#1 - Casuistry of xenarthrans treated at the Veterinary Hospital of the Federal University of Pará, Brazilian Amazon

Abstract in English:

The present study aimed to analyze the casuistry of xenarthrans treated at the veterinary hospital of the Federal University of Pará to quantify the most important cases for clinical care and identify the main species, sex, age group, origin, and destination of the animals admitted. The data were collected from January 2013 to August 2020. The variables analyzed were the number of animals by species, sex, age group, weight (kilograms), form of admission (rescue or guardianship), person responsible for the admission of the animal, origin, circumstances of admission, length of stay (days), and evolution or destination of the animal (release, captivity, death, euthanasia, or escape). Fisher’s exact test was used to determine whether disease incidence was related to sex and whether animal mortality was influenced by sex and disease. The chi-square test was used to determine whether mortality differed by age group. Ninety-two (92) xenarthrans were included: 66 Bradypus variegatus, 10 Choloepus didactylus, 15 Tamandua tetradactyla, and one Dasypus novemcinctus. The animals were mainly from the municipality of Castanhal (70%) and were acquired through rescue (88%, B. variegatus; 80%, C. didactylus; 100%, T. tetradactyla), a significant proportion of which was per natural persons (88%, B. variegatus; 80%, C. didactylus; 100%, T. tetradactyla). The most important circumstances that led to the admission of the animals were as follows: animals for examination (42%, n=39), young animals at risk (9%, n=8), hit by a vehicle (8%, n=7), respiratory disease (7%, n=6), fall from a tree (5%, n=5), and electrotrauma (5%, n=5). Release was the most common destination of animals after hospitalization (59% B. variegatus, 50% C. didactylus, and 66% T. tetradactyla). The sex of the animals did not affect the incidence of disease or mortality in any species. In contrast, mortality of B. variegatus was significantly higher in animals admitted with disease (p=0.000). Mortality in cubs of this species was higher than that in juveniles and adults (p=0.003).

O objetivo deste trabalho foi analisar a casuística dos Xenarthras atendidos no Hospital Veterinário da Universidade Federal do Pará (UFPA), a fim de quantificar as principais ocorrências para atendimento clínico, identificar as principais espécies, o sexo, a faixa etária, a procedência e destinação dos animais recebidos. Os dados foram obtidos entre janeiro de 2013 a agosto de 2020. As variáveis analisadas foram número de animais por espécie, sexo, faixa etária, peso (quilogramas), forma de recebimento (resgate, tutela), responsável pelo encaminhamento do animal ao hospital, procedência, circunstância de admissão, tempo de internação (dias) e evolução ou destinação do animal recebido (soltura, cativeiro, morte, eutanásia, fuga). O teste de Fisher foi utilizado para avaliar se a ocorrência de doenças estava relacionada ao sexo, bem como se a mortalidade animal foi influenciada pelo sexo e por enfermidades. O teste Qui-quadrado foi utilizado para avaliar se a mortalidade diferiu em relação à faixa etária. Um total de 92 animais Xenarthras foi admitido, composto por 66 Bradypus variegatus, 10 Choloepus didactylus, 15 Tamandua tetradactyla e um Dasypus novemcinctus. Os animais foram procedentes principalmente do município de Castanhal (70%), sendo o resgate a principal forma de aquisição dos animais (88%, B. Variegatus; 80%, C. Didactylus; 100%, T. tetradactyla), realizado em maior parte por pessoa física (88%, B. Variegatus; 80%, C. Didactylus; 100%, T. tetradactyla). O sexo dos animais não influenciou a ocorrência de doenças ou mortalidade em nenhuma espécie. Por outro lado, a mortalidade de B. variegatus foi significativamente maior naqueles animais admitidos com alguma enfermidade (p=0,000). A mortalidade em filhotes desta espécie foi maior quando comparada com as categorias jovem e adulta (p=0,003).


#2 - Parasitic macrofauna of Podocnemis unifilis from the Tocantins and Javaés Rivers, Eastern Amazon, Brazil

Abstract in English:

The parasitic fauna of Podocnemididae species is of great relevance to understanding the ecological relationships of the organisms in this genus. The parasites can have a symbiotic relationship with their hosts; however, environmental imbalances may change this relationship. Therefore, these parasites may become agents that generate zoonoses, especially in animals used by parasites as a food source. The aim of this study was to investigate the gastrointestinal parasitic fauna of specimens of Podocnemis unifilis. The animals were collected from the Tocantins and Javaés Rivers, both of which flow in the State of Tocantins, predominantly in the Cerrado territory. Samples of stomach and intestinal contents were fixed in 10% formalin buffered with sodium phosphate, transferred to 70% alcohol for analysis, and separated after 72 hours. Parasites were isolated and identified according to morphology and identification key. Four species of parasites were observed in 100% of specimens of P. unifilis, namely Nematophila grandis (Trematoda) and Paratractis hystrix, Paraorientatractis semiannulata, and Orientatractis leiperi (Nematoda). N. grandis was the only parasite that was not observed in the intestine; it appeared to adhere to the inner wall of the stomach. All nematodes were observed both in the stomach and intestine. These data expand the knowledge related to the P. unifilis parasitic fauna and the geographic distribution of these parasites in P. unifilis from the Tocantins and Javaés Rivers.

A fauna parasitológica das espécies de Podocnemididae é de grande relevância para o conhecimento das relações ecológicas desse grupo de vertebrados. Os parasitos podem ocorrer em simbiose com os seus hospedeiros sem causar danos, mas o desequilíbrio ambiental pode causar um desequilíbrio nessa relação e, assim, serem agentes geradores de zoonoses, especialmente em animais que são utilizados como recurso alimentar. Esta pesquisa fez o levantamento da fauna parasitária gastrointestinal de espécimes de Podocnemis unifilis. Os animais foram coletados no rio Tocantins e Javaés, ambos no Estado do Tocantins, em território predominante de Cerrado. As amostras de conteúdo estomacal e intestinal foram fixadas em formol a 10% tamponado em fosfato de sódio, transferidas para álcool 70% para análise e separação após 72 horas. Os parasitos foram separados e identificados de acordo com morfologia e chave de identificação. Foram encontradas quatro espécies de parasitos com prevalência em 100% dos espécimes de P. unifilis, sendo elas Nematophila grandis (Trematoda) e Paratractis hystrix, Paraorientatractis semiannulata e Orientatractis leiperi (Nematoda). Somente N. grandis não foi observado no intestino, sendo encontrado somente aderido à parede interna do estômago. Todos os nematoides foram observados no estômago e intestino. Esses dados ampliam o conhecimento sobre a fauna parasitária de P. unifilis, e aumentam o conhecimento sobre a distribuição geográfica desses parasitos em P. unifilis dos Rios Tocantins e Javaés.


#3 - Prevalence of canine atopic dermatitis at the Veterinary Hospital of the “Universidade Federal Rural da Amazônia” in Belém/Pará, Brazil

Abstract in English:

Canine atopic dermatitis (CAD) is a pruritic, chronic inflammatory disease, recurrent and genetically predisposed, which is the second most frequent allergic skin disorder, and ranks second among all the causes of pruritus in dogs worldwide. Given the absence of data on the occurrence of CAD in the northern region of Brazil, the aim of the current study was to conduct a survey to define the prevalence of canine atopic dermatitis attended at the Dermatology Department of the Mário Dias Teixeira Veterinary Hospital of the “Universidade Federal Rural da Amazônia” (HOVET-UFRA). To determine the prevalence of CAD, a retrospective survey was carried out of clinical records and results of dermatological examinations conducted at the Dermatology Department of HOVET-UFRA Belém, Pará from October 2018 to October 2019. During this period, 456 dogs were examined, of which 25.65% (117) were diagnosed with atopic dermatitis. Among the animals diagnosed, 62.4% (73) were females and 51.7% (29) were of the Shih-tzu breed. This level of atopic dermatitis is considered high. There are still no exact data on the incidence and prevalence of this dermatopathy, with described occurrence ranging from 3 to 15% of the canine population, a geographical relationship may be present. Although a sex-related predisposition has not been proven, a higher incidence of atopy in females is described, which indicated this may be the case. The most commonly diagnosed dogs in this study were the Shih-tzu breed. It is suspected that the regional popularity of some breeds, or the different genetic backgrounds in different geographical areas, may affect the predominance of CAD in some breeds. The results of the present study demonstrate the need for more research on the prevalence of canine atopic dermatitis, and better means of characterizing the population of atopic dogs in the region, so that it is possible to obtain a reliable epidemiological profile.

A dermatite atópica canina (DAC) é uma doença inflamatória crônica e pruriginosa recorrente e geneticamente predisposta, que se destaca como o segundo transtorno cutâneo alérgico mais frequente e ocupa o segundo lugar entre todas as causas de prurido em cães. Diante da ausência de dados da ocorrência de DAC na região Norte do Brasil, objetivou-se realizar um levantamento de dados para definir a prevalência de dermatite atópica canina atendida no Serviço de Dermatologia do Hospital Veterinário Mário Dias Teixeira da Universidade Federal Rural da Amazônia (HOVET-UFRA). Foi realizado um levantamento retrospectivo através dos registros clínicos e dos resultados de exames dermatológicos com base nos atendimentos dermatológicos no setor de Dermatologia do HOVET-UFRA Belém/Pará, de outubro de 2018 a outubro de 2019 para determinar a prevalência de DAC. Nesse período, foram atendidos 456 cães e destes, 25,65% (117) foram diagnosticados com dermatite atópica. Dentre os animais diagnosticados, 62,4% (73) eram fêmeas e 51,7% (29) eram da raça Shih-tzu. Neste estudo houve uma alta prevalência de cães com Dermatite atópica. Ainda não existem dados exatos sobre a incidência e a prevalência desta dermatopatia, com ocorrência descrita variando de 3 a 15% da população canina, podendo ter relação geográfica. Embora a predisposição sexual não tenha sido comprovada, descreve-se maior incidência de atopia em fêmeas, concordando com esta pesquisa. Os cães mais acometidos neste estudo foram da raça Shih-tzu. Suspeita-se que a popularidade regional de algumas raças ou os diferentes antecedentes genéticos em diferentes áreas geográficas afetam a predominância de algumas raças. Estes resultados mostraram que a prevalência de DAC em cães é a mais frequentemente diagnosticada no setor de Dermatologia do HOVET-UFRA, sendo mais prevalente em fêmeas e na raça Shih-tzu, podendo refletir um aspecto regional desta dermatopatia. Os resultados do presente estudo contribuíram para demonstrar a importância de mais pesquisas sobre a prevalência da Dermatite atópica canina e melhor caracterizar a população de cães atópicos na região, para que seja possível obter um perfil epidemiológico confiável.


#4 - Influence of seasonality on the physiological and seminal parameters of buffaloes in the western region of Pará

Abstract in English:

Awareness of the physiological changes that occur when animals are subjected to climatic changes that are considered stressful is essential to maintain animal welfare and to be able to exploit their reproductive potential efficiently and rationally. The present study was carried out to evaluate climatic variables’ influence on physiological parameters, and Murrah buffalo ejaculates reared in a humid tropical climate in the Amazon. The immediate analyzes pertinent to the physical and morphological characteristics of the ejaculates were carried out and corresponded in the rainy season (RS) volume of 3.4±2.0mL; the mass activity of 4.4±0.5; motility of 80.4±5.6%; vigor of 4.4±0.4; concentration of 657,300±237,865.1 x 106sptz/mL; major defects of 9.0±2.6%; minor defects of 11.2±3.9%; total defects 20.2±5.3% and sperm plasma membrane integrity (SPMI) 84.8±5.6%, whereas in the non-rainy season (nRS), the results were 4.0±2.1mL; the mass activity of 3.0±1.0; motility of 56.2±13.4%; vigor of 3.0±1.0; concentration of 586,000±291,925.9 x 106sptz/mL; major defects of 20.8±9.9%; minor defects of 27.5±6.3%; total defects 48.3±9.3% and SPMI of 57.9±12.4%. Furthermore, a statistical difference (P<0.05) was observed for the parameters mass activity, motility, vigor, major defects, minor defects, total defects, and sperm plasma membrane integrity between both periods. The data on heart frequency, superficial temperature (head, back, groin, and scrotal pouch) showed a statistical difference between both periods (P<0.05). To conclude is necessary specific management in the non-rainy season that thermal stress is not a determining factor in reducing the reproductive quality of buffaloes; it is necessary to use means to improve animal welfare; one alternative is to use baths regularly for these animals or provide constant access to areas of rivers or lakes, as well as shading, preventing the buffaloes from being directly exposed to the unfavorable thermal environment.

O conhecimento das alterações fisiológicas que ocorrem quando os animais são submetidos a alterações climatológicas consideradas estressantes é fundamental para manter o bem-estar animal, e poder explorar o seu potencial reprodutivo de forma eficiente e racional. O presente estudo foi realizado com o objetivo de avaliar a influência de variáveis climáticas sobre parâmetros fisiológicos e de ejaculados de búfalos, da raça Murrah, criados em clima tropical úmido da Amazônia. As análises imediatas pertinentes às características físicas e morfológicas dos ejaculados foram realizadas e corresponderam no período chuvoso (PCh) o volume de 3,4±2,0mL, turbilhonamento de 4,4±0,5; motilidade de 80,4±5,6%; vigor de 4,4±0,4; concentração de 657.300±237.865,1 x 106sptz/mL; defeitos maiores de 9,0±2,6%; defeitos menores de 11,2±3,9%; defeitos totais de 20,2±5,3% e integridade da membrana plasmática (IMP) de 84,8±5,6%, enquanto que no período não chuvoso (PnCh), os resultados foram de 4,0±2,1mL; turbilhonamento de 3,0±1,0; motilidade de 56,2±13,4%; vigor de 3,0±1,0; concentração de 586.000±291.925,9 x 106sptz/mL; defeitos maiores de 20,8±9,9%; defeitos menores de 27,5±6,3%; defeitos totais de 48,3±9,3% e IMP de 57,9±12,4%. Observou-se diferença estatística (P<0,05) para os parâmetros movimento de massa, motilidade, vigor, defeitos maiores, defeitos menores, defeitos totais e integridade da membrana plasmática entre os dois períodos. Dados de frequência cardíaca, temperatura superficial (cabeça, dorso, virilha e bolsa escrotal) diferiram estatisticamente entre os períodos (P<0,05). Conclui-se que se faz necessário usar de um manejo específico no período não chuvoso para que o estresse térmico não seja um fator determinante na redução da qualidade reprodutiva dos búfalos, para isto se faz necessário utilizar de meios para melhorar o bem-estar animal, sendo uma das alternativas fazer uso de banhos regularmente para estes animais, ou disponibilizar acesso constante destes a áreas de rios ou lagos, assim como sombreamentos, evitando que os búfalos fiquem expostos diretamente ao ambiente térmico desfavorável.


#5 - Detection of Treponema spp. in bovine digital dermatitis in the Amazon biome, Brazil

Abstract in English:

Bovine digital dermatitis (BDD) is a polybacterial claw disease that is endemic to dairy cattle kept in loose house systems, and treponemas are the main bacteria implicated in this disease. The objective of this study was to report the occurrence of Treponema spp. in BDD from crossbred dairy cattle (Holstein x Zebu) kept in a pasture in the Brazilian Amazon biome. The diagnostic of BDD was performed by inspecting the distal extremities of cattle during milking in one or more visits comprising 15 farms. In total, it could be inspected 1,847 cows from August 2016 to July 2017, and 25 lesions of BDD were diagnosed. The feet were scored (System M: M0 = no lesion, M1 = ulcer stage <2cm, M2 = ulcer stage >2cm, M3 = healing stage, M4 = chronic stage, M4.1 = chronic stage with ulcer area). Twenty four biopsy samples were taken from feet with BDD and five biopsy samples from feet with no lesions. The histopathology of stained tissues was performed by hematoxylin and eosin and Warthin-Starry method. The samples were also tested by nested PCR for the three previously isolated BDD Treponema phylogroups (T. medium/T. vincentii-like, T. phagedenis-like and T. putidum/T. denticola-like). Spirochetes were observed in 54.2% (13/24) of the lesions, and in 91.7% (22/24) of the samples were detected the DNA of this spirochete belonging to the treponema phylogroups implicated in BDD. In 25% (6/24) of the lesions were detected all the phylogroups. Forty percent (40%, 2/5) of the M0 samples were also positive for the nested Polymerase Chain Reaction (nested-PCR), as 8.3% (2/24) of the lesions were negative in both techniques employed. Treponema putidum/T. denticola-like was the most detected bacterial in all the stages, and active lesions (M2 and M4.1) presented a greater proportion of T. medium/T. vincentii-like and T. phagedenis-like, but no statistical differences were observed (p>0.05). It could be concluded that BDD lesions in crossbred dairy cattle kept to pasture in the Amazon biome were classified as “polytreponemal” infections and the phylogroup T. putidum/T. denticola-like was the most frequent in the lesions.

Dermatite digital bovina (DDB) é uma enfermidade polibacteriana dos dígitos endêmica em vacas leiteiras criadas em estábulos e as treponemas são as principais bactérias envolvidas. Este estudo teve como objetivo relatar a ocorrência de Treponema spp. em DDB em bovinos leiteiros mestiços (Holandês x Zebu) criados a pasto no bioma amazônico brasileiro. O diagnóstico da DDB foi realizado pela inspeção, em uma ou mais visitas, das extremidades distais das vacas durante a ordenha em 15 propriedades. No total, foram inspecionadas 1.847 vacas de agosto de 2016 a julho de 2017 e diagnosticou-se 25 lesões de DDB. As extremidades distais inspecionadas foram classificadas em escores (M0 = sem lesão, M1 = estágio ulcerado <2cm, M2 = estágio ulcerado >2cm, M3 = estágio em cicatrização, M4 = estágio crônico, M4.1 = estágio crônico com área ulcerada) e realizada 24 biópsias de dígitos com DDB e cinco biópsias de dígitos em estágio M0. Foram realizadas a histopatologia pelas colorações de hematoxilina e eosina e pelo método de Warthin-Starry, e a nested de reação em cadeia de polimerase (nested-PCR) para os três filogrupos de treponemas previamente isolados de DDB (Treponema medium/T. vincentii-like, T. phagedenis-like e T. putidum/T. denticola-like). Espiroquetas foram observadas em 54,2% (13/24) das lesões e em 91,7% (22/24) detectou-se o DNA de, pelo menos, um dos filogrupos de treponemas pesquisados. Em 25% (6/24) das lesões foram detectados o DNA dos três filogrupos. Em 40% (2/5) das amostras em estágio M0 também foram positivas na nested-PCR, assim como 8,3% (2/24) das lesões foram negativas em ambas as técnicas empregadas. T. putidum/T. denticola-like foi o filogrupo mais detectado em todos os estágios e lesões ativas (M2 e M4.1) apresentaram uma maior proporção para Treponema medium/T. vincentii-like e T. phagedenis-like, mas não se obteve diferença estatística na ocorrência dos filogrupos entre os estágios das lesões (P>0,05). Conclui-se que lesões de DDB em rebanhos leiteiros mestiços criados a pasto no bioma amazônico brasileiro são “politreponemais” e o filogrupo T. putidum/T. denticola-like é o mais frequente nas lesões.


#6 - Livestock rabies in Pará state, Brazil: a descriptive study (2004 to 2013)

Abstract in English:

Rabies is an important zoonosis to public health associated with lethal encephalitis and economic losses. Analysis of its spatial distribution is a meaningful tool in understanding its dispersion, which may contribute to the control and prophylaxis of the disease. This study analyzed the spatial-temporal distribution of rabies outbreaks in livestock in Pará state, Brazil, from 2004 to 2013. We used records of neurological syndromes obtained from the state’s livestock authority (Adepará). The analysis recorded 711 neurological syndromes reports in livestock, of which 32.8% were positive for rabies. In 8% of the neurological syndromes (n=57) was not possible to perform the analysis because of bad-packaging conditions of the samples sent. Outbreaks involved at least 1,179 animals and cattle were the most affected animal species (76.8%). The numbers of reported neurological syndromes and of rabies outbreak shad strong positive correlation and exhibited decreasing linear trend. Spatially, most outbreaks occurred in two mesoregions in Pará (Northeast and Southeast). One of the justifications for this spatial distribution may be related with the distribution of the animals in the state, since these mesoregions are the largest cattle producers in Pará and have most of their territory deforested for pasture implementation.

A raiva é uma zoonose importante para a saúde pública associada à encefalite letal e às perdas econômicas. A análise de sua distribuição espacial é uma ferramenta importante no entendimento de sua dispersão, o que pode contribuir para o controle e a profilaxia da doença. Este estudo analisou a distribuição espaço-temporal do surto de raiva em rebanhos no estado do Pará, Brasil, entre 2004 e 2013. Foram utilizados registros de síndromes neurológicas obtidas junto à agência de defesa agropecuária do estado (Adepará). A análise revelou 711 notificações de síndromes neurológicas em herbívoros, das quais 32,8% foram positivas para raiva. Em 8% das síndromes neurológicas (n=57) não foi possível realizar as análises devido às más condições das amostras enviadas. Surtos envolveram pelo menos 1.179 animais e os bovinos foram a espécie animal mais afetada (76,8%). Os números de síndromes neurológicas relatadas e de surtos de raiva apresentam forte correlação positiva e exibem tendência linear decrescente. Espacialmente, a maioria dos surtos ocorreu em duas mesorregiões no Pará (Nordeste e Sudeste). Uma das justificativas para essa distribuição espacial pode estar relacionada à distribuição dos animais no estado, uma vez que essas mesorregiões são os maiores produtores de gado do Pará e possuem grande parte do seu território desflorestado para implantação de pastagens.


#7 - Dirofilaria immitis infection in dogs in Algodoal Island, Brazilian Amazon

Abstract in English:

Dirofilaria immitis, a parasite that mainly infects domestic or wild canids, but can infect felines or humans as well, is frequent in many Brazilian areas. The main objective of this research was to determine the prevalence of natural canine infection at the Algodoal‑Maiandeua Island complex, in the coastal region of the state of Pará, Brazil. A total of 67 dogs were sampled for blood microfilariae detection and for D. immitis DNA detection. Microfilaria and D. immitis DNA could be detected in 35.8% (24/67) of the animals. In one dog’s sample no microfilariae were detected, but the PCR was positive, suggesting that either larvae recently were eliminated or adults died shortly before sample collecting. Therefore, it can be concluded that the occurrence of D. immitis is a health threat for domestic and wild canids at the Island of Algodoal, as well as for feline or human health.

Dirofilaria immitis, um parasito que infecta principalmente canídeos domésticos ou selvagens, embora também possa infectar felinos e humanos, é frequente em muitas áreas do Brasil. O objetivo deste estudo foi determinar a ocorrência da infecção natural em cães provenientes do complexo da Ilha de Algodoal-Maiandeua, região litorânea do estado do Pará, Brasil. Um total de 67 cães tiveram o sangue coletado para detecção de microfilárias de D. immitis e seu DNA. Microfilárias e o DNA de D. immitis foram detectados em 35,8% (24/67) dos animais. Na amostra de um animal, não foram observadas microfilárias, mas o seu DNA foi detectado, sugerindo que as larvas tenham sido recentemente eliminadas ou os adultos tenham morrido antes da coleta da amostra. Portanto, pode-se concluir que a ocorrência de D. immitis é uma ameaça à saúde de canídeos domésticos no complexo da Ilha de Algodoal-Maiandeua, bem como para felinos e seres humanos.


#8 - The urban and rural capybaras (Hydrochoerus hydrochaeris) as reservoir of Salmonella in the western Amazon, Brazil

Abstract in English:

The capybara (Hydrochoerus hydrochaeris) is the largest rodent in the world. In the state of Acre, Brazil, populations of capybaras have been increasing significantly. The role of capybaras in the transmission of certain bacterial zoonotic infections is not well understood, including bacteria of the genus Salmonella. Salmonella spp. generally cause enteritis or septicemia in mammals, however many mammalian species can carry the bacteria asymptomatically and shed it in their feces. To better understand the possible role of capybaras as reservoirs of Salmonella spp., we conducted a study of Salmonella within fecal samples from capybara in Acre. In a convenience sample, 54 capybaras from two urban and two rural areas of Acre were captured and kept for three to four days for sampling. None of the animals were symptomatic of any intestinal illness. Three separate fecal samples were collected from each animal, during their stays in captivity. Each sample was cultured for the presence of Salmonella spp. at the bacteriology laboratory of the Veterinary College of the Federal University of Acre. Samples were seeded in tetrationate pre‑enrichment broth and in pre‑enrichment broth peptone. After a 24 hour of incubation all samples were streaked on MacConkey Agar (MC) and Salmonella‑Shigella Agar (SS). Suggestive colonies were submitted to biochemical analysis. Salmonella compatible colonies according to biochemical profile were submitted to serotyping (Sorokit for Salmonella - Probac do Brasil). In addition, the first sample from each of the 54 capybara was tested for Salmonella spp. using PCR targeting gene hilA. Eight (5%) of the 162 samples examined by bacterial culture were positive for Salmonella spp., while four (7%) of the 54 examined by PCR were positive. From the eight positive animals on culture, five were from urban area and three from rural area. On PCR, only one positive animal was from urban area and four were from rural area. Overall, by either test, one of the 54 animals was positive. All samples were collected in free - living animals with no apparent clinical signs of salmonellosis, indicating the potential of capybara as reservoir on this ecosystem.

A capivara (Hydrochoerus hydrochaeris) é o maior roedor do mundo. No estado do Acre, Brasil, as populações de capivaras têm aumentado significativamente. O papel das capivaras na transmissão de certas infecções zoonóticas bacterianas não é bem compreendido, incluindo as bactérias do gênero Salmonella. Salmonella spp. geralmente causam enterite ou septicemia em mamíferos, porém muitas espécies de mamíferos podem carregar a bactéria de forma assintomática e eliminá-la em suas fezes. Para entender melhor o possível papel das capivaras como reservatórios de Salmonella spp., realizamos um estudo para identificação de Salmonella spp. em amostras fecais de capivaras no Acre. Em uma amostra de conveniência, 54 capivaras de duas áreas urbanas e duas áreas rurais do Acre foram capturadas e mantidas por três a quatro dias para amostragem. Nenhum dos animais era sintomático de qualquer doença intestinal. Três amostras fecais foram coletadas de cada animal, durante sua permanência em cativeiro. Cada amostra foi cultivada para a presença de Salmonella spp. no Laboratório de Bacteriologia Veterinária da Universidade Federal do Acre. As amostras foram semeadas em caldo de pré-enriquecimento tetrationato e em peptona de caldo de pré-enriquecimento. Após 24 horas de incubação, todas as amostras foram semeadas em ágar MacConkey (MC) e ágar Salmonella-Shigella (SS). Colônias sugestivas foram submetidas a análises bioquímicas. Colônias compatíveis com Salmonella de acordo com o perfil bioquímico foram submetidas à sorotipagem (Sorokit para Salmonella - Probac do Brasil). Além disso, a primeira amostra de cada uma das 54 capivaras foi testada para Salmonella spp. usando PCR, visando gene hilA. Oito (5%) das 162 amostras examinadas por cultura bacteriana foram positivas para Salmonella spp. Enquanto quatro (7%) das 54 examinadas pela PCR foram positivas. Dos oito animais positivos em cultura, cinco eram de área urbana e três de área rural. Na PCR, apenas um animal positivo era de área urbana e quatro de área rural. Considerando o diagnóstico conjunto por ambos os testes, PCR e cultura, um animal foi considerado positivo. Todas as amostras foram coletadas em animais livres, sem sinais clínicos aparentes de salmonelose, indicando o potencial da capivara como reservatório nesse ecossistema.


#9 - Risk factors associated to the prevalence of anti-Sarcocystis neurona, Neospora spp. and Toxoplasma gondii antibodies in horses from Roraima, Brazilian Amazon

Abstract in English:

Samples of 303 horses from 56 ranches of Rorainópolis municipality, state of Roraima, were evaluated by means of the Indirect Immunofluorescence Test (IFAT) to detect antibodies against Sarcocystis spp., Toxoplasma gondii and Neospora spp. A subset of positive sample (n=15) against Sarcocystis spp. was evaluated by Western Blotting (WB) with crude antigen of S. neurona. From the serological result, possible risk factors were evaluated against individual or farming variables. The prevalence of anti-Sarcocystis spp. antibodies was estimated to be 43.2% (37.6-49.0%), anti-Neospora sp. was 26.7% (21.9-32.2%), and anti-T. gondii was 18.5% (14.3-23.4%). Fourteen samples (14/15) evaluated by WB were positive for S. neurona antigens. From the ranches, 76.8% (43/56) presented at least one positive horse for Sarcocystis spp., 69.6% (39/56) for Neospora spp., and 55.4% (31/56) for T. gondii. Thirteen (14.3%) horses had antibodies against all agents, 50 (16.5%) had antibodies against Sarcocystis spp. and Neospora spp., 10 (3.3%) for Neospora spp. and T. gondii, and eight (2.6%) for Sarcocystis spp. and T. gondii. Associated variables (P≤0.05) for antibodies against Neospora spp. were not found in horses fed on rented pastures, not in horses without veterinary assistance and stables, and not in herds up to 5 horses; while they were associated for T. gondii by contact with cats, in the Lavradeiro breed with use of stables, in horses raise with cattle, and in herds up to 5 horses. There were no variables associated with the presence of antibodies against S. neurona. Antibodies against S. neurona, Neospora spp. and T. gondii were reported in horses from the state of Roraima, Northern Brazilian Amazon, highlighting to the elevate prevalence on ranches.

Amostras de sangue de 303 equinos provenientes de 56 propriedades do município de Rorainópolis, Roraima, foram avaliadas por Reação de Imunofluorescência Indireta (RIF) para pesquisa de anticorpos contra Sarcocystis neurona, Toxoplasma gondii e Neospora spp. Algumas amostras de soros positivos para Sarcocystis spp. foram avaliadas pelo Western Blotting (WB) utilizando antígenos crus de S. neurona. A partir dos resultados sorológicos, possíveis fatores de risco foram avaliados frente a variáveis individuais e de propriedade. A prevalência de anticorpos anti-Sarcocystis spp. foi estimada em 43,2% (37,6-49,0%), anti-Neospora sp. em 26,7% (21,9-32,2%) e anti-T. gondii de 18,5% (14,3-23,4%). Quatorze amostras (14/15) testadas por WB resultaram positivas para antígenos de S. neurona. Das propriedades, 76,8% (43/56) apresentaram pelo menos um equino positivo para Sarcocystis spp.; 69,6% (39/56) para Neospora spp. e 55,4% (31/56) para T. gondii. Dos equinos, 13 (4,3%) apresentarem anticorpos para os três agentes, 50 (16,5%) para Sarcocystis spp. e Neospora spp., 10 (3,3%) para Neospora spp. e T. gondii, e oito (2,6%) para Sarcocystis spp. e T. gondii. As variáveis associadas (P≤0,05) à presença de anticorpos foram: para Neospora spp. não pastejar em áreas alugadas, ausência de assistência veterinária na propriedade, sexo masculino, não estabular animais e plantel equino acima de 5 animais; enquanto para T. gondii foram o contato com felinos, animais da raça lavradeiro, animal estabulado, criação de bovinos na propriedade e plantel equino acima de 5 animais. Não houveram variáveis associadas a presença de anticorpos contra S. neurona. Relata-se no presente estudo a primeira detecção de anticorpos anti-S. neurona, Neospora spp. e T. gondii em equinos do estado de Roraima, localizado na Amazônia Setentrional Brasileira, ressaltando para a elevada frequência de fazendas com equinos soropositivos.


#10 - Histopathological evaluation of formalin toxicity in Arapaima gigas (Arapaimidae), the giant fish from Amazon

Abstract in English:

This study aimed to determine the lethal concentration and the structural and ultra-structural effects caused by the formalin exposure on juveniles of Arapaima gigas. Ninety fish (60.1± 2.5g and 20.2±0.9cm) were exposed to 0, 22, 44, 66, 88 and 110mg L-1 in order to determine the lethal concentration (LC50-96h) that was 36.4mg L-1 of formalin. Sublethal effects were evaluated using histopathological analysis on the gills and assessment of behavioral alterations and clinical signs. The LC50 of formalin for 24, 48 and 72h was 88.3, 64.7 and 56.8mg L-1 respectively. Clinical signs and behavioral changes were found: erratic swimming, lethargy, crowding on the water surface, loss of hydrodynamic equilibrium, spasms and agonistic confrontation, which were observed only at 88 and 110mg L-1. The histological alteration index (HAI) showed that 66, 88 and 100mg L-1 presented significant difference (p<0.05) in relation to unexposed fish, indicating that moderate damage to the gills of fish exposed to formalin had occurred. The mean values of alteration (MVA) for 22, 44, 66, 88 and 110mg L-1 were 1.14, 1.29, 1.51, 1.53 and 1.60 respectively, and differences in this index were only observed with 110 mgL-1 of formalin. It is therefore possible to conclude that sublethal concentrations of formalin (22.0mg L-1) did not compromise the health of juveniles of A. gigas. Finally, concentrations greater than to LC50-96h may be carefully used for short-term exposure, since the MVA for all concentrations tested only indicated localized lesions that did not compromise gills functionality of exposed fish.

O presente estudo teve como objetivo determinar a concentração letal e efeitos estruturais e ultraestruturais causados ​​pela exposição a formalina em juvenis de Arapaima gigas. Noventa peixes (60,1±2,5g e 20,2±0,9cm) foram expostos a 0, 22, 44, 66, 88 e 110mg L-1, para determinar a concentração letal (CL50-96h) de formalina que foi 36,4mg L-1. Os efeitos subletais foram avaliados por análises histopatológicas das brânquias e avaliação das alterações comportamentais e sinais clínicos. A CL50 de formalina para 24, 48 e 72horas foi de 88,3, 64,7 e 56,8; respectivamente. Sinais clínicos e alterações comportamentais encontradas foram: natação errática, letargia, aglomeração de peixes na superfície da água, perda de equilíbrio hidrodinâmico, espasmos e confronto agonísticos, observados apenas nas concentrações de 88 e 110mg L-1. O índice de alteração histológica (IAH) mostrou que as concentrações de 66, 88 e 100mg L-1 apresentaram diferenças significativas (p<0,05) em relação aos controles, indicando a ocorrência de danos moderados nas brânquias dos peixes expostos a formalina. Os valores médios de alteração (VMA) para as concentrações 22, 44, 66, 88 e 110mg L-1 foram 1,14, 1,29, 1,51, 1,53 e 1,60; respectivamente, e as diferenças na composição desse índice foram observados apenas na exposição com 110mg L-1 de formalina. Foi possível concluir que concentrações subletais de formalina (22,0mg L-1) não comprometem a saúde de juvenis de A. gigas. Concentrações de formalina acima da CL50-96h podem ser usadas cuidadosamente para banho de curto tempo, uma vez que o VMA para todas as concentrações testadas indicou apenas lesões localizadas que não comprometem a funcionalidade das brânquias dos peixes expostos.
