E-mail: karla.ufal@gmail.com The objective of this study was to produce and partially purify Malleo-protein from Burkholderia mallei samples isolates from Equidae in Brazil with potential for use in the diagnosis of glanders. The strain B. mallei phenotypically characterized and proven virulent was inoculated into broth Dorset-Henley to grow and metabolize. The proteins were separated by trichloroacetic acid precipitation and amonium sulfate precipitation. The PPD mallein was concentrated 1.0mg/mL and biologically tested in guinea pigs. It was effective in the development of delayed-type hypersensitivity and consequently to identify true-positive animals and to exclude of true negatives. There is the possibility for potential use in the glanders diagnosis in Equidae.

E-mail: karla.ufal@gmail.com Objetivou-se com este estudo produzir e purificar parcialmente a PPD-maleína a partir de amostras de Burkholderia mallei isoladas de equídeos no Brasil com potencial para uso no diagnóstico do mormo. As linhagens de B. mallei fenotipicamente caracterizadas e de virulência comprovada foram inoculadas em caldo Dorset-Henley para crescer e metabolizar. Em seguida, as proteínas foram separadas por precipitação com ácido tricloroacético e precipitadas com sulfato de amônia. As PPDs-maleínas foram concentradas em 1,0mg/mL e na avaliação realizada em cobaios foi eficaz no desenvolvimento da hipersensibilidade do tipo tardia e consequentemente na identificação de animais verdadeiro positivos e exclusão dos verdadeiro negativos, sendo uma possibilidade em potencial para utilização no diagnóstico do mormo.

E-mail: tcftavares@yahoo.com.br The aim was to determine whether hens immunized with an inactivated suspension of Leptospira and a solution of outer membrane proteins extracted from the serovar Hardjo, could produce specific polyclonal antibodies to Leptospira, detected in ELISA assay. Eight hens White Leghorn race with 25-weeks-old were immunized, three with an inactivated suspension of Leptospira, three with a solution of outer membrane proteins (OMP) extracted from the serovar Hardjo and two controls immunized with saline. Blood samples were collected fortnightly and eggs daily. The IgY was purified from the egg yolk using the method for the delipidation of dilution with water acidic and ammonium sulfate precipitation. The ELISA assay was performed to verify the specificity of the IgY, these was possible to observe the production of specific antibody to Leptospira both in serum and purified egg yolk. The specific antibody titers peaked in the fifth week post immunization. The production of polyclonal IgY was effective for producing high titers of specific antibodies.

Foram imunizados oito galinhas com 25 semanas de idade, da raça White Leghorn, sendo três imunizadas com uma suspensão de leptospiras inativadas, três com uma solução de proteínas de membrana externa extraída do sorovar Hardjo e duas controle. Coletas de sangue foram realizadas quinzenalmente e de ovos diariamente. A IgY foi purificada a partir da gema dos ovos utilizando para a delipidação o método de diluição em água ácida e a precipitação com sulfato de amônio. Nos testes ELISA realizados para verificar a especificidade da IgY, foi demonstrada a produção de anticorpos anti-Leptospira, tanto no soro quanto nas gemas purificadas. O pico de produção de anticorpos específicos ocorreu na 5º semana após a primeira imunização. Ficou demonstrada a possibilidade da indução da produção de anticorpos específicos em galinhas imunizadas com leptospiras do sorovar Hardjo inativadas, bem como, com proteínas de membrana externa (PME) extraidas desse sorovar. As galinhas imunizadas com uma suspensão de leptospiras inativadas ou com PME de Leptospira interrogans do sorovar Hardjo produziram anticorpos reativos a PME Hardjo detectáves por teste ELISA.

It has key functions on energetic metabolism, and cell death and survival. VDAC characterization is important for understanding mitochondrial interactions with cytosolic proteins, such as hexokinase (HK). HK-VDAC interaction supports preferential access to intramitochondrial ATP in neural cells. Brain HK interacts in different ways with VDAC. It results in two HK binding sites (A and B). VDAC isoforms differential metabolic roles may be explained by the presence of post-translational modifications. In this study we purified avian neuronal mitochondrial VDAC1. At same time we showed that VDACs 1 and 2 pI heterogeneity in rat and avian brains is due to phosphorylation. Purified VDAC had a molecular weight of 30 KDa. The purified VDAC submitted to phosphorylated protein staining on gel, was dephosphorylated. The knowledge of presence or absence of VDAC phosphorylation is important for understanding the molecular nature basis of A and B HK binding sites in brain mitochondria.

E-mail: mccesar@usp.br A VDAC é uma porina presente na MME cuja função é crucial no metabolismo energético, sobrevivência e morte celular. A caracterização da VDAC torna-se importante para a compreensão das inter-relações da mitocôndria com os diferentes componentes citosólicos, tais como a HK. A ligação HK-VDAC favorece a utilização do ATP intramitocondrial em células neuronais, a HK cerebral pode interagir de formas diferentes com a VDAC, o que resulta em diferentes sítios de ligação (sítios A e B). Os variados papéis metabólicos das isoformas da VDAC podem ser explicados pela presença de alterações pós-traducionais. No presente trabalho purificamos a VDAC1 mitocondrial neuronal proveniente de cérebro aviar. Paralelamente, comprovamos que a presença de múltiplas formas das VDACs 1 e 2 em cérebros murino e aviar, seja devida à presença de modificações pós-traducionais, nomeadamente a fosforilação. A proteína isolada apresentou peso molecular de 30KDa. Quando submetida à eletroforese e posteriormente à coloração para a identificação de fosfoproteínas, a mesma mostrou-se desfosforilada. O conhecimento da presença, ou ausência de fosforilação das VDACs, reside na importância de estabelecer-se as bases moleculares ligadas à existência de sítios A e B nas mitocôndrias neuronais.

[Comparison between the clinical and pathological picture in the experimental poisoning by crotalic venom.] Quadro clínico-patológico do envenenamento crotálico experimental em bubalinos comparado com o de bovinos. E-mail: diomedes@ufpa.br The objective of the study was to verify the sensibility of buffaloes to the poison of Crotalus durissus terrificus and to study the clinical-pathological picture in buffaloes in comparison with the one in cattle. The subcutaneous inoculation of the liofilized poison of the snake, diluted in 1ml of physiologic solution, was done in the area of the humerus-radio-ulnar joint of three buffaloes at doses of 0.015, 0.03 and 0.066mg/kg, and of two cattle at doses of 0.03 and 0.066mg/kg. The buffalo that received the 0.03mg/kg dose presented severe clinical signs but recovered six days later. The bovine that received the same dose, died after a clinical course of 22h56min. The 0.066mg/kg dose caused death of the bovine as also the buffalo, with a clinical course of 4h23min and 8h12min, respectively. The buffalo that received the 0.015mg/kg dose recovered, after a course of 48 hours. The buffalo that died, showed clinical signs from 3h58min on, and the buffaloes that showed symptoms from 17h25min and 24h00min after inoculation of the venom, but recovered. In the two cattle that died (with doses of 0.03 and 0.066mg/kg), the first clinical signs occurred 6h10min and 6h31min after the inoculation of the venom. The inoculation produced in the buffaloes and cattle nervous symptoms of flaccid paralysis. The main signs observed in the buffaloes as well as the cattle, were slight volume increase at the site of inoculation, respiration difficulties characterized by predominantly abdominal breathing, apathy, sialorreia, difficulty to get up when stimulated, evolution to sternal decubitus followed by lateral decubitus with peddling movements of the legs, and decrease of the reflexes related to the cranial nerves. The buffaloes showed also augmentation of the sustentation base, dragging of the hooves of the hind legs, slow and staggering gait, difficulty in apprehension of the food, The cattle showed additionally paralysis of the eyeballs, revealed through non-exhibition of the sclera during head rotation in latero-caudal direction. Laboratory exams revealed in the cattle and the buffaloes leucocytosis by neutrofilia, and in the biochemistry series, increase in the levels of alanine aminotransferase, aspartato aminotransferase, creatinaquinase and lactic dehydrogenase. There was no alteration in the urinalysis nor in the activation time of protrombine and in the time of partially activated tromboplastin. Necropsy only evidenced slight edema corresponding to the inoculation site in one bovine. Histopathological examination revealed picnosis of the epitelial cell nuclei of some kidney tubules in the cortex (in the buffalo and in one bovine) and slight vacuolation of hepatocites (in one bovine).

E-mail: diomedes@ufpa.br O estudo teve por objetivo verificar a sensibilidade dos bubalinos à peçonha de Crotalus durissus terrificus, estudar o quadro clínico-patológico e laboratorial nessa espécie e estabelecer comparações com o verificado em bovinos. A inoculação do veneno liofilizado de Crotalus durissus terrificus, diluído em 1ml de solução fisiológica, foi feita na região da articulação úmero-rádio-ulnar, por via subcutânea, em três bubalinos (doses de 0,015; 0,03; e 0,066mg/kg) e em dois bovinos (doses de 0,03 e 0,066mg/kg). O bubalino que recebeu a dose de 0,03mg/kg apresentou sinais clínicos graves recuperou-se seis dias após, e o bovino que recebeu a mesma dose morreu com evolução de 22h56min. A dose de 0,066mg/kg causou a morte tanto do bovino quanto do bubalino, com evolução clínica de 4h23min e 8h12min, respectivamente. O bubalino que recebeu a dose de 0,015mg/kg, recuperou-se com evolução de 48 horas. Os sinais clínicos tiveram início dentro de 3h58min no bubalino que morreu, e nos bubalinos que adoeceram mas se recuperaram, dentro de 17h25min e 24h00min após a inoculação do veneno. Nos dois bovinos que morreram (com doses de 0,03 e 0,066mg/kg), os primeiros sinais clínicos foram observados 6h10min e 6h31min após a inoculação do veneno. A inoculação do veneno produziu nos búfalos e bovinos um quadro nervoso de paralisia flácida. Os principais sinais observados tanto nos búfalos quanto nos bovinos, foram discreto aumento de volume no local da inoculação, dificuldade respiratória caracterizada por respiração predominantemente abdominal, apatia, sialorreia, dificuldade para se levantar quando estimulados, evolução para decúbito esternal permanente, seguido de decúbito lateral e movimentos de pedalagem, e diminuição dos reflexos relacionados aos pares de nervos cranianos. Nos bubalinos adicionalmente foi observado aumento da base de sustentação, arrastar das pinças dos membros posteriores, marcha lenta e cambaleante, dificuldade na apreensão dos alimentos; nos bovinos ainda foram observados paralisia do globo ocular, revelada através da não exposição da esclera durante a rotação da cabeça na direção latero-caudal. Tanto nos bovinos quanto nos bubalinos, verificou-se no leucograma, leucocitose por neutrofilia, e na bioquímica sérica, aumento nos níveis de alanina aminotransferase, aspartato aminotransferase, creatinaquinase e dehidrogenase láctica. Não houve alterações na urinálise, nem no tempo de ativação da protrombina e nem no tempo de tromboplastina parcial ativada. À necropsia evidenciou-se apenas discreto edema correspondente ao local da inoculação em um bovino. Os achados histopatológicos observados foram picnose nos núcleos de células epiteliais de alguns túbulos uriníferos no córtex renal (em um búfalo e um bovino) e fígado com leve vacuolização de hepatócitos (em um bovino).


#25 - Caseous lymphadenitis in slaughtered hair sheep in northeastern Brazil, 31(3):224-230

[Caseous lymphadenitis in slaughtered hair sheep in northeastern Brazil.] Linfadenite caseosa em ovinos deslanados abatidos em um frigorífico da Paraíba. E-mail: franklin.riet@pq.cnpq.br The objective of this paper is to report the prevalence and distribution of lesions of caseous lymphadenitis (CL) in hair sheep slaughtered in an abattoir in the state of Paraíba, northeastern Brazil. From 1,466 sheep slaughtered, 236 (15.9%) had gross lesions similar to that of CL. The prevalence was higher in females, 17.9% (135/754), than in males, 13.8% (101/732). Ninety seven (36.3%) out of 268 lesions were localized in the prescapular lymph node, 22.4% (60/268) in the parotideal, and 20.9% (56/268) in the prefemoral. Only 70 (26.1%) out of 268 lesions suggestive of CL were detected during ante mortem examination. Caseous lymphadenitis-like lesions were observed in all flocks examined from different municipalities in the states of Paraíba, Pernambuco and Bahia. Cultures and histologic examination were performed in samples from 51 lesions. Corynebacterium pseudotuberculosis was isolated in 74.5% (43/51) samples, Staphylococcus aureus in 7.8% (4/51) samples, Escherichia coli in 2% (1/51), and in 5.9% (3/51) samples cultures were negative. Histologic findings in 11 lymph nodes showed all characteristic lesions of caseous lymphadenitis: central area of necrosis formed by concentric lamellae, with large bacterial colonies and mineralization, surrounded by an inflammatory exudate with epithelioid macrophages and few neutrophils; lymphocytes and plasma cells were observed in the next layer, and fibrous tissue was surrounding the whole lesion. Lesions in the other 32 lymph nodes showed most, but not all lesions characteristic of the disease. Giant cells were observed in 21 lymph nodes. Histologic lesions in lymph nodes from which S. aureus was isolated were similar as those of CL. These findings suggest that CL lesions are characteristic, but not pathognogmonic, because they can be confused with lesions caused by other pyogenic bacteria or tuberculosis; therefore bacteriologic examination is necessary for the diagnosis of the disease.

E-mail: franklin.riet@pq.cnpq.br Este trabalho teve como objetivo determinar a prevalência e distribuição de lesões da linfadenite caseosa em ovinos deslanados abatidos no município de Mulungu, Paraíba. Dos 1466 ovinos abatidos, 236 (15,9%) apresentaram lesões macroscópicas semelhantes à linfadenite caseosa. A prevalência foi maior em fêmeas, 17.9% (135/754) que em machos, 13,8% (101/732). As principais lesões estavam localizadas nos linfonodos pré-escapulares em 36,3% (97/268) dos ovinos, nos parotídeos em 22,4% (60/268) e no pré-crural em 20,9% (56/268). Somente 70 (26,1%) de um total de 268 lesões foram detectadas no exame ante mortem. Foram encontradas lesões em todos os lotes provenientes de diversos municípios da Paraíba, Pernambuco e Bahia. Das 51 amostras em que foi feito o cultivo bacteriológico, em 74,5% (43/51) foi isolado Corynebacterium pseudotuberculosis. Em 7,8% (4/51) foi isolado Staphylococcus aureus, em 2% (1/51) Escherichia coli e 5,9% (3/51) amostras foram negativas. No exame histológico, em 11 linfonodos havia lesões da linfadenite caseosa caracterizadas por área de necrose central formada por lamelas concêntricas, com presença de grandes colônias bacterianas e focos de mineralização, rodeada por uma faixa de infiltrado inflamatório com macrófagos epitelióides e poucos neutrófilos. Na camada adjacente observaram-se linfócitos e plasmócitos e toda a lesão era delimitada por tecido conjuntivo fibroso. Nas outras 32 amostras foram observadas a maioria, mas não todas as lesões características da doença. Em 21 amostras foram encontradas células gigantes. As lesões histológicas dos linfonodos que foi isolado S. aureus foram semelhantes as da linfadenite caseosa. Conclui-se que as lesões histológicas de linfadenite caseosa são características, mas não patognomônicas, pois podem ser confundidas com lesões causadas por outros organismos piógenos e com tuberculose; portanto o isolamento bacteriológico é imprescindível para o diagnóstico definitivo da linfadenite caseosa.

[Comparação da microbiota de cascavéis (Crotalus durissus terrificus) de vida-livre e cativeiro.]Pesquisa Veterinária Brasileira 29(12):999-1003. E-mail: rseabra@cevap.org.br The study evaluated and compared the aerobic microbiota from the oral cavity, cloaca and venom of Crotalus durissus terrificus snakes, recently caught from the wild and kept under quarantine (WQ), individual captivity (IC) and collective captivity (CC). Antimicrobial drug effectiveness on isolated agents also was assayed. From group I, II and III were isolated, respectively, 29 (63.04%), 38 (90.48%) and 21 (42.86%) microorganisms from the cloaca; 15 (32.61%), 3 (7.14%) and 25 (51.02%) microorganisms from the oral cavity; and, 2 (4.35%), 1 (2.38%) and 3 (6.12%) microorganisms from venom. The most frequent bacteria were Pseudomonas aeruginosa, Proteus vulgaris and Morganella morganii, with sensitivity to amikacin, gentamicin, norfloxacin, sulfazotrin and tobramycin. Snakes kept in semi-open captivity exhibited the fewest microorganisms in oral cavities, perhaps due to the environment in captivity, with different temperature gradients, running water, absence of daily handling, circulating air, possibility of moving around, daily cleaning, and sunlight access.

E-mail: rseabra@cevap.org.br Este estudo avaliou e comparou a microflora aeróbica da cavidade oral, cloaca e veneno de serpentes Crotalus durissus terrificus recém-capturadas da natureza e mantidas sob quarentena (WQ), mantidas em cativeiro coletivo (CC) e em cativeiro individual (IC). A eficácia de drogas antimicrobianas de agentes isolados foi também avaliada. Foram isolados microorganismos dos grupos I, II e III respectivamente: 29 (63.04%), 38 (90.48%) e 21 (42.86%) da cloaca; 15 (32.61%), 3 (7.14%) e 25 (51.02%) da cavidade oral, e finalmente 2 (4.35%), 1 (2.38%) e 3 (6.12%) do veneno. As bactérias mais frequentes foram Pseudomonas aeruginosa, Proteus vulgaris e Morganella morganii, com sensibilidade para amikacina, gentamicina, norfloxacina, sulfazotrina e tobramicina. Serpentes mantidas no cativeiro semi-aberto mostraram menor número de agentes infecciosos em cavidade oral, talvez devido ao ambiente de cativeiro com diferentes gradientes de temperatura, água corrente, ausência de manejo diário, ampla circulação de ar, possibilidade de movimentação pelos animais, limpeza diária e acesso ao Sol.

Knowledge of the physiology of these animals and serum proteins concentration changes are important for early identification of major diseases which lead to states of hypoproteinemia and hyperproteinemia. The objective was to determine the concentration of total protein and serum protein electrophoresis profile of Crotalus durissus terrificus (rattlesnake) in captivity. Blood samples were taken from the ventral coccygeal vein of 21 adult and healthy snakes divided into groups: Group 1 with 12 males, weighing in average 588.89±193.55g, and Group 2 with nine females, weighing in average 708.33±194.04g. The total serum concentration of protein was determined by the method of refractometry and agarose gel electrophoresis. The total protein values in the serum for females was 4.82±0.72, for males 4.51±0.50 and males and females 4.64±0.61, identified by four fractions (g/dL): albumin, a, b and g-globulin. Additionally the albumin/globulin ratio was calculated. The female snakes showed higher values for the variables, albumin and the albumin/globulin (AG) differed significantly (P<0.05) from the group of male snakes, but there was no clinical significance.

The highest antibody titers predicted Australis as the infecting serogroup due to reactions against a reference strain of serovar Bratislava and a canine local isolate of serovar Australis, characterized as Leptospira noguchii. The data presented in this report highlight that a considerable fraction of capybaras in captivity may behave as reservoir for pathogenic leptospires emphasizing the occupational risk of those who deal with animal farming and slaughter.

[Clinical and pathological aspects of experimental Crotalus poisoning in cattle.] Aspectos clinicos e patológicos do envenenamento crotálico experimental em bovinos. Pesquisa Veterinária Brasileira 28(6):261-270. Projeto Sanidade Animal Embrapa/UFRRJ, Seropédica, RJ 23890-000, Brazil. E-mail: flaviograca@uol.com.br Crotalus poisoning was experimentally reproduced by subcutaneous inoculation of Crotalus durissus terrificus (South American rattlesnake) venom into 10 clinically healthy mixed bred 12 to 36-month-old cattle, weighing 125 to 449 kg. Two animals were used as controls. The animal that received a dose of 0.03mg/kg body weight died 7h40min after inoculation. A 0.015mg/kg dose provoked death in 4 out of 7 young oxen. Two animals given 0.0075mg/kg became slightly sick and recovered. Onset of symptoms occurred from 1h30min to 13h45min after inoculation. The clinical course varied from 5h25min to 45h for animals that died, and from 33h15min to 17 days for animals that recovered. The main nervous signs observed were diminished response to external stimuli, hypotonic reflexes, dragging of the hooves, apathy, difficulties in moving around obstacles, ocular globe paralysis, lateral and sternal decubitus, and tongue paralysis. Adipsia and sometimes petechiae in the conjunctival and vaginal mucosa were observed. A slight to moderate increase in bleeding time was noted in 6 animals, and a moderate increase in partial thromboplastin time was found in 7 others. Moderate leukocytosis with neutrophilia, relative lymphopenia, eosinopenia, and monocytosis was found. There was a significant increase in creatine kinase serum levels of a ten-fold order. No significant alterations were revealed by urinalysis. Necropsy revealed minimal edema at the inoculation site, few petechiae and equimoses in the epicardium, omentum, biliary vesicle and bladder mucosa of some animals. Histopathological examination revealed necrosis (hyalinization) of groups or isolated myocytes in different muscles examined, both near and far from the inoculation site, in all animals. The diagnosis of Crotalus poisoning and its differentiation from diseases causing paralysis and muscular necrosis in cattle in Brazil are discussed.

The prevalence of Salmonella carrier animals was 55.66%, being 17.6% of the animals Salmonella positive in lymph nodes, 18.3% in feces and 19.6% in both materials. Twenty-six different serovars were identified among 226 Salmonella isolates. The most prevalent serovars were: Typhimurium (24.3%), Agona (19.9%), Derby (13.2%) e Bredeney (12%). These results point out the need of control programs to reduce the prevalence of carrier pigs at slaughter.
