#11 - Infection of sparrows (Passer domesticus) and different mice strains with Lawsonia intracellularis, 33(3):372-378

E-mail: guedesufmg@gmail.com The susceptibility of sparrows (Passer domesticus) and strains of mice (Swiss, BALB/c, C-57 and DB-A) to Lawsonia intracellularis infection was studied. Thirty-two sparrows were inoculated with pure culture of L. intracellularis and eleven received sham inoculum. Feces were collected on -1, 7, 14 and 21 days post infection (dpi) for detection of L. intracellularis by PCR. After 21 days, all sparrows were euthanized and the tissues processed for histology and immunohistochemistry (IHC). One hundred sixty mice of four different strains (n=40, per strain) were used. For each mouse strain, 16 animals received mucosa homogenate from a pig infected with L. intracellularis, 16 received pure culture of L. intracellularis and eight animals received sham inoculum. Two control and four inoculated mice from each group were euthanized on 7, 14, 21 and 28 dpi. Sections of intestine were collected for histologic analysis and IHC and pooled feces were collected for L. intracellularis PCR. None of the sparrows had any histologic lesions characteristic of proliferative enteropathy or antigen labeling by IHC. All sparrow fecal samples were negative by PCR. All mice strains studied had histopathological lesions typical of PE and IHC labeling consistent with L. intracellularis infection, especially those animals inoculated with pure culture. The most severe lesions were observed in DB-A and Swiss mice. Fecal shedding was detected in all mice strains, with peak at 14 dpi. We conclude that sparrows do not seem to be relevant in the epidemiology of L. intracellularis. The results showed variations in the lesions among the four mice strains used.

Trinta e dois pardais foram inoculados com cultura pura de L. intracellularis e onze receberam placebo. As fezes foram coletadas nos dias -1, 7, 14 e 21 após a infecção (dpi) para a detecção de Lawsonia intracellularis por PCR. Após 21 dias, todos os pardais foram eutanasiados e os tecidos processados para a realização da histologia e imuno-histoquímica (IHQ). Cento e sessenta camundongos de quatro linhagens diferentes (n=40, por linhagem) foram utilizados. Para cada linhagem de camundongo, 16 receberam homogeneizado de mucosa preparado a partir de um suíno infectado com L. intracellularis, 16 receberam cultura pura de L. intracellularis e oito animais receberam placebo. Dois camundongos controle e quatro camundongos inoculados de cada grupo foram sacrificados aos 7, 14, 21 e 28 dpi. Seções de intestino foram coletadas para análise histológica e IHQ e amostras de fezes foram coletadas para a realização da PCR para detecção de L. Intracellularis. Nenhum dos pardais apresentou lesões histológicas características da enteropatia proliferativa ou marcação positiva por meio da IHQ. As amostras de fezes dos pardais foram negativas na PCR. Todas as linhagens de camundongos estudadas tinham lesões histopatológicas típicas de enterite proliferativa e IHQ positiva para a infecção por L. intracellularis, especialmente aqueles animais inoculados com a cultura pura. As lesões mais graves foram observadas em camundongos DB-A e Swiss. A eliminação fecal foi detectada em todas as linhagens de camundongos, com pico 14 dpi. Conclui-se que os pardais não são relevantes na disseminação da L. intracellularis. Os resultados mostraram variações nas lesões entre as quatro linhagens de camundongos utilizadas, indicando o potencial risco que os camundongos representam na transmissão de L. Intracellularis.

E-mail: rajao.ds@gmail.com Influenza A virus (IAV) is a respiratory pathogen of pigs and is associated with the porcine respiratory disease complex (PRDC), along with other respiratory infectious agents. The aim of this study was to diagnose and to perform a clinic-pathological characterization of influenza virus infection in Brazilian pigs. Lung samples from 86 pigs in 37 farrow-to-finish and two farrow-to-feeder operations located in the States of Minas Gerais, São Paulo, Paraná, Rio Grande do Sul, Santa Catarina, and Mato Grosso were studied. Virus detection was performed by virus isolation and quantitative real time reverse-transcription PCR (qRT-PCR). Pathologic examination and immunohistochemistry (IHC) were performed in 60 lung formalin-fixed paraffin-embedded tissue fragments. Affected animals showed coughing, sneezing, nasal discharge, hyperthermia, inactivity, apathy, anorexia, weight loss and growth delay, which lasted for five to 10 days. Influenza virus was isolated from 31 (36.0%) lung samples and 36 (41.9%) were positive for qRT-PCR. Thirty-eight (63.3%) lung samples were positive by IHC and the most frequent microscopic lesion observed was inflammatory infiltrate in the alveoli, bronchiole, or bronchi wall or lumen (76.7%). These results indicate that influenza virus is circulating and causing disease in pigs in several Brazilian states.

O objetivo deste estudo foi diagnosticar e realizar a caracterização clínica e patológica de casos/surtos de influenza em suínos brasileiros. Foram utilizadas amostras de tecido pulmonar de 86 suínos de 37 granjas de ciclo completo e duas unidades produtoras de leitões localizadas em Minas Gerais, São Paulo, Paraná, Rio Grande do Sul, Santa Catarina e Mato Grosso. A detecção viral em fragmentos pulmonares frescos foi realizada através do isolamento viral e da transcrição reversa-PCR em tempo real quantitativa (qRT-PCR). Exame patológico e imuno-histoquímica (IHQ) foram realizados em 60 amostras de pulmão fixadas em formalina 10% e embebidas em parafina. As amostras eram de animais apresentando tosse, espirros, secreção nasal, hipertermia, prostração, apatia, anorexia, perda de peso e ganho de peso reduzido, com duração entre cinco e 10 dias. O vírus influenza foi isolado de 31 (36,0%) amostras e 36 (41,9%) foram positivas na qRT-PCR. Na IHQ, 38 (63,3%) amostras foram positivas e a lesão mais frequentemente observada foi a presença de infiltrado inflamatório na parede e lúmen de vias aéreas (76,7%). Estes resultados indicam que o vírus influenza está circulando e causando lesões e doença respiratória em suínos de diversos Estados do Brasil.

E-mail: alage@vet.ufmg.br This paper describes the clinical, pathological, and microbiologic aspects of paratuberculosis (Johne’s disease) in a dairy Gyr herd in the State of Paraíba, northeastern Brazil. An eight years old cow with chronic unresponsive diarrhea was clinically examined and euthanized for pathological evaluation. Fecal samples from all 160 animals over 12 months of age from the herd were collected for isolation of Mycobacterium avium subsp. paratuberculosis. Clinically, the index case cow was severely dehydrated, cachectic, with profuse mucous diarrhea. The main post-mortem findings were emaciation and thickened intestinal wall. Microscopically, the intestinal lamina propria and submucosa were infiltrated by macrophages, epithelioid cells, and Langhans giant cells with numerous alcohol-acid resistant bacilli in the cytoplasm. Two fecal samples displayed growth in slants of Herrold’s egg-yolk agar supplemented with mycobactin J, 150 days after incubation. No growth was noticed in slants without mycobactin J. Microscopic examination of the isolated microorganisms stained by Ziehl-Neelsen revealed considerable amounts of alcohol-acid resistant bacilli, morphologically compatible with Mycobacterium spp. Based on the clinical signs, gross and histological lesions, growth time, bacterial morphology in Ziehl-Neelsen staining, and dependence of mycobactin J, the first diagnosis of paratuberculosis in Zebu cattle was made.

Uma vaca de oito anos que apresentava diarréia persistente, refratária a tratamento foi necropsiada para estudo anátamo-histopatológico. Também foram coletadas amostras de fezes de todos os 160 animais do plantel, com idade superior a 12 meses, para tentativa de isolamento de Mycobacterium avium subsp. paratuberculosis. Ao exame clínico, o animal caso índice apresentou caquexia, diarréia profusa e desidratação grave. À necropsia, o animal apresentou-se emaciado e, ao exame detalhado do trato digestivo, foi observado espessamento da parede e superfície mucosa do íleo e intestino grosso. À microscopia, verificou-se intensa infiltração de macrófagos espumosos associado a raras células epiteliódes e gigantes do tipo Langerhans na lâmina própria e submucosa. À coloração de Ziehl-Neelsen foram observadas miríade de bacilos álcool-ácido resistentes no citoplasma destas células. Houve crescimento de colônias bacterianas em duas das 160 amostras de fezes após 150 dias de incubação em tubos com meio Herrold’s egg-yolk suplementados com micobactina J e ausência de crescimento nos tubos com mesmo meio, mas sem suplementação. Os microrganismos isolados foram corados pelo Ziehl-Neelsen observando-se presença de grande quantidade de bacilos álcool-ácido resistente, com morfologia compatível ao gênero Mycobacterium. Baseado na história clínica, achados anátomo-histopatológicos e histoquímicos (Ziehl-Neelsen), e microbiológicos, firmou-se o primeiro diagnóstico de paratuberculose em Zebu na Paraíba.

Clinically, the pigs presented reduced growth rate, anorexia, lethargy, cough and dyspnea, especially after they were moved. High body temperature (40oC in average) was verified in some animals. Auscultation revealed moderate pulmonary crepitation and stertors. Pulmonary gross lesions were typical of lobular bronchopneumonia. Lung lesions were characterized by ventral-cranial consolidation in the majority of the cases. The color of affected pulmonary areas varied from diffuse dark red to mosaic pattern (dark red lobule intercalate by grayish lobule) or diffusely grayish. The majority of the lungs had mucopurulent exsudate in the bronchial lumen that also drained from the parenchyma cut surface. Upon microscopy, the changes were characterized by purulent and histiocytic bronchopneumonia with necrotic foci. In some animals, there was BALT hyperplasia associated with perivascular and peribronchial plasma cells and lymphocytes infiltration in most of these cases. Bordetella bronchiseptica and Streptococcus spp. were the most frequently isolated bacteria. Immunohistochemistry evaluation demonstrated Mycoplasma hyopneumoniae on the luminal surface of bronchial and bronchiolar epithelial cells, and the DNA of bacteria was detected by PCR. This is the first report of bronchopneumonia in wild boars associated with M. hyopneumoniae infection.
