Aspects dealt with include sampling collection, necropsy procedures, gross and histopathological findings, some of which are of little or no pathological significance but often observed in the routine diagnostic work and misinterpreted as real or important lesions.

#12 - Gastric ulcers in pigs affected with postweaning multisystemic wasting syndrome, p.601-605

E-mail: davetpat@ufrgs.br Samples of gastric lymph nodes and the stomachs from 24 pigs selected from herds affected by postweaning multisystemic wasting syndrome (PMWS) and sudden death associated with gastric ulcers were studied. Pigs were selected on the basis of unthriftiness, decreased feed intake, and wasting. The stomachs were opened, inverted, and classified into 0-3 score according the severity of the gross lesions present in pars oesophagica (non-glandulargastric mucosa). Selected samples were processed for paraffin embedding and stained with hematoxylin and eosin. Immunohistochemistry using anti-PCV2 (porcine circovírus type 2) antibody, anti-Helicobacter pylori antibody and a wide-spectrum anti-cytokeratin antibody was performed. Gross changes in pars oesophagea were classified according to the severity of lesions as score 3, 2, and 1 in 8, 6, 5 stomachs respectivelly. Microscopically, hyperplastic lymphoid follicles, lymphohistiocytic inflammatory infiltrates and focci of necrosis in the gastric mucosa were common findings. Large amounts of PCV2 antigen were observed in the cytoplasm and nuclei from intralesional cells and debris from the gastric glandular mucosal zone; however, in the fundus, anti-PCV2 immunostaining was restricted to the surface mucosal cells and foveolar compartment. All gastric lymph nodes were positive for PCV2 antigen. Anti-H. pylori immunostaining was seen in eleven cases, mainly in the antrum, on the mucosal surface and foveolar compartment. The association of the anti-PCV2 immunostaining with the glandular mucus-producing cells suggests a role for PCV2 as an additional factor for the swine ulcer development.

E-mail: davidbarcellos@terra.com.br The occurrence and bacteriology of purulent otitis in groups of pigs affected by three different clinical conditions are described: postweaning multisystemic wasting syndrome (PMWS), depressed growth (attrition) and normal growth (control). A total of 385 animals, 60-130 days of age, were examined. Diagnosis of PMWS was confirmed by the presence of clinical signs and pathological findings compatible with the disease, and through detection of viral antigens in tissues by immunohistochemistry. From 242 pigs with PMWS, 57 (23.5%) showed purulent lesions in the middle ear. Among 119 pigs with attrition, only 1 (0.7%) presented the lesion. In 24 control pigs, middle ear lesions were not detected. The most frequently isolated agents from the lesions were Arcanobacterium pyogenes (43%), á–hemolytic Streptococci (37.2%) and Pasteurella multocida (27.9 %) out of 86 materials. The frequent occurrence of purulent lesions in the middle ear of PMWS affected pigs suggests that PCV2 infection may increase the susceptibility to bacterial otitis. The low occurrence of this disease in piglets with attrition suggests that otitis does not represent a significant cause for depressed growth in pigs from growing and finishing age. The isolation of A. pyogenes, á-hemolytic Streptococci and P. multocida from most of the lesions emphasizes the importance of these organisms as causal agents of otitis media in swine.

E-mail: davetpat@ufrgs.br Swine producers have historically resected needle teeth of newborn pigs to prevent potential injuries to the sow mammary gland and faces of littermates. However, the possible impact of this practice on pig performance has been questioned. As part of a study, 280 PMWS affected piglets, with 40-70 days of age, were examined for the presence of dental lesions immediately after euthanasia. Most pigs were confirmed as PMWS by the detection of typical microscopic lesions and immunohistochemical pattern. At least one periapical abscess was observed in 58 piglets (20.7%), of which 12 had multiple abscesses. There were 22 (31.4%), 16 (22.9%), 4 (5.7%), 23 (32.9%), and 5 (7.1%) abscesses in 3rd upper incisives, 3rd lower incisives, upper canines, lower canines, and other teeth, respectively. The higher prevalence of dental abscesses observed in 3rd incisives and lower canines is probably associated with the wider area of grinding or clipping to which these teeth were subjected. Among bacteria isolated from samples collected from 65 abscesses, Streptococcus sp. was the most prevalent bacteria and was present in 21.48% and 27.7% of the aerobic and anaerobic isolates, respectively. In the second most frequent group were coryneform microorganisms, which showed higher rate of isolation under anaerobic atmosphere than in aerobic culture. There was preponderance of Gram-positive isolates. It could not be determined whether these bacterial co-infections predisposed pigs to development of PMWS-type lesions or if were subsequent to the PCV2 infection.

The chicken inoculated with live bacterial cultures were submitted to euthanasia 21 days later, and the cecum was examined histologically using hematoxylin-eosin, silver staining and immunohistochemistry with a polyclonal antibody anti-Brachyspira pilosicoli. With silver staining and immunohistochemistry, respectively, 21.59% and 70.96% of the chicken showed colonization of the cecal epithelium with B. pilosicoli. Differences in the type of colonization characterized by continuous adhesion, focal adhesion or free bacteria in the intestinal lumen were observed. Immunohistochemistry was more efficient to assess intestinal colonization, because 49.37% more colonized chicken could be detected with silver staining. With three strains, elongated figures inside the cytoplasm of epithelial cecal cells in the inoculated chicken were observed.

#16 - Serotypes of Haemophilus (Actinobacillus) pleuropneumoniae isolated from swine in Brazil

Abstract in English:

Thirty-three strains of Haemophilus (Actinobacillus) pleuropneumoniae biovar 1 were isolated from swine lungs with acute or chronic lesions of swine pleuropneumonia, with one exception, which was isolated from the nasal cavity of a pig from a problem herd. These strains originated from 14 herds, ten located in the State of Santa Catarina, three in Rio Grande do Sul and one in São Paulo. Serotyping was done by rapid slide agglutination (RSA), immunofluorescence (IF), counterimmunoelectrophoresis (CIE) and immunodiffusion (ID). Twenty-two (66%) of the 33 strains could be serotyped, 19 belonged to serotype 5 and three to serotype 3. All serotype 3 strains together with those not possible to be serotyped originated from swine of the sarne herd, Santa Catarina, one in which serotype 5 was also isolated. The IF and CIE were the most sensitive tests, while the ID was the least sensitive. Three strains could not be serotyped by the RSA because they auto-agglutinated.

Isolaram-se 33 amostras de Haemophilus (Actinobacillus) pleuropneumoniae, biovar 1, sendo que 32 originaram-se de pulmões de suínos com lesões macroscópicas agudas ou crônicas de Pleuropneumonia Suína (PPS) e uma amostra da cavidade nasal de um suíno de granja problema. Estas amostras foram coletadas em 14 propriedades, sendo dez localizadas no Estado de Santa Catarina, três no Rio Grande do Sul e uma em São Paulo. Para a sorotipagem utilizaram-se as técnicas de soroaglutinação rápida (SAR), imunofluorescência (IF), imunoeletroosmoforese (IEOF) e imunodifusão (ID). Das 33 amostras, 22 (66%) foram sorotipadas e, entre estas, 19 pertenciam ao sorotipo 5 e três ao sorotipo 3. As amostras do sorotipo 3 e aquelas não sorotipadas originaram-se de suínos de uma mesma propriedade do Estado de Santa Catarina, onde também isolou-se o sorotipo 5. Os testes de IF e IEOF foram os mais sensíveis e o de ID apresentou a menor sensibilidade. Três amostras não puderam ser sorotipadas pelo teste de SAR porque auto-aglutinavam.
