E-mail: joana.vet@gmail.com.br The study aimed to compare endometrial histology and plasmatic progesterone (P4) concentration of repeat breeds and healthy mares. The hypothesis was that there is a correlation between infertility and endometrial histology and P4 concentration in both groups. A total of 36 Campolina and Mangalarga Marchador mares in reproductive age (3-23 years) were used, 11 of them were healthy mares (Control group, 7 embryo recipient and 4 embryo donors), and 25 repeat breeders (10 embryo recipient and 15 embryo donors), classified as based on their reproductive history. Endometrial and blood samples were collected for respectively histological and plasma progesterone concentration evaluation. The endometrial samples obtained after biopsy were fixed in Bouin’s fluid, processed, included in paraffin, and stained with Hematoxylin-Eosin (HE) for histopathological examination. Plasmatic progesterone concentrations were evaluated by enzyme immunoessay (ELISA). There was no correlation between progesterone concentration and fertility. But there was a positive correlation between age and fertility, as older mares had major tendency of subfertility than younger ones. There was also a correlation between biopsy categories and fertility, as more histological alterations were found, higher were the chances for the mares to be subfertile. However not all mares classified as Category I and II maintained pregnancy until parturition. Other factors could influence pregnancy maintenance. In the same way, not all mares in Category III were infertile. The endometrial biopsy was shown to be an easy and cheap diagnostic technique with minimal discomfort to the animals and, together with other data, to be a very important component in the investigation of mare fertility.

Caracterização de seqüelas subseqüentes à punção folicular em bovinos. Pesquisa Veterinária Brasileira 23(3):119-124. Embrapa Gado de Leite, Juiz de Fora, MG 36038-330, Brazil. E-mail: jhmviana@cnpgl.embrapa.br The aim of this studywas to evaluate lesions of the genital tract in animais used as oocyte donors. Gir breed cows (n=20) used in different experiments involving the ovum pick-up technique were evaluated. At the end of each experimental period the animais were examined by rectal palpation, ultrasonography and vaginoscopy. A group of cows (n=8) were selected for histopathologic characterization of ovarian lesions. Ovaries were surgically removed, fixed and processed for histological examination. The cows evaluated underwent 9 to 42 puncture sessions, and yielded 10 to 719 oocytes. There was a low prevalence (5.26%) of inflammatory changes in the vagina mucosa. The occurrence of adherence was significantly correlated with the number of puncture sessions the animal underwent (r=0.31; P<0.05), but not with the total of oocytes collected (r=0.03; P>0.05). The opposite was observed reganding ovarian fibrosis, which showed a high correlation with oocyte yield (r=0.53; P<0.05). Histopathologic evaluation revealed scar tissue, inflammatory cell infiltration and presence of luteal tissue dispersed within the ovarian stroma.

[Characterization of tissue damages after ovum pick-up in bovine.] Caracterização de seqüelas subseqüentes à punção folicular em bovinos. Pesquisa Veterinária Brasileira 23(3):119-124. Embrapa Gado de Leite, Juiz de Fora, MG 36038-330, Brazil. E-mail: jhmviana@cnpgl.embrapa.br Objetivaram-se avaliar lesões no trato genital em animais utilizados como doadores de oócitos. Foram examinadas vacas da raça Gir (n=20) que participaram de diferentes experimentos envolvendo a técnica de punção folicular. Ao término de cada período experimental, os animais foram examinados por palpação retal, ultra-sonografia e vagi-noscopia. Um grupo de vacas (n=8) foi selecionado para a caracterização histopatológica das lesões ovarianas. Os ovários foram retirados cirurgicamente, fixados e processados, e as lâminas obtidas coradas e analisadas. Fóram avaliados animais submetidos a entre nove e 42 sessões de punção, e que produziram entre 10 e 719 oócitos. Observou-se uma baixa prevalência (5,26%) de processos inflamatórios na mucosa do fundo de saco vaginal. A ocorrência de aderências apresentou uma correlação positiva com o número de sessões de punção a que o animal foi submetido (r=0,31; P<0,05), mas não com o total de oócitos recuperados (r=0,03; P>0,05). Tendência oposta foi observada em relação à fibrose ovariana, que apresentou uma alta correlação com o número de oócitos recuperados (r=0,53; P<0,05). A avaliação histopatológica demonstrou a presença de tecido cicatricial, infiltrados de células inflamatórias e presença de tecido luteal disperso no estroma ovariano.

Genetic and antigenic analysis of Babesia bigemina isolates from five geographical regions of Brazil. Pesquisa Veterinária Brasileira 22(4):153- 160. [Análise genética e antigênica de isolados de Babesia bigemina das cinco regiões fisiográficas do Brasil.] Embrapa Gado de Corte, Rodovia BR 262 Km 4, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil. A molecular epidemiological study was performed with Babesia bigemina isolates from five geographical regions of Brazil. The genetic analysis was done with random amplification of polymorphic DNA (RAPD), repetitive extragenic palindromic elements-polymerase chain reaction (REP-PCR) and enterobacterial repetitive intergenic consensus sequences-polymerase chain reaction (ERIC-PCR) that showed genetic polymorphism between these isolates and generated fingerprinting. In RAPD, IL0872 and IL0876 primers were able to detect at least one fingerprinting for each B. bigemina isolate. The amplification of B. bigemina DNA fragments by REP-PCR and ERIC-PCR gave evidence for the presence in this haemoprotozoan of the sequences described previously in microorganisms of the bacterial kingdom. For the first time it was demonstrated that both techniques can be used for genetic analysis of a protozoan parasite, although the ERIC-PCR was more discriminatory than REP-PCR. Toe dendogram with similarity coeficiente among isolates showed two clusters and one subcluster. The Northeastern and Mid-Westem isolates showed the greatest genetic diversity, while the Southeastem and Southem isolates were the closest. Toe antigenic analysis was done through indirect fluorescent antibodytechnique and Westem blotting using a panei of monoclonal antibodies directed against epitopes on the merozoite membrane surface, rhoptries and membrane of infected erythrocytes. As expected, the merozoite variable surface antigens, major surface antigen (MSA)-1 and MSA-2 showed antigenic diversity. However, B cell epitopes on rhoptries and infected erythrocytes were conserved among all isolates studied. In this study it was possible to identify variable and conserved antigens, which had already been described as potential immunogens. Considering that an attenuated Babesia clone used as immunogen selected populations capable of evading the immunity induced by this vaccine, it is necessary to evaluate more deeply the cross-protection conferred by genetically more distant Brazilian B. bigemina isolates and make an evaluation of the polymorphism degree of variable antigens such as MSA-1 and MSA-2.

[Análise genética e antigênica de isolados de Babesia bigemina das cinco regiões fisiográficas do Brasil.] Embrapa Gado de Corte, Rodovia BR 262 Km 4, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil. Um estudo de epidemiologia molecular foi executado com isolados de Babesia bigemina das cinco regiões fisiográficas do Brasil. A análise genética foi feita com amplificação aleatória de DNA polimórfico (RAPD), reação da polimerase em cadeia com seqüências de elementos extragênicos repetitivos palindrômicos (REP-PCR) e reação da polimerase em cadeia com seqüências repetitivas enterobacterianas intergênicas de consenso (ERIC-PCR) que apresentaram polimorfismo genético entre os isolados e geraram marcadores. No RAPD com os oligonucleotídeos iniciadores IL0872 e IL0876, foi possível detectar pelo menos um marcador por isolado de B. bigemina. A amplificação de fragmentos de DNA de B. bigemina por REPPCR e ERIC-PCR demonstrou a presença dessas seqüências, descritas anteriormente somente em microrganismos bacterianos, nesse hemoprotozoârio, e, pela primeira vez, foi verificado que podem ser utilizadas para análise genética de um protozoário. O ERIC-PCR foi mais discriminatório que o REP-PCR. O dendograma formado com o coeficiente de similaridade entre os isolados evidenciou dois agrupamentos e um subgrupo. Os isolados do Nordeste e Centro-Oeste demonstraram maior diversidade genética, enquanto que os isolados do Sudeste e Sul foram os mais próximos. A análise antigênica foi executada por meio de imunofluorescência indireta e Western blotting usando um painel de anticorpos monoclonais direcionados a epitopos B na membrana dos merozoítos, roptries e membrana de eritrócitos infectados. Os antígenos variáveis da superfície dos merozoítos, antígeno principal da superfície do merozoíto (APSM)-1 e APSM-2 apresentaram diversidade antigênica. Entretanto, os epítopos de células B nas roptries e nos eritrócitos infectados foram conservados em todos os isolados. Nesse estudo foi possível identificar antígenos variáveis e conservados que anteriormente haviam sido descritos como potenciais imunógenos. Considerando que um clone atenuado de Babesia utilizado para imunização selecionou populações capazes de evadir a resposta imune à vacina, torna-se necessário avaliar mais detalhadamente a imunidade cruzada existente entre os isolados brasileiros mais distantes geneticamente e realizar uma avaliação do grau de polimorfismo dos antígenos variáveis APSM-1 e APSM-2.
