Clinical pathological data and analyses of mineral elements of cattle affected by epizootic botulism in the State of São Paulo

In arder to study laboratorial aspects of beef cow mortality, a syndrome popularly known as "doença da vaca caída", examens were made of blood, cerebrospinal fluid, serum, bone and liver samples from 32 naturally affected 4 to 9 year old cows, 27 belonging to the Nellore breed and5 were crossbred Nellore, all originating from farms located in municipalities near Botucatu, State of São Paulo. Laboratory determinations were analysed by descriptive statistics and included hematological values, total plasma protein, plasma fibrinogen, cerebrospinal fluid analysis, and concentration measurements of serum calcium, phosphorus, magnesium, sodium, potassium, chloride, total protein, albumin, globulin, alkaline phosphatase, aspartate aminotransferase, gama-glutamyltransferase and creatine kinase activities, included bane ash percentage and concentrations of calcium, phosphorus and magnesium, and also hepatic levels of copper, zinc, iron, manganese and cobalt. In addition, mouse bioassays and complement micro-fixation tests were performed to detect botulinum toxins in liver samples. The results indicated leukocytosis (13,3±3,9 x103/mm3) with neutrophilia (8,9±3,2 x103/mm3), hypocalcemia (7,8±1,7mg/dl), hypophosphatemia (3,6±1,6mg/dl), hypoalbuminemia (2,9±0,9g/cll), increased creatine kinase activity (691,0±829,7 UI/1), and reduced ash percentage (60,3±1,9%) and low phosphorus (17,2±0,4%) in bane. The other values were all within normal limits. The diagnosis of botulism, involving type C and D toxins, was confirmed as the cause of the mortality in the region of study, what is strongly consistent with the other laboratorial findings.

Com o objetivo de verificar alguns aspectos laboratoriais da mortalidade de vacas de corte, popularmente conhecida como "doença da vaca caída", estudaram-se amostras de sangue total, líquor, soro sanguíneo, osso e fígado colhidas de 32 fêmeas bovinas (27 da raça Nelore e 5 mestiças), entre 4 e 9 anos de idade, naturalmente acometidas e pertencentes a propriedades rurais localizadas em municípios próximos a Botucatu, Estado de São Paulo. Procederam-se as determinações e o estudo estatístico descritivo das variáveis: 1) hematológicas, de proteínas plasmáticas totais e fibrinogênio; 2) dos aspectos físicos, bioquímicos e celularidade no líquor; 3) de cálcio, fósforo, magnésio, sódio, potássio; cloretos, proteína total, albumina, globulina e atividades da fosfatase alcalina, aspartato aminotransferase, gama-glutamiltransferase e creatina quinase no soro sanguíneo; 4) de cinzas, cálcio, fósforo e magnésio no osso, e, 5) de cobre, zinco, ferro, manganês e cobalto no fígado, empregando-se as provas de inoculação em camundongos e de microfixação de complemento para a detecção da presença de toxinas botulínicas nas amostras de tecido hepático. Os resultados apontaramcomo alterações, leucocitose (13,3±3,9 x103/mm3) com neutrofilia (8,9±3,2 x103/mm3), hipocalcemia (7,8±1,7mg/ dl), hipofosfatemia (3,6±1,6mg/dl), hipoalbuminemia (2,9±0,9g/dl), elevação na atividade da creatina quinase (692,0±829,7 UI/1) e reduções na cinza (60,3±1,9%) e fósforo (17,2±0,4%) ósseos, como indicativos do estado carencial de fósforo, apresentando-se as demais variáveis dentro dos limites de variação considerados normais para a espécie. O diagnóstico de botullsmo, com envolvimento das toxinas C e D, foi confirmado como a causa da mortalidade investigada na área de abrangência do estudo, e encontra coerência com o conjunto de resultados.


Experimental poisoning by the seeds of Ricinus communis (Euphorbiaceae) in sheep

Abstract in English:

The oral administration in single doses of the seeds of Ricinus com munis, a shrub of the Euphorbiaceae family, caused poisoning characterized by gastroenteriritis in sheep. The whole seeds caused death of the four sheep which received 10 and 7.5 g/kg. The dose of 5 g/kg caused slight symptoms of poisoning and the dose of 2.5 g/kg no symptoms at all. The smashed seeds of R.communis caused death in two sheep which received 2.5 g/kg; the dose of 1.25 g/kg caused severe symptoms in one sheep; the dose of 1.25 g/kg given to four sheep and 0.625 g/kg given to two others did not cause any symptoms of poisoning. In the sheep which died, first symptoms were obsetved between 6h40min and 17h24min, and death occurred between 31h55min and 53h39min after the beginning of administration of the seeds. The clinical course of poisoning varied between 19h50min and 44h52min in these animals. The main symptoms were anorexia, apathy and colics, diarrhea with soft to liquid faeces of foetid odor with mucus and/or fibrine; finally the animals defecated in form of a jet of liquid faeces. The animals had increasing tachicardia. There was dryness of the skin with loss of its elasticity, and sunken eyballs indicating dehydration. In the final stage there was severe muscular weakness. The main post mortem findings were congestion and oedema of the gastrointestinal wall, from the abomasum to the proximal colon. The jejunum was extended and contained a large amount of a dear greenish liquid together with fibrine in form of cilinders, filaments and pseudomembranes; there were similar contents in the cecum and proximal colon. In those sheep which had received the whole seeds of R. communissome of them were found in the rumen. The main histological changes seen in the small intestine were necrosis of the upper layer of the mucosa, accompanied by congestion of the whole mucosa, as well as submucosa and serosa.

A administração por via oral das sementes de Ricinus communis, arbusto da família Euphorbiaceae, em doses únicas, causou quadro de intoxicação com alterações gastrintestinais em ovinos. As sementes inteiras causaram a morte dos ovinos que as ingeriram nas doses de 10 e 7,5 g/kg. A dose de 5 g/kg causou sintomas leves e a de 2,5 g/kg não provocou sintomas de intoxicação. As sementes esmagadas de R. communis causaram a morte dos dois ovinos que as ingeriram na dose de 2,5 g/kg. A dose de 1,25 g/kg causou sintomas acentuados em um ovino; em quatro outros, que ingeriram 1,25 g/kg e em dois que ingeriram 0,625 g/kg não causaram sintomas de intoxicação. Nos animais que morreram, os primeiros sintomas foram observados entre 6h40min e 17h24min e a morte sobreveio entre 31h55min e 53h39min após o início da administração das sementes. A evolução da intoxicação variou entre 19h50min e 44h52min nesses animais. Os principais sintomas foram anorexia, apatia e manifestações de cólica e diarréia com fezes mal cheirosas e pastosas a líquidas com muco e/ou fibrina; finalmente os animais defecavam em jatos. Havia taquicardia cada vez mais acentuada. Foram observados sinais de desidratação sob forma de ressequimento da pele e perda de sua elasticidade e retraimento do globo ocular. Na fase final os animais mostravam acentuada fraqueza muscular. Os achados de necropsia principais foram congestão e edema da parede do tubo digestivo, desde o abomaso até o cólon proximal. O jejuno estava dilatado contendo grande quantidade de líquido verde-claro, com presença de fibrina sob forma de cilindros, filamentos ou pseudomembranas. O conteúdo do ceco e cólon proximal tinha características semelhantes. Nos ovinos que receberam as sementes inteiras de R. communis, algumas delas foram encontradas no rúmen. As principais alterações histológicas foram observadas no intestino delgado sob forma de necrose na parte superior da mucosa, acompanhada de congestão da mucosa, submucosa e serosa.


Experimental poisoning by the seeds of Abrus precatorius (Leg. Papilionoideae) in rabbits

Abstract in English:

The ground or whole seeds of Abrus precatoríus L., given by stomach tube in single doses to rabbits, caused a clinic-pathological picture of enteritis. The ground seeds caused death of all rabbits which received 0.03125 g/kg or more; doses of 0.015625 g/kg, 0.007812 g/kg and 0.003906 g/kg caused death of always one of three rabbits which received one of the doses. The dose of 0.001953 g/kg caused only slight symptoms in one of three rabbits tested; doses of 0.0009765 g/kg and 0.00048825 g/kg, each given to two rabbits, did not cause any symptoms. The whole seeds caused death of two rabbits which received the doses of 0.75 g/kg and 0.375 g/kg; doses of 0.25 g/kg and 0.125 g/kg, each one given to three rabbits, caused death of two from each group. The dose of 0.625 g/kg given to six rabbits, caused death only of one. The dose of 0.03125 g/kg did not cause any symptoms of poisoning in six rabbits. The period between the administration of the seeds and death varied from 13h35min to 7 days in the experiments with the ground seeds, and from 29hl 4min to 59h40min in the experiments with the whole seeds. The period between the administration of the seeds and recovery varied from 3 and a half days to 17 days in the experiments with the ground seeds and from 2 to 16 days in the experiments with the whole seeds. All 44 rabbits used in these experiments received the seeds only once. The clinic-pathological picture of the poisoning, which was the sarne in the experiments with the ground and the whole seeds of A.precatorius, consisted in inappetence/ anorexia, few faeces with bolus altered in form and size, sometimes dark coloured, soft or liquid, of foetid odor, with mucus and seldom with fibrina. There were manifestations of colic. The most important post-mortem changes were found in the small intestine and cecum. The former was distended by large amounts of liquid and mucous contents. Its wall was congested throughout its length and sometimes edematous, and fibrine covered the mucosa. The cecum was distended by soft to liquid contents and its wall showed congestion/hemorrhages and edema. The wall of the proximal colon was hemorrhagic and mucus was sometimes found in the lumen. In the rabbits which received the whole seeds, some of them were found in the stomach or rectum. In most rabbits slight to moderate splenomegaly was observed. The most important histological changes were seen in the digestive tract, mainly in the small intestine and less pronounced in the cecum. Few or no changes were observed in stomach and colon. The changes found in the different parts of the intestine were similar and consisted in coagulative necrosis of the mucosa with presence of nuclear fragments, congestion/hemorrhages of the mucosa, submucosa, muscularis and serosa, as well as edema of the submucosa. Additionally in the small intestine large amounts of nuclear fragments were seen in the upper portion of the lymphoid follicles and in smaller amounts in its center. In the appendix vermiformis, and in many cases also in the rudimentar cecum, variable amounts of nuclear fragments were seen in the stroma of the mucosa which resulted from necrosis by caryorhexis of macrophages of the lymphoid follicles, which migrated towards the intestinal lumen. In most rabbits thers was congestion of the spleen and mesenteric lymphnodes.

A administração por sonda intragástrica das sementes trituradas ou inteiras de Abrus precatorius L., em doses únicas, provocou em coelhos um quadro clínicopatológico grave de enterite. As sementes moídas causaram a morte de todos os coelhos a partir da dose de 0,03125 g/kg; as de 0,015625 g/kg, 0,0078125 g/kg e 0,003906 g/ kg causaram a morte, para cada uma dessas dosagens, de um de três coelhos. A dose de 0,001953 g/kg causou só sintomas leves em um de três coelhos e as de 0,0009765 g/kg e 0,00048825 g/kg não causaram sintomas nos dois coelhos para cada dosagem. As sementes inteiras causaram morte dos dois coelhos que receberam as doses de 0,75 g/kg e 0,375 g/kg; as de 0,25 e 0,125 g/kg causaram a morte, para cada uma dessas dosagens, de dois de três coelhos. A dose de 0,0625 g/kg causou a morte de um de seis coelhos. A dose de 0,03125 g/kg não causou sintomas de intoxicação em seis coelhos. O período entre a administração das sementes e a morte variou, nos experimentos com as sementes moídas, de 13h35min a 7 dias e nos experimentos com sementes inteiras, de 29h14min a 59h40min. O período entre a administração das sementes e a recuperação, variou, nos experimentos com as sementes moídas, de 3 e meio dias a 17 dias e nos experimentos com as sementes inteiras, de 2 a 16 dias. Todos os 44 coelhos usados nesses experimentos receberam as sementes de A. precatorius somente uma vez. O quadro clínico-patológico da intoxicação era o mesmo para a intoxicação pelas sementes moídas ou inteiras de A. precatorius. Os sintomas de intoxicação consistiram em inapetência/anorexia, fezes escassas e com forma e tamanho das síbalas alteradas, às vezes escuras, pastosas, líquidas, fétidas, com muco e raramente com fibrina. Havia sinais de cólica bem manifestos. Os achados de necropsia mais importantes ocorreram no intestino delgado e ceco. O intestino delgado estava muito dilatado com abundante conteúdo líquido-mucoso. Havia congestão da parede em toda extensão; às vezes edema. Fibrina revestia a mucosa. O ceco estava dilatado com conteúdo pastoso-líquido, a parede apresentava congestão/hemorragias e edema. No colon proximal às vezes havia muco na luz e homorragias na parede. Nos coelhos que receberam as sementes inteiras, algumas delas eram encontradas no estômago ou no rato. Na maioria dos coelhos havia ainda discreta a moderada esplenomegalia. As alterações histológicas mais importantes foram verificadas no tubo digestivo. Os segmentos mais afetados foram o intestino delgado e, com menor intensidade, o ceco. No estômago e colon as alterações foram poucas ou ausentes. As lesões nas diversas partes do intestino foram muito semelhantes e consistiram em necrose de coagulação da mucosa com presença de fragmentos nucleares, congestão/ hemorragias na mucosa, submucosa, muscular e serosa, e edema da submucosa. Na superfície dos folículos linfoides do intestino delgado observou-se grande quantidade de fragmentas nucleares; no interior desses folículos o grau de fragmentação era menor. No apêndice vermiforme e com menor frequência no ceco rudimentar, observou-se quantidade variável de fragmentos nucleares no estroma da mucosa, resultante da necrose por cariorrexia de macrófagos dos folículos linfóides que após ativação migraram em direção a luz intestinal. Na maioria dos coelhos havia ainda congestão do baço e dos linfonodos mesentéricos.


Poisoning by Cestrum intemedium (Solanaceae) in cattle

Abstract in English:

A disease of cattle characterized by acute hepatic insufficiency was studied in western Santa Catarina, Brazil. The affected animals showed anorexia, rumen atony, dried feces with blood and mucus, congestion of sclera and conjunctiva, muscular tremors, they kept their head against obstacles, showed instability of gait and decubitus, and death occurred 12 to 48 hours after the onset of first symptoms. The post-mortem findings in 6 bovines were nutmeg appearance of the liver, dried contents of omasum, colon and rectum and hemorrhages in the mucous membrane of the large intestine and in the endocardium of the left ventricle. Histological examination showed necrosis, congestion and hemorrhages in the hepatic lobe. The disease was experimentally reproduced by administration of fresh leaves of Cestrum intermedium to 6 bovines. With a single dose of 20 g/kg there were no clinical manifestations. The administration of 25 g/kg to 2 bovines caused clinical signs and one of them died. A single dose of 30 g/kg given to 2 bovines caused clinical signs and death. It was concluded that C. intermedium is responsible for acute hepatic insufficiency of cattle in the western region of the state of Santa Catarina.

Uma doença com quadro clínico-patológico de insuficiência hepática aguda foi estudada em bovinos, na região Oeste de Santa Catarina. Os animais suspeitos de estarem intoxicados por "coerana" ou "mata-boi" (Cestrum intermedium), apresentavam anorexia, parada da ruminação, fezes em pequena quantidade, secas com muco e sangue, congestão da esclera e conjuntiva, tremores musculares, andar cambaleante, cabeça apoiada contra abstáculos, decúbito e óbito entre 12 a 48 horas após início dos sintomas. Através da necropsia de seis bovinos doentes, foram constatados fígado com nítido aspecto de noz-moscada, conteúdo do omaso, do cólon e do reto seco, e hemorragias na mucosa do intestino grosso e no endocárdio esquerdo. As lesões histológicas se caracterizavam principalmente por necrose, congestão e hemorragias no lóbulo hepático. A doença foi reproduzida experimentalmente pela administração de folhas verdes de C. intermedium a seis bovinos. Com dose única de 20 g/kg não houve alterações clínicas. A administração de dose única de 25 g/kg, a dois bovinos, produziu sinais clínicos da doença com morte em um dos animais. Em doses únicas de 30 g/kg em dois bovinos, houve manifestações clínicas nseguidas de morte. Conclui-se que C. intermedium é responsável pelo quadro ele insuficiência hepática aguda que acomete bovinos da região Oeste de Santa Catarina.


Poisoning by Amaranthus spp. (Amaranthaceae) in pigs in Rio Grande do Sul, Brazil

Abstract in English:

Two outbreaks of Amaranthus spp. poisoning occurring in young pigs in Southern Brazil in late summer and early autumn are described. In both instances large amounts of freshly harvested Amaranthus spp. in its seeding stage were fed to the pigs. In one farm the Amaranthus was identified as A. viridis. Fourteen out of 25 pigs (56%) in one farm and 17 out of 100 07%) in another died. Clinical sigos included anorexia, depression, incoordinated gait; muscle stiffness, lateral recumbency, followed by death which occurred from 4 to 48 hours after clinical sigos were observed. Necropsy findings included perirenal edema and edema of other sites such as ascites; hydrotorax, hydropericardium, of the mesentery and the subcutaneous tissue of the ventral abdominal wall. The kidneys were pale and soft with subcapsular petechiae. In one of the three cases submitted for post-mortem examination there was marked edema of the renal papilla. Main histopathological findings were toxic tubular nephrosis affecting mainly the proximal convoluted tubules and mild tubular regeneration. In one case few refringent crystals were seen in the lumina of proximal tubules and in two cases the degenerative renal lesions were associated with moderate mononuclear interstitial inflitrate. Extra-renal lesions consisting of fibrinonecrotic glossitis and esophagitis were found in one pig and attributed to uremia.

São descritos dois surtos de intoxicação por Amaranthus spp. em suínos jovens no fim do verão e início do outono, no Estado do Rio Grande do. Sul. Em ambos os estabelecimentos onde ocorreram os surtos, os suínos eram alimentados com grandes quantidades de Amaranthus spp. em frutificação, recém-colhido. Em um dos estabelecimentos o Amaranthus foi identificado como A. viridis. No estabelecimento 1, morreram 14 suínos de um lote de 25 (56%) e no estabelecimento 2 morreram 17 de um total de 100 suínos (17%). Os sinais clínicos incluíam anorexia, apatia, andar incoordenado, rigidez muscular, decúbito lateral e morte, que ocorria de 4 a 48 horas após a observação dos sinais clínicos. Os achados de necropsia incluíam edema perirrenal e edemas em vários outros locais, como ascite, hidrotórax, hidropericárdio, mesentério e tecido subcutâneo da parede abdominal ventral. Os rins estavam pálidos e macios, com petéquias na superfície subcapsular. Em um caso havia marcado edema das papilas renais. Os principais achados histopatológicos foram nefrose tubular tóxica afetando principalmente os túbulos contorcidos proximais e regeneração tubular discreta. Em um dos três casos foram observados cristais refringentes na luz dos túbulos proximais e, em dois casos, as lesões renais degenerativas estavam associadas a moderado inflitrado mononuclear intersticial. Lesões extra-renais consistindo de glossite e esofagite fibrinonecróticas foram encontradas em um suíno e atribuídas à uremia.


Occurrence of enterotoxigenic Escherichia coli and Salmonella sp in calves in the southern agreste region of the State of Pernambuco, Brazil

Abstract in English:

The diarrheic processes caused by pathogenic enterobacteria are of great economicsanitary importance, with limiting effects on cattle raising. In the present study 106 fecal samples from 52 diarrheic and 54 non-diarrheic calves aged 1 to 90 days were analyzed, in arder to determine the occurrence of enterotoxigenic Escherichia coli (ETEC) and Salmonella sp among calves reared in the agreste region of the state of Pernambuco and to determine the behavior of the isolates in response to 16 antimicrobial agents. The strains were cultured in selective media and submitted to biochemical characterization, serologic identification and detection of heat-stable enterotoxin. E. coli was isolated from all 106 calves studied and five of these strains (4,7%) were proclucers of heat-stable enterotoxin. Salmonellasp was isolated from six (5,7%) samples belonging to serovars Dublin, Muenchen and Infantis. The Salmonella strains were highly sensitive to the drngs tested, whereas the E. co/istrains were more resistent to tetracycline (50,9%), sulfonamide (40,6%), streptomycin (35,9%) and ampicillin (22,6%). Particularly outstancling in the general analysis was the multiresistance of E. coli (52,0%), with a predominance of the tetracycline-sulfonamide association. The present results serve as a warning regarding the incliscriminated use of antimicrobial agents. Low percentages of ETEC and Salmonella in calves show the necessity of other diagnostic procedures to clarify the etiology.

Os processos diarréicos acarretados por enterobactérias patogênicas têm sido incriminados como de grande importância econômica-sanitária, desempenhando um papel limitante na criação de bezerros. Com o objetivo de estudar a ocorrência de Escherichia coli enterotoxigênica e Salmonella sp em bezerros no agreste meridional de Pernambuco e o compottamento dos isolados frente aos antimicrobianos, foram analisadas 106 amostras fecais provenientes de 52 bezerros diarréicos e 54 não diarréicos de 1 a 90 dias de idade. O cultivo das amostras foi realizado em meios seletivos com posterior caracterização bioquímica, identificação sorológica e detecção de enterotoxina termoestável. Para verificação do comportamento dos isolados, foram selecionados 16 antimicrobianos. E. coli foi isolada dos 106 bezerros estudados, sendo cinco (4,7%) produtoras de enterotoxina termoestável e, Salmonella isolada de seis (5,7%) amostras pertencentes aos sorovares Dublin, Muenchen e Infantis. As amostras de Salmonella demonstraram uma elevada sensibilidade frente as drogas testadas, enquanto que as de E. coli revelaram maior resistência à tetraciclina (50,9%); sulfonamida (40,6%); estreptomicina (35,9%) e ampicilina (22,6%). No cômputo geral destaca-se a multirresistência de E. coli (52%), predominando a associação tetraciclina-sulfonamida. Face aos resultados auferidos adverte-se quanto ao uso indiscriminado de antimicrobianos. Os baixos percentuais de ETEC e Salinonella salientam a necessidade de outros procedimentos de diagnóstico visando esclarecer a etiologia das diarréias.
