Pesquisa Veterinária Brasileira 28(8):333-340. Departamento de Cirurgia, Faculdade de Medicina Veterinária e Zootecnia, Universidade de São Paulo, Av. Prof. Dr. Orlando Marques de Paiva 87, São Paulo, SP 05508-900, Brazil. E-mail: cassioaf@usp.br The aim was to evaluate the clinical results of cranial and dorsal acetabular denervation using curettage in dysplastic dogs. Ninty seven dogs without distinction of breed and sex, 1 to 7 years of age, were analyzed for diagnosis and treatment of hip dysplasia, based on physical examination, clinical signs and radiographic findings. For evaluation of results of the surgical denervation technique, clinical examinations were performed preoperatively (initial exam) and postoperatively at days 2, 7, 14, 21, 30, 60 180 and 360. All animals were evaluated for lameness, pain to movement and touch, muscular atrophy degree, pain sensivity to Ortolani’s test and assessment of life quality. The surgical denervation procedure decreases lameness, pain to movement and touch after 2 days of procedure, decreases muscular atrophy after 60 days of procedure, and improves quality of life from the owner’s and veterinarian’s point of view even after 1 year of the treatment. Dorsal acetabular denervation is a feasible surgical technique in treatment of pain secondary to hip dysplasia in dogs, with significant decrease of pain after 2 days of treatment, improvement of quality of life, decrease of lameness, and consequently joyful animals and owners extremely satisfied with the results obtained by the proposed treatment. The surgical technique must include the curettage of nerve fibers from the cranial-dorsal and dorsal region of the acetabular periosteum.

#143 - ELISA com MSP5 recombinante truncada para detecção de anticorpos contra Anaplasma marginale em bovinos, p.301-306

Abstract in English:

ABSTRACT.- Melo E.S.P., Araújo F.R., Ramos C.A.N., Soares C.O., Rosinha G.M.S., Elisei C. & Madruga C.R. 2007. [ELISA based on recombinant truncated MSP5 for detection of antibodies against Anaplasma marginale in cattle.] ELISA com MSP5 recombinante truncada para detecção de anticorpos contra Anaplasma marginale em bovinos. Pesquisa Veterinária Brasileira 27(7):301-306. Embrapa Gado de Corte, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil. E-mail: flabio@cnpgc.embrapa.br The objective of this study was the production and solubilization of recombinant truncated MSP5 of Anaplasma marginale and the evaluation of its performance in an enzyme-linked immunosorbent assay (ELISA), to detect antibodies against the rickettsia in cattle. The fragment of msp5 gene, except the hydrophobic N-terminal region, was amplified by PCR, cloned in pTrcHis-TOPO plasmid and expressed in Escherichia coli. Solubilization of the recombinant protein was evaluated in different pHs and concentrations of urea. The sensibility and specificity of the assay were evaluated with 66 sera from cattle experimentally-infected and 96 sera from cattle free of A. marginale defined by polymerase chain reaction for msp5 gene. Serum samples from 1,666 cattle from Brazil - states of Rio Grande do Sul (73), Mato Grosso do Sul (91), Pernambuco (86), Bahia (314) and Minas Gerais (267), Uruguay (32) and Costa Rica (803), were tested by ELISAs with recombinant truncated MSP5 and with recombinant MSP1a, and the agreement between both ELISAs was calculated. ELISA with recombinant truncated MSP5 protein detected infected animals with sensibility of 96.97% and specificity of 100%. In cattle experimentally-infected, the ELISA detected antibodies from the 12th day post-infection (DPI) to the end of the experiment, at the 37th DPI. The agreement between the ELISAs with truncated MSP5 and MSP1a antigens was 95.67%, with a kappa index of 0.81. Disagreement results showed significative difference (p <0.001). Antibodies for A. marginale were detected in animals of the all the region analyzed. The ELISA with recombinant truncated MSP5 showed a good performance in ELISA for detention of antibodies against A. marginale, with high sensitivity and specificity, representing an important tool for the diagnosis of anaplasmose bovine in epidemiological studies.

ABSTRACT.- Machado T.F.S., Ferrigno C.R.A., Stopiglia A.J. & Pinto A.C.B.C.F. 2007. [Anatomic features of the dorsal venous sinus in skull of brachycephalous dogs.] Revisão anatômica do seio venoso sagital dorsal no crânio de cães braquicéfalos. Pesquisa Veterinária Brasileira 27(5):194-198. Departamento de Cirurgia, Faculdade de Medicina Veterinária e Zootecnia, Universidade de São Paulo, Av. Prof. Orlando Marques de Paiva 87, São Paulo, SP 05508-900, Brazil. E-mail: tfsm@usp.br The dorsal venous sinuses of the Dura mater perform the cerebral and spinal cord drainage to keep homeostasis of the Central Nervous System. Injuries in the cerebral venous network may cause severe deficits, including hemiplegia, hemorrhage, coma and death. The main landmark for neurologic surgery is the Dura mater venous sinus. The aim of this study is to analyse the pathway of the Sinus sagittalis dorsalis in the brachycephalous skull, as the brachycephalous dogs have specific characteristics. The skulls were taken from 8 Boxer dogs and venous perfusion with latex and barium sulfate solution was performed. After the perfusion, radiographies and tomographies were obtained to relate the dorsal sagittal sinus and the skull averages. The cephalic index showed mean values of 91.24±8.34cm, and the cranio-facial index was 2.89±0.23cm. The venous sinus mensuration interface to the skull was: Área = 10.18±4.69mm2; D1 = 11.84±2.35mm; D2 = 19.57±2.61mm; D3 = 17.88±2.31mm; D4 = 25.32±5.68mm e D5 = 24.84±4.40mm.

E-mail: adrianabonfimr@yahoo.com.br The objective was to evaluate conventional and alternative anthelminthic action on the sensibility of gastrointestinal nematodes in goats. Hundred and twenty goats of both sexes were distributed into 10 groups of 24 animals each (12 males and 12 females) and submitted to treatments of specific anthelminthic composition. The commercial drugs used were moxidectin 0.2%, albendazole, levamisol chlorhydrate, ivermectin and an aqueous extract of the purgative potato (Operculina hamiltonii). The reduction test was applied through egg counts per gram of feces and larval culture to evaluate resistance. The fecal samples were collected in the day when medication was accomplished (day base), at 7, 14 and 21 days after treatment. The following results for the reduction of Trichostrongyloidea eggs were obtained: Treatment of the females with Moxidectina reduced the egg counts to 92.8, 88.7 and 89.8%, of the males to 92.6, 96.2 and 98.1%; with Levamisol of the females to 96, 97.1 and 91%, of the males to 85.7, 94,2 and 100%; with Albendazol of the females 65, 60.3 and 75.4%, of the males 88.8, 88.8 and 55.5%; with Ivermectin of the females to 92.2, 68.6 and 70.6%, of the males to 41.7, 73.6 and 59.7%; and with the purgative potato of the females to 31.8, 34.1 and 49.4%, of the males to 61.5, 80.7 and 50%. In larval culture Haemonchus, followed by Bunostomum, Trichostrongylus, and Oesophagostomum, were identified even after treatments.

#146 - Abordagem cirúrgica inédita para denervação acetabular em cães, p.61-63

Abstract in English:

ABSTRACT.- Ferrigno C.R.A., Schmaedecke A. & Ferraz V.C.M. 2007. [A new surgical approach to acetabular denervation in dogs.] E-mail: cassioaf@usp.br The purpose of this study is to demonstrate a new surgical approach for the selective denervation of the sensitive fibers of the acetabular periosteum, for the treatment of hip dysplasia in dogs, conducted in 189 joints. The surgical approach of the cranial and dorso-lateral regions of the acetabular joint in dogs, by moon-shaped incision of approximately 3cm in extension, starting from the greater trocanter of the femur, in direction of the ileum body. After dorsal opening of the middle gluteal muscle, the insertion of the deep gluteal muscle is inserted for the access to the mentioned regions. With the help of a curette, the periosteum of the cranial and dorso-lateral acetabular margin was removed until the bone cortex was exposed, in all patients. In all animals, the approach was possible, both in the cranial as well as in the caudal faces for the access and curettage of the acetabulum, with the technique described, with a skin incision of about 3.2cm. The curettage of the cranial portion of the acetabulum was, in all cases, realized without the visualization of the bone, but of easy achievement and without intercurrences of nervous or muscular injuries. In all the approaches, it was possible to visualize the articular capsule of the coxo-femoral joint, and in none of the animals, this structure was incised for a technique mistake. The surgical approach described in the present study is feasible for this kind of surgical procedure in dogs, promoting adequate access, with minimal invasion and with no complications of any nature.

ABSTRACT.- Ferrigno C.R.A.,Della Nina M.I. & Fantoni D.T. 2007. [A comparative study of osteosynthesis with plates and plates associated with grafts of bone morphogenic proteins (Gen-Techâ) in distal radio-ulna fractures in dogs with less than 6 quilograms.] E-mail: cassioaf@usp.br It is well known that bone morphogenic proteins (BMP) cause osteogenesis, yet clinical research must be performed in order to really show their benefits. Animals weighing less than 6 kg show well known difficulties regarding radius and ulna fracture repair mainly with bone non-union, due to poor vascularization of the distal portion of the radius. Therefore this study aimed to compare the velocity of bone callus formation in the treatment of fracture repair with plates and screws alone or with plates and screws plus BMP. Thirty three dogs with radius and ulna fractures were distributed into two groups, where animals of the control group received the conservative treatment performed with screws and plates alone, whilst the other group received the conservative treatment and BMP. The time of bone callus formation was evaluated comparatively through radiographic exams 30, 60, 90, 120, 180 and 210 days after the surgical procedure. Animals treated with BMP showed a healing time of 32±15 days, which was significantly different (p <0.001) from the control group which required 127±34 days. With the results obtained, it can be concluded that the distal radio-ulna fractures of dogs weighing less than 6 kg suffered a significant reduction of the bone callus formation time, which was around 90 days.

E-mail: analudmv@nin.ufms.br The presumptive diagnosis of bovine tuberculosis is based on analysis of the immune response to micobacterial antigens. This experimental simulation of sensibilization by Mycobacterium bovis and Mycobacterium avium in cattle aimed to verify the immune response by both the cervical comparative test and the evolution of the specific production of gamma-interferon, and also to identify interference of unspecified reactions by M. avium on the test results. The results support that the experimental animals started a response of delayed hypersensitivity to the inactivated bacilli, and that both diagnostic tests for bovine tuberculosis were efficient for the identification of animals sensitized with M. bovis and for discrimination of reactions generated by inoculation of cattle with M. avium.

The results support that the experimental animals started a response of delayed hypersensitivity to the inactivated bacilli, and that both diagnostic tests for bovine tuberculosis were efficient for the identification of animals sensitized with M. bovis and for discrimination of reactions generated by inoculation of cattle with M. avium.

Cel José Soares Marcondes 2357, Presidente Prudente, SP 19013-050, Brazil. E-mail: zampieri@ial.sp.gov.br The immunological reply of a population to an infectious agent can vary between races and handling of this population. Regional research becomes important, in order to know the interrelation between the agent and its host. In this way, the occurrence of immunoglobulins of class G, anti-Babesia bovis and anti-Babesia bigemina in the Nelore (Bos indicus) and Hostein breed (Bos taurus), was investigated in two regions of the State of São Paulo, 300 km distant from each other. For the indirect method of ELISA, 1,161 bovine serum samples were tested. The medium frequencies of antibodies showed that in the two regions exists an enzootic stability for B. bovis in both breeds studied; even so there was a tendency of marginal area for the Nelore breed in one of the regions. Regarding B. bigemina, in both regions exists enzootic stability for the Hostein and enzootic instability for the Nelore breed. Therefore, acute cases of the disease or specific outbreaks by B. bigemina infection in the Nelore breed may occur in these regions.

Departamento de Clínica, Cirurgia e Reprodução Animal, Faculdade de Medicina Veterinária, Universidade Estadual Paulista, Rua Clóvis Pestana 793, Araçatuba, SP 16050-680, Brazil. E-mail: vmflima@fmva.unesp.br A rapid indirect enzyme-linked immunosorbent assay (ELISA) was developed for measuring antibodies against Leishmania chagasi using total antigen from lysed promastigotes. Fifty symptomatic mixed breed dogs from a region of high incidence of visceral leishmaniasis in Brazil were examined. The results showed that in the positive animals, diagnosed by cytological examination, the ELISA using protein A assay system (mean optical density ± SD / 2.078 ± 0.631) detected more antibodies than the anti-IgG assay (mean optical density ± SD / 1.008 ± 0.437), while in the negative animals, the results by both systems were similar. These results suggest that the ELISA assay using protein A peroxidase conjugated could be useful to detect early infected animals in endemic areas, and thus help to control the spread of the infection.
