#1 - ‘Beer yeast’ (liquid brewery waste) for sheep and pig feeding: epidemiological and clinical aspects of the poisoning, safety margin and prevention

Abstract in English:

Considering the practice of using ‘brewer’s yeast’ (BY) – liquid brewery yeast waste from the brewing industry – in animal feed in some establishments in the south of the State of Rio de Janeiro in Brazil, this study was conducted to establish safety margins and to determine epidemiological and clinical aspects. In addition, this study suggested prophylactic measures that can prevent or minimise alcohol intoxication due to BY in sheep and swine. The main characteristics of the clinical features in both natural poisoning and controlled spontaneous ingestion in swine as well as intoxication by controlled ingestion of BY using a ruminal probe in sheep were found to be staggering gait, tripping and falls. Overall, 3.8 and 8.875mL/kg of ethanol content in the BY caused mild-moderate and severe clinical conditions, respectively, in swine and sheep. The following prophylactic measures are suggested: 1. BY must be adequately diluted in water, whey, or with BY that has been previously stored in the property (old BY); 2. It must be administered in proportion to the animals’ weight and size. 3. It must be administered continuously, without interruptions, and with the provision of other food in the trough (such as soybean meal or cornmeal and water ad libitum). In conclusion, although BY is increasingly used in sheep and swine breeding properties – in the south of the State of Rio de Janeiro – many owners use one or more of the suggested prophylactic measures. Hence, ethanol intoxications because of BY are infrequent and rarely cause deaths; therefore, BY can be used provided adequate prophylactic measures are followed.

Considerando-se à prática de utilizar “levedo de cerveja” (LC) – resíduo líquido da indústria cervejeira – na alimentação de animais, em alguns estabelecimentos no sul do Estado do Rio de Janeiro, esse estudo foi realizado para estabelecer as margens de segurança e os quadros epidemiológico e clínico, bem como sugerir medidas profiláticas que impeçam ou minimizem esse tipo de intoxicação alcoólica para ovinos e suínos. Verificou-se que o quadro clínico na intoxicação por ingestão controlada de LC por sonda em ovinos e na intoxicação natural e ingestão espontânea controlada em suínos, caracterizou-se principalmente por andar cambaleante, tropeços e quedas. Concentrações de 3,8 e 8,875mL/kg de etanol contido no LC, causaram quadros clínicos leve a moderado e grave em suínos e ovinos, respectivamente. Como medidas profiláticas sugere-se: diluição adequada do LC com água, soro de leite ou com o LC já estocado na propriedade (LC antigo); administração do LC proporcional ao peso/tamanho dos animais, administração contínua, sem interrupções, disponibilizar outro alimento no cocho, como farelo de soja ou fubá de milho e água à vontade. Conclui-se que apesar de o LC ser cada vez mais utilizado nas propriedades criadoras de ovinos e suínos no sul do Estado do Rio de Janeiro já que, muitos proprietários aplicam uma ou mais medidas profiláticas aqui sugeridas, as intoxicações pelo etanol, contido no LC são pouco frequentes e raramente ocorrem mortes, de forma que esse produto deve ser utilizado, desde que as medidas profiláticas sejam observadas.


#2 - Effects of autolyzed yeast on leukocyte oxidative metabolism and pneumonia occurrence in feedlot steers

Abstract in English:

Although yeast supplementation has shown an immunostimulant effect in feedlot cattle, the question remains whether it influences the occurrence of the bovine respiratory disease complex (BRD) in feedlot cattle. Therefore, our objective was to evaluate whether dietary supplementation with autolyzed yeast affects the efficiency of blood phagocytes, reducing inflammation and the occurrence of BRD in feedlot cattle. A randomized experimental trial was conducted with whole steers with half Angus blood for 106 days: control (n=10): diet without yeast; Y4 (n=12): yeast diet (4g per animal per day); and Y7 (n=7): yeast diet (7g per animal per day). On Days 0, 14, 42, 70, and 105, blood count, leukocyte oxidative metabolism, serum haptoglobin, frequency of nasal secretion and orbital temperature were evaluated. On slaughter day, a lung evaluation was performed. On Days 42 and 70 of feedlot finishing, the treated animals showed increased leukocyte oxidative metabolism (D42 P=0.04; D70 P=0.02) compared to the control group. These animals had a lower frequency of mucopurulent nasal secretion, lower orbital temperature and reduced pneumonia occurrence than the treated groups (P=0.05) with less lung lesion severity (P=0.01), allowing us to infer that the autolyzed yeast improves blood phagocytic function and decreases the inflammation and BRD of feedlot steers, especially at a dose of 4g.

Embora a suplementação com leveduras tenha demonstrado efeito imunoestimulante em bovinos, permanece-se a dúvida se ela influencia na ocorrência do complexo doença respiratória bovina (CDRB) em bovinos confinados. Portanto, objetivou-se avaliar se a suplementação dietética com levedura autolisada influencia a eficiência de fagócitos sanguíneos, reduzindo inflamações e a ocorrência de CDRB em bovinos confinados. Realizou-se um ensaio experimental randomizado com 36 novilhos inteiros 1/2 sangue Angus por 106 dias: controle (n=10): dieta sem levedura; Y4 (n=12): dieta com levedura (4g por animal por dia); e Y7 (n=7): dieta com levedura (7g por animal por dia). Nos dias 0, 14, 42, 70 e 105, hemograma, metabolismo oxidativo de leucócitos, haptoglobina sérica, frequência de secreção nasal e temperatura orbital foram avaliados. No dia do abate, foi realizada avaliação pulmonar. Nos dias 42, e 70 de terminação em confinamento, os animais tratados apresentaram aumento do metabolismo oxidativo dos leucócitos (D42 P=0,04; D70 P=0,02) em relação ao grupo controle. Esses animais apresentaram menor frequência de secreção nasal mucopurulenta, menor temperatura orbital, menor ocorrência de pneumonias (P=0,05) com menor gravidade das lesões pulmonares (P=0,01), permitindo inferir que a levedura autolisada na dieta melhorou a eficiência dos fagócitos sanguíneos, reduzindo inflamações e a ocorrência de CDRB, especialmente na dosagem de 4g.

E-mail: nevesrita@hotmail.com.br Otitis externa is a common complaint in dogs. Bacteria and yeasts are commonly involved and may perpetuate inflammatory reactions inside the ear canal. Otoscopy, cytological examination of secretion and microbiological culture embody forms of diagnosis. Cytology also has great use in accessing treatment evolution. Therapy usually consists of cleaning ear canals and subsequent use of antibiotics or antifungal products. As some of them may cause hypersensitivity and even ototoxicity, searching for new pharmacological bases is currently necessary and justifies this study, which aimed to evaluate in vitro and in vivo efficacy of tea tree essential oil for bacterial and yeast ear infections in dogs. Twenty-eight dogs from a particular shelter in Cuiabá (Mato Grosso, Brazil), presenting clinical signs of otitis externa, were enrolled in this clinical trial. In all of them, clinical and cytological evaluations, as well as culture and susceptibility testing of the affected ears were carried out. From each dog, one ear was treated with 5% tea tree essential oil lotion and the other with standard otic formulation, according to the type of infection (bacterial, yeast or both). In vitro susceptibility testings of all ear cultures, to the same drugs used in treatment, were also carried out. Culture results showed 62.5% bacterial and fungal infection, 33.9% bacterial infection and 3.6% fungal infection, from the 56 ear samples collected. The most common microorganisms isolated were Staphylococcus intermedius, Staphylococcus aureus, Proteus mirabilis and Malassezia pachydermatis. Gram‑positive bacteria were susceptible to gentamycin in 60.5% and resistant in 16.3% of the samples. Five percent tea tree essential oil formulation produced a 5mm clear zone of inhibition around the disks in one of the 63 samples evaluated. Pure (100%) tea tree essential oil formulation produced a 10mm clear zone of inhibition around the disks in four of the 63 samples evaluated, a 9mm zone in three samples, an 8mm zone in 16 samples, a 7mm zone in seven samples, a 6mm zone in two samples and there was no clear zone in 31 samples. Inhibition zones were produced by strains of Staphylococcus intermedius, Staphylococcus hyicus, Corynebacterium sp., Proteus mirabilis and Enterobacter sp. tea tree essential oil ear solution significantly induced remission of clinical signs both in bacterial and yeast ear infections. It also reduced as much Malassezia pachydermatis ear infection as the nystatin solution used in this study, while gentamycin solution showed better antibacterial effect. More studies should be conducted to evaluate in vitro diffusion properties of tea tree essential oil. Good antimicrobial spectrum and the absence of adverse reactions confirm the importance of developing a tea tree formulation as an alternative therapy for ear infections in dogs.

E-mail: nevesrita@hotmail.com.br Otite externa é queixa frequente em cães. Bactérias e leveduras estão comumente envolvidas e podem perpetuar as reações inflamatórias dentro do canal auditivo. Dentre as formas de diagnóstico, encontram-se a otoscopia, o exame citológico da secreção e a cultura microbiológica. Citologia também tem grande utilidade no acesso à evolução do tratamento. A terapia consiste de limpeza dos canais auditivos e posterior utilização de antibióticos ou produtos antifúngicos. Como alguns antimicrobianos utilizados no tratamento podem causar hipersensibilidade e até mesmo ototoxicidade, a busca por novas bases farmacológicas justifica a existência deste estudo, que teve como objetivo avaliar in vitro e in vivo a eficiência do óleo de Melaleuca alternifolia em otites bacterianas e fúngicas de cães. Vinte e oito cães, de um abrigo particular, apresentando sinais clínicos de otite externa, foram incluídos neste estudo clínico. Todos passaram por avaliação clínica, citologia e cultura de material das orelhas afetadas. De cada animal, uma orelha foi tratada com óleo de Melaleuca 5% e a outra com formulação ótica padrão, de acordo com a afecção (bacteriana, fúngica ou mista). As culturas também foram submetidas a testes de susceptibilidade in vitro aos mesmos agentes utilizados no tratamento in vivo. Os resultados da cultura mostraram 62,5% de infecção mista (bacteriana e fúngica), 33,9% de infecção bacteriana e 3,6%, de infecção fúngica a partir das 56 orelhas. Os micro-organismos mais isolados foram Staphylococcus intermedius, Staphylococcus aureus, Proteus mirabilis e Malassezia pachydermatis. As bactérias GRAM-positivas foram sensíveis à gentamicina em 60,5% e resistentes em 16,3% das amostras. A formulação com 5% de óleo essencial de Melaleuca produziu uma zona de inibição de 5mm em torno dos discos em uma das 63 amostras avaliadas. A formulação pura (100%) do mesmo produto produziu uma zona de 10mm de inibição em quatro das 63 amostras analisadas, uma zona de 9 mm em três amostras, uma zona de 8mm em 16 amostras, uma zona de 7mm em sete amostras, uma zona de 6mm em duas amostras e não havia nenhuma zona clara em 31 amostras. Zonas de inibição foram produzidas por estirpes de Staphylococcus intermedius, Staphylococcus hyicus, Corynebacterium sp., Proteus mirabilis e Enterobacter sp. Clinicamente, a formulação com o óleo essencial de melaleuca induziu significativamente uma melhora nas manifestações, tanto nas otites bacterianas quanto nas causadas por leveduras, sendo estatisticamente semelhante ao tratamento com nistatina (nas otites por levedura), mas menos eficaz que a solução de gentamicina nas otites bacterianas. Mais estudos devem ser realizados para avaliar as propriedades de difusão in vitro do óleo essencial de melaleuca. O bom espectro antimicrobiano, a boa resposta clínica e a ausência de reações adversas confirmam a possibilidade de desenvolvimento de formulação ótica com o óleo essencial de melaleuca, como uma alternativa para a terapia de infecções do ouvido em cães.


#4 - Ethanol poisoning in cattle fed with malted barley waste with brewer’s yeast

Abstract in English:

An outbreak of ethanol poisoning that affected a dairy cattle herd fed with the brewery by-product known as malt bagasse, wet brewery residue, malted barley waste or “barley”. The outbreak began about 24 hours after a new product of the by-product was offered to cattle that had an alcoholic odor. Chromatographic and microbiological analysis of this by-product sample confirmed the presence of ethanol and Saccharomyces spp., respectively, indicating the addition of another by-product brewery, brewer’s yeast or yeast. The main clinical signs observed were diarrhea, salivation, staggering gait and decubitus. Morbidity was 12.2% (5/41) and mortality was 2.4% (1/41). A cow that died after a 3-day of clinical course was necropsied. No significant macroscopic lesions were observed, but in the histopathology, there was acute, multifocal, moderate necrosupurative rumenitis with secondary bacterial and fungal colonization, indicating concomitant ruminal acidosis. In the chromatographic analysis of samples of rumen and liver contents of this bovine, variable amounts of ethanol were detected. The data from the present study indicate that the possibility of ethanol intoxication should be considered in cattle with neurological and digestive signs fed with RUC when added to brewer’s yeast.

O presente trabalho descreve um surto de intoxicação por etanol que afetou um rebanho bovino de aptidão leiteira alimentado com o subproduto de cervejaria denominado bagaço de malte, resíduo úmido de cervejaria (RUC), resíduo de cevada maltada ou simplesmente “cevada”. O surto iniciou cerca de 24 horas após ao fornecimento de uma nova partida do subproduto que apresentava odor alcoólico. Análise cromatográfica e microbiológica de amostra deste subproduto confirmou a presença de etanol e Saccharomyces spp., respectivamente, indicando a adição de outro subproduto de cervejaria, a levedura de cerveja ou levedo. Os principais sinais clínicos observados foram diarreia, salivação, andar cambaleante e decúbito. A morbidade foi de 12,2% (5/41) e mortalidade de 2,4% (1/41). Uma vaca que morreu após um curso clínico de 3 dias foi necropsiada. Não foram observadas lesões macroscópicas significativas, mas na histopatologia havia rumenite necrosupurativa aguda, multifocal, moderada, com colonização bacteriana e fúngica secundária, indicando acidose ruminal concomitante. Em análise cromatográfica de amostras de conteúdo ruminal e fígado deste bovino foram detectadas quantidades variáveis de etanol. Os dados do presente estudo indicam que a possibilidade de intoxicação por etanol deve ser considerada em bovinos com sinais neurológicos e digestivos alimentados com RUC quando a este acrescentado levedura de cerveja.

E-mail: mariporsani@hotmail.com Recently, glutamine and β-glucan have been demonstrated to play an important role in modulation of the immune system and in promoting intestinal health benefits. The aim of this study was to investigate the effect of this intervention on inflammatory responses and intestinal health in mice orally pretreated with soluble Saccharomyces cerevisiae derived 1,3/1,6-β-glucan (80mg/kg) with or without glutamine (150mg/kg) and then challenged with cytarabine (Ara-C) (15mg/kg). Improvements in villi and crypts were not observed in the β-glucan group. The intestinal morphometry in the glutamine group showed the best results. β-glucan in combination with glutamine presented the highest values of IL-1β and IL-10 and lowest values for leukocytes and INF-γ. Based on these results, combined β-glucan and glutamine pretreatment reduced intestinal inflammation and improved the immune response after Ara-C challenge.

O objetivo deste estudo foi investigar o efeito dessa intervenção sobre as respostas inflamatórias e saúde intestinal de camundongos pré- tratados por via oral com 1,3/1,6-β-glucano (80mg/kg) derivado de Saccharomyces cerevisiae com ou sem glutamina (150mg/kg) e posteriormente desafiados com citarabina (Ara-C) (15mg/kg). Melhoras em vilosidades e criptas não foram observadas no grupo de tratamento com β-glucano. A morfometria intestinal no grupo de tratamento com glutamina apresentou os melhores resultados. O grupo em que foi utilizado β-glucano em combinação com glutamina apresentou os maiores valores de IL-1β e IL -10 e valores mais baixos para os leucócitos e INF-γ. Com base nestes resultados, o pré-tratamento de β-glucano combinado com glutamina reduziu a inflamação intestinal e melhorou a resposta imune após o desafio com Ara-C.

E-mail: rinaldo.mota@hotmail.com The aim of this study was to evaluate in vitro sensibility of yeast isolated from milk of dairy cows with mastitis against some commercial disinfectants used in pre and post-dipping in dairy farms of the State of Pernambuco. After identification, 12 yeast isolates were tested with the following disinfectants: iodine at 0,57%, chlorine at 2,5%, chlorhexidine, lactic acid and quaternary ammonium at 2,0%, at different exposure times. It was observed that 100% of the isolates were sensitive to chlorhexidine at all exposure times. Iodine has obtained the second best result with the following percentages: 83,33% were susceptible at 15’’ exposure, 91,7% in 30’’ and 100% were sensitive at 60’’ 300’’ and 600’’ times. For lactic acid, the results were 41,67% sensitive at 15’’ of exposure, 58,33% at 30’’, 66,7% at 60’’ and 300’’ and 75% at 600’’. In the analysis of the results for quaternary ammonium, it was observed that 66,67% were sensitive in 15’’and 30’’ times and 91,67% at 60’’, 300’’ and 600’’. Regarding the chlorine activity, only 16,67% were sensitive at the 15’’ and 30’’ times, and 25% at 60’’, 300’’ and 600’’. It was concluded that chlorhexidine and iodine have disinfectant activity significantly superior to chlorine, against to yeast involved in infectious processes of the mammary gland in cattle. It is necessary to perform a periodic assessment of the disinfectant activity from products used in the pre and post-dipping routine against dairy farms microorganisms most commonly involved in cases of mastitis (bacterial or fungal) which is an effective alternative to reduce the frequency of mastitis in cattle.

Observou-se que 100% dos isolados mostraram-se sensíveis ao clorexidine, em todos os tempos de exposição. O iodo obteve o segundo melhor resultado com os seguintes percentuais: 83,33% foram sensíveis em 15” de exposição, 91,67% em 30” e 100% foram sensíveis nos tempos 60”, 300” e 600”. Para o ácido láctico, os resultados foram: 41,67% sensíveis em 15” de exposição, 58,33% em 30”, 66,67% em 60” e 300” e 75% em 600”. Na Análise dos resultados para amônia quaternária, observou-se que 66,67% foram sensíveis nos tempos 15” e 30”, e 91,67% em 60”, 300” e 600”. Em relação ao princípio ativo cloro apenas 16,67% foram sensíveis no tempo de 15” e 30”, e 25% em 60”, 300” e 600”. Conclui-se que o clorexidine e o iodo apresentam atividade desinfetante significativamente superior ao cloro, frente a leveduras envolvidas nos processos infecciosos da glândula mamária em bovinos. É necessário realizar uma avaliação periódica da atividade desinfetante dos produtos utilizados na rotina do pré e pós-dipping nas propriedades leiteiras frente aos microrganismos mais comumente envolvidos nos casos de mastite (micótica ou bacteriana), pois esta é uma medida eficaz para reduzir a frequência de casos de mastite nos rebanhos.

E-mail: aemurakami@uem.br An experiment was carried out to evaluate the effect increasing levels of yeast wall and broiler breeders’ age levels on lymphoid organs weight, cellular immune response and hematological parameters in broilers. A total of 3,360 Cobb broilers were allotted, in a completely randomized design and a 2x5 factorial arrangement, and two controls, compound of two broiler breeders age (34 and 57 weeks of age) and five yeast wall levels (zero, one, two, three and four kg of yeast wall/ton of diet). Broiler breeders’ age affected all studied variables. The inclusion of 3kg of yeast wall/ton of meal increased, at 57 weeks age broiler breeders, more intense inflammatory reaction when compared to control diet; however the circulated heterophils and lymphocytes numbers were not increased. In conclusion, the use of yeast wall, associated or not to sorghum on broilers diet still needs complementary studies, for example, purified components of yeast wall (MOS and ß-glucano).

E-mail: aemurakami@uem.br Um experimento foi conduzido para avaliar o efeito de níveis crescentes de parede de levedura e idade das matrizes reprodutoras sobre o peso dos órgãos linfóides, a resposta imune celular e o perfil hematológico de frangos de corte. Foram utilizados 3.360 pintos de corte da linhagem Cobb, distribuídos em delineamento inteiramente casualisado, em esquema fatorial 2x5, mais dois controles, sendo duas idades de matrizes (34 e 57 semanas de idade) e cinco níveis de suplementação de parede de levedura (zero, um, dois, três e quatro kg de parede de levedura/tonelada de ração). A idade das matrizes influenciou a resposta de todas as variáveis. A inclusão de 3kg de parede de levedura/tonelada de ração promoveu, na progênie de reprodutoras de 57 semanas, reação inflamatória mais intensa quando comparada a dieta controle, no entanto não houve aumento significativo no número de heterófilos e linfócitos circulantes. Conclui-se que a utilização da parede de levedura associada ao sorgo ou não em rações de frangos de corte ainda necessita de estudos complementares, que incluam, por exemplo, os componentes purificados da parede de levedura (MOS e ß-glucano).

Screening of antigenemia and isolation of Cryptococcus neoformans and C. gattii from cloaca and crop of birds in the state of Paraná, Brazil. Pesquisa Veterinária Brasileira 28(7):341-344. E-mail: camilelug@gmail.com Cryptococcus neoformans and C. gattii are associated with dry bird excreta but rarely recovered from birds’ digestive tract. The objective of the present study was (1) to verify the existence of C. neoformans and C. gattii in crop and cloaca of wildlife and captivity birds hypothesizing about a possible primary source of this yeast in the excreta, and (2) to determine the fungi’s invasive capability in avian species through latex agglutination. For that purpose, 172 cloacal and 77 crop samples of domestic pigeon, Passerine, and Psittacine birds were collected. None of these samples was positive, suggesting that the yeast is not saprobiotic in the digestive tract of these birds. Only one out of 82 serum samples collected from pigeons and Psittacine birds was positive (title 1:2) showing that Cryptococcus sp. probably has a low invasive capability in birds, and is thus considered only a dry excreta colonizer.
