#11 - Caseous lymphadenitis in slaughtered hair sheep in northeastern Brazil, 31(3):224-230

E-mail: franklin.riet@pq.cnpq.br The objective of this paper is to report the prevalence and distribution of lesions of caseous lymphadenitis (CL) in hair sheep slaughtered in an abattoir in the state of Paraíba, northeastern Brazil. From 1,466 sheep slaughtered, 236 (15.9%) had gross lesions similar to that of CL. The prevalence was higher in females, 17.9% (135/754), than in males, 13.8% (101/732). Ninety seven (36.3%) out of 268 lesions were localized in the prescapular lymph node, 22.4% (60/268) in the parotideal, and 20.9% (56/268) in the prefemoral. Only 70 (26.1%) out of 268 lesions suggestive of CL were detected during ante mortem examination. Caseous lymphadenitis-like lesions were observed in all flocks examined from different municipalities in the states of Paraíba, Pernambuco and Bahia. Cultures and histologic examination were performed in samples from 51 lesions. Corynebacterium pseudotuberculosis was isolated in 74.5% (43/51) samples, Staphylococcus aureus in 7.8% (4/51) samples, Escherichia coli in 2% (1/51), and in 5.9% (3/51) samples cultures were negative. Histologic findings in 11 lymph nodes showed all characteristic lesions of caseous lymphadenitis: central area of necrosis formed by concentric lamellae, with large bacterial colonies and mineralization, surrounded by an inflammatory exudate with epithelioid macrophages and few neutrophils; lymphocytes and plasma cells were observed in the next layer, and fibrous tissue was surrounding the whole lesion. Lesions in the other 32 lymph nodes showed most, but not all lesions characteristic of the disease. Giant cells were observed in 21 lymph nodes. Histologic lesions in lymph nodes from which S. aureus was isolated were similar as those of CL. These findings suggest that CL lesions are characteristic, but not pathognogmonic, because they can be confused with lesions caused by other pyogenic bacteria or tuberculosis; therefore bacteriologic examination is necessary for the diagnosis of the disease.

Dos 1466 ovinos abatidos, 236 (15,9%) apresentaram lesões macroscópicas semelhantes à linfadenite caseosa. A prevalência foi maior em fêmeas, 17.9% (135/754) que em machos, 13,8% (101/732). As principais lesões estavam localizadas nos linfonodos pré-escapulares em 36,3% (97/268) dos ovinos, nos parotídeos em 22,4% (60/268) e no pré-crural em 20,9% (56/268). Somente 70 (26,1%) de um total de 268 lesões foram detectadas no exame ante mortem. Foram encontradas lesões em todos os lotes provenientes de diversos municípios da Paraíba, Pernambuco e Bahia. Das 51 amostras em que foi feito o cultivo bacteriológico, em 74,5% (43/51) foi isolado Corynebacterium pseudotuberculosis. Em 7,8% (4/51) foi isolado Staphylococcus aureus, em 2% (1/51) Escherichia coli e 5,9% (3/51) amostras foram negativas. No exame histológico, em 11 linfonodos havia lesões da linfadenite caseosa caracterizadas por área de necrose central formada por lamelas concêntricas, com presença de grandes colônias bacterianas e focos de mineralização, rodeada por uma faixa de infiltrado inflamatório com macrófagos epitelióides e poucos neutrófilos. Na camada adjacente observaram-se linfócitos e plasmócitos e toda a lesão era delimitada por tecido conjuntivo fibroso. Nas outras 32 amostras foram observadas a maioria, mas não todas as lesões características da doença. Em 21 amostras foram encontradas células gigantes. As lesões histológicas dos linfonodos que foi isolado S. aureus foram semelhantes as da linfadenite caseosa. Conclui-se que as lesões histológicas de linfadenite caseosa são características, mas não patognomônicas, pois podem ser confundidas com lesões causadas por outros organismos piógenos e com tuberculose; portanto o isolamento bacteriológico é imprescindível para o diagnóstico definitivo da linfadenite caseosa.

The main objective was to compare the methods of isolation of mycobacteria of the Mycobacterium avium Complex (MAC), Ziehl-Neelsen (ZN) staining, histopathological examination and immunohistochemistry (IHC) for the diagnosis of granulomatous lymphadenitis caused by MAC. A total of 431 samples of lymph nodes from 394 swine carcasses kept in 12 cold storage rooms of southern Brazil, which had the Service of Federal Inspection (SFI), were analyzed. Lymph nodes with granulomatous lesions were submitted to histological examination, ZN staining and IHC with monoclonal antibody produced with cellular extract of M. avium. The agreement between the exams was measured by the Kappa test at a 95% confidence level. The macrocospic examination done by SFI correctly identified 90.3% of granulomatous lesions, when compared with histological examination, and the majority of carcasses (92.5%) showed the lesions only in lymph nodes of the alimentary chain. Histological examination confirmed the presence of granulomatous lesions in 90.3% of the lymph nodes. The agreements between histological examination, ZN staining (Kappa: 0.342), IHC and the isolation of MAC (Kappa: 0.102) were low, however high between IHC and histology with typical presence of granulomas in the lymph nodes (Kappa: 0.973). IHC together with histological examination revealed to be efficient for identification of the granulomatous lymphadenitis caused by MAC.

E-mail: armando@cnpsa.embrapa.br An epidemiological study in 60 units of swine termination (UT), pertaining to producers integrated with the main Agribusiness of the States of Santa Catarina and Rio Grande do Sul. These units had been selected due to the occurrence of lymphadenitis in slaughtered pigs. The batch size studied in each UT varied from 22 to 960 pigs. The animals were considered positive or negative for lymphadenitis, following the evaluation routine of the Service of Federal Inspection (SIF) at slaughter. For data collection at the UTs, a questionnaire with 156 variables was applied at a single visit to the farms. In the 60 UT studied, the percentage of positive animals for lymphadenitis was 10.37%. Among the variables analyzed, nine risk factors were identified (explicative variables) and associated with the occurrence of lymphadenitis (objective variables). The explicative variables were: bad hygiene of the feeders and drinking fountains, untreated drinking water, bad conservation of the facilities, use of the same truck for ration and animal transportation, production of ration inside the farm, free access of other animals to the ingredients of ration, storage of ready usable ration in boxes or bags and handling of the production of animals in an continuous system.

#14 - Histologic, histochemical and ultrastructural study of livers and lymph nodes with foamy macrophages (foam cells), 18(1):29-34

[Histologic, histochemical and ultrastructural study of livers and lymph nodes with foamy macrophages (foam cells).) Estudos histológico, histoquímico e ultra-estrutural de fígados e linfonodos de bovinos com presença de macrófagos espumosos ("foam cells"). Pesquisa Veterinária Brasileira 18(1):29-34. Depto Patologia Clínica Veterinária, Faculdade de Veterinária, UFRGS, Cx. Postal 15094, Porto Alegre, RS 91540-000, Brazil. Samples of liver, kidney, spleen, and hepatic, mesenteric, retromandibular, prescapular and mediastinic lymph nades, collected from 12 groups of cattle of different ages from the State of Mato Grosso, were studied. A total of 120 bovines was examined. According to their history, the animals were kept in pastures where Brachiaria decumbens and Brachiaria brizantha were the predominant plants. Grossly there was a yellowish liver which was more evidente after 24 hr. fixation with formalin. Hepatic and mesenteric lymph nades had normal size, howeve1; on the cut surface there were whitish grooves, extending from the cortex to the medulal; and multiple small white nodular areas in the medulal: Beside these areas there were red, sometimes hemorrhagic, foci. Microscopically all animals had cells with foamy citoplasm in the liver and in the hepatic and mesenteric lymph nades, of ten forming multinucleated cells. In the lymph nodes these infiltrates were adjacent to necrotic, hemorrhagic areas. In the liver the foamy cells were irregularly disseminated throughout the parenchyma or formed nodules around the central vein. These cells did not stain with PAS but weakly with oil red. Ultrastructurally foamy cells and hepatocytes showed negative images of crystals involved partially or totally by membranes. Mediastinic, retromandibular and prescapular lymph nades of the sarne animals showed no gross and microscopic alterations.

Os animais haviam sido mantidos em pastos onde Brachiaria decumbens e Brachiaria brizantha eram as forrageiras predominantes. Macroscopicamente o fígado desses animais mostrava coloração amarelada, mais evidente após 24 horas de fixação em formol a 10%. Nos linfonodos hepáticos e mesentéricos foram evidenciadas, na superfície de corte, estriações esbranquiçadas de forma radiada na cortical e medular com pequenas áreas brancas nodulares multifocais principalmente na medular. Em muitos casos, associado com essas áreas foram vistos focos de aspecto hemorrágico. Microscopicamente foram encontradas, no fígado, linfonodos hepáticos e mesentéricos, células com citoplasma espumoso, muitas das quais multinucleadas. Nos linfonodos hepáticos e mesentéricos estes infiltrados estavam associados a áreas de necrose e hemorragia. No fígado, as células de citoplasma espumoso estavam presentes em todo parênquima, de forma irregular, geralmente formando nódulos ao redor da veia centrolobula1: Estas células não se coravam pelo ácido periódico de Schiff (PAS) e apenas fracamente na coloração de gordura pelo Oil Red O. Na ultraestrutura as células de citoplasma espumoso apresentavam fendas, parcial ou totalmente delimitadas por membrana, que representam a imagem negativa de cristais, presentes também no citoplasma dos hepatócitos.


#15 - Histochemical diagnosis of actinomycotic-like granulomas in cattle from southern Brazil

Abstract in English:

The etiology of actinomycotic-like lesions from 254 lymph nodes of cattle from southern Brazil was determined by histochemistry. The diagnosis of these lesions in hematoxylin-eosin (H-E) preparations was based upon the pyogranulomatous inflammatory reaction with centrally located sulphur grains. The lesions stained by the MacCallum-Goodpasture technique showed 60.36% Gram-positive coccoid bacteria (Staphylococcus), 36.62% Gram-negative cocco-bacillus (Actinobacillus lignieresz) and 2.72% Gram-positive filamentous branching elements (Actinomyces bovis). Fite-Faraco stain demonstrated the acid-fast properties and Gomori stain, the morphological features to distinguish between Actinomyces bovis .and Nocardia sp. Gram-positive cocei were diagnosed as botryomycosis which occurred predominantly in prescapular, parotid and femoral lymph nodes. The presence of Gram-negative coccobacillary elements indicated Actinóbacillus sp., whereas the finding of Gram-positive filamentous branching elements indicated Actinomyces sp. These latter two were more frequently found in retropharyngeal and sublingual lymph nodes. Small differences in the morphology of the bacterial colonies may help to establish the etiological diagnosis in H-E preparations. The Gram method, however, allows an accurate distinction between Actinobacillus lignieresi and Actinomyces bovis in cases where fresh material for bacteriological isolation is no longer available. Of 467 other cases originally diagnosed as tuberculosis in gross inspection at the slaughter house, 10.23% were histologically confirmed as actinomycotic-like granulomas. This certainly causes economic losses due to the condemnation of carcasses.

Foi determinada a etiologia dos "granulomas actinomicóides" através da histoquímica em 254 materiais de linfonodos de bovinos do Rio Grande do Sul. O diagnóstico de granuloma actinomicóide foi baseado na inflamação piogranulomatosa, contendo, na porção central, os grânulos de enxofre, observados na coloração pela hematoxilina e eosina (H-E). Ao material com essa lesão foi aplicada a técnica de MacCallum-Ooodpasture que evidenciou uma incidência de 60,36% de bactérias cocoides Gram-positivas (Staphylococcus), 36,62% de coco-bacilos Gram-negativos (Actinobacillus lignieresz) e de 2,72% de elementos filamentosos, ramificados, Gram-positivos (Actinomyces bovis) como constituintes do grão de enxofre. Nos casos de actinomicose aplicaram-se as técnicas de Fite-Faraco e Prata de Gomori para a diferenciação entre Actinomyces bovis e Nocardia sp. Os casos de botriomicose foram vistos principalmente nos linfonodos pré-escapulares, parotídeos e femurais; os de actinobacilose e actinomicose nos retrofaringes e sublinguais. Pequenas diferenças do aspecto das colônias podem auxiliar no diagnóstico etiológico nas preparações coradas pela H-E, mas o emprego das técnicas de Gram permite a diferenciação entre Actinobacillus lignieresi e Actinomyces bovis quando não se dispõe de culturas. Adicionalmente observou-se pelo estudo histológico de outros 467 casos, que 10,23% deles foram diagnosticados como granuloma actinomicóide, porém interpretados macroscopicamente, na inspeção, como tuberculose o que certamente leva a prejuízos econômicos pela condenação das carcaças.


#16 - Serologic and allergic monitoring of experimental infections in goats by Corynebacterium pseudotuberculosis

Abstract in English:

The six month serologic and allergic monitoring of 26 experimentally infected goats, by intradermic inoculation of 5, 20 and 100 Corynebacterium pseudotuberculosis cells, showed the development of both humoral and cellular imune responses at the sarne time. Only one goat did not develop antibodies, but showed a strong allergic reaction, during 22 weeks post-infection. The antibodies, investigated by the synergistic hemolysis inhibition test (Knight 1978), were first detected at the 3rd week and were most elevated at the 7th week post-infection. In 8 animals the antitoxins were persistent until the 29th week; in 7 animals the antibodies could not be detected between the 12th and the 24th week and in other 9 goats between the 5th and 8th week post-infection. Three animals did not show antibodies in any phase after the infection. The allergic reaction, evaluated by the lymphadenization test (Langenegger et al. 1987) in six-week intervals, showed the highest value between the 4th and the 6th week post-infection and then decreased slowly, following generally the antibody level. One third of the infected goats lost the antitoxin titre and the allergic reaction at the 9th and 10th week, having selfcures. A gradual decrease of the antitoxins titre and the allergic sensitization was observed until the total disappearance in some goats with chronic lymphnode abscesses which did not ulcerate, despite live C. pseudotuberculosis still being in the lesion.

O monitoramento sorológico e alérgico de 26 caprinos infectados experimentalmente por inoculação intradérmica de 5, 20 e 100 bactérias de Corynebacterium pseudotuberculosis, durante 6 meses, revelou que as respostas imunológicas humoral e celular desenvolveram-se paralelamente, exceto em um animal no qual não foram detectadas antitoxinas, porém forte reação alérgica até a 22ª semana pós-infecção. A pesquisa de antitoxinas feita através da técnica da Inibição da Hemálise Sinérgica (Knight 1978), revelou a presença de anticorpos a partir da 3ª e atingiu maior concentração na 7ª semana pós-infecção. Em 8 animais a presença destas persistiu até a 29ª semana; em 7 desapareceu entre a 12ª e 24ª e em 9 entre a 5ª e 8ª semana. Três animais não reagiram com título igual ou superior à diluição 1:20. A linfadenização (Langenegger et al. 1987), feita em intervalos de 6 semanas, revelou reações maiores entre a 4ª e 6ª semana pós infecção. Depois a sensibilidade alérgica decrescia, pouco a pouco, acompanhando grosseiramente o declínio do nível das antitoxinas. Em um terço dos animais houve autocuras a julgar pela regressão total das antitoxinas e perda da sensibilidade alérgica a partir da 9ª e 10ª semana pós-infecção. Em alguns caprinos cujas lesões ganglionares se tornavam crônicas sem ulcerar, observou-se também progressiva regressão, tanto das antitoxinas quanto da reação alérgica, até o seu desaparecimento, apesar da presença de C. pseudotuberculosis na lesão.


#17 - Prevalence and comparative diagnosis of caseous lymphadenitis in goats from Rio de Janeiro State

Abstract in English:

The prevalence of caseous lymphadenitis in 13 goat herds with 760 adult animals (58.4 herd/media) in the State of Rio de Janeiro was evaluated by clinical, serological and allergical diagnostic procedures. Three herds (23.0%) were free and 10 (77.0%) were infected by Corynebacterium pseudotuberculosis and the infection rate varied from 3.6% to 100% in these herds. From the 760 goats, 171 (22.5%) showed lesions and/or serological and/or allergical imune response. Separately, the clinical examination based on inspection and palpation rçvealed 93 (12.2%) cases of lymphnode abscedation and/or scares of prior ruptured lesions. The serological test made by the synergistic hemolysis inhibiton test showed antibodies in 146 (19.2%) goats with titers varying from 1:20 to 1:1256, and the allergical sensitization measured by the lymphadenization test presented positive reaction up to 1.5 mm increase in the thickness of skin-folds in 168 (22.1%) animals. The allergy test was the most sensitive and specific diagnostic procedure and proved to be a practical method to detect pré-clinical infections in the field.

Foram examinados 13 rebanhos de caprinos leiteiros no Estado do Rio de Janeiro, constituídos por 760 (média 58,4/rebanho) animais de raças puras e mestiças. Utilizou-se o exame clínico para localizar lesões suspeitas através de inspeção e palpação dos linfonodos superficiais; o exame sorológico para evidenciar antitoxinas pela técnica da inibição da hemólise sinérgica e o teste alérgico da linfadenização para avaliar a imunidade mediada por células. A pesquisa revelou que três (23,0%) rebanhos estavam indenes e 10 (77 ,0%) infectados por Corynebacterium pseudotuberculosis, variando a incidência de 3,6% a 100% de infecções nos rebanhos. Dos 760 caprinos, 171 (22,5%) apresentaram lesões e/ou reações positivas nos testes utilizados. Individualmente, o exame clínico revelou em 93 (12,2%) dos caprinos a presença de linfonodos abscedados e/ ou cicatrizes na pele de ulcerações anteriores. O exame sorológico acusou a presena de antitoxinas com título igual ou superior a 1:20 em 146 (19,2%) dos caprinos e o teste alérgico revelou reações positivas (acima de 1,5 mm) em 168 (22,1 %) dos animais. O teste alérgico, além de se mostrar mais sensível e específico para o diagnóstico da linfadenite do que a pesquisa de antitoxinas e o exame clínico, é sobretudo um método prático para a detecção de infecções precoces e confirmar suspeitas clínicas, a nível de campo.


#18 - Levels of immunoprotection induced in goats against Corynebacterium pseudotuberculosis using several types of vaccines

Abstract in English:

In order to evaluate the immune response of goats against Corynebacterium pseudotuberculosis, 23 groups, comprising 196 animals, were immunized with several antigenic preparations obtained from the bacteriae, at different time intervals, and using various kinds of adjuvants. The results indicated that the rate of immunoprotection in the immunized animals challenged after 120 days was below 60%. The animals challenged between 8 and 10 months showed protection levels ranging from 44.5 to 22.3%. The lowest rate of incidence of the disease appeared in the anima1s challenged between 30 and 60 days after the last immunization.

Com o objetivo de observar a imunoproteção desenvolvida em caprinos contra Corynebacterium pseudotuberculosis esses animais foram submetidos a diferentes esquemas de imunização, sendo utilizados inóculos de diferentes preparados antigênicos desta bactéria em intervalos de tempo variados e com diversos adjuvantes. Os resultados revelaram que o percentual de imunoproteção nos animais imunizados e desafiados 180 dias após estava abaixo de 60%. Já os animais desafiados dez meses após a primeira inoculação apresentaram níveis de imunoproteção entre 44,5 e 22,3%. O menor índice da doença foi observado nos animais desafiados entre 30 e 60 dias após a última inoculação do preparado antigênico.


#19 - Reproduction of caseous lymphadenitis in goats by small numbers of Corynebacterium pseudotuberculosis

Abstract in English:

Caseous lymphadenitis was reprodued experimentally by intradennic inoculation of 5, 20 and 100 washed Corynebacterium pseudotuberculosis bacteria in 26 normal goats. Between day 2 and 9 after inoculation, there was local erythema, papula and skin ulceration and, at day 15, a small enlargement of the local lymph node. Between day 45 and 180, independent of the infective dose rate, the infection developed to an abscess in the local lymph node of about one third of the goats. A similar number showed a pronounced enlargement of the local lymph node, but these later became normal again. The remainder of the goats showed an early return to normal after the initial signs of infection. All 26 goats, except 2, presented an antitoxin titre during some phase of the infection. The experimental·infection was shown to develop as the natural. It was concluded that selfcures, demonstrated experimentally, must also occur in the field, making clinical and immunological diagnosis difficult.

A linfadenite caseosa foi reproduzida experimentalmente através da inoculação intradérmica de 5, 20 e 100 bactérias lavadas de C. pseudotuberculosis em 26 caprinos normais. A infecção manifestou-se, inicialmente, por formação de eritema, pápula, pústula e ulceração local da pele entre o 2º e o 9º dia pós-inoculação e no 15º dia por um ligeiro aumento do linfonodo satélite. Posteriormente, entre o 45º e o 180º. dia, independentemente da dose infectante usada, a infecção evoluiu para abscedação do linfonodo local em cerca de 1/3 dos caprinos. Em outro número similar, observou-se acentuado aumento do volume do linfonodo mas que depois regrediu e, finalmente, nos restantes houve rápida normalização após os sinais iniciais da infecção. Dos 26 caprinos apenas 2 não apresentaram antitoxinas no soro durante nenhuma fase da infecção. Foi demonstrado com o presente experimento que a infecção evoluiu simulando a natural e concluiu-se que as curas espontâneas observadas também devem ocorrer nas infecções de campo, dificultando o diagnóstico clínico e imunológico.


#20 - Evaluation of an inactivated vaccine against caseous lymphadenitis of goats kept under extensive management

Abstract in English:

The experiment was carried out during a 2-year period (Oct. 1981 to Oct. 1983), using 163 goats of both sexes with 6 to 12 months of age, The objective was to evaluate the immunogenicity of a vaccine against caseous lymphadenitis, in northeastem Bahia. The vaccine was developed, utilizing the local strain 1002 Curaçá of Corynebacterium pseudotuberculosis isolated from na abscess of a goat. For the preparation, tryptose broth, enriched with yeast extract and lactalbumin, was used. Inactivation was effected with 0.5% formaldehyde, and aluminum gel phosphate was used as an adjuvant. The cell volume varied from 0.65 to 0.7%. The experiment consisted of the following treatments: A1 - animals vaccinated every 6 months; A2-animals vaccinated once a year; and A3 - animals not vaccinated. The goats were vaccinated by subcutaneous inoculation of 3 ml vaccine into each axillar region, and post-mortem examination was made at the end of the experiment. The results showed that 72.1% of the control animals (A3) had moderate to severe lesions in different lymphnodes, and in some cases in the internai organs. Of the vaccinated animals, only 25.9% and 18.9%, at 6 and 12 months interval respectively, showed discrete lesions which were restricted to no more than three lymphnodes per animal. There was a signif cant difference (P<0,05) between the vaccinated and the non-vaccinated animals, but no difference between treatment A1 and A2. It can be estimated that the vaccination gave a protection of 77% against the disease.

Foram utilizados 163 caprinos de ambos os sexos, de 6 a 12 meses de idade, com o objetivo de avaliar a imunogenicidade de uma vacina contra a linfadenite caseosa, no Nordeste do Estado da Bahia. A vacina foi elaborada com a cepa 1002 Curaçá de C. pseudotuberculosis, isolada de abscesso de caprino, cultivada em caldo tryptose, enriquecida com extrato de levedura mais lactalbumina e inativada com formaldeído a 0,5% durante 7 dias, utilizando-se como adjuvante o gel fosfato de alumínio. O volume celular variou de 0,65 a 0,70%. O experimento constituiu-se dos seguintes tratamentos: A1 - animais vacinados semestralmente; A2 - vacinados anualmente; A3 - animais não vacinados. Os animais vacinados foram inoculados via subcutânea com 3 ml da vacina em cada região axilar e, ao final do experimento, foram necropsiados. Os resultados mostraram que 72,1 % dos caprinos do grupo testemunha A3 se infectaram naturalmente e apresentaram lesão moderada a severa em vários linfonodos e, em alguns casos, nos órgãos internos. Dos vaciandos, apenas 25,9 e 18,9%, respectivamente, para os revacinados com 6 e 12 meses de intervalo, apresentaram lesões discretas que se restringiram a não mais de três linfonodos por animal. Houve diferença significativa (P<0,05) entre os animais vacinados e não vacinados mas não houve diferença entre os tratamentos A1 e A2. Pode-se estimar que a vacina proporcionou 77% de proteção contra a doença.
