#11 - Hematological parameters of Nile Tilapia (Oreochromis niloticus L.) under air exposure stress, 32(Supl.1):99-107

E-mail: diasroberta5@gmail.com The present study evaluated the hematological and biochemical parameters of adult tilapia (Oreochromis niloticus) under the influence of the physiological stress factor in animals submitted to air exposure during fattening in raceway system. Blood cell count, hemoglobin, hematocrit, mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), leukogram, differencial leukocyte count, platelet, glucose, total protein, cholesterol, triglycerides and electrolytes (calcium, chloride, sodium and potassium) were analyzed. The results showed a uniform distribution for red blood cells, hematocrit, hemoglobin, secondary Wintrobe indices, total protein, glucose, cholesterol, and serum ions, indicated by a relatively low variation coefficient. There was positive correlation only for total white blood cells, organic defense cells (neutrophils and lymphocytes), glucose, cholesterol, sodium and calcium. As for the leukocytes (WBC), as the animals were exposed to aeration, the number of leukocytes gradually decreased (leukopenia), simultaneously occurring neutrophilia and lymphopenia. The glycemic index was a good indicator of physiological stress due to hyperglycemia (82.0+20.88mg/dL) demonstrated in the treatments. The air exposure constituted an imbalance factor at the ion homeostasis and at the endogenous cholesterol synthesis. Besides, the recovery time did not result in the complete physiological rehabilitation face the imposed challenge.

E-mail: diasroberta5@gmail.com No presente trabalho avaliaram-se os parâmetros hematológicos e bioquímicos de exemplares adultos de tilápias (Oreochromis niloticus) sob a influência do fator estresse fisiológico em animais submetidos à exposição ao ar durante a engorda em sistema raceway. Foram analisados o eritrograma, teor de hemoglobina, volume globular, o volume corpuscular médio (VCM), a hemoglobina corpuscular média (HCM), a concentração de hemoglobina corpuscular média (CHCM), o leucograma, contagem diferencial de leucócitos, o plaquetograma, a glicose, a proteína total, o colesterol, o triglicerídeo e os eletrólitos (cálcio, cloretos, sódio e potássio). Os resultados revelaram que houve uma homogeneidade de distribuição para hemácias, volume globular, hemoglobina, índices hemantimétricos, proteína total, glicose, colesterol, e íons séricos, indicados pelos valores relativamente baixos do coeficiente de variação. Houve correlação positiva somente para leucócitos totais, células de defesa orgânica (neutrófilos e linfócitos), glicose, colesterol, sódio e cálcio. Quanto ao leucograma, à medida que os animais foram expostos ao ar, o número de leucócitos diminuiu gradativamente (leucopenia), ocorrendo simultaneamente neutrofilia e linfopenia. O índice glicêmico constituiu um bom indicador de estresse fisiológico, devido à hiperglicemia (82,0+20,88mg/dL) demonstrada nos tratamentos. A exposição ao ar constituiu um fator de desequilíbrio na homeostase iônica, e na síntese de colesterol endógeno. Entretanto, o tempo de recuperação não ocasionou a completa reabilitação fisiológica frente ao desafio imposto.

E-mail: bcs85@hotmail.com The efficacy of a polyvalent bacterin vaccine against Aeromonas hydrophila, Pseudomonas aeroginosa and Enterococcus durans administered by different routes in Nile tilapia was assessed by analyzing hematological and immunological parameters 7 and 21 days after vaccination. Treatments consisted of: non-vaccinated tilapia; tilapia vaccinated by intraperitoneal injection with 2x108 formalin-inactivated bacteria·mL-1; tilapia vaccinated orally with 2x107 formalin-inactivated bacteria·g-1, feed for 5 days; tilapia vaccinated by immersion bath in 2x107 formalin-inactivated bacteria·mL-1, for 20 minutes. Vaccinated fish groups presented higher hematocrit, number of erythrocytes and leukocytes than the non-vaccinated group. Serum agglutination titer of intraperitoneally vaccinated fish was higher on both evaluation periods for the three bacteria strains. Only on day 21 post-vaccination fish from the oral and immersion vaccination groups presented higher serum agglutination titer than the non-vaccinated fish for A. hidrophyla and E. durans. Serum antimicrobial activity in vaccinated fish was higher for P. aeroginosa and E. coli than in non-vaccinated fish on both evaluation periods. The different vaccine administration routes stimulated hematological and immunological responses in Nile tilapia 21 days post-vaccination, but intraperitoneal vaccination presented higher total number of leukocytes, lymphocytes and serum agglutination titer.

E-mail: bcs85@hotmail.com Este estudo avaliou a eficácia da administração de vacina polivalente (bacterina), contra Aeromonas hydrophila, Pseudomonas aeroginosa e Enterococcus durans, por diferentes vias de aplicação em tilápia do Nilo, analisando parâmetros hematológicos e imunológicos no 7º e 21º dia após vacinação. Os tratamentos consistiram de: tilápias não vacinadas; tilápias vacinadas via injeção intraperitoneal (i.p.) contendo 2x108 bactérias inativadas·mL-1; tilápias alimentadas com ração contendo vacina na proporção de 2x107 bactérias inativadas·g-1, durante 5 dias; tilápias vacinadas por banho de imersão em 2x107 bactérias inativadas·mL-1, durante 20 minutos. Os peixes vacinados apresentaram maior porcentagem de hematócrito, número de eritrócitos e leucócitos durante o período do experimento, em relação aos não vacinados. O título de aglutinação do soro dos peixes vacinados intraperitonealmente foi superior nos dois períodos de avaliação para todas as bactérias. Apenas no 21º dia os peixes vacinados oralmente e por imersão apresentaram título de aglutinação superior aos peixes não vacinados para A. hidrophyla e E. durans. O soro dos peixes vacinados apresentou maior atividade antimicrobiana para P. aeroginosa e E. coli, do que os peixes não vacinados, nos dois períodos. As diferentes vias de administração da vacina estimularam a resposta hematológica e imunológica da tilápia do Nilo após 21 dias de vacinadas, sendo que o número total de leucócitos, linfócitos, título aglutinante do soro dos vacinados i.p. foi maior do que os demais.

[The influence of the aquatic environment in tanks sequetially interconnected with PVC pipes on the gill epithelium and lamellas of tilapia (Oreochromis niloticus).] Alterações do epitélio branquial e das lamelas de tilápias (Oreochromis niloticus) causadas por mudanças do ambiente aquático em tanques de cultivo intensivo. Pesquisa Veterinária Brasileira 29(4):303-311. Laboratório de Neurogastroenterologia Experimental, Universidade Paranaense, Praça Mascarenhas de Moraes 4286, Umuarama, PR 87502-210, Brazil. Gill filaments of four specimens from four tanks (T1, T2, T3 e T4) sequentially interconnected were submitted to histological routine to obtain 5-mm-thick cuts that were stained with HE or submitted to histochemistry reactions PAS + diastase solution or Alcian Blue pH 2.5 or Alcian Blue pH 1.0. Considering the intermediary, apical and basal regions of the filaments, the lamellar area was measured and the amount of mucous cells was counted. It was verified that oxygen, pH, and temperature decreased progressively as the water flew from one tank to another. Thus, an increase was realized of the amount of mucous cells and the lamellar area in T2, as well as a progressive decrease of these measures on the tanks which received water from T2. Moreover, detachment of the gill epithelium, cellular hyperplasia in the interlamellar space, and telangectasias were observed in the fishes from T2, T3 e T4. It was concluded that the aquatic environment in tanks sequentially interconnected with PVC pipes suffers alterations from one tank to another, as that physico-chemical fluctuations reflect on the behavior of the gill epithelium through variations of the lamellar area and the amount of mucous cells.

University of Trás-os-Montes and Alto Douro, Center of Studies on Technological of Environmental and Life Sciences (CETAV), Apartado 1013, Vila Real 5001-801, Portugal. E-mail: fontain@utad.pt Nile tilapia, Oreochromis niloticus, of both sexes were reared in freshwater and exposed to 0.5, 1.0 and 2.5mg L-1 of waterborne copper for a period of 21 days. Liver and gill samples were collected after 21 days of exposure to copper and lesions were analyzed by light microscopy. The main histopathological changes observed in gills exposed to the highest concentration were edema, lifting of lamellar epithelia and an intense vasodilatation of the lamellar vascular axis. Although less frequent, lamellar fusion caused by the filamentar epithelium proliferation and some lamellar aneurisms were also found. The liver of control group exhibited a quite normal architecture, while the fish exposed to copper showed vacuolation and necrosis. These hepatic alterations were more evident in fish exposed to 1.0 and 2.5mg L-1 copper concentrations. The number of hepatocytes nucleus per mm2 of hepatic tissue decreased with the increase of copper concentration. In contrast, the hepatic somatic index was high in fish exposed at 2.5mg L-1 of copper. In short, this work advance new knowledge as influence of copper in the gill and liver histology of O. niloticus and demonstrated that their effects could be observed at different concentrations.
