#131 - Occurrence of virulence-related sequences and phylogenetic analysis of commensal and pathogenic avian Escherichia coli strains (APEC), p.533-540

E-mail: wds@unicamp.br The presence of iron uptake (irp-2, fyuA, sitA, fepC, iucA), adhesion (iha, lpfAO157/O141, lpfAO157/O154, efa, toxB) and invasion (inv, ial-related DNA sequences and assignment to the four main Escherichia coli phylogenetic groups (A, B1, B2 e D) were determined in 30 commensal E. coli strains isolated from healthy chickens and in 49 APEC strains isolated from chickens presenting clinical signs of septicemia (n=24) swollen head syndrome (n=14) and omphalitis (n=11) by PCR. None of the strains presented DNA sequences related to the inv, ial, efa, and toxB genes. DNA sequences related to lpfAO157/O154, iucA, fepC, and irp-2 genes were significantly found among pathogenic strains, where iucA gene was associated with septicemia and swollen head syndrome and fepC and irp-2 genes were associated with swollen head syndrome strains. Phylogenetic typing showed that commensal and omphalitis strains belonged mainly to phylogenetic Group A and swollen head syndrome to phylogenetic Group D. Septicemic strains were assigned in phylogenetic Groups A and D. These data could suggest that clonal lineage of septicemic APEC strains have a multiple ancestor origin; one from a pathogenic bacteria ancestor and other from a non-pathogenic ancestor that evolved by the acquisition of virulence related sequences through horizontal gene transfer. Swollen head syndrome may constitute a pathogenic clonal group. By the other side, omphalitis strains probably constitute a non-pathogenic clonal group, and could cause omphalitis as an opportunistic infection. The sharing of virulence related sequences by human pathogenic E. coli and APEC strains could indicate that APEC strains could be a source of virulence genes to human strains and could represent a zoonotic risk.

By fixing and dissecting pieces, it was found that phrenic nerves (F) frequently come from the ventral branches of the 5th (C5) and 6th (C6) cervical spinal nerves (Ec), at right (46.67%) and at left (43.33%). The F often form a lumbocostal trunk, sternal branches at right (40.00%) and lumbar, costal and esternal branches at left (36.68%). The lumbar branches of F innervate frequently at left (96.67%) the homolateral pillar of the diaphragma, and at right (50.00%) they give fillets to Vena cava caudalis. The costal branches of the F innervate at left (90.00%) and at right (76.66%) the dorsal and ventral regions of the pars costalis. The sternal branches of the F innervate at right (100.00%) and at left (83.33%) the pars sternalis and the ventral region of the pars costalis at the same side. The intercostal nerves (VII to XII pairs, 63.33%) contribute to innervate the diaphragm of Santa Inês sheep.

The expression of cell proliferation markers PCNA and especially Ki-67, has also proved to have a strong prognostic value in this tumor class. The expression of these markers was related to the clinical-pathological characteristics of 73 surgically removed mammary tumors in female dogs by immunohistochemistry. There was no statistical correlation between these markers and death by neoplasm, survival time and disease-free interval. However, the loss of e-cadherin expression and marked Ki-67 expression (p=0.016) were considered statistically significant for the diagnosis (p=0.032). When evaluated as independent factors, there was evidence of the relationship between the loss of e-cadherin expression and high PCNA expression with changes in the body status (divided into obese, normal and cachectic) of female dogs (p=0.030); there was also evidence of the relationship between pseudopregnancy and e-cadherin alone (p=0.021) and for ulceration and PCNA alone (p=0.035). The significant correlation between the markers expression and these well known prognostic factors used individually or in combination suggests their prognostic value in canine mammary tumors.

The sampling was performed in two stages. Blood samples were collected from 1930 females aged ³24 months from 295 non vaccinated herds. Serum samples were tested for antibodies against BoHV-1 using an indirect ELISA. The epidemiological questionnaire was applied on all the selected farms and aimed to obtain epidemiological data. Hundred ninety of the 295 herds were positive for BoHV-1, presenting prevalence of positive herds of 64.41% [58.65-69.87%]. The number (³23) of females aged 24 months (OR=2.22; IC: 1.09-4.51), purchased cattle (OR=2.68; IC: 1.48-4.82), use of common grass (OR=5.93; IC: 1.31-26.82), history of abortion in the last 12 months (OR=2.37; IC: 1.09-5.16) and presence of wildlife animals (OR=8.86; IC: 1.11-70.73) were identified as risk factors for BoHV-1 infection in the multivariate logistic regression. These results indicate that BoHV-1 infection is widespread in the studied region and that factors related to the herd characteristic and management are associated with the infection.

Pesquisa Veterinária Brasileira 28(1):1-14. Centro de Saúde e Tecnologia Rural, Universidade Federal de Campina Grande, Patos, PB 58700-000, Brazil. E-mail: franklin.riet@pq.cnpq.br Plants causing hepatic lesions in ruminants and horses constitute one important group of poisonous plants in Brazil. These plants can be placed in three major groups: plants causing acute liver necrosis; plants causing liver fibrosis; and plants causing hepatogenous photosensitization. For some of these plants the toxic principles are known. Cestrum parqui and Xanthium cavanillesi that cause acute liver necrosis contain carboxy-atractylosides. Senecio spp., Crotalaria spp., and Echium plantagineum that cause liver fibrosis contain pyrrolizidine alkaloids. As for the group of plants causing hepatogenous photosensibilization, Myoporum spp. contain furanosesquiterpenes, Lantana spp contain triterpenes, and Brachiaria spp. and Panicum spp. contain steroidal saponins. The clinical and pathologic features of the toxicosis caused by these phytotoxins, general mechanisms of production for the production of the clinical signs and the methods for diagnosis of hepatic failure in farm animals are reviewed.

Postal 47, Eldorado do Sul, RS 92990-000, Brazil. E-mail: proehe@ufrgs.br Pseudorabies or Aujeszky’s disease (AD), caused by pseudorabies virus (PRV) is a major concern in swine production. In the state of Rio Grande do Sul, Brazil, AD was only detected in 1954, in cattle. In 2003 two outbreaks of encephalitis occurred on the northern region of the state, close to the border with the state of Santa Catarina. Pseudorabies virus (PRV) was isolated from distinct farms within the region and subjected to antigenic and genomic analyses. These isolates were compared with prototype strains NIA-3 and NP. Antigenic characterization with a panel of monoclonal antibodies (Mabs) directed to viral glycoproteins (gB, gC, gD and gE,) was performed by an imunoperoxidase monolayer assay (IPMA) on infected cell monolayers. Genomic characterization was carried out by restriction enzyme analysis (REA) of the whole DNA viral genome with Bam HI. The antigenic profile of the eight isolates from Rio Grande do Sul as well as strains NIA-3 and NP were similar. REA analysis revealed that all isolates from Rio Grande do Sul displayed a genomic type II arrangement, a genotype often found in other outbreaks of AD previously reported in other Brazilian states. The results obtained suggest that the eight isolates examined here were similar.

In the state of Rio Grande do Sul, Brazil, AD was only detected in 1954, in cattle. In 2003 two outbreaks of encephalitis occurred on the northern region of the state, close to the border with the state of Santa Catarina. Pseudorabies virus (PRV) was isolated from distinct farms within the region and subjected to antigenic and genomic analyses. These isolates were compared with prototype strains NIA-3 and NP. Antigenic characterization with a panel of monoclonal antibodies (Mabs) directed to viral glycoproteins (gB, gC, gD and gE,) was performed by an imunoperoxidase monolayer assay (IPMA) on infected cell monolayers. Genomic characterization was carried out by restriction enzyme analysis (REA) of the whole DNA viral genome with Bam HI. The antigenic profile of the eight isolates from Rio Grande do Sul as well as strains NIA-3 and NP were similar. REA analysis revealed that all isolates from Rio Grande do Sul displayed a genomic type II arrangement, a genotype often found in other outbreaks of AD previously reported in other Brazilian states. The results obtained suggest that the eight isolates examined here were similar.

On each farm the animals were divided into two groups. For one group a commercial mineral mixture was used, and to the other group a selective mineral mixture containing only Na, P, Cu and Co was offered. The cattle were weighed every 30 days, and the mineral supplement intake was measured monthly. The daily weight gain of the animals receiving the selective mineral mixture was greater in three farms. Only on the second farm the group receiving the commercial mineral mixture had a superior daily weight gain. The daily intake of the selective mineral mixture was lower in all groups and was attributed to the high amount of NaCl in these mixtures. The selective mineral supplementation was 3 up to 7 times more economic than the conventional supplementation with a “complete” mineral mix. Throughout the experimental period the animals did not show any direct or indirect clinical signs of mineral deficiency. The results of these experiments confirms the hypothesis that selective mineral supplementation – i.e. supplementation of the mineral element(s) deficient – was correct and results in expressive reduction of the cost with the mineral supplementation for beef cattle.

The other kidney was microscopically examined after hematoxylin-eosin, silver impregnation and immunohistochemical stains. Leptospires, all of them belonging to the Ballum serogroup, were isolated from 16 (39%) out of 41 samples. The presence of the agent was recorded in 18 (44%) and in 19 (46%) out of 41 silver impregnated and immunohistochemically stained samples respectively. Additionally, leptospires were detected in high number on the apical surface of epithelial cells and in the lumen of medullary tubules and they were less frequently seen on the apical surface of epithelial cells or in the lumen of the cortical tubules, which represents an unusual finding in carrier animals. Microscopic lesions consisting of focal mononuclear interstitial nephritis, glomerular shrinkage and desquamation of tubular epithelial cells were observed in 13 of 19 infected and in 10 of 22 non-infected mice; differences in presence of lesions between infected and non-infected animals were not statistically significant (P=0,14). The three techniques, culture, silver impregnation and immunohistochemistry, had a high agreement (k³0.85) and no significant differences between them were detected (P>0.05). In addition, an unusual location of leptospires in kidneys of carrier animals was reported, but a relationship between lesions and presence of leptospires could not be established.

#140 - Mastitis caused by Mycoplasma bovis in dairy cattle, 21(4):143-145

[Mastitis caused by Mycoplasma bovis in dairy cattle] Mastite bovina por Mycoplasma bovis em rebanhos leiteiros. Pesquisa Veterinária Brasileira 21(4):143-145. Depto Medicina Veterinária Preventiva, Universidade Estadual de Londrina, Cx. Postal 6001, Londrina, PR 86051-970, Brazil. In this study 713 cows were examined. The animals were from three dairy farms in northern Paraná and the southwest of the State of São Paulo. From these cows, 137 had mastitis. On the three farms, 8 cows (1-12%) with Mycoplasma bovis mastitis were detected. Four were treated with tylosin and oxytetracyçlin and three with enrofloxacin There was no response to the treatments, and these animals were culled during the lactation period. One cow treated with enrofloxacin almost totally recovered milk production, but elimination of M. bovis continued during the lactation, and there was no bacteriological cure. This cow had a normal milk production in the next lactation period, without elimination of M. bovis. Culling of positive animals, the bacteriological study anel correct application of preventive practices for contagious mastitis controlled the dissemination of M. bovis to other animals.

[Mastitis caused by Mycoplasma bovis in dairy cattle] Mastite bovina por Mycoplasma bovis em rebanhos leiteiros. Pesquisa Veterinária Brasileira 21(4):143-145. Depto Medicina Veterinária Preventiva, Universidade Estadual de Londrina, Cx. Postal 6001, Londrina, PR 86051-970, Brazil. Foram examinadas 713 vacas de três rebanhos leiteiros localizados na região norte do Estado do Paraná e sudoeste do Estado de São Paulo, das quais 137 apresentaram mastite. Nas três propriedades foram detectados oito animais (1, 12%) com mastite clínica por Mycoplasma bovis. Destes animais, quatro tratados com oxitetraciclina e tilosina e três com enrofloxacina, não responderam ao tratamento e foram descartados no decorrer da lactação. Uma vaca medicada com enrofloxacina recuperou quase que totalmente a secreção láctea mas a eliminação de M. bovis persistiu por toda lactação. Esta vaca apresentou cura bacteriológica na lactação seguinte. O descarte dos animais positivos, monitoramento bacteriológico e a aplicação correta das me9idas de prevenção para as mastites contagiosas controlaram a disseminação de M. bovis nos rebanhos.
