[Experimental poisoning by Bothropoides jararaca and Bothrops jararacussu in sheep: Clinic-pathological and laboratory aspects.] Projeto Sanidade Animal Embrapa/UFRRJ, Seropédica, RJ 23890-000, Brazil. E-mail: paulaaragao.vet@gmail.com The purpose of this study was to establish the clinic-pathological and laboratory changes in sheep inoculated with Bothropoides jararaca and Bothrops jararacussu venom to provide subsidies for the differential diagnosis of snake bites. The liofilized venoms were diluted in 1 ml saline and administrated subcutaneously to four sheep. Three of the animals died, and the one that received 0.5mg/kg (B. jararaca venom) recovered. First symptoms were observed from 7 minutes to 1 hour after inoculation, and the clinical course varied from 7 hours and 9 minutes to 21 hours and 59 minutes. The symptoms, independent of the dosage, were swelling of the inoculation site, increased bleeding time and capillary filling, tachycardia, dyspnea, pale mucous membranes and diminished reaction to external stimuli. Laboratory tests revealed pronounced normocytic and normochromic anemia, trombocytopenia, slight reduction of fibrogen and total plasmatic protein, in two animals diminished hematocrit, besides pronounced increase of creatinaquinase and lactic dehydrogenase. At necropsy, the main findings at the inoculation site and adjacent tissues were extensive hemorrhages in the sheep inoculated with jararaca venom, and predominantly edema in the two animals inoculated with jararacussu venom. In two sheep which received jararacussu venom, acute pulmonary edema was observed. Hemorrhage and edema as the main histopathological changes, besides necrosis of muscle fibers and vessels at the inoculation site and adjacent tissue was observed. The renal tubular necrosis was attributed to shock. The volume increase at the inoculation site and surroundings was mainly due to hemorrhage (B. jararaca) or edema (B. jararacussu).

Pesquisa Veterinária Brasileira 30(9):717-728. Projeto Sanidade Animal Embrapa/UFRRJ, Seropédica, RJ 23890-000, Brazil.

#22 - Experimental poisoning by sodium monofluoroacetate in cattle: Clinical and pathological aspects, 30(7):533-540

Abstract in English:

ABSTRACT.- Nogueira V.A., França T.N., Peixoto T. C., Caldas S.A., Armién A.G. & Peixoto P.V. 2010. [Experimental poisoning by sodium monofluoroacetate in cattle: Clinical and pathological aspects.] Projeto Sanidade Animal Embrapa/UFRRJ, Seropédica, RJ 23890-000, Brazil. E-mail: vivianmedvet@yahoo.com.br Sodium monofluoroacetate (MF) was identified, by cromatography, in three of the 12 plants that cause sudden death in cattle in Brazil, including Palicourea marcgravii, the most important plant of this group. A special kind of hydropic-vacuolar degeneration (HVD) of the distal convoluted uriniferous tubules was considered typical for the poisoning by some authors. The objective of this study was to verify if the ingestion of MF causes similar clinical signs and lesion in cattle poisoned by plants that cause sudden death. Six cows received orally 0.5 and 1.0mg/kg of M diluted in 50mL of distilled water. Clinically the animals presented tachycardia, engorgement and positive pulse of the jugular vein, abdominal breathing, swaying gait, subtle unbalance, and recumbency with head placed the on the flank. In the agonal phase all the animals in lateral recumbence presented muscular fasciculation, tonic contraction, peddling movements, opisthotonus, nystagmus, and died between 2 and 14 minutes. At postmortem examination, the heart auricles, jugular and pulmonary veins were moderately distended and engorged with blood. Slight to moderate edema of the subserosa was seen around the gall bladder and the duodenum near to the pancreas. At light microscopy, HVD associated with nuclear picnosis of the epithelial cells of the distal convoluted uriniferous tubules was present in all animals. Ultrastructurally, the cell lesion represents formation of cytosolic vacuoles, likely due to accumulation of water. Coagulation necrosis of individual or groups of hepatocytes and slight hepatic congestion secondary to the venous stasis were also observed. Hydropic-vacuolar degeneration has been observed in cases of poisoning by many substances, which cause acute tubular nephrosis, however not restricted to the distal renal tubules and without nuclear picnosis. This study demonstrates that the HVD in the kidney can be caused by MF and, in analogy, the compound should be considered responsible for the death of cattle that ingest toxic plants which cause sudden death in Brazil. Our results can be supportive to studies that will focus on the degradation of MF by rumen bacteria, what might have economic implications, as at least 500.000 cattle die annually by sudden death causing toxic plants in Brazil.

#23 - Evaluation of renal biopsy techniques in sheep, 30(3):260-266

E-mail: nespoli@ufmt.br Due to lack of studies about renal biopsies in sheep, a comparative study was performed for three renal biopsy techniques in this species. In this study, nine crossbred (Santa Inês) ewe lamb (26.64 kg ±4,86) were used in serial procedures with one week consecutive intervals. The clinical aspects, clinical pathological findings, renal sample weights, histology quality, number of glomeruli, and the presence of artifacts in the histology slices were evaluated using the techniques of percutaneous blind biopsy, ultrasound guided biopsies and of videolaparoscopic procedure. No relevant hematological or biochemical alterations were observed in the animals subjected to renal biopsies and the clinical manifestations detected were slight and transitory, except for one case of urethral obstruction by blood clot. The blind technique was related to more frequent and severe cases of hematuria, with more severe damage to the renal tissue and to the only case of obstruction of the urinary flow. In the videolaparocopic technique, the average weight of the samples was superior and hematuria was slight and transitory. A direct relation was seen between occurrence of severe hematuria and presence of transitional epithelium in the samples and the number of trials used for obtainment of fragments.

The classical alterations of minera-lization and, occasionally, ossification of the cardiovascular system, as well the lesions of kidneys, lungs, stomach, among other organs, were reproduced by the subcutaneous administration of an oily solution of cholecalciferol (non-activated vitamin D3).

#25 - Evaluation of hepatic biopsy techniques in sheep, 30(1):29-36

Abstract in English:

ABSTRACT.- Néspoli P.B., Gheller V.A., Peixoto P.V., França T.N., Carvalho A.U., Godoy de Araújo D.K. & Malm C. 2010. [Evaluation of hepatic biopsy techniques in sheep.] E-mail: nespoli@ufmt.br Due to lack of information about the subject and lack of studies that confront the techniques of liver biopsy in sheep, a comparative study between eight biopsy techniques in this species was developed. In this study, eight crossbred (Santa Inês) ewe lambs (17.95 kg ± 2.71) were used in serial procedures for eight consecutive weeks. Liver samples obtained through six percutaneous techniques and two videolaparoscopic techniques were analyzed. The clinical aspects, clinical pathological findings, hepatic sample weights, histology quality, number of centrilobular veins and portal spaces, and the presence of artifacts in the histology slices were recorded. For the percutaneous techniques, two types of needles were used, modified Menghini (M) and semi-automatic tru-cut (T). They were used in combinations with blind biopsies, guided by ultrasound (US) and monitored by video laparoscopy (VL) biopsies. In the videolaparoscopy procedures, Blakesley biopsy forceps and the method of liver fragment resection by VL were used. No relevant hematological or biochemical alterations were found and the clinical manifestations were slight and transitory. In a general way, the use of US and VL in percutaneous biopsies did not yield a significant improvement in weight and histology quality of the liver samples; however, the videolaparoscopic techniques allowed a wider access to the liver than the percutaneous techniques. The quality of the samples obtained through both VL techniques were equivalent; however, the resection technique enabled the collection of heavier samples, but determined the formation of perihepatic adherences. More difficulty in fragment retrieval, more fluctuation in weight and less quality were found in samples obtained with M needles when compared to T needles.

Abstract in English:

ABSTRACT.- Peixoto P.V., Nogueira V.A., Gonzaléz A.P., Tokarnia C.H. & França T.N. 2009. Accidental and experimental salinomycin poisoning in rabbits. Pesquisa Veterinária Brasileira 29(9):695-699. Projeto Sanidade Animal Embrapa/UFRRJ, Seropédica, RJ 23890-000, Brazil. E-mail: pfpeixoto@terra.com.br An outbreak of salinomycin poisoning in rabbits is described. At least 27 out of 2,000 rabbits reared on a farm died after the coccidiostatic drug sulfaquinoxaline was substituted by salinomycin in the feed. An average of 26.9ppm salinomycin was detected in the ration given to the rabbits. Clinical signs included anorexia, apathy and bradykinesia, which progressed to incoordination and recumbency. Gross lesions consisted of pale areas in the skeletal muscles. The histopathological findings showed severe necrotic degenerative myopathy in association with infiltration of neutrophils and macrophages. One rabbit exhibited similar alterations in the myocardium. Mineralization was observed in the affected skeletal muscles in some cases. In order to verify if the poisoning was due to salinomycin, 20 rabbits were divided into five groups and a ration containing the drug at doses of 10, 25, 50, 75 and 100ppm was given. The administration of doses higher than 50ppm resulted in manifestation of the clinical signs seen in the outbreak of poisoning. It was concluded, that probably an error related to the mixture of salinomycin in the feed was the cause of deaths in the spontaneous outbreak of poisoning on the rabbit farm.

E-mail: ticianafranca@terra.com.br An outbreak of monensin poisoning in sheep in the State of Rio de Janeiro is described. From 180 animals, eight died after they had been fed with ration containing the ionophore. The poisoning had a variable course and was clinically characterized by apathy, heart arrhythmia, myoglobinuria, incoordenation, incapacity of getting up, and sternal decubitus; one sheep aborted. The macroscopic lesions consisted of pale areas in the myocardium, hydroperitoneum, hydrothorax, and pulmonary edema. Histopathological examination revealed degenerative-necrotic alterations in heart and skeletal muscles. In the myocardium lesions were more severe and were characterized by multifocal necrosis with substitution of the myofibres by fibrous tissue and interstitial mononuclear infiltration. Proliferation of satellite cells and mononuclear inflammatory reaction in skeletal muscles were also verified. It seems that the exaggerated addition of sodic monensin, eventually associated with improper homogenization of monensin in the ration, was responsible for the excessive ingestion of monensin by some animals.

Abstract in English:

ABSTRACT.- Helayel M.A., França T.N., Seixas J.N., Nogueira V.A., Caldas S.A. & Peixoto P.V. 2009. [Sudden death in cattle caused by ingestion of Pseudocalymma elegans (Bignoniaceae) in the county of Rio Bonito, Rio de Janeiro.] E-mail: michel_abdallavet@yahoo.com.br Natural poisoning by Pseudocalymma elegans, a plant that causes “sudden death” and only occurs in the State of Rio de Janeiro, is described in a cow. This condition was experimentally reproduced in three calves and three rabbits with the plant collected in the area where the deaths had occurred. Postmortem and histopathological examinations of the natural case in the cow did not reveal significant alterations. Oral administration of 1g/kg of the sprouts of P. elegans caused death of the calf 5 hours and 30 minutes after beginning of the administration. The dose of 0.5 g/kg caused death of another calf with onset of symptoms after 3 hours and 24 minutes and a course of 73 hours and 12 minutes. Animals that ingest borderline doses may show a longer course with a characteristic clinical picture of heart impairment. The dose of 0.25g/kg did not cause death, but only clinical signs. Heart palpitation and arrhythmia, increase of breathing frequency, reluctance in moving, positive venous pulse, ingurgitated jugular veins, falling down and peddling movements shortly before death, were observed. At postmortem examination the lesions were those of acute heart insufficiency, and histopathological examination revealed the typical kidney lesion (hydropic-vacular degeneration in the distal convoluted tubules) seen in poisoning by plants that cause “sudden death”. In the experimentally poisoned rabbits the clinical course was less than 1-2 minutes. In the kidney of two rabbits there was also the characteristic microscopic lesion. The presence of the plant in the county of Rio Bonito had not been known by the veterinarians. It is concluded that the plant could be more toxic than described before.

ABSTRACT.- Nogueira V.A., França T.N. & Peixoto P.V. 2009. [Ionophore poisoning in animals.] Intoxicação por antibióticos ionóforos em animais. Pesquisa Veterinária Brasileira 29(3):191-197. Projeto Sanidade Animal Embrapa/UFRRJ, Seropédica, RJ 23890-000, Brazil. E-mail: pfpeixoto@terra.com.br The therapeutic use of ionophores in veterinary medicine has grown in the last years, with resultant increase in the risk of poisoning in animals. Ionophores are used as food additives as coccidiostacts in several animal species and growth promoter and bloat prevention in ruminants. The most often used ionophores are monensin, lasalocid, narasin and salinomycin. There is a great variation in the susceptibility to the toxic effect of ionophores in different animal species. Poisoning can occur when the dosage is too high or when not correct doses for a certain animal species are given. Cases of poisoning have been described in sheep, swine, horses, dogs and poultry. For horses ionophores are extremely toxic. The use of ionophores is only safe when used accordingly to the instructions of the manufacturer and especially for each animal species. In this paper the most important data regarding clinical-pathological and pathogenic aspects, and also the conditions in which the poisoning may occur are critically reviewed.

#30 - Proliferative enteropathy (Lawsonia intracellularis) outbreak in rabbits in Brazil, p.447-451

Abstract in English:

ABSTRACT.- Peixoto P.V., Ribeiro C.T., Bezerra Jr P.S., Driemeier D. & França T.N. 2008. Proliferative enteropathy (Lawsonia intracellularis) outbreak in rabbits in Brazil. Pesquisa Veterinária Brasileira 28(10):447-451. Departamento de Nutrição Animal e Pastagem, Instituto de Zootecnia, Universidade Federal Rural do Rio de Janeiro, Seropédica, RJ 23890-000, Brazil. E-mail: pfpeixoto@terra.com.br An outbreak of Lawsonia intracellularis infection in rabbits, which occurred in 1988 in Rio de Janeiro state, Brazil, is reported. The disease had an acute course (24-48 hours) with clinical signs characterized by brownish or green diarrhea and dehydration. Occasionally, the animals died one day after the onset of diarrhea, without showing any other clinical signs. At necropsy, the ileum was prominent, firm and had a thickened wall; it was dilated in the caudal direction and had a somewhat reticulated appearance, perceptible through the serosa. The thickened mucous membrane had finely corrugated aspect and a shiny surface. The ileocecal valve and surrounding areas were slightly edematous and irregular. The Peyer’s patches were sometimes more evident. There was moderate enlargement of the mesenteric lymph nodes. The histological examination revealed different degrees of hyperplasia of the epithelial cells of intestinal crypts consisting of poorly differentiated, hyperchromatic cells with high mitotic index, arranged in a pseudostratified layer which, in some cases, reached the apical portions of the villi. The inflammatory infiltrate between the hyperplastic epithelial cells was composed of lymphocytes, plasma cells, macrophages, some eosinophils and globular leukocytes. Silver impregnation revealed large numbers of bacteria with morphology of the genus Lawsonia in the apical pole of cryptal enterocytes. These bacteria reacted positively to a Lawsonia intracellularis polyclonal antibody by the avidin-biotin immunohistochemistry method.
