E-mail: zep.filho@hotmail.com An experimental model of bovine pneumonic mannheimiosis (BPM) was used to evaluate the nasal and nasopharynx bacterial species of calves during the course of the disease and for checking the diagnostic efficiency of nasal swab (NS) and nasopharingeal swab (NPS) microbiological exams. A total of 28 calves were randomized into four experimental groups (G1-G4). NS and NPS were obtained 7 days before and 12 (G1), 24 (G2), 48 (G3) e 72 (G4) hours after intrabronchial inoculation of Mannheimia haemolytica. After the induction of BPM, M. haemolytica biotype A was the predominant isolated bacterium in NS and NPS in all evaluated sampling times, except for one NS (harvested 24 hours). There were no significant statistical differences for the rates of Pasteurella multocida isolation in NS and NPS, harvested before and after the induction of BPM. However, this bacterium was isolated more frequently after the induction of BPM, mainly in NPS. Therefore, the microbiological NS and NPS exams were important auxiliary tests for diagnosing BPM.

E-mail: riet@cstr.ufcg.edu.br An outbreak of nutritional muscular dystrophy is reported in the semiarid region of northeastern Brazil affecting 3-4 months old Dorper sheep. The animals, weighing 30-40 kg, were fed ad libitum with milk, concentrated ration, Tifton hay, and a mineral mixture. Six out of 70 lambs were affected and died in the first 48 hours after the onset of the outbreak. Clinical signs were fever of 40-41ºC, incoordination followed by paralysis and recumbence, depression, prostration with decreased pupillary and corneal reflexes, decreased tonus of the tongue and maxilla, salivation, submaxillar edema, and increased cardiac and respiratory rates. Death occurred after a clinical manifestation period of 6-12 hours. At necropsy of three animals, skeletal muscles were pale, the liver was yellowish and enlarged, the parotid, submaxillary, retropharyngeal, prescapular and mediastinal lymph nodes were enlarged with red surface, and red areas were observed on the lung surface. On histology, segmental muscular necrosis was observed in all skeletal muscles examined. The liver had centrilobular fatty degeneration, and congestion was observed in the lung and lymph nodes. From 48-96 hours after the begin of the outbreak, another 3 animals were affected. They were treated with Vitamin A, E and D complex; two of them died and one survived. On the same farm, a flock of 20 Santa Inês sheep of the same age as the affected animals, fed with the same food, but no milk, and another 900 sheep of different ages were not affected. The over nutrition of sheep with fast growing rates, and the stress caused by two days of water restriction to improve milk consumption had been predisposing factors for the occurrence of the disease. It is also possible that some of the minerals supplemented interfered with selenium availability.

Vaccination is widely applied to minimize losses induced by BoHV-1 infections; however, vaccination of dams during pregnancy with modified live virus (MLV) vaccines has been occasionally associated to abortions. We have previously reported the development of a BoHV-1 recombinant virus, constructed with basis on a Brazilian BoHV-1 (Franco et al. 2002a) from which the gene coding for glycoprotein E (gE) was deleted (gE-) by genetic manipulation. Such recombinant has been previously evaluated in its potential as a differential vaccine (gE- vaccine) that allows differentiation between vaccinated and infected animals. Here, in the first part of the present study, the safety of the gE- vaccine during pregnancy was evaluated by the intramuscular inoculation of 107.4 tissue culture 50 % infective doses (TCID50) of the virus into 22 pregnant dams (14 BoHV-1 seronegative; 8 seropositive), at different stages of gestation. Other 15 pregnant dams were kept as non-vaccinated controls. No abortions, stillbirths or fetal abnormalities were seen after vaccination. Seroconversion was observed in both groups of previously seronegative vaccinated animals. In the second part of the study, the potential of the gE- vaccine virus to spread among beef cattle under field conditions was examined. Four heifers were inoculated intranasally with a larger amount (107,6 TCID50) of the gE- vaccine (to increase chances of transmission) and mixed with other sixteen animals at the same age and body condition, in the same grazing area, at a population density equal to the average cattle farming density within the region (one cattle head per 10,000 m2), for 180 days. All animals were monitored daily for clinical signs. Serum samples were collected on days 0, 30, 60 and 180 post-vaccination. Seroconversion was observed only in vaccinated heifers. These results indicate that, under the conditions of the present study, the gE- vaccine virus did not cause any noticeable harmful effect on pregnant dams and on its offspring and did not spread horizontally among cattle.

Antigenic characterization with a panel of monoclonal antibodies (Mabs) directed to viral glycoproteins (gB, gC, gD and gE,) was performed by an imunoperoxidase monolayer assay (IPMA) on infected cell monolayers. Genomic characterization was carried out by restriction enzyme analysis (REA) of the whole DNA viral genome with Bam HI. The antigenic profile of the eight isolates from Rio Grande do Sul as well as strains NIA-3 and NP were similar. REA analysis revealed that all isolates from Rio Grande do Sul displayed a genomic type II arrangement, a genotype often found in other outbreaks of AD previously reported in other Brazilian states. The results obtained suggest that the eight isolates examined here were similar.

E-mail: adivaldo@ufrrj.br The cinetic of growth of Borrelia burgdorferi was studied during a 3-month period, using the following 8 culture media: (1) rabbit serum BSK, (2) swine serum BSK, (3) swine serum BSK+5 fluorouracil, (4) PMR, (5) CTB, (6) Dubos, (7) Brucella broth and (8) BHI. All media were prepared aseptically and were maintained in culture tubes of 10 ml capacity. For each medium, the inoculum was standardized to contain initially 102 spirochetes for each 0.1 ml of culture. The growth was monitorized by counting the total number of spirochetes in 0.1ml of medium in a dark field microscope, using a 10x30 mm cover slip. For the first 12 days, counting was done each 24 hours, and afterwards once a week during 14 weeks. There occurred growth of B. burgdorferi in all tested media, with the best performance of three of them: BSK with rabbit serum, BSK swine serum + 5 fluorouracil, and CTB medium. Growth of B. burgdorferi was seen from the 4th week on, reaching its maximum within 8-12 weeks, depleting the culture medium after this time. Cystic forms of B. burgdorferi were observed with all tested media.

#46 - Perinatal mortality of lambs in the county of Uruguaiana, Rio Grande do Sul

The following results reflect the lamb mortality rates of two neighbouring flocks of sheep in the district of Uruguaiana, Rio Grande do Sul. Post mortem examination was performed on 200 lambs, in order to establish the causa mortis and its relationship to sex, weight and climatic conditions. Perinatal mortality is a complex problem that does not have one specifc cause, but is the result of a variety of interrelating factors which are believed to differ from flock to flock. In this study, the non-infectious causes were found to be more important than the infectious ones. Based on the criteria used, the mortality was olassified as follows: complex of starvation/exposure to climatic conditions 157 (78.5%); dystocia, 21 (10.5%); predators, 9 (4.5%); stillbirth, 5 (2.5%); traumatism, 5 (2.5%); congenital defects, 1 (0.5%); neonatal infections, 1 (0.5%); without diagnosis, 1 (0.5%). There were a total of 119 males and 81 females. The mean weight of all lambs subrnitted to necropsy was 3,127 g, ranging from 1,400 to 5,100 g. The highest rate of mortality in relation tó weight was recorded in the group weighing between 3,000 and 3,500 g, 31 % of the total number of animals examined.

Estes resultados refletem a situação da mortalidade perinatal que ocorreu em dois rebanhos de propriedades vizinhas no município de Uruguaiana, Rio Grande do Sul, onde foram necropsiados 200 cordeiros no sentido de se tentar estabelecer o diagnóstico da causa mortis e, quando possível, correlacioná-la com o sexo, peso e as condições climáticas. A mortalidade perinatal de cordeiros é um problema complexo que não tem apenas uma causa específica, sendo, pois, o resultado do envolvimento de muitos fatores, os quais, acredita-se, se podem diversificar de rebanho para rebanho. Durante este estudo, as causas não infecciosas foram mais importantes que as infecciosas. Conforme os critérios adotados, a mortalidade ficou distribuída nos seguintes grupos: complexo da inanição/exposição às condições climáticas 157 (78,5%); distocia, 21 (10,5%); predadores, 9 (4,5%); natimortos, 5 (2,5%); traumatismo, 5 (2,5%); sem diagnóstico, 1 (0,5%). No total, foram 119 machos e 81 fêmeas. A média dos pesos de todos os cordeiros necropsiados foi de 3 .127 g, variando de 1.400 a 5.100 g. A maior freqüência de mortalidade em relação ao peso foi entre 3 .000 a 3.500 g, perfazendo 31 % dos animais examinados.
