6 - Chemical and structural composition of black pigmented supragingival biofilm of bovines with periodontitis

Abstract in English:

Bovine periodontitis is a multifactorial disease primarily associated with a potentially pathogenic microbiota housed in the oral biofilm of animals. Biofilms are organized structures, in which the constituents coexist in symbiosis, already described as a predisposing factor to periodontitis in other species. The objective of the present study was to characterize the structure and chemical aspects of the bovine black pigmented supragingival biofilm using scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS), respectively, and determine its relationship with bovine periodontitis. Eleven premolar teeth from different animals were evaluated; five non-pigmented samples and six samples with black pigmented biofilms were initially submitted to SEM, and three areas of these samples were selected for EDS. The structure of the pigmented biofilm was more complex and irregular because of a higher content of mineral elements. The semi-quantitative EDS data indicated an association of iron (p<0.014) and magnesium (p<0.001) with the occurrence of periodontitis, whereas carbon, phosphorus, calcium, manganese, sodium, and potassium were not associated with the disease. Carbon (p<0.039), manganese (p<0.007), and iron (p<0.015) were associated with pigmentation, whereas phosphorus, calcium, and magnesium were not associated with it. Spearman correlation test showed the relationships between calcium and phosphorus, and iron and silicon. The strong association of iron in the pigmented supragingival biofilm and with the occurrence of periodontitis suggests the presence of microorganisms that use this element in their metabolism and that are also associated with bovine periodontitis. This study suggests that the pigmented deposits in the crown of the teeth of cattle are an true biofilm with the deposition of iron, and it indicates that the presence of iron and magnesium in these formations may be involved in the metabolism of some microorganisms associated with the etiology of bovine periodontitis.

A periodontite bovina é uma infecção multifatorial associada primariamente à microbiota potencialmente patogênica presente no biofilme bucal. Biofilmes são estruturas organizadas, nas quais os constituintes convivem em simbiose, descritos em outras espécies como um fator predisponente à periodontite. O objetivo do presente estudo foi caracterizar estrutural e quimicamente o biofilme supragengival pigmentado de preto em bovinos, utilizando-se as técnicas de microscopia eletrônica de varredura (MEV) e espectroscopia de dispersão de energia (EDS), respectivamente, correlacionando os elementos identificados à ocorrência de periodontite e pigmentação. Foram avaliados 11 dentes primeiro-molares; cinco amostras sem pigmentação visível e seis amostras com biofilme pigmentado de preto, que foram submetidas inicialmente à MEV; posteriormente foram selecionadas três áreas aleatórias de cada dente para realização da EDS. A estrutura do biofilme pigmentado revelou formações irregulares e mais complexas, provavelmente devido ao maior acúmulo de elementos minerais. Os resultados semi-quantitativos da EDS apontaram associações entre a presença de ferro (p<0,014) e magnésio (p<0,001) com a ocorrência de periodontite. Carbono, fósforo, cálcio, manganês, sódio e potássio não apresentaram associação com a periodontite. Em relação à pigmentação, carbono (p<0,039), manganês (p<0,007) e ferro (p<0,015) foram os elementos estatisticamente significantes, enquanto fósforo, cálcio e magnésio não apresentaram associação com a pigmentação. O teste de correlação de Spearman demonstrou associações entre os elementos cálcio e fósforo, e ferro e silício. A forte associação do ferro presente no biofilme supragengival com a ocorrência de periodontite, sugere a presença de micro-organismos que utilizam este elemento em seu metabolismo e que possivelmente tenham envolvimento com o desenvolvimento da periodontite bovina. Os resultados inéditos do presente trabalho sugerem que os depósitos pigmentados que se formam na coroa dos dentes de bovinos são um biofilme verdadeiro com deposição de ferro, e indicam que a presença de ferro e magnésio nestas formações pode estar envolvida no metabolismo de alguns dos principais micro-organismos associados à etiologia da periodontite bovina.


#7 - Subgingival bacterial microbiota associated with ovine periodontitis

Abstract in English:

Periodontitis is an inflammatory response in a susceptible host caused by complex microbiota, predominantly composed of Gram-negative anaerobic bacteria. Aiming to characterize the subgingival bacterial microbiota associated with ovine periodontitis, polymerase chain reaction (PCR) was performed in subgingival periodontal pocket samples of 14 sheep with severe periodontitis and in subgingival sulcus biofilm of 14 periodontally healthy sheep in search mainly of Gram-negative and Gram-positive microorganisms considered important periodontopathogens. The most prevalent bacteria in the sheep with periodontal lesions were Tannerella forsythia (78.6%), Treponema denticola (78.6%), Fusobacterium nucleatum (64.3%), and Porphyromonas gingivalis (50%), whereas in the healthy sheep, F. nucleatum (42.8%) was the most often detected bacterium. Statistically significant differences were observed for Campylobacter rectus, Enterococcus faecium, Prevotella nigrescens, T. forsythia, and T. denticola (p<0.05) in the sheep with periodontitis in the comparison between groups. Aggregatibacter actinomycetemcomitans, Enterococcus faecalis, and Porphyromonas gulae were not detected in any of the samples analyzed. In conclusion, C. rectus, E. faecium, P. nigrescens, T. forsythia, and T. denticola were associated with severe lesions caused by ovine periodontitis, and F. nucleatum was the most prevalent microorganism in the subgengival sulcus biofilm of healthy sheep.

Periodontite é a resposta inflamatória de um hospedeiro suscetível causada por complexa microbiota, composta predominantemente por bactérias anaeróbias Gram-negativas. Com o objetivo de caracterizar a microbiota bacteriana subgengival associada à periodontite ovina foi realizada a reação em cadeia da polimerase (PCR) de amostras de biofilme subgengival de 14 ovinos com a enfermidade e 14 ovinos periodontalmente saudáveis, com destaque para micro-organismos Gram-negativos e Gram-positivos considerados importantes periodontopatógenos. As bactérias mais prevalentes em 14 animais com lesões periodontais foram Tannerella forsythia (78,6%), Treponema denticola (78,6%), Fusobacterium nucleatum (64,3%) e Porphyromonas gingivalis (50%). Entretanto, nos 14 ovinos sem lesões periodontais, F. nucleatum (42,8%) foi a bactéria mais detectada. Associação estatisticamente diferente foi observada para Campylobacter rectus, Enterococcus faecium, Prevotella nigrescens, T. forsythia e T. denticola (p<0,05) nos ovinos com periodontite em comparação entre os dois grupos. Aggregatibacter actinomycetemcomitans, Enterococcus faecalis e Porphyromonas gulae não foram detectados em nenhuma das amostras pesquisadas. Conclui-se que C. rectus, E. faecium, P. nigrescens, T. forsythia e T. denticola estão associados às lesões resultantes da periodontite ovina com manifestação clínica grave e F. nucleatum o micro-organismo mais prevalente no biofilme subgengival de animais periodontalmente sadios.

The first clinical signs of this disease in the herd – visible nodular swollen mandible - were observed about one month after the sheep began grazing on a recently renovated Panicum maximum cv. Massai pasture; the animals were also supplemented with elephant grass (Pennisetum purpureum). From the 545 adult Santa Ines, Dorper, Texel and crossbred sheep, 20 (3.7%) showed facial bulging, particularly of the mandible. These 20 sheep, which were generally over 36 months of age, had low body condition scores as well as dull and ruffled coats; many of them had loose or missing pre, upper or lower molars, showed the formation of abscesses and fistulas, exhibited pain upon palpation and had difficulty chewing. In the same herd, the prevalence of periodontal lesions at the premolars and molar teeth was evaluated via post-mortem examination of 39 young and 17 adult sheep. In 51.3% (20/39) of adolescents and in 100% of adults, periodontal lesions were found at least at one tooth of the dental arch, with unilateral or bilateral periodontal lesions at the maxillary and mandibular teeth. Histopathological analysis of 13 sheep revealed pyogranulomatous inflammation. The second and third maxillary premolars were the teeth that were most affected in young animals, whereas the third pre-molar and the molars, of the maxilla and mandible, were most often affected in adults. These lesions led to severe bone destruction, alterations in the dental arch, occlusion, tooth loss and periodontal abscesses. The epidemiological and clinic-pathological characteristics of this periodontitis outbreak, described here for the first time in sheep in Brazil, were similar to those for bovine periodontitis (“cara inchada”).

De 545 ovinos adultos, das raças Santa Inês, Dorper, Texel e seus mestiços, 20 (3,7%) apresentaram abaulamento facial, predominantemente mandibular. Os 20 animais, na sua maioria com idades acima dos 36 meses, apresentavam baixo escore corporal, pelos arrepiados e sem brilho, alguns com afrouxamento e perda dos dentes pré-molares e molares inferiores e superiores, formação de abscesso e fístula, demonstrando dor à palpação e dificuldade na mastigação. No mesmo rebanho, a prevalência de lesões periodontais nos dentes pré-molares e molares foi avaliada por meio do exame post-mortem em 39 ovinos jovens e 17 adultos. Assim, 51,3% (20/39) dos jovens e todos os adultos apresentavam lesões periodontais em pelo menos um dos dentes da arcada, com ocorrência uni ou bilateral nos maxilares e mandibulares. A análise histopatológica de 13 animais revelou processo inflamatório piogranulomatoso. Os dentes mais afetados nas arcadas dos animais jovens foram os segundo e terceiro pré-molares maxilares. Já nos adultos foram o terceiro pré-molar e os molares, tanto maxilares quanto mandibulares. As lesões causaram severa destruição óssea, alteração na arcada dentária, na oclusão, perda dental e ocorrência de abscessos periodontais. As características epidemiológicas e clínico-patológicas do surto de periodontite em ovinos, aqui descritos originalmente nesta espécie animal no País, assemelham-se às da periodontite bovina (“cara inchada”).

This study aimed to detect species of the genera Porphyromonas and Prevotella, which occurr in periodontal pockets of cattle with lesions deeper than 5mm (n=26) and in gingival sulcus of animals considered periodontally healthy (n=25). Presence of the microorganisms was evaluated by independent-culture medium diagnostic method, using polymerase chain reaction (PCR) with specific primers of Porphyromonas asaccharolytica, P. endodontalis, P. gingivalis, P. gulae, Prevotella buccae, P. intermedia, P. loescheii, P. melaninogenica, P. nigrescens, P. oralis and P. tannerae. The species P. endodontalis (80.7%), P. melaninogenica (73.1%) and P. intermedia (61.5%) were the most predominant in samples of cattle with periodontitis. Regarding non-injured gingival sulcus of cattle, P. endodontalis (40%) and P. loeschei (40%) prevailed. Porphyromonas gingivalis, P. gulae and Prevotella tannerae were not detected in the 51 samples studied. Data evaluation by T test, enabled to verify that ocorrence of Porphyromonas asaccharolytica (p=0.000003), P. endodontalis (p=0.0023), Prevotella buccae (p=0.0017), P. intermedia (p=0.0020), P. melaninogenica (p=0.00006) and P. oralis (p=0.0028) is correlated with bovine periodontitis.

Já no sulco gengival de bovinos sem lesões prevaleceram P. endodontalis (40%) e P. loeschei (40%). This study aimed to detect Treponema species in periodontal pockets of cattle with lesions deeper than 5mm in the gingival sulcus of 6 to 24-month-old animals considered periodontally healthy. We used paper cones to collect the materials, after removal of supragingival plaques, and kept frozen (at -80°C) up to DNA extraction and polymerase chain reaction (PCR) using T. amylovorum, T. denticola, T. maltophilum, T. medium and T. vincentii primers. In periodontal pocket, it was possible to identify by PCR directly, the presence of Treponema amylovorum in 73% of animals (19/26), T. denticola in 42.3% (11/26) and T. maltophilum in 54% (14/26). Among the 25 healthy sites, it was possible to identify T. amylovorum in 18 (72%), T. denticola in two (8%) and T. maltophilum in eight (32%). Treponema medium and T. vincentii were not detected over all 51 evaluated samples. The presence of Treponema amylovorum, T. maltophilum and, in particular, the widely recognized T. denticola in subgingival microflora brings an original and potencially important contribution in studies of the bovine periodontitis.

Na bolsa periodontal de 73% (19/26) dos animais foi possível detectar diretamente, pela PCR, a presença de Treponema amylovorum, de 42,3% (11/26) T. denticola e de 54% (14/26) T. maltophilum. Dos 25 sítios sadios, em 18 (72%) foi possível identificar T. amylovorum, em dois (8%) T. denticola e em oito (32%) T. maltophilum. Treponema medium e T. vincentii não foram detectados nas 51 amostras avaliadas. A presença de Treponema amylovorum, T. maltophilum na microbiota subgengival, e em especial do amplamente reconhecido periodontopatógeno T. denticola, traz uma contribuição original de importância potencial nos estudos da periodontite bovina.
