#1 - Epigenetic characterization of the H19/IGF2 locus in calf clones placenta

Somatic Cell Nuclear Transfer (SCNT-Cloning) is a promising technique in many areas and is based on genetically identical individuals. However, its efficiency is low. Studies suggest that the leading cause is inadequate epigenetic reprogramming. This study aimed to characterize the methylation pattern of the exon 10 regions of the IGF2 gene and the Imprinting Control Region (ICR) of the H19 gene in the placenta of cloned calves. For this study, female and male cloned calves presenting different phenotypes were used. Genomic DNA from these animals’ placenta was isolated, then treated with sodium bisulfite and amplified to the ICR/H19 and IGF2 loci. PCR products were cloned into competent bacteria and finally sequenced. A significant difference was found between controls and clones with healthy phenotypes for the ICR/H19 region. In this region, controls showed a hemimethylated pattern, as predicted in the literature due to this region has an imprinted control, while clones were showed less methylated. For the IGF2, no significant differences were found between controls and clones. These results suggest that different genomic regions in the genome may be independently reprogrammed and that failures in reprogramming the DNA methylation patterns of imprinted genes may be one of the causes of the low efficiency of SCNT.

A Transferência Nuclear de Células Somáticas (TNCS-Clonagem) é uma técnica promissora em várias áreas, e se baseia na produção de indivíduos geneticamente idênticos. No entanto, sua eficiência é baixa. Estudos sugerem que a principal causa seja uma reprogramação epigenética inadequada. O objetivo desse trabalho é caracterizar o padrão de metilação da região éxon 10 do gene IGF2 e da Região Controladora de Imprinting (ICR) do gene H19 na placenta de bezerros clonados. Para a execução do trabalho foram selecionados clones bovinos fêmeas e machos, apresentando diferentes fenótipos. O DNA da placenta desses animais foi extraído, e em seguida foi tratado com bissulfito de sódio e amplificado para os loci ICR/H19 e IGF2. Os produtos da PCR foram clonados em bactérias competentes e, por fim, sequenciados. Foi encontrada uma diferença significativa entre os controles e os clones com fenótipos saudáveis para a região da ICR/H19. Nesta região, os controles tiveram um padrão hemimetilado, como previsto pela literatura, devido essa região ser imprinted. Enquanto os clones encontravam-se menos metilados. Para a região do éxon 10 do IGF2, não foi encontrada diferença significativa entre controles e clones. Estes resultados sugerem que as diferentes regiões do genoma podem se reprogramar independente umas das outras e que falhas na reprogramação do padrão de metilação do DNA de genes imprinted podem ser uma das causas da baixa eficiência da TNCS.


#2 - Pathological features of cloned calves that died in the neonatal period

Abstract in English:

Somatic-cell nuclear transfer is a cloning technique that enables the creation of a viable embryo from a donor adult to produce a genetically identical individual. This technique opens numerous potential possibilities for medicine and animal reproduction. However, several reports have documented cloning-related issues. Embryo and fetal losses remain significantly higher than in other techniques, and there is a high incidence of dystocia and hydrops, which decreases efficiency and increases costs. Animals delivered at term often exhibit a syndrome known as macrosomia and experience difficulties in adapting to life outside the uterus, and death is a common outcome. In the present study, 41 cloned calves that died in the neonatal period were subjected to gross and histopathological examination. Most important gross lesions were found in the liver (enlargement, congestion, yellowish color), kidneys (brownish color at surface and cut, and cysts), lungs (atelectasis, parenchymal consolidation, and secretions in bronchi and bronchioles), and heart (concentric and eccentric hypertrophy, hematic cysts, persistence of ductus arteriosus). Primary microscopic findings were seen in the liver, kidneys, and lungs from neonatal calves. In the liver, 85% of the animals exhibited hepatic degeneration. The presence of a brownish pigment within the cortical tubules of the kidneys was found in approximately 90% of the samples; the presence of this pigment has not been previously reported in cloned calves. In the lungs, a large number of animals exhibiting lesions characteristic of pneumonia (55%). These changes were the pivotal causes of death, mainly due to problems in adapting to life outside the uterus and opportunistic infections in the neonatal period. Further investigation focusing on pathological anatomical changes is necessary to map these abnormalities in cloned animals.

A transferência nuclear de células somáticas ou clonagem é uma técnica que permite produzir um indivíduo geneticamente igual a um outro indivíduo adulto. Esta técnica abre inúmeras possibilidades para a medicina e para a reprodução animal. Porém, existem inúmeros relatos de problemas associados à clonagem. A taxa de perda nos períodos embrionário e fetal ainda é muito alta quando comparada a outras biotécnicas; além disso, há uma maior incidência de hidropsias e distocias, diminuindo a eficiência e aumentando o custo da técnica. Os animais que vem a termo frequentemente apresentam uma síndrome chamada de macrossomia, e apresentam dificuldades de adaptação à vida extrauterina e, por isso, o óbito é um desfecho comum. No presente trabalho realizou-se necropsia e coleta de fragmentos de órgãos para avaliação histopatológica de 41 bezerros com óbito neonatal. As lesões macroscópicas mais importantes foram encontradas no fígado (hepatomegalia, congestão e coloração amarelada), rins (coloração amarronzada na superfície e ao corte, e cistos), pulmões (atelectasia, parênquima consolidado, e secreções nos brônquios e bronquíolos), e coração (hipertrofia concêntrica e excêntrica, cistos hemáticos e persistência de ducto arterioso). As principais alterações microscópicas observadas foram presença de pigmento acastanhado no interior dos túbulos corticais renais (aproximadamente 90% dos animais), degeneração hepática (85% das amostras avaliadas) e lesões características de pneumonia (55% dos animais). A pigmentação acastanhada no interior dos túbulos corticais é uma alteração que ainda não havia sido relatada anteriormente em animais clonados. As alterações observadas nestes órgãos foram determinantes para o óbito, e devem ter ocorrido sobretudo devido a problemas na adaptação ao ambiente extrauterino e em decorrência de infecções adquiridas no período neonatal. Os achados encontrados no presente trabalho denotam a necessidade de investigação anatomopatológica detalhada de animais clonados inviáveis, na tentativa de mapear as anormalidades apresentadas por eles.

E-mail: verechia@gmail.com The transgenic application of green fluorescent protein (GFP) as fetal cell marker on cattle cloned placenta could provide an exclusive model for studying the morphologic and immunologic maternal-fetal interactions, providing information about its mapping, distinguishing the fetal from maternal cells. This model will have direct application, mainly because these animals present problems during its development. With this model’s support, we intend to verify the substances transport between mother and fetus during endocytosis, through the immunolocalization of protein named caveolae. For these, we used 06 cloned bovine and 30 cattle samples of artificial insemination (AI) with 90 days of pregnancy, which had been their development interrupted by humanitarian slaughter of the recipient and recovery of the pregnant uterus. We collected the placentome and the chorion. A part of the samples was cut and fixed, by immersion, on a solution containing 4% of parafomaldehyde or 10% of formaldehyde on a sodium phosphate buffer (PBS), at 0,1M pH 7.4, Zamboni solution (4% of paraformaldehyde, 15% of picric acid, on sodium phosphate buffer 0,1M pH 7.4), metacarn (60% of metanol, 30% of chloroform, and 10% glacial acetic acid), for morphologic and immunohistochemistry verification for caveolinas proteins -1 and -2 (CAV -1 and CAV- 2). The caveolins -1 were found in fetal and maternal villi, but its strongest staining was observed in the endometrial stroma. The caveolins -2 had positive staining in trophoblast and chorioallantoic membrane, and specifically in giant trophoblastic binucleated cell. Therefore the results were compared between cloned cattle and from AI or natural mating, for assisting on detection of the reason of many placental alterations, embryonic losses, spontaneous abortion, post-natal mortality and large offspring syndrome on laboratory-manipulated animals. The result suggests that the proteins caveolins -1 and -2 (CAV-1 and CAV-2) are part of the caveolae composition and important structures related to the molecule transfer to the fetus, nourish it through endocytosis and pinocytosis.

E-mail: verechia@gmail.com A utilização da transgenia com a proteína fluorescente verde (GFP) como marcador de células de origem fetal nas placentas de clones bovinos servirá de modelo inédito para estudo morfofisiológico e imunológico da interação materno-fetal, visto que possibilitará o seu mapeamento, diferenciando as células fetais das maternas. Tal modelo terá aplicação direta, principalmente porque estes são animais que apresentam problemas em relação ao seu desenvolvimento. Com o auxílio deste modelo, pretende-se verificar o transporte de substâncias entre a mãe e o feto via endocitose, pela imunolocalização das proteínas chamadas de caveolinas. Para tanto foram utilizados 06 bovinos clonados e 30 bovinos de inseminação artificial (IA) com idade até 90 dias de gestação, os quais tiveram seu desenvolvimento interrompido mediante abate humanitário das receptoras e ovariosalpingohisterectomia, com posterior recuperação do útero gestante. Foram coletados os placentônios e o cório. Uma parte das amostras foi recortada e fixada, por imersão, em solução de parafolmaldeído a 4% ou formoldeído a 10% em tampão fosfato de sódio (PBS) a 0,1M pH 7.4, solução de Zamboni (4% de paraformoldeído, 15% de ácido pícrico, em tampão fosfato de sódio a 0,1M pH 7.4), metacarn (60% de metanol, 30% de clorofórmio, e 10% de ácido acético glacial), para verificação da morfologia e realização de imuno-histoquímica para as proteínas caveolinas -1 e -2 (CAV -1 e CAV-2). As caveolinas -1 foram localizadas nos vilos fetais e maternos, mas sua marcação mais forte foi observada no estroma endometrial. As caveolinas -2 tiveram marcação positiva no trofoblasto e membrana córioalantoide, e, especificamente em célula trofoblástica gigante binucleada. Sendo assim, os resultados mostram que a proteína CAV-1 teve uma maior expressão em relação à proteína CAV-2 e que as proteínas CAV-1 e -2 são parte da composição das cavéolas, sendo estruturas importantes e relacionadas com a transferência de moléculas para o feto, realizando a nutrição do mesmo mediante endocitose e pinocitose.

Os CNC foram divididos segundo o período de gestação em Grupo I (2-3 meses, n=9), Grupo II (4-6 meses, n=9), Grupo III (7-8 meses, n=10) e Grupo IV (9 meses, n=7). Os CC formaram o Grupo V (9 meses, n=4). As carúnculas foram observadas macroscopicamente (número e dimensões: comprimento, largura e altura), microscopicamente e submetidas à análise estatística (5% de significância). Observaram-se três tipos de fusões carunculares macroscópicas: ovais (morfologicamente normais); duas carúnculas adjacentes unidas e do tipo lobuladas, caracterizadas por regiões com várias carúnculas unidas apresentando falsa fusão ou deformação do parênquima caruncular. O comprimento das carúnculas foi de 1,55±0,57; 2,45±0,55; 4,66±2,00 e 5,72±1,90cm para os Grupos I, II, III e IV, respectivamente. Quanto à altura, as carúnculas apresentaram um crescimento linear durante a gestação e foram de 0,40±0,15; 0;57±0,21; 1,00±0,48 e 1,80±0,91cm, para os respectivos Grupos I, II, III e IV. A largura das carúnculas foi semelhante entre os Grupos I e II (0,97±0,30 e 1,42±0,71cm) e os Grupos III e IV (2,68±1,22 e 3,52±1,16cm). Quando o Grupo V foi comparado ao Grupo IV, as carúnculas do Grupo V apresentaram maior comprimento (5,72±1,90 vs. 7,88±2,13cm) e largura (3,52±1,16 vs. 4,93±1,46cm), porém foram semelhantes em altura (1,80±0,91 e 2,25±0,67cm). Verificou-se que em gestações de CC, as carúnculas apresentaram maior desenvolvimento que em gestações de CNC. As carúnculas fusionadas apresentaram medidas estatisticamente semelhantes às isoladas em todos os parâmetros e grupos. Sob microscopia de luz, observou-se a formação de um eixo estromal, da base da carúncula ao ápice da fissura fusional, de constituição histológica semelhante ao estroma endometrial. Também foram ineditamente definidos três formatos microscópicos: fusão propriamente dita com eixo único, evidente abaixo da fissura fusional; pseudofusão com eixo duplo em forma de “H” e a falsa fusão com ausência do eixo. Os dois primeiros formatos referem-se às carúnculas ovais e lobuladas e o último à falsa fusão com deformação do parênquima caruncular. O eixo fusional aumentou de tamanho durante a gestação entre os Grupos I, II, III e IV. O Grupo V apresentou maior comprimento e largura de eixo quando comparado ao Grupo IV. Sendo assim, em gestações de CC a destruição do epitélio lateral das carúnculas está associada a uma incompetência na interdigitação materno-fetal, que comprometem a fusão cotiledonária. Sugere-se que, em gestações provenientes de CC, o aumento do tamanho das fusões carunculares possivelmente associa-se a um mecanismo compensatório para as trocas metabólicas entre mãe e feto, em razão do menor número de carúnculas isoladas.

E-mail: fverechia@dracena.unesp.br The objective of the study was to compare the characteristics of the caruncular fusion in gestations of non-cloned and cloned conceptuses. The non-cloned conceptuses were divided according to the gestation period: Group I (2 to 3 months; n=9), II (4 to 6; n=9); III (7 to 8; n=10) and IV (9 n=7). The cloned conceptuses formed the Group V: 9 months; n=4. The caruncles were observed macroscopically (number and dimensions: length, width and height), microscopically and submitted to statistical analysis (5% of significance). We observed three types of macroscopic caruncular fusions: oval (morphologically normal); two united adjacent caruncles and the lobulated type, characterized by regions with several united caruncles presenting a false fusion or deformation of the caruncular parenchyma. The length of the caruncles was 1.55±0.57; 2.45±0.55; 4.66±2.0 and 5.72±1.90cm for the groups I, II, III, IV respectively. As for the height, the caruncles presented a lineal growth during the gestation: 0.40±0.15; 0.57±0.21; 1.0±0.48 and 1.80±0.91cm, for the respective groups I, II, III and IV. The width of the caruncles was similar between the groups I and II (0.97±0.30 e 1.42±0.71cm) and the groups III and IV (2.68±1.22 and 3.52±1.16cm). When the group V was compared to the IV, the caruncles of the group V presented a larger length (5.72±1.90 vs. 7.88±.13cm) and width (3.52±1.16 vs. 4.93±1.46cm), however they were similar in height (1.80±0.91 and 2.25±0.67cm). We verified that in gestations of cloned conceptuses the caruncles presented a larger development than in gestations of non-cloned conceptuses. The fusioned caruncles presented measurements statistically similar to the isolated ones in all the parameters and groups. Under light microscopy, we observed the formation of a stromal axis from the basis of the caruncle to the apex of the fusional fissure, with the histological constitution similar to the endometrial stroma. Three microscopic shapes were also unpublished defined: true fusion with a single axis evident below the fusional fissure; pseudofusion with a double axis in “H” shape and false fusion with absence of the axis. The first two formats were associated to the oval and lobulated caruncles and the last one to the false fusion with deformation of the caruncle parenchyma. The fusional axis increased in size along the gestation among the groups I, II, III and IV. The group V presented a larger length and width of the axis when compared to the group IV. Thus, in gestations of cloned conceptuses a destruction of the lateral epithelium of the caruncles is associated to an incompetence in the maternal-fetal interdigitation, that compromises the cotyledonary fusion. We suggest that, in gestations derived of cloned conceptuses, the increase of the size of the caruncular fusions is possibly associated to a compensatory mechanism for the metabolic exchanges between mother and fetus, in reason of the smallest number of isolated caruncles.


#5 - Occurrence of virulence-related sequences and phylogenetic analysis of commensal and pathogenic avian Escherichia coli strains (APEC), p.533-540

E-mail: wds@unicamp.br The presence of iron uptake (irp-2, fyuA, sitA, fepC, iucA), adhesion (iha, lpfAO157/O141, lpfAO157/O154, efa, toxB) and invasion (inv, ial-related DNA sequences and assignment to the four main Escherichia coli phylogenetic groups (A, B1, B2 e D) were determined in 30 commensal E. coli strains isolated from healthy chickens and in 49 APEC strains isolated from chickens presenting clinical signs of septicemia (n=24) swollen head syndrome (n=14) and omphalitis (n=11) by PCR. None of the strains presented DNA sequences related to the inv, ial, efa, and toxB genes. DNA sequences related to lpfAO157/O154, iucA, fepC, and irp-2 genes were significantly found among pathogenic strains, where iucA gene was associated with septicemia and swollen head syndrome and fepC and irp-2 genes were associated with swollen head syndrome strains. Phylogenetic typing showed that commensal and omphalitis strains belonged mainly to phylogenetic Group A and swollen head syndrome to phylogenetic Group D. Septicemic strains were assigned in phylogenetic Groups A and D. These data could suggest that clonal lineage of septicemic APEC strains have a multiple ancestor origin; one from a pathogenic bacteria ancestor and other from a non-pathogenic ancestor that evolved by the acquisition of virulence related sequences through horizontal gene transfer. Swollen head syndrome may constitute a pathogenic clonal group. By the other side, omphalitis strains probably constitute a non-pathogenic clonal group, and could cause omphalitis as an opportunistic infection. The sharing of virulence related sequences by human pathogenic E. coli and APEC strains could indicate that APEC strains could be a source of virulence genes to human strains and could represent a zoonotic risk.

#6 - Antigenic differences of clones of Moraxella bovis

Abstract in English:

Some antigenic differences of clones of Moraxella bovis recovered from a 6 months old bullock with infectious keratoconjunctivitis were studied through different techniques. The clones were classified biochemically by standard procedures. Seven polyclonal sera were produced in rabbits and tested against 12 clones by double immunodiffusion. Six monoclonal antibodies produced against M. bovis GF9 were used to quantify titles contained in five clones through the hemagglutination inhibition technique. Six clones were used in an ELISA with the panel of monoclonal antibodies. Immunodiffusion detected that only one clone was different from the others, while hemagglutination showed that the five clones studied were different. ELISA showed no clear cut differences among the clones. It can be concluded that the M. bovis population present in the animal studied is antigenically heterogeneous and that the hemagglutination inhibition technique may detect these differences.

Foram estudadas diferenças antigênicas de clones de Moraxella bovis recuperados de um terneiro com 6 meses de idade, acometido pela primeira vez de ceratoconjuntivite infecciosa bovina. Sete soros policlonais produzidos em coelhos foram testados contra 12 clones por imunodifusão dupla. Seis anticorpos nionoclonais produzidos a partir da amostra GF9 de M. bovis foram usados para verificar títulos de cinco clones através da técnica da inibição da hemoaglutinação. O teste ELISA foi utilizado para detectar reações de seis clones. Através da imunodifusão verificou-se que apenas um clone foi diferente dos outros. Contudo, pela técnica de inibição da hemoaglutinação observou-se que os cinco clones estudados eram diferentes. O teste ELISA não demonstrou diferenças claras entre os clones.
