#631 - Intoxicação experimental por Sida carpinifolia (Malvaceae) em ovinos, p.15-20

E-mail: alseitz@terra.com.br. Seven sheep received dry crushed Sida carpinifolia L.f. One of them died at 18 and other at 53 days of the experiment. Four others were euthanatized and necropsied at 30, 45, 75 and 100 days. For one sheep the supply of S. carpinifolia was interrupted on the 80th day of the experiment, and 70 days later the animal was euthanized and necropsied. The minimal amount of the dry plant consumed was 11 g/kg and the maximum was 30 g/kg. The progression of clinical findings was similar in six animals with slight diarrhea at 20 days of experiment. Neurological signs were observed at 25 days and included ataxia with dysmetria, muscle tremors of the head, atypical postural reactions, frequent falls, sluggish of movements, difficulty in grazing and swallowing. These signs were enhanced when the animals were forced to walk. Four of the animals presented progressive emaciation. The sheep whose supply of the plant was interrupted recovered gradually, and 11 days after the animal returned to normal. During necropsy, only enlarged mesenteric lymph nodes were observed. The histological alterations were more significant in the central nervous system, with multiple and severe cytoplasmic distention and vacuolation which affects specially Purkinje cells of the cerebellum, neurons of cerebral cortex, thalamus, midbrain and the ventral horn of spinal cord. Axonal spheroids in the brain, more frequently in the granular layer of cerebellum were also observed. The cytoplasmic vacuolation was also found in pancreatic acinar cells, renal tubules, thyroid follicular epithelium, hepatocytes and macrophages of lymphoid organs. The ultrastructural lesions observed were cytoplasmic vacuolation, some surrounded by membranes in Purkinje cells of cerebellum and thyroid follicular cells. The sheep, which had S. carpinifolia withdrawn from its diet for 70 days, had no significant histological alterations.

On each farm the animals were divided into two groups. For one group a commercial mineral mixture was used, and to the other group a selective mineral mixture containing only Na, P, Cu and Co was offered. The cattle were weighed every 30 days, and the mineral supplement intake was measured monthly. The daily weight gain of the animals receiving the selective mineral mixture was greater in three farms. Only on the second farm the group receiving the commercial mineral mixture had a superior daily weight gain. The daily intake of the selective mineral mixture was lower in all groups and was attributed to the high amount of NaCl in these mixtures. The selective mineral supplementation was 3 up to 7 times more economic than the conventional supplementation with a “complete” mineral mix. Throughout the experimental period the animals did not show any direct or indirect clinical signs of mineral deficiency. The results of these experiments confirms the hypothesis that selective mineral supplementation – i.e. supplementation of the mineral element(s) deficient – was correct and results in expressive reduction of the cost with the mineral supplementation for beef cattle.

#633 - Febre catarral maligna em bovinos no Rio Grande do Sul: transmissão experimental para bovinos e caracterização do agente etiológico

E-mail: claudioslbarros@uol.com.br Two oubreaks of malignant catarrhal fever (MCF) occurring in cattle on two farms (A and B) in the municipality of Santiago, state of Rio Grande do Sul (RS), Brazil, and the transmission of the disease to susceptible calves as well as the detection of ovine herpesvirus-2 (OvHV-2) in tissues of affected cattle are reported. The two epizootics occurred from November 2001 to February 2002 (Farm A) and in January-February 2003 (Farm B). Numbers of cattle at risk, morbidity and letality rates were respectively 170, 10.59% and 83.33% for Farm A and 500, 2.4% and 100% for Farm B. Contact between affected cattle and sheep was detected in both farms, but lambing ewes were present only in farm A. Duration of clinical courses, gross findings and histopathology were the same for the affected cattle in both farms. Most affected cattle died or were euthanatized in extremis after a clinical course of 2-8 days. Clinical signs included fever (40.5 and 41.5°C), nasal and ocular discharge, corneal opacity, conjunctivitis, drooling, erosions and ulcerations of the mucosae, diarrhea, hematuria, and neurological disturbances. Eleven necropsies (9 on Farm A, 2 on Farm B) were performed. Gross lesions included erosions and ulcers affecting the mucosae of nasal turbinates, oral cavity, gastrointestinal and urogenital tracts; hemorrhage and necrosis of the tip of the buccal papillae, lymph node enlargement, multifocal white foci in renal cortex, and hyperemia of leptomeninges. Microscopically, there were arteritis and fibrinoid degeneration in medium and small arteries and arterioles of multiple organs and tissues, necrosis and inflammation in several mucosal surfaces, keratitis, conjunctivitis, uveitis, intersticial nephritis, and encephalitis. Transmission experiments were attempted in five calves (E1-E5) by inocculating each of them intravenously with 500 ml of whole heparinized blood from a MCF affected cow. The transmission was suscessful in at least three (E1-E3) of the experimental calves which became sick after an incubation period of 15-27 days. Four experimental calves either died or were euthanatized in extremis after a clinical course which varied from 3 days to 8 weeks. The remaining experimental calf (E5) recovered from a mild disease and was euthanatized 14 weeks after inocculation. Necropsies were performed in all five calves. Clinical signs, necropsy and histopathological findings of three calves (E1-E3) were characteristic of MCF. OvHV-2 viral DNA was detected by the polimerase chain reaction (PCR) test in paraffin embedded tissues from seven cattle out of the 11 spontaneous MCF cases and from three experimental calves (E1-E3). PCR tests resulted negative in the remaining four of the 11 spontaneous MCF cases tested and in two (E4,E5) of the five experimental calves. Immunohistochemistry performed in sections of lymphoid tissue from calf E4 failed to detect BVD virus antigen. The experimental transmission of MCF and the characterization of the etiological agent as OvHV-2 were successfully attempted in cattle for the first time in Brazil.

The animals were daily fed with fresh and chopped Andropogon gayanus or Pennisetum purpureum; the concentrate ration was formulated without a mineral mixture and was offered together with the forage of 300 up to 500 g/animal/day. Sixteen goats were divided into two groups of 8 animals each (4 small and 4 larger goats). For one group a commercial mineral mixture was used, and to the other group a selective mineral mixture containing only Na, P and Cu was offered. The animals were weighed every 30 days and the mineral supplement intake was measured monthly. Blood samples were collected at the beginning and the end of the experimental period. There was no difference in daily weight gain when the selective or commercial mixture was given for both groups. The selective mineral supplementation was 2.3 (for the smaller animals) and 3.9 (for larger animals) times more economic than the conventional supplementation with a “complete” mineral mix. The two groups showed recovery of hair quality and had an increase of hair pigmentation around the eyes, which was attributed to the correction of copper deficiency. In the larger animals the intake of both supplements increased the red blood cells counts, and the haematocrit and haemoglobin values. The forage intake was sufficient to meet the Ca and Co needs of the animals, and for P the concentrate intake was sufficient to overcome the demand for this element. The requirement of zinc was supplied through the forage and the concentrate ration. Thus, when an adequate concentrate and a good quality forage is given, only the deficient mineral element(s) should be supplemented. In this specific case only Na and Cu should be given. The result of this experiment confirms the hypothesis that selective mineral supplementation – i.e. supplementation of the mineral element(s) deficient in the diet – was correct and can result in reduction of the cost with the mineral supplementation of a herd.

A review.] Controle biológico de helmintos parasitos de animais: estágio atual e perspectivas futuras. Pesquisa Veterinária Brasileira 23(3):93-100. Postal 486, Belo Horizonte, MG 31270-901, Brazil.E-mail:mmot@icb.ufmg.br Biological control is a non-chemical alternative method with its main goal to reduce the amount of parasite population using natural antagonists. The administration of nematophagous fungi to animais has been considered an alternative in gastrointestinal helminthiasis prophylaxis. The nematophagous fungi produce trap-shaped structures, which are responsible for capturing and destroying the free-living stages of nematodes. The genera Arthrobotrys, Duddingtonia and Monacrosporium has been shown efficacy in laboratory and field experiments against cattle, equine, ovine and swine parasites. Severa) fungi formulations have been evaluated, but there is so far no commercial product available. The association of research groups with industry could improve the successful implementation of this control method.

A administração de fungos nematófagos aos animais domésticos é considerada uma promissora alternativa na profilaxia das helmintíases gastrintestinais parasitárias. Os fungos nematófagos desenvolvem estruturas em forma de armadilhas, responsáveis pela captura e destruição dos estágios infectantes dos nematóides. Os fungos dos gêneros Arthrobotrys, Duddingtonia e Monacrosporium têm demonstrado eficácia em experimentos laboratoriais e no campo no controle de parasitos de bovinos, eqüinos, ovinos e suínos. Diversas formulações fúngicas têm sido avaliadas, no entanto, ainda não há nenhum produto comercial disponível. A associação dos grupos de pesquisa e o envolvimento das indústrias poderão colaborar para o sucesso na implementação desta forma de controle.

Caracterização de seqüelas subseqüentes à punção folicular em bovinos. Pesquisa Veterinária Brasileira 23(3):119-124. Embrapa Gado de Leite, Juiz de Fora, MG 36038-330, Brazil. E-mail: jhmviana@cnpgl.embrapa.br The aim of this studywas to evaluate lesions of the genital tract in animais used as oocyte donors. Gir breed cows (n=20) used in different experiments involving the ovum pick-up technique were evaluated. At the end of each experimental period the animais were examined by rectal palpation, ultrasonography and vaginoscopy. A group of cows (n=8) were selected for histopathologic characterization of ovarian lesions. Ovaries were surgically removed, fixed and processed for histological examination. The cows evaluated underwent 9 to 42 puncture sessions, and yielded 10 to 719 oocytes. There was a low prevalence (5.26%) of inflammatory changes in the vagina mucosa. The occurrence of adherence was significantly correlated with the number of puncture sessions the animal underwent (r=0.31; P<0.05), but not with the total of oocytes collected (r=0.03; P>0.05). The opposite was observed reganding ovarian fibrosis, which showed a high correlation with oocyte yield (r=0.53; P<0.05). Histopathologic evaluation revealed scar tissue, inflammatory cell infiltration and presence of luteal tissue dispersed within the ovarian stroma.

Foram examinadas vacas da raça Gir (n=20) que participaram de diferentes experimentos envolvendo a técnica de punção folicular. Ao término de cada período experimental, os animais foram examinados por palpação retal, ultra-sonografia e vagi-noscopia. Um grupo de vacas (n=8) foi selecionado para a caracterização histopatológica das lesões ovarianas. Os ovários foram retirados cirurgicamente, fixados e processados, e as lâminas obtidas coradas e analisadas. Fóram avaliados animais submetidos a entre nove e 42 sessões de punção, e que produziram entre 10 e 719 oócitos. Observou-se uma baixa prevalência (5,26%) de processos inflamatórios na mucosa do fundo de saco vaginal. A ocorrência de aderências apresentou uma correlação positiva com o número de sessões de punção a que o animal foi submetido (r=0,31; P<0,05), mas não com o total de oócitos recuperados (r=0,03; P>0,05). Tendência oposta foi observada em relação à fibrose ovariana, que apresentou uma alta correlação com o número de oócitos recuperados (r=0,53; P<0,05). A avaliação histopatológica demonstrou a presença de tecido cicatricial, infiltrados de células inflamatórias e presença de tecido luteal disperso no estroma ovariano.

E-mail: pvpeixoto@uol.com.br The reproduction performance of about 1,200 beef cows, raised on pasture and given three different mineral supplements, was evaluated during 5 years in Cássia county, Minas Gerais, Brazil. The cows were divided into three groups: For one group (A) a comercial mineral mixture was used in 1997 and 1998; for the second group (B) common salt and dicalcium phosphate 1:1 was offered in 1999; and the third group (C) received only common salt during the sarne year. ln 2000 and 2001, ali the cows received exclusively common salt. There was no decrease in i:eproduction efficiency when only common salt was given; on the contrary, the reproduction performance of the inseminated e pregnant cows, thát was respectively 92.5 and 78.2 % (1997) and 92.2 and 80.5 % (1998), increased to 94.5 and 85.7 % (2000) and 96.7 and 89.7 % (2001). The decrease of reproduction performance of the cattle in 1999 was probably due to the severe drought during this year. The authors emphasize the importance of a selective mineral supplementation, i.e. to supplement cattle with mineral mixtures including only the elements which the animais can not obtain from the pasture. Theselective mineral supplementation requires the assistance of professionals with training and experience in clinics, pathology and nutrition of ruminants. These professionals should be able to accompany and evaluate the nutritional condition of a herd when a selective mineral mixture is used. Selective mineral supplementation could represent a two or threefold economy of the total cost of conventional supplementation with "complete" mineral mixtures. The authors discuss ifthe improvement of the reproduction performance of the cattle used in this fiel d experiment was due to a better management of the herd or to possible antagonism between the minerais.

Enquanto que a mistura A (sal mineral comercial) foi oferecida nos anos de 1997 e 1998, a mistura B (cloreto de sódio 50% e fosfato bicálcico 50%) foi disponibilizada, em 1999, para as matrizes mantidas em um dos setores da fazenda e a mistura C (apenas cloreto de sódio), foi oferecida, neste mesmo ano, para os lotes cria dos em outros dois setores. Nos anos de 2000 e 2001, a mistura C foi ofertada a todas matrizes. Não houve queda na eficiência reprodutiva pela suplementação exclusiva com cloreto de sódio, pelo contrário, as percentagens de matrizes inseminadas e matrizes prenhes, que eram, respectivamente, de 92,5 e 78,2 (1997) e 92,2 e 80,5 (1998) alcançaram níveis de 94,5 e 85,7 (2000) e 96,7 e 89,7 (2001). A redução nos índices de matrizes inseminadas e de prenhez verificadas no ano de 1999 foi atribuída à baixa precipitação pluviométrica registrada naquele ano. Os autores ressalvam, porém, que a suplementação seletiva, isto é, aquela baseada na exclusiva administração do (s) mineral (is) que está (ão) faltando em uma determinada fazenda, só deve ser implantada mediante avaliação e acompanhamento clínico-nutricional do rebanho, o que exige apoio de profissional com adequados conhecimentos sobre nutrição, deficiências minerais e clínica de ruminantes. Por outro lado, a suplementação seletiva, pode representar uma despesa 2 a 3 vezes menor do que a verificada com a "mineralização" convencional do rebanho. Discute-se se a melhoria dos índices reprodutivos deveu-se à adequação do manejo ou à cessação de possível antagonismo entre os minerais.

The susceptibility of antimicrobials was studied in Gram positive and catalase negative cocci (21 samples of Lactococcus garvieae and 6 Enterococcus gallinarum), isolated from the milk of cows with subclinical mastitis, belonging to six buffalo herds in the State of Rio de Janeiro. The test used was diffusion of disks in agar Müller Hinton, according to recommendations of the National Committee for Clinicai Laboratory Standards - NCCLS. There were tested disks with ampicillin (10mg), cefalotin (30mg), cefotaxime (30mg), cefoxitin (30mg), doranfenicol (30mg), eritromycin (15mg), gentamycin (10mg), nitrofurantoin (300mg), norfloxacin (10mg), penicillin (1 O IU), tetracydin (30mg) and vancomycin (30mg). The results showed that with Lactococcus garvieae, the most efficient antimicrobial was nitrofurantoin, revealing 85.71% sensibility, followed by cefotaxime (61.90%), vancomycin (52.38%), norfloxacin (47.62%) and cefalotin (47.62%). The highest resistance was developed against penicillin and ampicillin, with 95.24% resistance for the two antimicrobials. The susceptibility profile developed by the strains of Enterococcus gallinarum showed low sensibility against the tested antimicrobials; the highest resistance observed was against eritromycin and gentamycin, with 33.34% sensibility for both. The antimicrobial evaluation showed 100% resistance against vancomycin and tetracyclin, followed by cloranfenicol, penicillin, ampicillin, cefoxitin, cefotaxim, norfloxacin and nitrofurantoin; all of them showed a resistance of 83.33% with the samples tested.

Estudou-se o perfil de susceptibilidade a antimicrobianos em cocos Gram-positivos catalase negativos (21 amostras de Lactococcus garvieae e 6 de Enterococcus gallinarum), isoladas do leite de fêmeas com mastite subdínica e pertencentes a uma população composta por seis rebanhos bubalinos localizados no Estado do Rio de Janeiro. O teste utilizado foi o da difusão de discos em agar Müller Hinton, segundo recomendações do National Committee for Clinicai Laboratory Standards - NCCLS, tendo sido testados discos com ampicilina (10mg), cefalotina (30mg), cefotaxima (30mg), cefoxitina (30mg), cloranfenicol (30mg), eritromicina (15mg), gentamicina (10mg), nitrofurantoína (300mg), norfloxacina (10mg), penicilina (10 UI), tetracidina (30mg) e vancomicina (30mg). Os resultados evidenciaram que em se tratando de Lactococcus garvieae, o antimicrobiano mais eficiente foi o nitrofurantoína com 85,71% de sensibilidade, seguido da cefotaxima (61,90%), vancomicina (52,38%), norfloxacina (47,62%) e cefalotina (47,62%). A maior resistência foi desenvolvida frente a penicilina e ampicilina, com 95,24% de resistência para os dois antimicrobianos testados. O perfil de susceptibilidade desenvolvido pelas amostras de Enterococcus gallinarum, mostrou baixa sensibilidade frente aos antimicrobianos testados, onde os maiores índices foram observados frente eritromicina e gentamicina, com 33,34% de sensibilidade para ambos; quanto à resistência desenvolvida, foi possível observar 100% de resistência com relação a vancomicina e tetraciclina, seguindo-se cloranfenicol, penicilina, ampicilina, cefoxitina, cefalotina, cefotaxima, norfloxacina e nitrofurantoína, todas evidenciando uma resistência de 83,33% das amostras testadas.

E-mail: diomedes@ufpa.br Eight lactating buffaloes of the Murrah breed were used from 60 days before up to 60 days after calving, in order to evaluate alterations in the concentration of blood glucose, total plasmatic protein, haematocrit and the presence of ketonic bodies in the urine. The 5 to 10-year old buffaloes were clinically healthy and had had at least two lactations. During the experimental period the animais were maintained on Brachiaria brizantha pasture and · supplemented with a mineral mixture and water. The media concentrations of blood glucose and the media values of the haematocrit diminished significantly after calving (p<0,05). The concentrations of total plasmatic protein did not show significant variation during the pre and post-parturient period. The ketonic bodies in the pre-parturient period were only detected in the urine of one buffaloe, but after the 32nd day of lactation ketonic bodies were detected in ali animais. There was a direct relationship between the color of the urine positive by the Rothera test and blood glucose concentrations. lt can be concluded that at the beginning of lactation the buffaloes had an energetic deficit, characterized by a decline of blood glucose concentrations and the presence of ketonic bodies in the urine, and that lactation caused a progressive decline of the hematocrit, but that the concentration of total plasmatic protein did not vary during the pre and post-parturient period.

As concentrações médias de glicose sangüínea e o valor médio do hematócrito diminuíram significativamente no pós-parto (p<0,05). As concentrações de proteína plasmática total não sofreram variações significativas do pré para o pós-parto. No período préparto os corpos cetônicos só foram detectados na urina de uma búfala; entretanto, a partir do 32º dia de lactação foram detectados em todos os animais. Houve uma relação direta entre a coloração da urina positiva para o teste de Rothera e as concentrações de glicose sangüínea. Pode-se concluir que na fase inicial da lactação as búfalas utilizadas sofreram um déficit energético, caracterizado pela diminuição nas concentrações sangüíneas de glicose e presença de corpos cetônicos na urina, e que a lactação causou um declínio progressivo no hematócrito, enquanto que as concentrações da proteína plasmática total não sofreram variações do pré para o pós-parto.

The aim of the present work was to evaluate the association among the kinetics of humoral immune response, the recovery ofviable leptospiras and the intensity ofthe tissue lesions in mice selected for high (H) and low (L) production of antibodies (selection N-A) and in mice from the outbreed Balb/c Iine, inoculated with pathogenic Leptospira interrogans serovar icterohaemorrhagiae. Toe H and L lines showed modifications in some steps ofthe immune response, mainly in relation to the macrophagic activity, representing extreme phenotypes found in heterogeneous natural populations. Mice were sacrificed in eight moments after the infection. Analysis of the results revealed that from the 7th day after the infection, on Iine H mice presented antibodies titles significantly higher as compared to the L line mice, maintaining the multispecific effect Balb/c tine mice showed intermediàte results between the two Iines. Antibodies production worked as a timiting factor to the infection, becausewhen a greater leptospiras recovery was obtained, at the initial phase of the infection, the titles of antibodies were elevated. Inflammatory and degenerative process led to similar lesions in the organs ofinfected mice. Only a variation in the degree oftissue compromising was observed according to the tine. H tine mice showed more extensive lesions than L and Balb/c lines mice. For the Balb/c tine mice, the lesions were moderate. As a mie, H and Balb/c mice lines showed a Th2 profile of immune response, with higher indexes of antibody production and gravity ofthe lesions, while L line mice presented a Th 1 profile ofimmune response.

O presente trabalho teve por finalidade associar a cinética da resposta humoral à recuperação de leptospiras viáveis e à intensidade das lesões teciduais em camundongos geneticamente selecionados para bons (High) e maus (Low) produtores de anticorpos (seleção IV-A), além de camundongos outbreed, Balb/c, inoculados com amostra patogênica de Leptospira interrogans sorovar icterohaemorrhagiae. As linhagens High e Low (seleção IV-A) apresentam modificações em alguns compartimentos da resposta imune, principalmente em relação à atividade macrofágica, representando fenótipos extre mos encontrados em populações naturais heterogêneas. Os camundongos foram sacrificados em oito momentos após a infecção. A análise dos resultados revelou que a partir do 7º dia após a infecção, os camundongos da linhagem High apresentaram elevação nos títulos de anticorpos estatisticamente significantes quando comparados aos camundongos da linhagem Low, mantendo assim o efeito multiespecífico. Os camundongos Balb/c apresentaram resultados intermediários entre as duas linhagens. A produção de anticorpos colaborou como fator limitante à infecção, pois quando obtevese maior recuperação de leptospiras, na fase inicial da infecção, os títulos de anticorpos encontravam-se em elevação. As lesões observadas nos órgãos de camundongos infectados consistiram basicamente nos mesmos processos inflamatórios e degenerativos, que não se alteraram, variando apenas o grau de comprometimento tecidual, de acordo com a linhagem. A linhagem high apresentou lesões mais extensas que as apresentadas pelas linhagens low e Balb/c, sendo que nesta última as lesões foram moderadas. De forma geral a linhagem High e Balb/c apresentaram um perfil de resposta Th2, com o maior índice de produção de anticorpos e gravidade das lesões, enquanto a linhagem Low apresentou um perfil de resposta Th 1.
