E-mail: acynnara@gmail.com The objective of this study was to determine normal standards to ruminal fluid characteristics of Santa Inês sheep under pasture conditions in the State of Pernambuco. Fifty samples were collected, using an esophageal tube, during winter (rainy season) and summer (dry season). The predominant colors were an olive green in the rainy season and a nut-brown in the dry season. The smell was aromatic, but was stronger in winter. A slight viscous consistence was found in most of the samples, with a greater proportion in winter. The sedimentation and flotation time was 6.73min (±1.63) in the rainy period and 3.15min (±0.72) in the dry period. In the biochemical tests, the average values found in winter and summer were, respectively: pH 6.76±0.21 and 6,59±0.14; methylene blue reduction, 3.20min (±0.76) and 7.76min (±3.00); chloride, 28.14±4,16mEq/L and 24.97±5.65mEq/L; acidity, 21.90±4.38UC and 13.68±2.97UC. Ruminal microbiotic analyses revealed abundant and moderate density of protozoa in winter and summer, respectively. The motility was very active and there were almost 90% of live protozoa in both seasons. Protozoa numbers were 425.373±217.258/mL in winter and 155.375±83.113/mL in summer. There was a mixed population of bacteria with prevalence of Gram-negative forms in both seasons.

All the animals presented clinical systemic manifestations, and in the inoculated glands with more intensity from 24 hours on after the inoculation. There were significant alterations (P<0.05) in the production and physical-chemical composition of the milk in relation to the controls, with reduction of volume and fat. The pH, the chlorates and CCS reached very high indexes, accompanied by the CMT test reaction. The bacteriological exam revealed the presence of S. aureus during the phase of infection. After the treatment of the sheep 36 hours PI, one animal came to death 48h PI, and the others showed clinical recovery; however there was no physiologic re-establishment of the inoculated udders which lost their function.

Ten 1 to 2 year old male goats, cross bred (SRD), with 25 kg body weight, in good health condition and housed in a stall, were rumen-fistulated. After establishing normal pattern for the variables studied, the acidosis was induced with the administration of 15g sucrose/kg body weight. Clinical and laboratorial observations were accomplished at intervals of 4, 8, 12, 16, 24, 32, 48, 72, 96, 120 and 144 hours, after the induction (PI). Already 4 hours PI the animals showed clinical signs of apathy, anorexia, increased heart frequency, ruminal atony, abdominal distention and diarrhea. The characteristics of the ruminal fluid suffered alterations, as reduction of the pH (below 5.0), and its color became a milky aspect, acid smell and aqueous consistency. The time of sedimentation activity (TAS) was reduced and the dynamics of the fauna and flora were committed, with the prevalence of Gram-positive bacteria. At the end of the observation period the analyzed variables were not yet fully re-established in some animals.

E-mail: proehe@ufrgs.br Bovine herpesvirus type 1 (BoHV-1) is recognized as a major cause of respiratory, reproductive disease and abortion in cattle. Vaccination is widely applied to minimize losses induced by BoHV-1 infections; however, vaccination of dams during pregnancy with modified live virus (MLV) vaccines has been occasionally associated to abortions. We have previously reported the development of a BoHV-1 recombinant virus, constructed with basis on a Brazilian BoHV-1 (Franco et al. 2002a) from which the gene coding for glycoprotein E (gE) was deleted (gE-) by genetic manipulation. Such recombinant has been previously evaluated in its potential as a differential vaccine (gE- vaccine) that allows differentiation between vaccinated and infected animals. Here, in the first part of the present study, the safety of the gE- vaccine during pregnancy was evaluated by the intramuscular inoculation of 107.4 tissue culture 50 % infective doses (TCID50) of the virus into 22 pregnant dams (14 BoHV-1 seronegative; 8 seropositive), at different stages of gestation. Other 15 pregnant dams were kept as non-vaccinated controls. No abortions, stillbirths or fetal abnormalities were seen after vaccination. Seroconversion was observed in both groups of previously seronegative vaccinated animals. In the second part of the study, the potential of the gE- vaccine virus to spread among beef cattle under field conditions was examined. Four heifers were inoculated intranasally with a larger amount (107,6 TCID50) of the gE- vaccine (to increase chances of transmission) and mixed with other sixteen animals at the same age and body condition, in the same grazing area, at a population density equal to the average cattle farming density within the region (one cattle head per 10,000 m2), for 180 days. All animals were monitored daily for clinical signs. Serum samples were collected on days 0, 30, 60 and 180 post-vaccination. Seroconversion was observed only in vaccinated heifers. These results indicate that, under the conditions of the present study, the gE- vaccine virus did not cause any noticeable harmful effect on pregnant dams and on its offspring and did not spread horizontally among cattle.

#205 - Aspectos epidemiológicos da seneciose na região sul do Rio Grande do Sul, Brasil

E-mail: fernando@alternet.com.br Seneciosis is the main cause of livestock mortality due to poisonous plants in the State of Rio Grande do Sul, Brazil. This paper presents epidemiological data of 24 outbreaks in cattle and one in horses, diagnosed by the Regional Diagnostic Laboratory at Pelotas University in southern Rio Grande do Sul, from 1998 to 2000. Additionally, data of 54 outbreaks which occurred in 1978-1997 were analyzed. The farms where outbreaks occurred in 1998-2000 were visited to obtain clinical and epidemiological data and to verify the presence of Senecio spp. Eleven outbreaks (45.83%) affected cattle up to 3 years of age, and 13 (54.16%) cattle over 3 years. Nine (37.5%) outbreaks affected females and 15 (62.5%) males. Ten (41.66%) outbreaks occurred in spring, 4 (16.66%) in summer, 5 (20.83%) in autumn, and 5 (20.83%) in winter. Morbidity rate was estimated with 4.92% and case fatality with 95.59%. The predomionating Senecio species were S. brasiliensis on 12 farms (57.14%), S. selloi on 10 (47.61%), S. oxyphyllus on 6 (28.57%), S. heterotrichius on 3 (14.28%), and S. leptolobus on 1 farm (4.76%). The main clinical signs were progressive emaciation, incoordination, diarrhea, tenesmus, rectal prolapse and aggressiveness. The clinical manifestation periods of affected cattle observed during the farm visits, or of cattle sent for post-mortem examination,were 24-96 hours in 4 outbreaks (16.66%), 4-7 days in 7 (29.16%), 1-2 weeks in 4 (16.66%), 2-3 weeks in 2 (8.33%), 1-2 months in 2 (8.33%), and 2-3 months in 1 outbreak (4.16%). In 4 outbreaks (16.66%) the clinical manifestation period was not established. In outbreaks with longer clinical manifestation periods some animals showed photosensitization. The main necropsy findings were a hard and enlarged liver, distended gall bladder, edema of the mesenterium and abomasum, and increased amount of liquid in the cavities. Histopathological findings were fibroplasia, megalocytosis and biliary ductal proliferation of the liver, and spongy degeneration of the cerebral white matter. Samples of different Senecio species, in different seasons, were analyzed for detection of pyrrolizidine alkaloids (PAs) by thin layer chromatography. Retrorsine was found in Senecio brasiliensis, S. heterotrichius, S. selloi and S. oxyphyllus. In S. brasiliensis and S. heterotrichius one and two more non-identified PAs were detected, respectively. The total PAs concentration by spectrophotometric method was 0.25% for S. brasiliensis, 0.19% for S. heterotrichius, 0.03% for S. oxyphyllus, and 0.03% for S. selloi. The highest PAs concentration occurred in winter (June/July). No alkaloids were found in samples of S. leptolobus. These results show that S. brasiliensis is the most important cause of seneciosis in southern Rio Grande do Sul. Additional data obtained dealt with 54 outbreaks of PAs poisoning in 1978-1997. Tracheal tissues and swabs were collected from 10 different flocks of layers and broilers displaying respiratory signs of disease. Samples were processes for virus isolation in embryonated eggs and the membranes examined by histopathology. In addition, specimens were examined by polymerase chain reaction (PCR). Three flocks had ILTV positive chickens by virus isolation and PCR. These results confirm the occurrence of ILTV in chickens in Brazil.

A comparative study was made regarding the clinical and hematological alterations caused by isolates of Babesia bigemina from southeastern, northeastern and northern Brazil in experimentally infected Nelore calves. Eighteen calves between 7 and 9 months of age, without antibodies against Babesia sp and raised free from ticks, were used. Three animais were previously inoculated with 2.0x109 parasitic erythrocytes (PE) for each stabilate. The other 15 calves were subdivided into three groups, with five animais each, that were subinoculated with 1.0x1010 PE of the respective isolates. The clinical and hematological alterations were evaluated by the determination of parasitaemia, haemogram, plasmatic fibrinogen,reticulocyte count, descriptive analysis of the bone marrow and erythrocytic osmotic fragility, for 30 days, totalizing seven moments of observation. The follow-up of the immunological response by the indirect fluorescent antibody test was carried out daily until the 10th day after inoculation (DAI) and after that, on the 151 20th, 25th and 30th DAI. A mild clinical manifestation of the disease was observed. The laboratory findings revealed low leveis of parasitaemia; decrease of the erythrogram values; absence of reticulocytes, initial decrease in the total count of leukocytes, neutrophils and lymphocytes with a posterior elevation of the number of these cells; hypercellularity of the erythrocytic series and decrease of the myeloid: erythroid relation which was more accentuated between the 8th and 12th DAI, and an increase of the erythrocytic osmotic fragility in the groups inoculated with the Southeast and Northeast isolates. None of the three isolates of B. bigemina gave rise to the clinical characteristic form of the disease, although they induced an humoral immune response.

Foram utilizados 18 bezerros com idade entre sete e nove meses, isentos de anticorpos contra Babesia sp. e criados livres de carrapatos. Três animais foram previamente inoculados com 2,0x109 eritrócitos parasitados (EP) para cada isolado. Os outros 15 bezerros foram subdivididos em três grupos de cinco animais, que foram subinoculados com 1,0x1010 EP dos respectivos isolados. Foram avaliadas as alterações clínicas e hematológica por meio da determinação da parasitemia, do hemograma, do fibrinogênio plasmático, da contagem de reticulócitos, da análise descritiva da medula óssea e da fragilidade osmótica eritrocitária, no decorrer de 30 dias, perfazendo um total de sete momentos de observação. O acompanhamento da resposta imunológica pelo teste de imunofluorescência indireta foi realizado diariamente até o 10º dia pós-inoculação (DPI) e posteriormente no 15º, 20º, 25º e 30º DPI. Clinicamente, observou-se uma manifestação muito branda da doença. Os achados laboratoriais revelaram baixos níveis de parasitemia; decréscimo nos valores do eritrograma; ausência de reticulócitos; diminuição inicial na contagem total dos leucócitos, neutrófilos e linfócitos com posterior elevação do número destas células; hipercelularidade da série eritrocítica e decréscimo da relação mielóide:eritróide mais acentuada entre o 8º e 12º DPI e um aumento da fragilidade osmótica eritrocitária nos grupos inoculados com os isolados sudeste e nordeste. Nenhum dos três isolados de B. bigemina desencadeou a forma clínica característica da enfermidade, apesar de induzirem uma resposta imune humoral.

Ruminal acidosis is due to excessive ingestion of carbohydrates of rapid fermentation without previous adaptation of the microorganisms, causing severe metabolic disturbances to the animals. The objective of the present study was to assess the neutrophilic oxidative metabolism in sheep treated with sodium monensin in experimentally induced ruminal lactic acidosis. A total of 18 mate sheep, half-bred (Ideal x Merino), fistulated in the rumen, were used; nine of them received 33 mg/kg of the ionophore diet per day, for 30 days; the others were controls. The acidosis was induced by supplying 15g of sucrose/kg of body weight. The clinical evaluation and the rumen and blood samples were obtained before (Oh) and at 6, 12, 24 and 48 hours post-induction. In both groups, all the animals presented clinical manifestations of ruminal lactic acidosis 6 hours after the induction. From this period on, a significant pH decrease (P<0.05) was observed in the ruminal fluid, which reached levels below 5. There were relevant differences (P<0.05) between the groups 12 hours after the induction, when the sheep treated with monensin had higher values than those of the control group. During this period, the oxidative metabolism of the neutrophils remained inhibited, and the reestablishment of this function only occurred in the sheep which received monensin. Blood pH, plasmatic glucose and the ionizable calcium suffered alterations within its levels. The seric cortisol concentration rose significantly (P<0.05) in both groups, although differences (P<0.05) between them were found at the end of the observation period. The treatment with monensin did not influence the oxidative metabolism of the neutrophils inhibited by the lactic acidosis; however, a faster recovery of this metabolism was verified in the animals treated with the ionophore.

A acidose láctica ruminal é causada pela ingestão excessiva de carboidratos de fermentação rápida sem uma prévia adaptação dos mocroorganismos, podendo com isso gerar distúrbios metabólicos graves aos ruminantes. Este trabalho tem por objetivo estudar o metabolismo oxidativo dos neutrófilos em ovinos tratados com a monensina sódica na acidose láctica ruminal induzida experimentalmente. Foram empregados 18 ovinos, machos, mestiços (Ideal x Merino), fistulados no rúmen; dos quais nove receberam 33 mg/kg da dieta do ionóforo ao dia, durante 30 dias, os demais ovinos pertenceram ao grupo controle. A acidose foi induzida fornecendo 15g de sacarose/kg de peso corporal. A avaliação clínica e as amostras de rúmen e sangüíneas foram obtidas antes (momento controle) 0h e às 6h, 12h, 24h e 48h pós-indução. Em ambos os grupos os animais apresentaram manifestações clínicas de acidose láctica ruminal 6 horas pós-indução. A partir deste período se observou uma diminuição significativa (p< 0,05) do pH no fluido ruminal, que alcançou valores inferiores a cinco, e diferenças significativas (p< 0,05) entre os grupos a partir das 12h pós-indução foram constatadas, nos quais os ovinos tratados com a monensina apresentaram valores superiores em relação aos do grupo controle. Durante este período ocorreu uma inibição do metabolismo oxidativo dos neutrófilos nos animais estudados, ocorrendo restabelecimento desta função somente nos ovinos que receberam a monensina. O pH sangüíneo, a glicose plasmática e o cálcio ionizável sofreram alterações nos seus índices. A concentração do cortisol sérico elevou-se de forma significativa (p< 0,05) em ambos os grupos, porém diferenças (p< 0,05) entre eles foram constatadas ao final no período de observação. O tratamento com a monensina não influenciou o metabolismo oxidativo dos neutrófilos, inibido pela acidose láctica; porém se verificou uma recuperação mais rápida deste metabolismo nos animais em que receberam o ionóforo.

E-mail: fernando@alternet.com.br This study aimed to determine the phenology of Senecio brasiliensis, S. oxyphyllus, S. heterotrichius and S. selloi, and their relationship with cattle poisoning in the southern region of the State of Rio Grande do Sul, Brazil. The phenology was studied during two years in the rural area of the municipalities of Bagé and Capão do Leão. These phenological observations were made at monthly intervals during the vegetative phase, and every 15 days during the reproductive period. The plants were observed from emergence until the dispersai of seeds, considering their vigor and their relationship with environmental factors. The results indicate that whenever the environmental conditions, like moisture and light, were favorable, Senecio spp emerge. The vegetative phenophases are practically constant during ali life cycle of the plant and the whole yea1: Unfavorable environmental factors like water stress, soil management and damage by insects, associated or not, can alter the cycle of the plants and determine their permanence in the environment. The majority of the species studied behaved like annual and monocarpic plants. According with the permanence during the two years of observation, the most persistent species in the environmentwas S. heterotrichius (15% of the plants persisted during the two year period), followed by S. selloi (2,8%) and S. brasiliensis (0,9%). S. oxyphyllus did not persist for more than one year.

O estudo fenológico foi feito durante dois anos nos municípios de Bagé e Capão do Leão. As leituras foram mensais durante o período vegetativo e quinzenais no período reprodutivo das espécies, para observação desde sua emergência até dispersão de sementes, avaliando-se o vigor, e relacionando essas variáveis com fatores ambientais. Os resultados permitiram concluir que durante todo o ano há emergência de plantas de Senecio spp, desde que haja condições ambientais favoráveis, como umidade e luz, e as fenofases vegetativas são praticamente constantes durante todo o ciclo da planta. Fatores ambientais desfavoráveis como o déficit hídrico, o manejo do solo e o dano de insetos, associados ou não, podem alterar o ciclo das plantas e serem determinantes para a sua permanência no ambiente. A maioria dos exemplares, das quatro espécies, comportou-se como anual e monocárpica. A espécie mais persistente no ambiente foi S. heterotrichius (15% das plantas persistiram durante os dois anos de estudo), seguida de S. selloi (2,8%} e S. brasiliensis (0,9%). S. oxyphyllus não permaneceu no ambiente por mais de um ano.


#210 - Eradication of Herpesvirus Bovine-1 (BHV-1) from a dairy cattle herd with a semi-intensive management., 18(2):65-68

[Eradication of Herpesvirus Bovine-1 (BHV-1) from a dairy cattle herd with a semi-intensive management.] Erradicação do Herpesvírus Bovino- 1 (BHV-1) de um rebanho bovino leiteiro em manejo semi-intensivo. Pesquisa Veterinária Brasileira 18(2):61-68. Instituto de Zootecnia, Cx. Postal 60, Nova Odessa, SP 13460-000, Brazil. To eradicate the BHV-1 from a dairy cattle herd without vaccination, a preliminar serological survey was performed in 154 animals, that revealed 15.6% seropositives for BHV- 1. The test used was serum neutralization with the microtitration technique. The seropositive nonpregnant cows were immediately eliminated from the herd and the pregnant cows were isolated and eliminated after parturiation. The calves with colostral antibodies were kept in the herd. There were no seropositive animals between the 6 to 12 months old calves and the heifers. The animals were monitored serologically every 3 months for 21 months and afterwords in 2 semestral intervals. The seropositive nonpregnant, pregnant and the nursing cows were the source of BHV-1 infecction. In order to maintain a BHV-1 free herd, measurements of control had been taken, as the utilization of virus-free semen, quarantine when animals were introduced into the herd, and semestral serological surveys. Through these measurements the farm has been BHV-1 free for 18 months.

Com o objetivo de erradicar o BHV-1 de um rebanho bovino leiteiro de alto valor genético sem a utilização de vacina, foi realizado um exame sorológico prévio em 154 animais, onde constatou-se 15,6% de reagentes ao BHV-1. A técnica utilizada foi a soroneutralização em microplacas. Dentre os animais soropositivos, as vacas vazias foram descartadas imediatamente e as prenhes isoladas e descartadas após o parto. Os bezerros apresentaram anticorpos colostrais até os seis meses de idade, motivo pelo qual não foram descartados; nos bezerros de 6 a 12 meses de idade e nas novilhas não foram diagnosticados animais soropositivos. Os animais foram examinados trimestralmente, por 21 meses, seguido de mais duas coletas semestrais. As vacas secas, prenhes e em lactação, soropositivas, revelaram ser a fonte de infecção do BHV-1. A manutenção de rebanho livre é possível, desde que sejam adotadas medidas como a utilização de sêmen livre de BHV-1, realização de quarentena no ingresso de animais e exames sorológicos anuais visando impedir a reintrodução do vírus. Com o conjunto destas medidas adotadas, a fazenda encontra-se há 18 meses livre do BHV-1.
