E-mail: faleirosufmg@gmail.com The purpose of this study was to investigate histological changes in dairy cows’ hooves with or without injuries from naturally acquired laminitis. Cull cows with no clinical signs of hoof abnormalities (G1, n=9) and those with macroscopic lesions associated with laminitis without (G2, n=23) or with lameness (G3, n=7) were used in the study. After slaughter, samples of dermo-epidermal junctions of sole, axial and dorsal regions of the hoof were obtained and histologically processed using HE and PAS staining. Congestion, hemorrhage and inflammatory infiltrate in the dermis of sole, axial and dorsal regions were blindly and semiquantitatively evaluated by the same researcher. Inflammatory infiltrate was evaluated in the dermal laminae of axial and dorsal regions. The morphology of epidermal cells and the presence of irregularities in three regions of the basement membrane (BM) length were examined using PAS staining. Scores of lesions in different regions of the hoof in the same group and in different groups for each region of the hoof were compared using non-parametric analyses. Inflammatory infiltrate in the dermis of all regions of the hoof was detected in all groups with no significant statistical difference. Cows with no clinical signs of hoof abnormalities secondary to laminitis (G1) have inflammation scores and epidermal cell changes similar to those of groups with laminitis injuries, suggesting the existence of a prodromal phase for this disease in bovines. BM had irregularities with a variable intensity along its length, however, with no difference among groups. The pattern of BM irregularities found has not been reported so far and does not resemble the BM collapse described in horses and cattle with induced acute laminitis. Is it concluded that even in the absence of macroscopic hoof signs associated to laminitis, dairy cows have histological injuries compatible with inflammation of the dermo-epidermal junction as in affected animals. Basement membrane of cows with or without laminitis associated lesions had irregularities with an irregular distribution along its length which need to be further studied.

Utilizaram-se animais de descarte sem lesões macroscópicas no casco (G1 n=9) e com lesões macroscópicas associadas à laminite sem (G2 n=23), ou com claudicação (G3 n=7). Após o abate, amostras da junção derme-epiderme das regiões solear, axial e dorsal do casco foram obtidas e processadas, histologicamente, nas colorações de HE e PAS. Avaliou-se, semiquantitativamente, às cegas e por um mesmo pesquisador, congestão, hemorragia e infiltrado inflamatório na derme das regiões solear, axial e dorsal. Avaliou-se infiltrado inflamatório na lâmina dermal das regiões axial e dorsal. A morfologia das células epidermais e a presença de irregularidades em três regiões do comprimento da membrana basal (MB) foram examinadas na coloração de PAS. Os escores das lesões nas diferentes regiões do casco no mesmo grupo e nos diferentes grupos para cada região do casco foram comparados através de análises não paramétricas (P<0,05). Infiltrado inflamatório na derme de todas as regiões do casco foi detectado em todos os grupos sem diferença estatística (P>0,05). Vacas sem lesões macroscópicas secundárias à laminite (G1) apresentaram escores de inflamação e alteração de células epidermais semelhantes aos dos grupos com lesões de laminite, sugerindo a existência de fase prodrômica para a doença em bovinos. A MB apresentou irregularidades com intensidade variável ao longo de seu comprimento (P<0,05), porém, sem diferença entre grupos (P>0,05). O padrão de irregularidades na MB encontrado não foi relatado até o momento e não se assemelha ao colapso de MB descrito em equinos e bovinos com laminite induzida. Concluiu-se que, mesmo na ausência de lesões macroscópicas no casco causadas por laminite, vacas leiteiras apresentam lesões histológicas compatíveis com inflamação da junção derme-epiderme como em animais afetados. A membrana basal de bovinos sem e com lesões de laminite apresenta irregularidades com distribuição irregular ao longo de seu comprimento que precisam ser melhor estudadas.

In this study, leukocyte infiltration in hoof laminar tissue was investigated in horses subject to intestinal obstruction using immunohistochemistry to detect calprotectin, and zymography to detect neutrophil gelatinase associated lipocalin (NGAL). There were four groups: the Control Group (Gc), with seven horses, without surgical procedures; the Sham-operated Group (Gi), with five horses that were subjected to surgical procedure without intestinal obstruction; the No Treat group (Gnt), with four horses subjected to intestinal obstruction (jejunal distention using an intraluminal balloon) without treatment; and Treated group (Gt), with four horses subjected to intestinal obstruction and treated with hydrocortisone. Positive calprotectin imunostaining was detected in all experimental groups, with increase cell counts in horses of the distended group compared with the control group. NGAL expression was increased in Gd compared with Gc e Gi. The Gt did not differ from the others. In conclusion, small intestine distension can promote leukocyte infiltration in equine hoof laminar tissue, and NGAL zymography was considered a useful method for leukocyte tissue detection in horses. New studies will be conducted to verify the possible beneficial anti-inflammatory effects of hydrocortisone in hoof of horses with intestinal obstruction.

Neste estudo foram pesquisadas a presença de calprotectina por meio da imunoistoquímica, e de lipocalina associada à gelatinase de neutrófilos (NGAL), por zimografia, no tecido laminar do casco de equinos após obstrução intestinal. Os animais foram divididos em quatro grupos: Grupo controle (Gc), contendo sete animais normais, sem procedimento cirúrgico; Grupo Instrumentado (Gi), contendo cinco animais, os quais passaram por todo o procedimento cirúrgico sem sofrerem obstrução intestinal; Grupo Não Tratado (Gnt), contendo quatro equinos submetidos a obstrução intestinal do jejuno por distensão de balão intraluminal, sem tratamento; e Grupo Tratado (Gt), contendo quatro equinos submetidos a obstrução intestinal, e tratados preventivamente com hidrocortisona. Houve imunomarcação de calprotectina em todos os grupos experimentais, com aumento nos equinos do grupo distendido em relação ao Gc. Com relação ao NGAL, houve aumento também do Gnt e do Gi em relação ao Gc. O Gt não diferiu dos demais. Conclui-se que a distensão do intestino delgado pode promover acúmulos de leucócitos nos cascos de equinos e que o NGAL é um método viável para se detectar infiltração neutrofílica em equinos. Novos estudos deverão ser realizados para se verificar possível benefício anti-inflamatório da hidrocortisona no casco de equinos com obstrução intestinal.

E-mail: lucianelaskoski@hotmail.com The gastrointestinal diseases of horses are aggravated by complications such as laminitis. The laminitis etiopathogeny are connected with lamellar basement membrane degradation by matrix metalloproteinases (MMPs). Inflammatory cytokines and leukocytes enzymes can active MMPs. The object of this study was to evaluate morphological changes on lamellar tissue of horses with colic syndrome and its association with clinical and laboratorial parameters. It was observed intensive destruction of lamellar architecture, mainly on animals with severe physical and laboratorial alterations, such as delayed capillary refill time, congested mucous membrane, tachycardia, hemoconcentration and low count of platelet and leukocytes. The results sign to the most likely moment of development of lamellar tissue injuries in horses with colic syndrome, which can be adopted preventive measures against laminitis.

A ativação das MMPs pode ocorrer devido à liberação local de citocinas inflamatórias ou enzimas provenientes de leucócitos infiltrados no tecido laminar. O objetivo deste trabalho foi avaliar as alterações morfológicas do tecido laminar de equinos com síndrome cólica letal e sua provável associação com parâmetros clínicos e laboratoriais. Observou-se intensa destruição da arquitetura laminar, principalmente nos animais com alterações físicas e laboratoriais mais acentuadas, como tempo de preenchimento capilar prolongado (TPC), membranas mucosas congestas, taquicardia, hemoconcentração e redução nas contagens de plaquetas e leucócitos. Os resultados sinalizam o provável momento do desenvolvimento de lesões do tecido laminar em equinos com síndrome cólica, no qual é possível adotar medidas preventivas contra a laminite.

Thirty hours after the overload, the horses were randomly distributed in four groups (n=5) and were submitted to four IV treatments every 12 hours: 10ml of saline (GC), ketoprofen 2.2mg/kg (GK), phenylbutazone 4.4mg/kg (GF), and flunixin meglumine 1.1mg/kg (GFM). Blood samples were collected at various times after the overload (0-72 h). Plasma TYR and PEA concentrations ranged from 0.18 to 164.2mg/L, and differed significantly with time (p<0.01), but did not differ in the treatments. Plasma concentrations of TRP differed between times and treatments. The GC was significantly major than other treatments at 48h and 60h after the overload, and the plasma concentration of TRP in groups GK and GFM was significantly lower than in groups GF and GC at 72 h (p=0.0012). We concluded that the anti-inflammatory drugs evaluated do not interfere in the plasma concentration of TYP and PEA. For TRP, ketoprofen and flunixin meglumine was effective to reduce de plasmatic concentration of this amine.
