E-mail: rseabra@cevap.org.br The study evaluated and compared the aerobic microbiota from the oral cavity, cloaca and venom of Crotalus durissus terrificus snakes, recently caught from the wild and kept under quarantine (WQ), individual captivity (IC) and collective captivity (CC). Antimicrobial drug effectiveness on isolated agents also was assayed. From group I, II and III were isolated, respectively, 29 (63.04%), 38 (90.48%) and 21 (42.86%) microorganisms from the cloaca; 15 (32.61%), 3 (7.14%) and 25 (51.02%) microorganisms from the oral cavity; and, 2 (4.35%), 1 (2.38%) and 3 (6.12%) microorganisms from venom. The most frequent bacteria were Pseudomonas aeruginosa, Proteus vulgaris and Morganella morganii, with sensitivity to amikacin, gentamicin, norfloxacin, sulfazotrin and tobramycin. Snakes kept in semi-open captivity exhibited the fewest microorganisms in oral cavities, perhaps due to the environment in captivity, with different temperature gradients, running water, absence of daily handling, circulating air, possibility of moving around, daily cleaning, and sunlight access.

E-mail: rseabra@cevap.org.br Este estudo avaliou e comparou a microflora aeróbica da cavidade oral, cloaca e veneno de serpentes Crotalus durissus terrificus recém-capturadas da natureza e mantidas sob quarentena (WQ), mantidas em cativeiro coletivo (CC) e em cativeiro individual (IC). A eficácia de drogas antimicrobianas de agentes isolados foi também avaliada. Foram isolados microorganismos dos grupos I, II e III respectivamente: 29 (63.04%), 38 (90.48%) e 21 (42.86%) da cloaca; 15 (32.61%), 3 (7.14%) e 25 (51.02%) da cavidade oral, e finalmente 2 (4.35%), 1 (2.38%) e 3 (6.12%) do veneno. As bactérias mais frequentes foram Pseudomonas aeruginosa, Proteus vulgaris e Morganella morganii, com sensibilidade para amikacina, gentamicina, norfloxacina, sulfazotrina e tobramicina. Serpentes mantidas no cativeiro semi-aberto mostraram menor número de agentes infecciosos em cavidade oral, talvez devido ao ambiente de cativeiro com diferentes gradientes de temperatura, água corrente, ausência de manejo diário, ampla circulação de ar, possibilidade de movimentação pelos animais, limpeza diária e acesso ao Sol.

E-mail: hlangoni@fmvz.unesp.br Nowadays, farmers have great interest in milk production within an organic system due to the consumers’ claim for natural products. The purpose of this paper was to study the cytological and microbiological quality of milk produced by four farms in the county of Botucatu, SP, which use organic system management. A total of 150 mammary gland milk samples were tested by CMT and Somatic Cell Count (SCC/mL milk), microbiological examination, and Colony Former Unit Count (CFU mesophile microrganisms/mL milk). From these individual milk samples, 66 (44.0%) were CMT positive. Corynebacterium bovis was isolated from 37.9% milk samples, Staphylococcus aureus from 18.2%, S. epidermidis from 15.2%, Streptococcus uberis from 3.0%, and S. dysgalactiae from 3.0%. Association of at least two different bacteria species was observed in 7.6% samples. In respect to the whole milk of each farm, the SCC/mL values were in normal limits on three of them (i.e. lower than 400x103), and considering the CFU/mL, on three farms high levels were observed (8,5x105; 1,5x106; 4,1x105). From the individual milk samples, contagious mastitis pathogens were isolated predominantly, but environmental microorganisms were detected from the whole milk, as Escherichia coli and Pseudomonas aeruginosa, probably due to contamination during and/or after milking. The results suggest that more than mastitis problems, the four farms studied have hygienic deficiencies, what strengthens the importance of sanitary education to achieve a better milk hygiene quality.

E-mail: hlangoni@fmvz.unesp.br A produção de leite no sistema orgânico tem despertado o interesse dos produtores rurais, pelo aumento de consumo de produtos naturais. Estudaram-se os aspectos citológicos e microbiológicos do leite no sistema orgânico de produção em quatro propriedades no município de Botucatu, SP, utilizando métodos como CMT, exame microbiológico das amostras positivas, contagem de células somáticas (CCS/mL de leite) e Contagem de Unidades Formadoras de Colônias (UFC de microrganismos mesófilos/mL de leite) em amostras individuais de leite em animais com pelo menos um teto positivo ao CMT. Foi também realizado a CCS/mL de leite e UFC/mL de leite, e exame microbiológico de amostras de leite do conjunto (tanque) de cada propriedade. Das 150 glândulas mamárias examinadas, 66 (44,00%) amostras foram positivas ao CMT, com isolamento de Corynebacterium bovis em 37,90%, Staphylococcus aureus (18,20%), S. epidermidis (15,20%), Streptococcus uberis (3,00%) e S. dysgalactiae (3,00%), e isolamento de mais de um agente bacteriano em 7,60% das amostras. Os valores de CCS/mL das amostras do leite de conjunto estiveram dentro dos limites de normalidade em três das quatro propriedades (< 400x103), por outro lado considerando a UFC/mL em três das quatro propriedades observou-se altos índices (8,5x105; 1,5x106; 4,1x105). Obteve-se o isolamento de microrganismos ambientais, como Escherichia coli e Pseudomonas aeruginosa, sugerindo a contaminação do leite durante ou após a ordenha, o que reforça a importância de atividades de educação sanitária para obtenção higiênica do leite.

Pesquisa Veterinária Brasileira 29(11):899-904. Setor de Patologia Veterinária, Faculdade de Veterinária, Universidade Federal do Rio Grande do Sul, Av. Bento Gonçalves 9090, Porto Alegre, RS 91540-000, Brazil. E-mail: davetpat@ufrgs.br This retrospective study included clinical and pathological findings from 15 cattle affected by rabies. Thirteen of the 15 cases were confirmed by direct immunofluorescence. Cattle between 4 months and 8 years of age were affected. Clinical course ranged from 3 to 7 days. Paralytical form was the most common clinical picture and included incoordination, paresis, and paralysis of the pelvic members, besides recumbence, paddling, and death. The main histopathological findings were lymphoplasmacytic meningoencephalitis associated with characteristic Negri bodies in 86.6% of the cases. All cases showed anti-rabies immunostaining, which were most prominent in the brainstem including medulla oblongata, pons, and midbrain, besides trigeminal ganglion. Positive labeling was present within axons, dendrites, and perikarya of neurons as aggregates of granules or round formations associated with varying numbers of inclusion bodies. Immunostaining was also observed in the Purkinje neurons and their processes in the molecular layer, in the neurons of of the brainstem, and deep layer of the telencephalic cortex. Immunohistochemistry may be an important auxiliary tool for the diagnosis of rabies, especially in circumstances in which refrigeration cannot be adequately maintained, and in cases characterized by nonsuppurative meningoencephalitis with absence of inclusion bodies.

A forma paralítica foi a mais frequente e incluiu incoordenação, paresia e paralisia dos membros pélvicos, decúbito, movimentos de pedalagem e morte. Os principais achados histopatológicos foram meningoencefalite linfoplasmocitária associada com corpúsculos de Negri em 86,6% dos casos. Todos os casos foram positivos na imuno-histoquímica para raiva, cujas reações foram mais evidentes no tronco encefálico, incluindo bulbo, ponte e mesencéfalo, além de gânglio trigêmeo. A imuno-histoquímica demonstrou o vírus da raiva em axônios, dendritos e pericário de neurônios, como agregados de grânulos ou em formações arredondadas associadas com números variáveis de corpúsculos de inclusão virais nos neurônios. Houve também marcação nos neurônios de Purkinje e de seus processos na camada molecular, nos núcleos do tronco encefálico e camada profunda do córtex telencefálico. A imuno-histoquímica pode ser importante ferramenta diagnóstica no diagnóstico da raiva, especialmente em situações nas quais não é possível manter refrigeração adequada das amostras e em casos com meningoencefalite não-supurativa e ausência de corpúsculos de inclusão.

E-mail: alschild@terra.com.br An outbreak of cutaneous pythiosis is described in cattle from southern Rio Grande do Sul, Brazil. Cattle were introduced into flooded pastures and developed ulcerative and wet cutaneous lesions on distal limbs and Planum nasale. Histologically, the lesions were characterized by multiple granulomas with intralesional hyphae, better seen in the methenamine silver stain, and surrounded by abundant fibrous tissue. Diagnosis was based on epidemiology, gross and histological lesions, and by positive immuno-histochemical reaction with anti-Pythium insidiosum polyclonal antibody. Morbidity was 23.8% and lesions were observed 15-90 days after cattle were introduced in the flooded area. Affected cattle had spontaneous healing without treatment. It is suggested that the disease is more frequent than what was earlier thought, mainly in flooded areas of the Southern region of the Rio Grande do Sul State.

Os animais foram introduzidos em área alagada e desenvolveram lesões cutâneas ulcerativas e úmidas, de tamanhos variados, localizadas na região distal dos membros e no chanfro nasal. Histologicamente, as lesões eram caracterizadas por múltiplos granulomas com hifas intralesionais, melhor observadas pela coloração de metenamina nitrato de prata de Gomori, e circundadas por abundante tecido conjuntivo fibroso. O diagnóstico foi realizado com base na epidemiologia, lesões macroscópicas e histológicas e pela reação imuno-histoquímica positiva com anticorpo policlonal anti-Pythium insidiosum. A morbidade foi de 23,8% e os animais adoeceram 15-90 dias após a introdução na área alagada. Em todos os casos as lesões evoluíram para a cura sem tratamento. Sugere-se que a doença possa ser mais frequente em bovinos do que se supõe, principalmente em áreas alagadas da região Sul do Rio Grande do Sul.

E-mail: erika.branco@ufra.edu.br Buffalo is an important livestock resource, with a great participation in agricultural systems, providing milk, meat, and work power. Umbilical cord is responsible for maternal-fetal nutrients exchange during pregnancy, and its alterations can compromise the fetal development. We investigated ten pregnant uteruses collected from cross-bread buffaloes in different stages of gestation. Pregnancy and fetal age was determined by measuring the apex sacral length and development period was calculated by previously published formula. Umbilical cords were measured for length determination. Umbilical cord vascular net and anastomosis were observed by injection of Neoprene latex. Histological sections of the umbilical cord were studied after stain with HE, picrossirius, toluidine blue, orceine, and PAS reaction. Buffaloes’ umbilical cord was formed by two central arteries, an allantois duct and two peripheral veins. The artery wall was composed by large quantity of collagen, elastic fibers, fibroblasts and large number of vasa vasorum. The allantois duct was located between the arteries and presented a great number of small nourishing vessels. Small nourishing vessels should be carefully considered to avoid to be mistaken to the arterials and veins vasa vasorum. Medium length of umbilical cord from buffalos was 11.8cm (minimum of 6.8cm and maximum of 17.4cm).

O Cordão umbilical é responsável pela troca de nutrientes materno-fetais durante a gestação, e suas alterações podem comprometer o desenvolvimento fetal. Nós investigamos dez úteros gravídicos de búfalos de raças cruzadas em fases diferentes de gestação. O período de gestação e a idade fetal foram determinados pelo comprimento ápice sacral, aplicando fórmulas previamente estabelecidas. Posteriormente mediu-se o comprimento do cordão umbilical. A rede vascular do cordão umbilical e anastomoses foram observadas por injeção ou látex de neoprene. O cordão umbilical foi estudado a partir de cortes histológicos, corados por HE, picrossirius, azul de Toluidina, orceína e reação histoquímica de PAS. O cordão umbilical de búfalos é formado por duas artérias centrais, ducto alantóide e duas veias periféricas e apresentam forma de ampulheta. A parede da artéria umbilical é composta por grande quantidade de fibras colágenas e elásticas, fibroblastos e um grande número de vasa vasorum. O ducto alantóide fica alocado entre as artérias e apresenta um grande número de pequenos vasos nutritivos. Os vasos nutritivos devem ser cuidadosamente identificados para evitar-se confundi-los com vasa vasorum. O comprimento médio do cabo de cordão umbilical dos búfalos era 11.8cm (mínimo de 6.8cm e máximo de 17.4cm).

[Physical, biochemical and cytological characteristics of the equine temporomandibular joint synovial fluid.] Características fisicoquímicas e citológicas do líquido sinovial da articulação temporomandibular em eqüinos. Pesquisa Veterinária Brasileira 29(10):829-833. Hospital Veterinário de Grandes Animais, Faculdade de Agronomia e Veterinária, Universidade de Brasília, Brasília, DF 70636-200, Brazil. E-mail: robertagodoy@unb.br Foram estudadas as características fisicoquímicas e citológicas do líquido sinovial da articulação temporomandibular de dez eqüinos hígidos. Verificou-se que o líquido é viscoso, amarelo claro a citrino, límpido e livre de partículas à temperatura ambiente. Houve contaminação da amostra por sangue em três amostras que se apresentaram amarelo avermelhadas a vermelhas e de aspecto turvo. A taxa de glicose variou entre 100 e 250 e a concentração protéica não ultrapassou 3,8g/dL. O número médio de células nucleadas foi de 417 células/µL, com predominância de grandes células mononucleares e linfócitos. As mensurações das características pesquisadas no líquido sinovial da articulação temporomandibular de eqüinos são de execução simples e passíveis de implantação na rotina de atendimentos clínico-cirúrgicos.

E-mail: robertagodoy@unb.br Physical, biochemical and cytological characteristics of the temporomandibular joint synovial fluid were studied in ten clinically normal horses. It is a viscous, pale yellow, clear fluid and without flocculent material at room temperature. There was blood contamination in three samples, they presented red-yellow to red and cloudy. The range of glucose levels were 100 to 250 and its protein concentration was up to 3,8g/dL. Nucleated cells mean number was 417 cells/µL, with predominating large mononuclear cells and lymphocytes. Equine temporomandibular synovial fluids can be easily evaluated, being feasible in clinical and surgical routine, and the information may be useful to the diagnosis, treatment and prognosis of animals with temporomandibular alterations.


#527 - Intoxicações natural e experimental por salinomicina em coelhos, 29(9):695-699

ABSTRACT.- Peixoto P.V., Nogueira V.A., Gonzaléz A.P., Tokarnia C.H. & França T.N. 2009. Accidental and experimental salinomycin poisoning in rabbits. Pesquisa Veterinária Brasileira 29(9):695-699. Projeto Sanidade Animal Embrapa/UFRRJ, Seropédica, RJ 23890-000, Brazil. E-mail: pfpeixoto@terra.com.br An outbreak of salinomycin poisoning in rabbits is described. At least 27 out of 2,000 rabbits reared on a farm died after the coccidiostatic drug sulfaquinoxaline was substituted by salinomycin in the feed. An average of 26.9ppm salinomycin was detected in the ration given to the rabbits. Clinical signs included anorexia, apathy and bradykinesia, which progressed to incoordination and recumbency. Gross lesions consisted of pale areas in the skeletal muscles. The histopathological findings showed severe necrotic degenerative myopathy in association with infiltration of neutrophils and macrophages. One rabbit exhibited similar alterations in the myocardium. Mineralization was observed in the affected skeletal muscles in some cases. In order to verify if the poisoning was due to salinomycin, 20 rabbits were divided into five groups and a ration containing the drug at doses of 10, 25, 50, 75 and 100ppm was given. The administration of doses higher than 50ppm resulted in manifestation of the clinical signs seen in the outbreak of poisoning. It was concluded, that probably an error related to the mixture of salinomycin in the feed was the cause of deaths in the spontaneous outbreak of poisoning on the rabbit farm.

E-mail: pfpeixoto@terra.com.br Relata-se, pela primeira vez, um surto de intoxicação por salinomicina em coelhos. De 2000 animais, no mínimo 27 morreram após troca do coccidiostático sulfaquinoxalina pela salinomicina. A análise de parte da ração detectou 26,9ppm de salinomicina. Os sinais clínicos observados foram anorexia, apatia e lentidão com evolução para incoordenação dos movimentos e decúbito. As lesões macroscópicas consistiram de áreas pálidas na musculatura esquelética. O exame histopatológico evidenciou miopatia degenerativo-necrótica. Adicionalmente, verificou-se reação inflamatória constituída por neutrófilos e macrófagos. Um coelho apresentou lesões similares no miocárdio. Em alguns casos, mineralização estava presente nos músculos esqueléticos afetados. Vinte coelhos experimentais foram divididos em 5 grupos que receberam 10, 25, 50, 75 e 100ppm de salinomicina por via oral, com a finalidade de reproduzir a intoxicação. Os animais que receberam a partir de 50ppm de salinomicina apresentaram sinais clínicos semelhantes aos observados no surto espontâneo. Nossos resultados indicam que, provavelmente, erro na mistura da substância à ração causou a morte dos coelhos.

E-mail: glaukommers@yahoo.com Four suspect and two confirmed cases of gastrointestinal pythiosis were found in a retrospective study of biopsy and necropsy cases of dogs received in the Laboratório de Patologia Veterinária at the Universidade Federal de Santa Maria. The two diagnosed and published cases have had the etiology confirmed by culture and zoosporogenesis induction or by nested-PCR. On this study, the etiologic diagnosis of four suspect cases was confirmed by immunohistochemistry using anti-Pythium insidiosum policlonal antibody. Epidemiology, clinical signs, gross and microscopic lesions, histochemistry, immunohistochemistry, and differential diagnoses are reported and discussed.

Os dois casos diagnosticados e publicados tiveram a etiologia confirmada através da cultura e indução de zoosporogênese ou por nested-PCR. Neste estudo utilizou-se a técnica de imuno-histoquímica com anticorpo policlonal anti-Pythium insidiosum para confirmação da etiologia dos quatro casos suspeitos. A epidemiologia, sinais clínicos, lesões macroscópicas e microscópicas, características histoquímicas e imuno-histoquímicas e diagnósticos diferenciais são relatados e discutidos.

A plaque reduction neutralization test (PRNT) was applied to search for antibodies to Orthopoxvirus; the vesicular/pustular fluids and scabs were examined by PCR, electron microscopy (EM) and by inoculation in VERO cells for virus isolation. Antibodies to Orthopoxvirus were detected in most cases. The PCR test indicated a high nucleotide homology among the isolates and the vaccinia viruses (VACV) used as controls. By EM, typical orthopoxvirus particles were observed in some specimens. The agents isolated in tissue culture were confirmed as vaccinia-like viruses by EM and PCR. The HA gene of the vaccinia-like Cantagalo/IOC virus isolated in our laboratory was sequenced and compared with other vaccinia-like isolates, showing high homology with the original Cantagalo strain, both strains isolated in 1999 from dairy cattle. Antibodies to Orthopoxvirus were detected in one wild rodent (genus Akodon sp.) collected in the northwestern region of the state, indicating the circulation of poxvirus in this area. Nonetheless, PCR applied to tissue samples collected from the wild rodents were negative. Vesicular/pustular lesions in people in close contact with animals have been also recorded. Thus, the vaccinia-like virus infections in cattle and humans in the state seem to be an expanding condition, resulting in economic losses to dairy herds and leading to transient incapacitating human disease. Therefore, a possible immunization of the dairy cattle in the state should be carefully evaluated.

The least efficient was ampicillin, which was resistant to 56.5% of the isolates taken from region A, 72.8% from region B, and 71.8% from region C. These results indicate the need for periodic testing of sensitivity in vitro, as these variations can compromise the treatment of animals as well as control programs for bovine mastitis caused by Staphylococcus spp.
